Description
B鎖の29位がリジンの代わりにグルタミン酸;
B鎖の3位がアスパラギンの代わりにリジン。
A鎖:21位がアスパラギンの代わりにグリシン;
B鎖:さらに、31位および32位の2個のアルギニン。
B鎖の最後のアミノ酸(スレオニンB30)が除去されている;
グルタミン酸および16-C脂肪酸がリジンB29に結合している。
活性化合物含有層の成分を溶媒に溶解および/または懸濁する工程、
溶液または懸濁液を指定される厚さで担体に塗布する工程;および
その後乾燥させる工程
を含む。
さらなる活性化合物含有層の成分を溶媒に溶解および/または懸濁する工程、
溶液または懸濁液を指定される厚さで乾燥した活性化合物層に塗布する工程、および
このようにして得られたさらなる活性化合物層を乾燥させる工程
によって、既に乾燥した活性化合物層に塗布される。そのため、さらなる活性化合物層のための溶液または懸濁液が塗布される乾燥した活性化合物層は平らな担体を構成する。
カバー層の成分を溶媒に溶解および/または懸濁する工程、
カバー層のための溶液または懸濁液を最後の乾燥した活性化合物層に塗布する工程、および
カバー層を乾燥させる工程
を含む。
カバー層の成分を溶媒に溶解および/または懸濁する工程;
溶液を指定される厚さで平らな担体に塗布する工程;
カバー層を乾燥させる工程;
活性化合物含有層の成分を溶媒に溶解または懸濁する工程;
活性化合物含有溶液を前もって製造されたカバー層に塗布する工程;
および
このようにして得られた活性化合物層を乾燥させる工程
を含む。
インスリンまたはインスリンアナログの放出および経粘膜透過を、会社Skin Ethics、Lyon、フランスのヒト上皮皮膚モデルを使用したインビトロモデルで調査した。このモデルでは、細胞株TR146のヒトケラチノサイトを、化学的に定義された培地において、角質層を有さず、よって、少なくとも組織学的に口腔粘膜に相当する上皮組織が空気との液体界面上に形成するように、ポリカーボネートフィルタ上で培養した。
Claims (12)
- 口腔粘膜を介した抗糖尿病ペプチドの送達用のフィルム状剤形であって、少なくとも1種の抗糖尿病ペプチドを含有し、異なる粘度の少なくとも2種のヒドロキシプロピルメチルセルロースの混合物を含む、少なくとも1つの活性化合物含有層を含み、前記少なくとも2種のヒドロキシプロピルメチルセルロースの混合物は、第1のヒドロキシプロピルメチルセルロースと第2のヒドロキシプロピルメチルセルロースの混合物であり、ここで、第1のヒドロキシプロピルメチルセルロースは、高粘度のヒドロキシプロピルメチルセルロースであり、第2のヒドロキシプロピルメチルセルロースは、中粘度のヒドロキシプロピルメチルセルロースであり、前記高粘度のヒドロキシプロピルメチルセルロースは、20℃の温度において水中2%溶液で測定される、約3000mPa・s~約5600mPa・sの粘度を有し、前記中粘度のヒドロキシプロピルメチルセルロースは、20℃の温度において水中2%溶液で測定される、約75mPa・s~約140mPa・sの粘度を有する、前記フィルム状剤形。
- 前記少なくとも2種のヒドロキシプロピルメチルセルロースの混合物は、第1のヒドロキシプロピルメチルセルロース、第2のヒドロキシプロピルメチルセルロースおよび第3のヒドロキシプロピルメチルセルロースの混合物であり、ここで、第1のヒドロキシプロピルメチルセルロースは、高粘度のヒドロキシプロピルメチルセルロースであり、第2のヒドロキシプロピルメチルセルロースは、中粘度のヒドロキシプロピルメチルセルロースであり、第3のヒドロキシプロピルメチルセルロースは、低粘度のヒドロキシプロピルメチルセルロースであり、前記低粘度のヒドロキシプロピルメチルセルロースは、20℃の温度において水中2%溶液で測定される、約3mPa・sの粘度を有する、請求項1に記載のフィルム状剤形。
- 少なくとも1種の抗糖尿病ペプチドはインスリン、インスリンアナログ、インクレチンおよびインクレチン模倣物で構成される群から選択される、請求項1または2に記載のフィルム状剤形。
- カバー層をさらに有する、請求項1~3のいずれか1項に記載のフィルム状剤形。
- カバー層はエチルセルロースを含む、請求項4に記載のフィルム状剤形。
- カバー層中のエチルセルロースの含量は、カバー層の総重量基準で、少なくとも50重量%であるが、最大で99重量%である、請求項5に記載のフィルム状剤形。
- 請求項1~6のいずれか1項に記載の抗糖尿病ペプチドの経粘膜送達用のフィルム状剤形を製造する方法であって、以下の工程:
- 活性化合物含有層の成分を溶媒に溶解および/または懸濁する工程、
- 溶液または懸濁液を指定される厚さで担体に塗布する工程;
および
- その後乾燥させる工程
を含む、前記方法。 - - カバー層の成分を溶媒に溶解および/または懸濁する工程、
- カバー層のための溶液または懸濁液を担体に塗布する工程、および
- その後カバー層を乾燥させる工程
をさらに含む、請求項7に記載の方法。 - 活性化合物含有層の成分および/またはカバー層の成分を溶解または懸濁するための溶媒は、水および水とエタノールの混合物を含む群から選択される、請求項7または8に記載の方法。
- 乾燥は80℃以下の温度で行われる、請求項7~9のいずれか1項に記載の方法。
- 活性化合物含有層の成分を含む溶液または懸濁液の塗布は前もって製造された活性化合物含有層または前もって製造されたカバー層に行われる、請求項7~10のいずれか1項に記載の方法。
- 剤形を分離するまたは所定の幅および長さのストリップへ切断する工程;ならびに
その後パッケージングする工程
をさらに含む、請求項7~11のいずれか1項に記載の方法。
