インビトロ細胞ベースの候補材料のアレルギー阻害活性の分析
A.実験方法
(1)細胞培養:ヒト肥満細胞株(HMC-1)を、37℃および5% CO2で、10% FBSが補われたIMDM中において培養した。候補材料をDMSO中に溶解し、次いで、0.22μmフィルターを使用して濾過した。得られた候補材料をDMSOで希釈し、HMC-1細胞をそれで処理した。
実験方法
A.実験動物:Daehan Biolink (Eumseong, Chungbuk)から得られた5週齢雄BALB/cマウスを、環境条件:温度22±3℃;相対湿度50±20%;換気頻度10〜15回/時間;照明12時間;および照度150 Lux〜300 Lux下に設定された、Hoseo University Safety Evaluation Centerのクリーンな動物飼育室(小さな動物室#2-302)中に2週間適応させ、次いで実験に使用した。
実験方法
A.飼育環境
(1)環境条件:本実験において、実験動物を、環境条件:温度22±3℃;相対湿度50±20%;換気頻度10〜15回/時間;照明期間12時間;および照度150 Lux〜300 Lux下に設定された、Hoseo University Safety Evaluation Centerのクリーンな動物飼育室(小さな動物室#2-302)中において飼育した。
(1)実験群の構成
(A)投与の約24時間前に、背部上の体毛を、体毛除去部位がその全表面積の約10%以上を占めように除去し、皮膚を傷つけないように注意を払った。
(A)全身症状観察:全ての動物を投与後30分〜4時間集中的に観察し、全身症状を14日間の間1日1回観察した。観察時に、死および臨床症状を個別に記録した。
実験方法
A.組織サンプルの調製:マウス皮膚組織を液体窒素中に凍結し、次いで、乳鉢中において細かくすりつぶした。液体窒素を断続的に添加し、組織が解凍されるのを防いだ。細かくすりつぶされた皮膚組織を、溶解Mバッファー中に置き、組織サンプル溶解物を調製した。
ドッキング計算の結果として修正された最も高い最終スコアを有する50個の化合物についての結果が得られ、その分子構造を表に示した。

カスパーゼ-1は、アトピー性皮膚炎原因物質である、TSLPの分泌を誘発する。TSLP調節物質を探るために、アトピー性皮膚炎の原因細胞である肥満細胞を、PKC活性化剤PMAおよびカルシウムイオノフォアA23187(PMACI)で刺激した。刺激された肥満細胞からのTSLP分泌は、非刺激細胞(ブランク)からのそれと比較して有意に増加することが分かり、様々な候補材料の効果を調べた。結果として、5182436、5211084、および5356744物質は、肥満細胞からのTSLP分泌を有意に阻害したことが分かった(図2を参照のこと)。
有効な材料の細胞傷害性を調べるために、MTT-アッセイを行い、MTT-アッセイ結果は、IPS-07002(5211084)およびIPS-07003(5356744)は10μMの濃度で細胞傷害性を示さなかったことを示した。しかしながら、IPS-07001(5182436)は10μMの濃度で細胞傷害性を示したことが確認された。様々な濃度で繰り返し行った実験の結果として、IPS-07001(5182436)は1μMまでは細胞傷害性を示さなかった(図3を参照のこと)。
反復実験を行い、非細胞傷害性濃度でのTSLP産生の調節に対するIPS-07001の効果を調べ、反復実験結果から、IPS-07001は0.011μMの濃度でTSLP産生を有意に調節したことが確認された。IPS-07001はまたTSLP mRNA発現を有意に阻害した(図4を参照のこと)。
カスパーゼ-1活性に対する#5182436の調節効果を調べるために、カスパーゼ-1アッセイを、細胞抽出物を使用して行った。結果として、IPS-07001は濃度依存様式でカスパーゼ-1活性を有意に阻害したことが分かった。加えて、IPS-07001は活性カスパーゼ-1の発現を阻害したことが分かった(図5を参照のこと)。
カスパーゼ-1活性に対するIPS-07001の調節効果を調べるために、カスパーゼ-1アッセイを行った。結果として、IPS-07001は濃度依存様式でカスパーゼ-1活性を有意に阻害したことが分かった(図6を参照のこと)。
IPS-07001(#5182436)は、様々な炎症性サイトカインの産生およびmRNA発現を有意に阻害した(図7を参照のこと)。
ヒスタミン、IL-4、およびIgEレベルを、DNCB適用balb/cマウス血清中において測定した。IPS-07001(#5182436)は0.011μMの濃度で血清ヒスタミン、IL-4およびIgEレベルを有意に調節したことが分かった(図8を参照のこと)。
インビボアトピー性皮膚炎モデルにおいて、カスパーゼ-1活性に対するIPS-07001(#5182436)の調節効果を調べるために、カスパーゼ-1アッセイを、DNCB適用balb/cマウスの背部皮膚を使用して行った。結果として、IPS-07001(#5182436)は濃度依存様式でカスパーゼ-1活性を有意に阻害したことが分かった(図9を参照のこと)。
炎症性サイトカインレベルを、DNCB適用balb/cマウスの背部皮膚中において分析した。IPS-07001(#5182436)は0.011μMの濃度でIL-4およびIL-6レベルを有意に調節したことが分かった(図10を参照のこと)。
炎症性サイトカインmRNA発現がDNCB適用balb/cマウスの皮膚組織中において調節されたかどうかの分析の結果として、IPS-07001(#5182436)はアトピー性皮膚病巣中のTARC、TNF-aおよびIL-6 mRNA発現を調節したことが分かった(図11を参照のこと)。
A.目視観察
乾癬およびアトピー性皮膚炎は、皮膚の乾燥を示す代表的な疾患として公知である。これらの中で、特に、アトピー性皮膚炎は、皮膚の乾燥に加えて、過角化、紅斑、浮腫、重篤な痒み、滲出物、おでき、および痂皮を特徴とし、かつ、皮膚炎症など、例えば、急性期における水疱および慢性期における皮膚の肥厚のような様々な症状を伴う、皮膚疾患である(Bieber T. Atopic dermatitis. N Engl J Med. 2008;358:1483-94)。従って、本実験において、紅斑、乾燥皮膚、浮腫および表皮剥離、びらん、ならびに苔癬化に主に焦点を絞って、皮膚臨床症状を観察した。観察の結果として、明瞭な乾燥、紅斑、表皮剥離、および苔癬化が、正常対照と比較して、DNCBのみで処置された群において観察された。対照的に、紅斑を除いて、DNCB誘発皮膚症状が、デキサメタゾン処置群においてほとんど排除された。有意な視覚的変化は、試験材料処置群において0.01μMおよび0.1μMで観察されなかったが、紅斑、表皮剥離および苔癬化が、DNCB群と比較して1μMで低減した(図12を参照のこと)。
誘発されたアトピーを有するDNCB群において、組織学的観察は、マウスの皮膚組織の過角化を示した。DNCB群の表皮厚みは87.1±13.0μmであり、これは、正常対照群のそれ、即ち、24.5±10.3μmよりも有意に高く、アトピー性皮膚炎に典型的な過角化の発生を示した。陽性対照としてのデキサメタゾン処置群は39.9±9.4μmの表皮厚みを示し、表皮層の厚みの明瞭な減少を示した。0.01μM、0.1μM、および1μMの濃度にて試験材料で処置された群において、表皮厚みは、それぞれ、70.8±4.3μm、74.4±7.1μm、および60.0±8.2μmへ減少した(図13を参照のこと)。
アトピー性皮膚炎中の肥満細胞の脱顆粒によって分泌されるヒスタミンは、痒みの主な原因として作用しそれによって皮膚バリアを損傷しそして皮膚炎症を誘発することによって、アトピー性皮膚炎の進行において主要な役割を果たすことが公知である(Lee, E.J., G.E. Ji and M.K. Sung. 2010. Inflamm. Res. 59(10):847-854)。3週間の試験材料の適用後、0.1μM試験材料適用群(25.8±3.5, p<0.01)および1μM試験材料適用群(23.8±5.1, p<0.01)は統計的に有意な減少を示した。これらの結果はDNCB処置群のそれと類似し、これは、試験材料が肥満細胞の数を低減することによって痒みの緩和に有効であることを示している(図14を参照のこと)。
血清IgEの増加はアトピー性皮膚炎の指標であることが公知である。特に、IgEは、臨床的重症度と密接な相関関係を有することが公知である(M. Ban and D. Hetich, Toxicol. Lett, 118, 129 (2001)。アトピー性皮膚炎はIgE抗体応答を増加させる(Matsuda, H., Watanabe, N., Geba, G. P., Sperl, J., Tsudzuki, M. and Hiroi, J.: Int. Immunol. 9, 461 (1997)。加えて、増加したIgE産生は、増加した抗体応答および増加したIgE依存性ヒスタミン硝子体活性をもたらし、それによってヒスタミン分泌を促進する。ヒスタミンは、好酸球の浸潤を誘導すること、ならびに急性過敏症および痒みを引き起こすことが公知である(H. C. Sung, W. J. Lee, S. J. Lee, and D. W. Kim, Kor. J. Dermatol, 44, 1051 (2006)。従って、IgEの減少は、アトピー性皮膚炎の緩和の主要指標であると考えられる。本実験において、DNCB群(790.0±151.8)は、正常対照と比較してIgEレベルの70%以上の増加を示し、一方、デキサメタゾン処理群(703.8±86.5)は、僅かに減少したIgEレベルを示したが、統計的有意性は有さなかった。それぞれの濃度にて試験材料で処置された群において、0.1μM試験材料適用群(632.6±138.8, p<0.05)および1μM試験材料適用群(537.9±132.9, p<0.01)は、IgEレベルの統計的に有意な減少を示した(図15を参照のこと)。
IPS-07001の毒性パターンおよび毒性強度を同定するために、0μMおよび200μMの単回経皮投与を、雌および雄SDラットの両方へ与え、次いで、死んだおよび瀕死の動物の存在および非存在、臨床症状、ならびに体重変化を14日間観察した。観察が完了した後、SDラットを剖検し、異常を各個体について調べ、それによって以下の結果が得られた。
死んだ動物は、試験期間中、全ての試験材料投与群において観察されなかった(表7を参照のこと)。
IPS-07001の投与後、皮膚病変を含む臨床症状は、0および200μM/kg bwが投与された群の雄および雌ラットの両方において観察されなかった(表8を参照のこと)。皮膚紅斑が、0μM/kgおよび200μM/kg群の各々の1匹のラットにおいて観察されたが、これは試験材料によって引き起こされたとは考えられず、症状は投与後1日以内に排除された。
試験材料が投与された雌および雄ラットの全ての群が、投与後の第1日に体重の減少を示した。これらの結果は、試験材料処置のプロセスにおける環境変化に起因すると考えられ、正常な体重増加パターンが第2日から示された。(表9および10)。

観察期間の完了後、視覚的病理検査を、生きている動物に対して行い、個体はいかなる異常も示さなかった(表11を参照のこと)。

試験材料の経皮投与による致死用量(LD50)は、雌および雄ラットの両方において200μM/kg/4ml bw以上であった。
A.サイトカインおよびケモカイン抗体マイクロアレイタンパク質チップ
DNCBのみで処理された皮膚組織ならびにDNCBおよびIPS-07001で処理された皮膚組織の各々を破壊することによって得られた組織溶解物を、蛍光物質(Cy3またはCy5)で標識し、タンパク質発現プロファイリングを、抗体チップを使用して行った。抗体マイクロアレイタンパク質チップ分析を、ProteoChip上に固定化された抗体マップを参照して行った。26抗体固定化抗体チップ上の発現レベルを比較することによって得られた蛍光画像を図16-1に示す。INR分析結果についてのグラフを図16-2に示す。
DNCB単独処理皮膚組織ならびにDNCBおよびIPS-07001処理皮膚組織を破壊することによって得られた組織溶解物を、SDS-PAGEへ供し、次いで、PVDF膜へ転写し、特定のタンパク質の発現レベルを、特異的結合性抗体を使用して調べた。
07001(式1)のスクリーニング後、07001の構造と類似した構造を有する化合物についての文献検索からIPS-07004を見つけ出し、Key Organics (London, UK)から購入した。
組換えカスパーゼ-1および薬物を反応させ、次いで、カスパーゼ-1アッセイキット(R&D Systems Inc., Minneapolis, MN, USA)を使用して活性を測定した。
実験動物
6週齢雌SPF(特定病原体除去)C57BL/6マウスをDuPont Biotechから購入した。温度21±3℃、湿度50±20%、および明/暗サイクル12h/12hで維持されたHoseo University Safety Evaluation Centerの動物室中において1週間の順化期間後、マウスに飼料および水を自由に摂取させた。本動物実験における全ての手順を、Hoseo Universityの動物実験倫理委員会(HTRC-15-19、およびHTRC-16-22(2))によって承認された後に行った。
大腸炎誘発因子としてのデキストラン硫酸ナトリウム(Mpbio. com製のcat no 160110)を、飲料水中3%濃度へ希釈し、マウスへ経口投与し、腸炎を誘発した。マウスを6つの群へ分割し、DWを対照の動物へ投与し、1.0 mg/kgおよび10 mg/kgの試験材料を5週間の間1日1回実験群の動物へ経口投与した。粗製オリーブ油中に溶解されたスルファサラジン(100 mg/kg)を、陽性対照の動物へ1日1回経口投与した。急性症例において、疾患進行レベルを5日間の投与後に調べた。慢性症例において、疾患進行レベルを5日間の投与後15日および30日に調べた(参考文献:Toxicology Reports 2 (2015) 10391045 -Caryophyllene attenuates dextran sulfate sodium-induced colitis in mice via modulation of gene expression associated mainly with colon inflammation)。
体重を試験期間中5日間以上の間に1回測定した。測定された体重を、正常対照と比較して差異がなかった場合は0ポイント、15%の体重減少率については1ポイント、510%については2ポイント、1015%については3ポイント、および>15%については4ポイントとして定量化した。糞便観察を行い、下痢の発生を調べ、その結果をスコアとして定量化した(0、正常な便;1、やや柔らかい便;2、非常に柔らかい便;3、非常に柔らかい便(規則形状無し);4、水様便)。
直腸組織および血液を、剖検時間のそれぞれについて5匹の個体から収集した。Yuhanケタミン50(登録商標)(Yuhan Corporation, Korean)およびRompun(Bayer, USA)を3:1の比で混合し、次いで、筋肉注射によって動物を麻酔するために使用した。それぞれの麻酔された動物を開腹術へ供し、腹大動脈から血液を収集し、血液収集後、腹大動脈を切断し、静脈切開へ供し、死なせ、直腸組織を収集した。収集された組織の長さおよび重量を測定し、組織1 cm当たりの重量を計算した。収集された組織の一部を生検に使用し、その一部を分析前に液体窒素中に凍結して-70℃で保存した。
組織学的評価のために、直腸の一部の組織を、4%中性緩衝ホルマリン(4% NBF)中に固定し、続いて一般的な組織学的検査を行い、次いで、ミクロトームを使用して4μmの厚みへ薄切片化し、ヘマトキシリン&エオシン染色をそれにおいて行った。炎症の程度を光学顕微鏡下での観察に基づいて5つの段階(正常-0、最小限-1、軽度-2、中程度-3、および重度-4)へ分割した。
IPS-07004の場合、HaCat細胞ベースのアッセイにおいて細胞傷害性は観察されず、化合物はHMC-1細胞中において組換えカスパーゼ-1活性およびカスパーゼ-1活性の両方を阻害し、それによってアトピーを阻害することが分かった(図22〜24を参照のこと)。
腸炎に対するIPS-07001およびIPS-07004の阻害効果を、上記実験方法によって構築されたDSS誘発動物モデルを使用して観察した。用量を1.0 mg/kgおよび10 mg/kgへ分割し、経口投与を行った。効能を、腸の長さに対する腸の重量の比率(wt/cm)によって分析し、重量変化、下痢、下血などによってさらに分析した。実験の結果として、2つの物質が、DSS対照と比較して10 mg/kgの用量で優れた腸炎阻害活性を示すことが分かった(図18〜20、25、27および28を参照のこと)。重量変化は正常群のそれと類似し、下血はDSS誘発群と比較して有意に減少したことが観察された(図20、26、および27を参照のこと)。この腸炎阻害機構は、インフラマソームの重要なタンパク質であるカスパーゼ-1を阻害することによって、炎症誘発機構を制御すると考えられる。
Claims (21)
- 75 mg〜350 mgの式4によって示される化合物、またはその薬学的に許容される塩を含む、請求項1に記載の薬学的組成物。
- 患者の肥満細胞からの胸腺間質性リンホポイエチン(thymic stromal lymphopoietin)(TSLP)分泌を阻害する、請求項1に記載の薬学的組成物。
- 患者のカスパーゼ-1活性を阻害する、請求項1に記載の薬学的組成物。
- 患者のIL-4および/またはIL-6レベルを減少させる、請求項1に記載の薬学的組成物。
- 患者の血清IgEレベルを減少させる、請求項1に記載の薬学的組成物。
- 患者の表皮厚みを減少させる、請求項1に記載の薬学的組成物。
- 患者の皮膚組織中の肥満細胞の数を減少させる、請求項1に記載の薬学的組成物。
- 75 mg〜350 mgの式4によって示される化合物、またはその薬学的に許容される塩を含む、請求項9に記載の薬学的組成物。
- 経口投与されるように用いられる、請求項9に記載の薬学的組成物。
- 炎症性腸疾患が潰瘍性大腸炎である、請求項9に記載の薬学的組成物。
