Description
まず、バイオマスを用意する。本発明に用いられるバイオマスの種類は特に限定されず、当該技術分野で通常用いられる生物由来のバイオマスを全て用いることができる。具体的には、例えば特許文献2に記載のバイオマス(木材、藁や籾殻などの植物繊維素、紙、パルプ類のほか、これらの粉砕物など)が挙げられる。
・図1の上段(No.1):ブタノールを30mL、硫酸を50mL添加して前処理を行った。このときのブタノール比率は37.5%(=[30/(30+50)]×100)である。
・図1の中段(No.2):ブタノールを40mL、硫酸を40mL添加して前処理を行った。このときのブタノール比率は50%(=[40/(40+40)]×100)である。
・図1の下段(No.3):ブタノールを50mL、硫酸を30mL添加して前処理を行った。このときのブタノール比率は62.5%(=[50/(50+30)]×100)である。
次に、上記のようにして得られた固体画分に糖化酵素を加えて糖化させる。本発明に用いられる糖化酵素は、当該技術分野で通常用いられるものであれば特に限定されず、例えば、セルラーゼなどが挙げられる。
上記のようにして得られた糖類をエタノール発酵すると、エタノールが得られる。エタノール発酵の方法は特に限定されず、通常用いられる方法を採用することができる。例えばエタノール発酵に用いられる微生物は、糖類を代謝してエタノール発酵を行うことができるものであれば特に限定されず、出芽酵母などの酵母、大腸菌などが挙げられる。
本実施例ではモノアルコールとしてブタノールを用いて前処理を行ったときにおける固体画分の重量を測定した。前処理による固体画分の重量は、前処理によるバイオマスの分解効率を表している。すなわち、固体画分の重量が少ない程、バイオマスが効率的に分解されていることを示唆している。
本実施例では、上記実施例1によって得られた各固体画分を酵素糖化させたときのグルコース濃度を測定した。固体画分から得られるグルコース濃度が高い程、固体画分が効率的に分解されていることを示唆している。
・カラム:Agilent CP−Sil 8CB−MS(30m×0.25mm)
・キャリアーガス:ヘリウム
・注入温度:230℃
・オーブンの温度:0〜2分までは80℃とし、2分以降は、1分当たり15℃ずつ330℃まで上昇させた。
本実施例では、上記実施例1によって得られた固体画分(ブタノール濃度25%を使用したもの)を用い、酵素糖化、およびエタノール発酵を行ったときのエタノール生成量を測定した。
本実施例では、上記実施例1〜3で用いたブタノール以外のモノアルコールを用いて前処理を行ったときにおける固体画分の重量を比較検討した。
本実施例では、上記実施例4によって得られた各固体画分を酵素糖化させたときのグルコース濃度を、前述した実施例2と同様にして測定した。
前述した実施例で用いたバイオマス(ソルガム)は、セルロース、ヘミセルロース、リグニン(酸不溶性リグニンと酸可溶性リグニン)を主成分として含む。本実施例では、エタノール、1−プロパノール、2−プロパノール、1−ブタノール、1−ペンタノールの各種モノアルコール(濃度は全て12.5%)を用い、実施例1と同様にして前処理を行ったバイオマス(ソルガム)の成分を、国立再生可能エネルギー研究所(National Renewable Energy Laboratory、NREL)に記載の下記手順に基づいて各成分に分画して組成分析を行い、比較検討した。これらの測定は3回行い、その平均値と標準偏差を調べた。

=グルコース濃度(g/L)×0.1÷300×(180−18)÷180×100
・ヘミセルロース(%)
=キシロース濃度(g/L)×0.1÷300×(150−18)÷150×100
Claims (2)
- バイオマスを前処理し、酵素糖化により糖類を製造する方法であって、
前記前処理を、無機酸;並びに2以上、8以下の炭素を含むモノアルコールの存在下(但し、脂肪族アルコール、水、および二酸化硫黄の溶液を除く。)、常圧(加熱による陽圧を含む)で、100〜200℃の温度に加熱して行うと共に、
前記酸と前記モノアルコールの合計に対する前記モノアルコールの容量比率は、10%以上、40%未満であることを特徴とする糖類の製造方法。 - 請求項1に記載の製造方法を用いて化成品を製造する方法。
