Abstract
Description
従来の毛髪ミネラル検査で用いられていた、原子吸光法、吸光光度法、中性子放射化分析法、又はICP−MS法を用いた元素の定量は、試料の煩雑な前処理やサンプル調製に高度なテクニックを必要とする。そのため、これら高度なテクニックを備えた技術者でなければ、正確なデータを得ることが難しく、また、前処理に時間と経費を強いられるという課題があった。さらに、汚染を避けるために、高価な設備であるクリーンルームなどを設置する必要があるという課題もあった。
特許文献1、2には、蛍光X線分析により、毛髪に含有されるミネラルの含有率、生体液に含有されるミネラル金属の含有比率及び/又は含有量を測定することが記載されている。しかしながら、特許文献1、2には、蛍光X線分析による元素分析の結果から、これら生体試料中のミネラルの含有量とともに、アミノ酸の含有量を検出するという技術的思想は、一切記載も示唆もなされていない。
上記特許文献1、2には、蛍光X線分析により、生体液に含有される元素を定量分析するにあたり、生体液に含まれるいずれの元素を内部標準の基準元素にすべきかを示唆する記載は一切なされていない。
特許文献1に記載のように、毛髪の場合は、毛髪を構成するケラチンに含まれる硫黄を内部標準の基準元素として採用することができたが、血液などの生体液には、不溶性のケラチンは含まれていないため、生体液の場合に、いずれの元素を基準にすべきなのかが明確ではなかった。
一方で、蛍光X線分析による血液の分析結果において、硫黄が検出され、その硫黄が、水溶性のアミノ酸であるメチオニン由来であることを見い出した。
蛍光X線分析装置を用い、被検者が採取可能な被検者由来の生体液に含まれるミネラルの、前記生体液に含まれる内部標準の基準元素としてのメチオニン由来の硫黄に対する含有比率を測定し、
測定された前記含有比率と、前記硫黄及び前記ミネラルの標準溶液を用いて蛍光X線分析を行って作成しておいた検量線とを用いて、前記生体液に含まれる前記硫黄及び前記ミネラルの含有量を算出し、
算出された前記硫黄の含有量に基づいて、前記生体液に含まれるメチオニンの含有量を算出し、
前記含有比率を測定する際に、
前記生体液を1滴又は複数滴だけ採取した試料を用いて、前処理を行うことなく、前記蛍光X線分析装置による分析を行うことを特徴としている。
また、上記生体液の検査方法(1)によれば、前記生体液を1滴又は複数滴だけ採取した試料を用いることにより、前記被検者から前記生体液を採取する際の負担を減らすことができる。また、前記試料の前処理を行うことなく、前記蛍光X線分析装置による分析を行うので、高度なテクニックがなくても簡便に分析を行うことができる。
同一の前記被検者から定期的に採取された前記生体液に含まれる前記ミネラルの、前記生体液に含まれる前記メチオニン由来の硫黄に対する含有比率を定期的に測定し、
定期的に算出された前記生体液に含まれる前記ミネラル及び前記メチオニンの含有量の経時変化を、前記被検者の健康状態を維持又は改善するための指標の一つとすることを特徴としている。
算出された前記生体液に含まれる前記ミネラル及び前記メチオニンの含有量と、健常人の基準含有量とを比較して、過不足に関する評価を行うことを特徴としている。
前記生体液が、被検者自身が採取した血液、尿、唾液、汗、及び涙のうちの少なくとも一つを含むことを特徴としている。
前記生体液が、被検者自身が採取した血液、尿、唾液、汗、及び涙のうちの少なくとも2種以上を含むことを特徴としている。
前記ミネラルが、マグネシウム、リン、塩素、カリウム、鉄、銅、亜鉛、ゲルマニウム、及び臭素のうちの少なくとも2つ以上を含むことを特徴としている。
また本発明に係る生体液の検査方法(7)は、上記生体液の検査方法(1)〜(6)のいずれかにおいて、
被験者自身が自己採取用キットを用いて前記生体液を採取することを特徴としている。
また本発明に係る生体液の検査方法(8)は、上記生体液の検査方法(1)〜(6)のいずれかにおいて、
被検者自身がろ紙、薄膜、又はフィルムからなる点滴材料に前記生体液を採取することを特徴としている。
また本発明に係る生体液の検査方法(9)は、上記生体液の検査方法(1)〜(8)のいずれかにおいて、
前記生体液の検査結果に基づいて、被検者毎の報告書を作成し、
該報告書には、
前記生体液に含まれる前記ミネラル及び前記メチオニンの検査結果とともに、前記蛍光X線検査装置を用いて検査された、被検者由来の毛髪に含まれるミネラルの検査結果が含まれていることを特徴としている。
また本発明に係る生体液の検査方法(10)は、上記生体液の検査方法(1)〜(3)のいずれかにおいて、
前記生体液の検査結果に基づいて、被検者毎の報告書を作成し、
該報告書には、
前記生体液に含まれる前記ミネラル及び前記メチオニンの検査結果とともに、前記蛍光X線検査装置を用いて検査された、被検者由来の毛髪に含まれるミネラルの検査結果が含まれ、
前記生体液の検査結果には、少なくとも血液の検査結果が含まれ、
前記毛髪に含まれるミネラルの検査結果には、前記血液の恒常性機構により一定に維持されるミネラルの検査結果が含まれていることを特徴としている。
図1は、実施の形態に係る生体液の検査方法を説明するためのフローチャートである。
まず、ステップS1では、蛍光X線分析装置を用い、被検者が採取可能な被検者由来の生体液に含まれるミネラルの、前記生体液に含まれるメチオニン由来の硫黄に対する含有比率を測定する。前記含有比率は、蛍光X線分析により検出されたピークの半値幅や高さ、面積などの値から求めた比率であり、換言すれば、信号比率ともいう。
また、蛍光X線分析を行う生体液は、被検者自身が採取した血液、尿、唾液、汗、及び涙のうちの少なくとも2種以上を含んでもよい。これら生体液は、例えば、自己採取用キットなどを用いて、被検者自身で採取することが可能である。
作成しておいた前記検量線のデータは、蛍光X線分析装置、又は該蛍光X線分析装置に接続されたコンピュータ装置に登録しておき、これら装置を用いて、測定された含有比率を、検量線の式に当てはめることにより、前記生体液に含まれる硫黄及びミネラルの含有量を算出するようにしてもよい。
本実施例では、血液中の元素として、マグネシウム(Mg)、リン(P)、硫黄(S)、塩素(Cl)、カリウム(K)、鉄(Fe)、及びゲルマニウム(Ge)を蛍光X線分析装置で分析した。
図2に示すスペクトル図では、リン(P)、硫黄(S)、塩素(Cl)、カリウム(K)、鉄(Fe)、及びゲルマニウム(Ge)のピークが検出されている。硫黄(S)は、メチオニン由来の硫黄である。
図3に示すスペクトル図では、マグネシウム(Mg)のピークが検出されている。軽金属(軽元素)であるマグネシウムを測定する場合は、発生する蛍光X線が微弱であるため、減衰しないように装置内部を真空にすることが好ましい。
検量線の作成には、マグネシウム(Mg)、リン(P)、塩素(Cl)、カリウム(K)、鉄(Fe)、及びゲルマニウム(Ge)などの各ミネラルの標準溶液と、硫黄(S)の標準溶液が用いられる。
例えば、硫黄の標準溶液を、血液中での硫黄の濃度範囲を考慮して、3〜5段階の濃度に調製し、これら調製した標準溶液を、例えば、ろ紙、又はスライドガラスなどに所定量だけ滴下し、乾燥させる。乾燥後の残留物を、蛍光X線分析装置により分析して、各濃度における蛍光X線強度を測定し、蛍光X線強度を縦軸にとり、硫黄の含有量(濃度)を横軸にプロットすることにより、検量線が作成される。図10に示す硫黄の検量線についても、良好な直線性が確認された。
図11は、実施例2における被検者唾液に含まれるミネラルの蛍光X線分析によるスペクトル図の一例である。横軸はエネルギー、縦軸は蛍光X線強度を示す。
図11に示すスペクトル図では、リン(P)、硫黄(S)、塩素(Cl)、カリウム(K)、及びゲルマニウム(Ge)のピークが検出されている。硫黄(S)は、メチオニン由来の硫黄である。被検者唾液では、被検者血液で検出された鉄が検出されていないが、他のミネラルは同様に検出された。
図12は、実施例3における被検者尿に含まれるミネラルの蛍光X線分析によるスペクトル図の一例である。横軸はエネルギー、縦軸は蛍光X線強度を示す。
図12に示すスペクトル図では、リン(P)、硫黄(S)、塩素(Cl)、カリウム(K)、及びゲルマニウム(Ge)のピークが検出されている。硫黄(S)は、メチオニン由来の硫黄である。被検者尿では、被検者血液で検出された鉄が検出されていないが、他のミネラルは同様に検出された。
「血液による検査結果」欄には、ミネラルの検査結果一覧表と、ミネラルの過不足に関する評価コメントとが記載されている。ミネラルの検査結果一覧表には、分析されたミネラルの種類毎に、測定値(ppm)と基準値(ppm)とが示され、さらに、測定値が基準値と比較して低いか、基準内か、又は高いかを示す横棒グラフが示されている。
このような報告書を作成すれば、血液などの生体液に含まれる各種ミネラル及びメチオニンの含有量と、健常人の基準含有量とが比較できるので、ミネラル等の摂取過不足に関して、健常人と比較した相対的な評価を適切に行うことができる。これにより、血液に含まれるミネラル及びメチオニンの含有量の過不足を、健康状態を維持又は改善するための指標の一つとして利用することが可能となる。
Claims (10)
- 蛍光X線分析装置を用い、被検者が採取可能な被検者由来の生体液に含まれるミネラルの、前記生体液に含まれる内部標準の基準元素としてのメチオニン由来の硫黄に対する含有比率を測定し、
測定された前記含有比率と、前記硫黄及び前記ミネラルの標準溶液を用いて蛍光X線分析を行って作成しておいた検量線とを用いて、前記生体液に含まれる前記硫黄及び前記ミネラルの含有量を算出し、
算出された前記硫黄の含有量に基づいて、前記生体液に含まれるメチオニンの含有量を算出し、
前記含有比率を測定する際に、
前記生体液を1滴又は複数滴だけ採取した試料を用いて、前処理を行うことなく、前記蛍光X線分析装置による分析を行うことを特徴とする生体液の検査方法。 - 同一の前記被検者から定期的に採取された前記生体液に含まれる前記ミネラルの、前記生体液に含まれる前記メチオニン由来の硫黄に対する含有比率を定期的に測定し、
定期的に算出された前記生体液に含まれる前記ミネラル及び前記メチオニンの含有量の経時変化を、前記被検者の健康状態を維持又は改善するための指標の一つとすることを特徴とする請求項1記載の生体液の検査方法。 - 算出された前記生体液に含まれる前記ミネラル及び前記メチオニンの含有量と、健常人の基準含有量とを比較して、過不足に関する評価を行うことを特徴とする請求項1記載の生体液の検査方法。
- 前記生体液が、被検者自身が採取した血液、尿、唾液、汗、及び涙のうちの少なくとも一つを含むことを特徴とする請求項1〜3のいずれかの項に記載の生体液の検査方法。
- 前記生体液が、被検者自身が採取した血液、尿、唾液、汗、及び涙のうちの少なくとも2種以上を含むことを特徴とする請求項1〜3のいずれかの項に記載の生体液の検査方法。
- 前記ミネラルが、マグネシウム、リン、塩素、カリウム、鉄、銅、亜鉛、ゲルマニウム、及び臭素のうちの少なくとも2つ以上を含むことを特徴とする請求項1〜5のいずれかの項に記載の生体液の検査方法。
- 被験者自身が自己採取用キットを用いて前記生体液を採取することを特徴とする請求項1〜6のいずれかの項に記載の生体液の検査方法。
- 被検者自身がろ紙、薄膜、又はフィルムからなる点滴材料に前記生体液を採取することを特徴とする請求項1〜6のいずれかの項に記載の生体液の検査方法。
- 前記生体液の検査結果に基づいて、被検者毎の報告書を作成し、
該報告書には、
前記生体液に含まれる前記ミネラル及び前記メチオニンの検査結果とともに、前記蛍光X線検査装置を用いて検査された、被検者由来の毛髪に含まれるミネラルの検査結果が含まれていることを特徴とする請求項1〜8のいずれかの項に記載の生体液の検査方法。 - 前記生体液の検査結果に基づいて、被検者毎の報告書を作成し、
