Description
[1]高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の1つまたは複数の株、およびコレステロール改変剤を含む、組成物。
[2]高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の1つまたは複数の株、およびコレステロール改変剤を含む、個体のコレステロール吸収の改変に使用するための、組成物。
[3]高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の1つまたは複数の株、およびコレステロール改変剤を含む、高コレステロール、心疾患、糖尿病または肥満の処置に使用するための、組成物。
[4]上記1つまたは複数の株が、ラクトバチルス・プランタルム(Lactobacillus plantarum)2828(ECGC13110403)、ラクトバチルス・プランタルム(Lactobacillus plantarum)2830(ECGC13110402)、およびラクトバチルス・プランタルム(Lactobacillus plantarum)2691(ECGC13110401)、およびラクトバチルス・アシドフィルス(Lactobacillus acidophilus)(ATCC43121)またはそれらの変異株から選択される少なくとも1つの株を含む、上記[1]から[3]のいずれかに記載の組成物。

本発明の第1の態様によれば、高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の1つまたは複数の株、およびコレステロール改変剤を含む組成物が提供される。
ここで、本発明の実施形態を、以下の図面の詳細な説明を単なる例として用いて説明する。
これらの実験の基礎は、新規なGOSを生成するために、微生物においてβ−ガラクトシダーゼを可逆的に使用することであった。β−ガラクトシダーゼは、普通はラクトースを消化するはずである。しかし、基質および温度に関して反応条件を変化させることによって、酵素は可逆的に作用し、ラクトース(GOS)のオリゴ糖版を産生する。
1.様々なプロバイオティクスの乳酸桿菌を集め、GOSを産生するそれらの能力について試験し、β−ガラクトシダーゼ活性を測定すること、
2.逆酵素手順を使用してプレバイオティクスGOSを産生すること、
3.新規な分子をスケールアップして、インビトロ試験を行うこと、
4.プロバイオティクスおよびシンバイオティクスを試験する一連の「腸モデル」実験において、プレバイオティクスが存在しない状態および存在する状態で、乳酸桿菌の生存および増殖を比較すること、
5.乳酸桿菌のための被包材料としてGOSを使用できる可能性を評価すること、ならびに
6.被包材料の送達特性を試験すること。
コレステロール%×乾燥重量(g)−1=(B−T/B×100)/W
式中、B=接種されなかった対照中のコレステロール含量(mg/l−1)、T=培養培地中のコレステロール(mg/l−1)およびW=細胞(12時間のインキュベーション後の乾燥重量(g))。
実験1と並列して、乳酸桿菌の718の株の初期ハイスループットスクリーニング(HTS)試験を実施して、異なるタイプの胆汁酸塩に対する耐性を評価した。次に、胆汁酸塩ヒドロラーゼ活性を確立するために、さらなる実験を実施して、286の耐性株における胆汁の沈殿を調査した。
生物学的材料の寄託は、英国National Collection of Type Cultures(NCTC)の英国特許出願の目的で行われた。NCTCは、ブダペスト条約の下で認知されている国際寄託当局(IDA)である。
Claims (11)
- 高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の株およびオリゴ糖を含む組成物であって、前記株が、ラクトバチルス・プランタルム(Lactobacillus plantarum)2830(ECGC13110402)を含む、組成物。
- 高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の株およびオリゴ糖を含む、個体のコレステロール吸収の改変に使用するための組成物であって、前記株が、ラクトバチルス・プランタルム(Lactobacillus plantarum)2830(ECGC13110402)を含む、組成物。
- 高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の株およびオリゴ糖を含む、高コレステロールの処置に使用するための組成物であって、前記株が、ラクトバチルス・プランタルム(Lactobacillus plantarum)2830(ECGC13110402)を含む、組成物。
- 前記株が、105cfu/g〜1012cfu/gの範囲の量で存在する、請求項1から3のいずれかに記載の組成物。
- 被包されている、請求項1から4のいずれかに記載の組成物。
- 食料品の形態である、請求項1から5のいずれかに記載の組成物。
- 栄養補助食品として使用するための、請求項1から6のいずれかに記載の組成物。
- ビタミン、ミネラル、植物性化学物質、抗酸化剤、およびそれらの組合せから選択される1つまたは複数の活性成分をさらに含む、請求項1から7のいずれかに記載の組成物。
- プレバイオティクスをさらに含み、またはプレバイオティクスを含む場合、二次プレバイオティクスをさらに含む、請求項1から8のいずれかに記載の組成物。
- 高い胆汁酸塩ヒドロラーゼ活性を有する乳酸桿菌の株を、生物学的に有効な量のオリゴ糖と混合するステップを含む、請求項1から9の組成物を生産する方法であって、前記株が、ラクトバチルス・プランタルム(Lactobacillus plantarum)2830(ECGC13110402)を含む、方法。
- 前記組成物が、ラクトバチルス・プランタルム(Lactobacillus plantarum)2828(ECGC13110403)、およびラクトバチルス・プランタルム(Lactobacillus plantarum)2691(ECGC13110401)から選択される1つまたはそれを超える株をさらに含む、請求項10に記載の方法。
