Description
本出願は、2016年6月6日に出願された「キメラ抗原受容体T細胞技術および他の細胞治療のための微小流体ウイルス導入」と題する米国仮特許出願第62/346,031号、および2016年11月14日に出願された「細胞形質導入のためのシステムおよび方法」と題する米国仮特許出願第62/421,784号の利益および優先権を主張するものであり、これら両者の全体を本明細書の一部として援用する。
図7A~7Dに示された構成を有する細胞形質導入システムを、当該システムの形質導入効率を評価するために、従来のスピノキュレーション形質導入技術と比較して試験した。図8は、この実験の結果を示す。
Claims (11)
- 第1の流体マニホルドに結合された少なくとも1つの第1の流れチャンバを画定する第1の基板と、
細胞随伴層を画定する第2の基板であって、前記細胞随伴層は、
少なくとも1つの第2の流れチャンバと、
複数の細胞随伴キャビティであって、各細胞随伴キャビティは、一端が前記少なくとも1つの第2の流れチャンバの中へと開口し、前記第2の基板を通って延出し、少なくとも1つの細胞を保持する大きさである、複数の細胞随伴キャビティ―と、
実質的に前記第2の基板の平面内にある前記少なくとも1つの第2の流れチャンバへの少なくとも1つの入口と、
実質的に前記第2の基板の平面内にある前記少なくとも1つの第2の流れチャンバからの少なくとも1つの出口と
を含む、第2の基板と、
前記第1の基板と前記第2の基板との間に配置された第1の膜であって、前記第1の膜は、細胞の通過を防止しかつウイルス粒子の通過を可能とするため、0.1ミクロン~1.0ミクロンの第1の直径を有する、第1の複数の細孔を備える、第1の膜と、
第2の流体マニホルドに結合された少なくとも1つの第3の流れチャンバを画定する第3の基板と、
前記第2の基板と前記第3の基板との間に配置された第2の膜であって、前記第2の膜は、ウイルス粒子の通過を防止するために十分小さいが、細胞培地の通過を可能にするために十分に大きい第2の直径であって、前記第1の直径よりも小さい第2の直径を有する、第2の複数の細孔を備える、第2の膜、
を含む装置であって、
前記装置は、前記第1および第2の流体マニホルドを更に含み、
前記第1の流体マニホルドの第1の端部は、前記第1の基板によって画定される前記少なくとも1つの第1の流体チャンバに結合し、
前記第2の流体マニホルドの第1の端部は、前記第3の基板によって画定される前記少なくとも1つの第3の流体チャンバに結合し、
前記第1の流体マニホルドの第2の端部は、前記第2の流体マニホルドの第2の端部に流体的に結合されて、流体が前記第1の流体マニホルド、前記第1の膜、前記複数の細胞随伴キャビティ、前記第2の膜、前記第2の流体マニホルドを通って循環し、前記第1の流体マニホルドに戻ることができるようになっている、装置。 - 前記第1の流体マニホルドおよび前記第2の流体マニホルドのうち少なくとも1つは、垂直流マニホルドを含む、請求項1に記載の装置。
- 前記第1の流体マニホルドおよび前記第2の流体マニホルドのうち少なくとも1つは、水平流マニホルドを含む、請求項1に記載の装置。
- 前記第1の流体マニホルドの前記第2の端部と前記第2の流体マニホルドとの間にバルブにより結合された廃棄チャネルを更に含み、前記バルブは、前記第1の流体マニホルドの前記第2の端部から排出される流体流れを廃棄物リザーバへと選択的に迂回させるように構成される、請求項1に記載の装置。
- 前記第1の流体マニホルドの前記第2の端部へと流体をポンプ輸送するように構成された第1のポンプを更に含む、請求項1に記載の装置。
- 前記第2の流体マニホルドの前記第2の端部に流体をポンプ輸送するように構成された第2のポンプを含み、前記第2のポンプは、前記第1のポンプと同じポンプであるか、または前記第1のポンプとは異なる、請求項5に記載の装置。
- 前記第1の基板は前記少なくとも1つの第1の流体チャンバの遠位端に結合された出口を更に含む、請求項1に記載の装置。
- 前記細胞随伴キャビティは、前記少なくとも1つの第2の流体チャンバの遠位端に向かう方が、前記少なくとも1つの第2の流体チャンバの近位端に向かうよりも高い密度を有する、請求項1に記載の装置。
- 前記細胞随伴キャビティの各々は、それぞれの細胞随伴キャビティの側壁に配置された一対のエレクトロポレーション電極を含む、請求項1に記載の装置。
- 少なくとも1つの第1の流れチャンバを第1の流体マニホルドに結合する少なくとも1つの第1の流れチャンバを画定する第1の基板と、
細胞随伴層を画定する第2の基板であって、前記細胞随伴層は、
少なくとも1つの第2の流れチャンバと、
メッシュまたは多孔質ゲルから形成された少なくとも1つの細胞随伴領域であって、前記少なくとも1つの細胞随伴領域の各々は、対応する第2の流れチャンバに隣接して配置され、遺伝子導入剤を含浸される、細胞随伴領域と、
前記第2の基板を通って延び、且つ前記第2の基板に対して実質的に垂直に配向された第2の複数の流体通路であって、前記第2の複数の流体通路の各々は、一端において、前記少なくとも1つの第2の流れチャンバの1つの中に開口する、第2の複数の流体通路と
を含む、第2の基板と、
前記第1の基板と前記第2の基板との間に配置された第1の膜であって、前記第1の膜は、細胞の通過を防止しかつウイルス粒子の通過を可能とするため、0.1ミクロン~1.0ミクロンの第1の直径を有する、第1の複数の細孔を備える、第1の膜と、
第2の流体マニホルドに結合された少なくとも1つの第3の流れチャンバを画定する第3の基板と、
前記第2の基板と前記第3の基板との間に配置された第2の膜であって、前記第2の膜は、ウイルス粒子の通過を防止するために十分小さいが、細胞培地の通過を可能にするために十分に大きい第2の直径であって、前記第1の直径よりも小さい第2の直径を有する、第2の複数の細孔を備える、第2の膜、
を含む装置。 - 細胞形質導入の方法であって、
細胞を少なくとも1つの第1の流れチャンバに導入することであって、少なくとも1つの第1の流れフィールドは、それぞれの第1の流れチャンバの領域に広がり、且つ複数の遺伝子情報導入剤が含浸されているそれぞれのメッシュまたはゲルに隣接している、導入することと、
第1の流体を、前記少なくとも1つの第1の流れフィールドに対して実質的に垂直な第1の方向に、且つ前記メッシュまたはゲルを通して流し、それによって前記導入された細胞を前記メッシュまたはゲルの中に取り込み、且つ第1の時間前記遺伝子情報導入剤と緊密に接触させ、それによって前記遺伝子情報導入剤に担持された前記遺伝子情報を随伴細胞の中に導入することを可能にすることと、
前記少なくとも1つの第1の流れチャンバの反対側のメッシュまたはゲルの側を通しての、前記細胞および前記遺伝子情報導入剤の通過を防止することと、
前記第1の流体の流れの方向を第2の時間だけ反転させ、それによって前記細胞を前記メッシュまたはゲルから放出し、前記細胞を洗浄することと、
前記放出された細胞を、収集のために前記少なくとも1つの第1の流れチャンバから流出させることと
を含む、方法。
