Description
バイオリアクタは、幾つもの目的で生物学的反応を発生させ及び維持するために有用である。生物学的反応は、温度及び/又は圧力の変化に敏感な場合がある。更に、生物学的反応が進行するにつれて、反応自体が、溶存酸素含有量及び/又はpHなどの、バイオリアクタ容器内の種々のパラメータを変化させることがあり得る。したがって、生物学的反応の多数の変数を監視することは重要であり得る。
バイオリアクタ容器が、提供される。バイオリアクタ容器は、内側及び外側並びに内側から外側まで延在する開口を有するバイオリアクタ袋壁を含み、内側は、反応混合物に接触し反応混合物を収容するように構成される。バイオリアクタ容器はまた、開口の近接に設けられたポートを含み、ポートは、バイオリアクタ袋に固定して取り付けられるフランジを含み、その結果、流体シールが、フランジの円周に沿って維持される。バイオリアクタ容器はまた、ポート内に配設された複数のセンサを含む。
図1は、本発明の実施形態が特に有用である使い捨てバイオリアクタ袋の一例を示す。単回使用、すなわち使い捨て、反応チャンバは、当技術分野において公知であり、例えば、生物学的反応のために頻繁に使用される。
Claims (19)
- 内側及び外側を有するバイオリアクタ袋であって、前記内側から前記外側まで延在する開口を有し、前記内側が反応混合物流体に接触し、前記反応混合物流体を収容するように構成される前記バイオリアクタ袋と、
前記バイオリアクタ袋の前記開口に近接する袋壁に固定するように取り付けられたポートであって、前記ポートはフランジと、チューブバーブを有するチューブとを含み、前記チューブは第1センサ入口及び第2センサ入口を有するものである、前記ポートと、
前記第1センサ入口及び前記第2センサ入口のうち、一方の入口に挿入されポート内に収容された第1のセンサと、他方の入口に挿入されポート内に収容された第2のセンサと、
を含むバイオリアクタ容器であって、
前記固定する取り付けは、前記フランジの円周に沿って流体シールが維持されるように実施されており、
前記第1センサ入口および第2センサ入口を有する前記チューブは、挿入されたセンサが前記バイオリアクタ袋の内部の反応混合物流体に接触可能となるように構成され、かつ、一方向弁を有し、
前記第1センサ入口及び前記第2センサ入口は、各々が離間されて隣接するように配置されている
前記バイオリアクタ容器。 - 前記ポートが、前記壁の前記内側に固定するように取り付けられる、請求項1に記載のバイオリアクタ容器。
- 前記ポートが、前記壁の前記外側に固定するように取り付けられる、請求項1に記載のバイオリアクタ容器。
- 前記第1のセンサ及び/又は前記第2センサは、マルチセンサである、請求項2に記載のバイオリアクタ容器。
- 前記第1のセンサ及び/又は前記第2センサは、pH、反応混合物の濃度、溶存酸素濃度、伝導率、温度、及び圧力からなるグループから選択される変数を検出するように構成される、請求項1に記載のバイオリアクタ容器。
- 前記バイオリアクタ容器が、単回使用バイオリアクタ容器である、請求項1に記載のバイオリアクタ容器。
- 前記バイオリアクタ容器の前記内側が、無菌である、請求項1に記載のバイオリアクタ容器。
- 前記フランジの前記バイオリアクタ容器の内側への取り付けは、接着層を介して実施される、請求項2に記載のバイオリアクタ容器。
- 前記フランジの前記バイオリアクタ容器の内側への取り付けは、融着(熱溶接)により実施される、請求項2に記載のバイオリアクタ容器。
- 前記ポート及び前記バイオリアクタ容器が、プラスチック材料を含む、請求項1に記載のバイオリアクタ容器。
- 単一の開口を有する反応チャンバを形成する工程、
前記反応チャンバの前記単一の開口に近接する壁にポートを接着する工程であって、前記ポートは、第1センサ入口および第2センサ入口を有するものである、工程、
前記ポートが有する前記第1センサ入口および前記第2センサ入口の、一方の入口に第1のセンサを挿入して収容し、他方の入口に第2のセンサを挿入して収容する工程、
を含む単回使用反応容器を製造する方法であって、
前記ポートは、挿入したセンサが前記単回使用反応容器の内部の流体に接触可能となるように構成され、かつ、一方向弁を有し、
前記第1センサ入口及び前記第2センサ入口は、各々が離間されて隣接するように配置される、
前記方法。 - 前記ポートはフランジを有し、前記単一の開口の前記壁への前記ポートの接着は、前記フランジと前記壁の間に接着層を適用して実施される、請求項11に記載の方法。
- 前記ポートはフランジを有し、前記単一の開口の前記壁への前記ポートの接着は、前記フランジと前記壁の間の融着(熱溶接)により実施される、請求項11に記載の方法。
- 前記接着が、前記ポートを前記反応チャンバの内側に取り付けることを含む、請求項11に記載の方法。
- 前記接着が、前記ポートを前記反応チャンバの外側に取り付けることを含む、請求項11に記載の方法。
- 前記ポート及び前記反応チャンバが、プラスチックから形成される、請求項11に記載の方法。
- 前記第1のセンサ及び/又は前記第2センサは、pH、濃度、溶存酸素濃度、伝導率、温度、及び圧力からなるグループから選択されるプロセス変数を検出するように構成される、請求項11に記載の方法。
- 前記第1のセンサ及び/又は前記第2センサは、マルチセンサである、請求項11に記載の方法。
- 前記反応チャンバが、無菌の反応チャンバである、請求項11に記載の方法。
