Description
ワクチンは予防の礎として残されているが、これらの新たな病原体に対する有効なワクチンを開発するにはかなりの時間を要する。抗菌剤(抗ウイルス、特にインフルエンザウイルスノイラミニダーゼ阻害剤および抗生物質を含む)が利用可能であるが、それらの有効性は、病原体が変異し、薬物耐性となる能力によって損なわれる可能性がある6,7。新しい治療アプローチの必要性があるのは明らかである。
「Engineering Multivalent Sialic Acid Recognition using the CBM40 module from Vibrio cholerae Sialidase」(2008年5月17日公開、http://www.biochem.emory.edu/conferences/glycot/Images/GlycoTProgram-Posters.pdfを参照)と題されるポスターは、多価性であることでシアル酸に対する親和性が増加した試薬の開発について記載している。しかしながら、そのポスターは、このような試薬が、病原体によって引き起こされる疾患および/または状態の治療に有用な免疫調節化合物として任意の用途を有することについては開示していない。
シアル酸に結合する能力を示す分子は、宿主細胞の表面上のシアル酸を含有する受容体の存在を利用する病原体による感染を中和または予防するために使用され得ることが知られている。例えば、オルトミクソウイルス科またはパラミクソウイルス科に属するウイルス、およびある種の連鎖球菌属(Streptococcus)細菌などの呼吸器病原体は、種々の哺乳動物組織における特定の細胞型に結合し、入り込むための細胞表面のシアル酸を含有する受容体を利用する。シアル酸部分および/またはそれを含む分子(例えば、細胞表面受容体)に結合する化合物、例えば、タンパク質およびペプチドは、細胞に結合または付着する手段として、細胞表面のシアル酸受容体を利用する病原体によって引き起こされるまたはそれに寄与される疾患の治療および/または予防に使用され得る。理論に束縛されるものではないが、シアル酸結合分子は、結合および/または付着から病原体を妨げるように、病原体と細胞表面のシアル酸受容体の間の相互作用を干渉し、および/または阻害し、妨げおよび/またはブロックすることができる。
本発明者らは、ここで、病原体と、例えば、シアル酸を含有する細胞表面受容体の間の相互作用を干渉またはブロックし、妨げおよび/もしくは阻害することに加えて(または、おそらくはその結果としてもしくはそれと組み合わせて)、ある種のシアル酸結合分子が免疫調節特性を有することを発見した。
したがって、第一の態様において、本発明は、対象における免疫応答の調節またはプライミングに使用するためのシアル酸結合分子を提供する。さらに、本発明は、対象における免疫応答を調節またはプライミングする方法において使用するためのシアル酸結合分子を提供する。
さらに、本発明は、対象における免疫応答を調節する医薬を製造するためのシアル酸結合タンパク質の使用を提供することができる。また、本発明は、対象における免疫応答を調節する方法を提供することができ、該方法は、必要とする対象に本発明のシアル酸結合タンパク質の免疫調節(または免疫刺激)の量(amount)または量(quantity)を投与することを含む。
例えば、本発明は、例えば、ケモカインおよび/もしくはサイトカイン分子ならびに/またはそれらをコードするおよび/もしくはそれに関連する任意の遺伝子/転写因子を含む1つまたは複数の免疫調節化合物の発現、機能および/もしくは活性を増加または増強するシアル酸結合分子を提供し得る。
さらに、本発明のシアル酸結合分子は、感染部位への免疫細胞の動員、増殖および/または移動を調節する(例えば、誘導するまたは増加する)ことができる。例えば、本発明のシアル酸結合分子が投与された対象において、免疫細胞の動員、増殖および移動のプロセスは、ある程度および/またはより迅速に悪化され得る。
さらに、本発明は、対象における1つまたは複数のケモカインおよび/またはサイトカインの発現、機能および/または活性を刺激、増強または増加する医薬の製造においてシアル酸結合タンパク質の使用を提供することができる。さらに、本発明は、対象における1つまたは複数のケモカインおよび/またはサイトカインの発現、機能および/または活性を刺激、増強または増加する方法を提供することができ、該方法は、それを必要とする対象に本発明のシアル酸結合タンパク質の免疫調節量(amount)または量(quantity)を投与することを含む。

新規な遺伝子操作されたタンパク質を用いて宿主受容体を標的にすることによるインフルエンザの予防
方法
ウイルス。インビトロ研究に関して、以下のインフルエンザウイルスを使用した:A/WSN/1933(H1N1)、A/プエルトリコ/8/1934(A/PR/8/1934、H1N1)、A/Udorn(ウドーン)/1972(H3N2)、およびB/香港/1973は、Madin−Darbyイヌ腎細胞(MDCK、アメリカン・タイプ・カルチャー・コレクション、Manassas、VA)中で増殖させた。MDCK細胞におけるウイルス感染性をプラークアッセイによって決定し、log10PFU/mlとして表した。インビボ研究に関して、2つの菌株を使用した:マウスに適合されたA/WSN/1933(H1N1)株はMDCK細胞内で増殖し、マウスに適合されたA/カリフォルニア/04/2009(H1N1)ウイルス30は発育鶏卵で増殖させた。A/カリフォルニア/04/2009(H1N1)について50%マウス致死的投薬量(MLD50)を決定するために、4匹の雌性6週齢のBALB/cマウス(Jackson Laboratories、Bar Harbor、ME)をイソフルランで軽く麻酔し、50μLの10倍連続希釈した、PBS中のウイルスを用いて鼻腔内接種した。MLD50値は、21日間の観察期間後に決定された。
インフルエンザウイルスは、表面の赤血球凝集素(HA)糖タンパク質を介して、気道上皮に存在するシアル酸受容体に結合し、そのイベントは、ウイルスのエンドサイトーシスを引き起こす12。2009年のパンデミックH1N1ウイルスなどのヒトインフルエンザウイルスは、上気道に存在するα−2,6連結したシアル酸受容体を認識し、一方、H5N1などのトリインフルエンザウイルスは、α−2,3連結したシアル酸受容体を主に認識するが、このような受容体はまた、ヒトの下気道に存在する13,14。最近出現したヒトH7N9ウイルスは、受容体の両タイプを認識する点で異例であり、したがって、持続的なヒトからヒトへの伝染およびパンデミック能力の可能性がある15,16。本発明者らは、特定のタンパク質を用いて気道におけるこのような受容体をマスキングすることが、感染を妨げるための新規な治療経路を提供し得るという仮説を立てた。多数のシアル酸結合タンパク質が知られているが、ほとんどはシアル酸に対する親和性が低く、例えば、HAモノマーはその受容体に対して約2.5mMの親和性であるが、三量体であることを通じて、およびウイルス表面上に高コピー数で存在させることによって親和性を高める17。天然を模倣し、本発明者らは、ビブリオ・コレラNanHシアリダーゼ18由来のシアル酸結合ドメイン(VcCBM、図1a)またはストレプトコッカス・ニューモニエNanAシアリダーゼ由来の相同なドメイン(SpCBM、図1b)のいずれかを用いて多価形態を遺伝子操作した。多価は、タンデムリピート8を介して、またはシュードモナス・エルギノーサシアリダーゼ19由来のオリゴマー化ドメイン(TD)(図1c)に1つもしくは2つのタンデムに連結されたCBMドメインであって、三量体を形成するために自己会合するドメインを融合させることによって達成された。VcCBMとSpCBMの多価形態は、表面プラズモン共鳴(SPR)によって測定したとき、解離定数が1nM未満である結合力を介して親和性を上げる(図1d、表1)。グリカンアレイスクリーニングは、VcCBM8およびSpCBM(図5)がα−2,3またはα−2,6連結したシアル酸を認識することを示し、結晶学的分析は、両方のドメインがシアリルラクトースを用いた研究において末端シアル酸のみを認識することを明らかにする18。

マウスガンマヘルペスウイルス68(MHV−68)を用いて感染させたマウスにおける遺伝子操作されたタンパク質Vc2CBMTDとSp2CBMTDの予防効果
導入
マウスガンマヘルペスウイルス68(MHV−68)は、ヒトに感染するカポジ肉腫関連ヘルペスウイルス(KSHV)とエプスタイン−バーウイルス(EBV)に密接に関連している2天然に存在するげっ歯類の呼吸器病原体1である。ガンマヘルペスウイルスは、それらのエンベロープ上に、ウイルスの細胞から細胞への伝達に関与している多数の表面タンパク質、糖タンパク質H(gH)および糖タンパク質L(gL)および糖タンパク質150(gp150)を提示する。EBVと同様に、MHV−68は、感染から後何ヶ月も後に発生し得る、リンパ増殖性疾患3と関連付けられる。しかしながら、EBVおよび他の多くのガンマヘルペスウイルスとは異なり、MHV−68は、インビトロで上皮細胞内で複製し、マウスの実験室系統に感染し、したがって、ガンマヘルペスウイルスの研究4のために優れたモデルを提供する。治療については、抗ウイルス剤は、通常、ほとんどのヘルペスウイルス感染の治療に提案されている。
インビボ研究。マウスの研究は、エジンバラにある感染症センターでインフルエンザ研究のための動物実験施設で行われ、Animals(Scientific Procedures)Act 1986に従ったUKホームオフィスライセンスの下で実施された。マウスは、ハロタン(Rhone Merieux Ltd,Harlow,Essex,UK)を用いて麻酔され、その後、50μgのVc2CBMTDまたはSp2CBMTDのいずれかの鼻腔内投与が−7日、−3日および−1日に3回与えられ、続いて、40μlのPBS中の4×105個のMHV68でチャレンジされた。感染の5日後、マウスを処分し、ウイルス力価の決定とサイトカイン分析のために死後にそれらの肺を回収した。
ウイルス力価。感染性ウイルス力価は、MDCK細胞において標準的なプラークアッセイによって決定された。マウスにおける肺ウイルス力価は、感染の5日後に決定された。
サイトカイン分析。清澄化されたマウス肺ホモジネート由来のMIP−2、TNF−α、IL−6、IL−1β、IFN−γ、GM−CSFおよびIL−2のサイトカイン分析は、製造業者の指示書(RD Biosystems,UK)に従って、Quantikine ELISAキットを用いて行われた。
本発明者らは、非シアル酸結合呼吸器病原体であるMHV−68ウイルスを用いてチャレンジした場合、BALB/cマウスにおける肺のウイルス負荷と免疫応答における六量体mCBM、Vc2CBMTDおよびSp2CBMTDの効果を検討した。生物製剤は、ウイルスチャレンジ前に、3回の投薬量(50μg、−7、−3、−1日)として鼻腔内投与された。全ての処理されたマウスは、未処理の感染マウスと比較して、感染の5日後の肺ウイルス力価において一対数減少を示した(図12a)。炎症性メディエーターIL−1β、MIP−2(IL−8のマウスホモログ)、IFN−γ、TNF−α、GM−CSF、IL−6およびIL−2をモニターするためにELISAを使用したマウスの肺ホモジネートのさらなる分析は、処理と未処理の感染マウス間において差異を示した。両方の生物製剤は、未処理の感染マウスと比較して、有意により低いレベルのIL−6、MIP−2およびIFN−γを示したが、TNF−α、IL−2およびIL−1βのレベルは感染マウスのみに類似していた(図12b)。このデータは、mCBMsの高度な予防的治療が、IL−6およびIFN−γなどのMHV−68感染によって刺激される特定の炎症性メディエーターのレベルの低下によって見られるように、免疫応答を調節することにより感染に応答する宿主を調製し得ることを示唆する。mCBMsは、潜在的に有害なケモカインの発現を妨げながら、MHV−68ウイルスチャレンジに対する免疫応答を増強するために、初期炎症プロセスを誘導する可能性がある。さらなる研究は、動物モデルに投与した場合、mCBMの免疫応答プロファイルのタイプを理解する必要がある。

材料および方法
ウイルス。インフルエンザA/Anhui/1/2013(H7N9)ウイルスは、世界保健機関の監視ネットワークを介して得られた。A/Anhui/1/2013(H7N9)ウイルスは、10日齢の発育鶏卵(卵)の尿膜腔に臨床試料を培養することにより、患者から単離され、実験に使用したウイルスのストックは卵中に調製された(継代歴:E2/E2)。動物の50%が死亡したマウス致死的投薬量(MLD50)は、21日後、6週齢の雌性BALB/cマウスにおいて決定された(体重、18〜20g;Jackson Laboratories,Bar Harbor,Maine)。重篤な疾患を示し、最初の体重の>25%を失った動物を安楽死させた。
H7N9ウイルス高投薬量による再感染。A/Anhui/1/2013(H7N9)インフルエンザウイルスによる致死的感染から生存した全ての動物は、感染後の21日に25MLD50のウイルスを用いて再感染させた。マウスは、感染の臨床徴候と生存について毎日観察され、体重変化は、指定された日にモニターされた。
A/Anhui/1/2013(H7N9)インフルエンザウイルスで致死的にチャレンジされたBALB/cマウスの生存におけるSp2CBMTDタンパク質の効力。本発明者らは、A/Anhui/1/2013(H7N9)インフルエンザウイルスで致死的にチャレンジされたマウスの生存および臨床兆候における、Sp2CBMTDタンパク質の単回または反復投与の効果を評価した(表2a)。単回投薬量、2回および3回投薬量(100μg/マウス/日)として投与した場合に、Sp2CBMTDは、未感染マウスの体重変化と死亡を引き起こさなかった。本発明者らは、Sp2CBMTDがこれらの実験で使用した最高投薬量でマウスに対して非毒性であると結論付けた。
A/Anhui/1/2013(H7N9)ウイルスによるマウスの致死的チャレンジに対するSp2CBMTDを用いた反復投薬の有効性を評価するために、タンパク質の2回投薬がD−3とD−1に投与され、3回投薬がD−7、D−3、D1に投与された(図16)。Sp2CBMTDを2回および3回適用した場合、全ての投薬処方計画について100%生存であった。
A/Anhui/1/2013(H7N9)インフルエンザウイルスで致死的にチャレンジされたBALB/cマウスの体重変化におけるSp2CBMTDタンパク質の効力。100μg/マウス/日の最大Sp2CBMTD投薬量を動物に投与した場合に、体重変化はほとんどないまたは全くなかった。一方、0.1μg/マウス/日の投薬量は、体重減少に最小の効果を与え、この投薬量で処理された動物は、最も顕著な体重減少を経験した(表3a)。Sp2CBMTDの投与時期は、生存および体重減少に最も有益な効果の提供に重要である。Sp2CBMTDタンパク質による処理がウイルス接種の24時間後に開始された場合、動物は、最も顕著な重量変化を示した。全体的に、観察された体重変化は、H7N9感染後に観察された生存転帰と相関した。
・A/Anhui/1/2013(H7N9)インフルエンザウイルスによる致死的チャレンジ前のSp2CBMTDの反復投与は、最も有益な結果を与え、使用された最低投薬量(0.1μg/マウス/日)でさえ、BALB/cマウスの100%の生存をもたらした。これは、使用された投薬量の減少の可能性を示す。
・投与時期は、致死的マウスモデルにおけるSp2CBMTDタンパク質処理の有効性と関連する:タンパク質をA/Anhui/1/2013(H7N9)インフルエンザウイルス接種前に投与した場合に、完全な保護(100%生存率)が達成された。
・Sp2CBMTDの用量依存的効果は、H7N9致死的マウスモデルにおいて観察され、したがって、インフルエンザウイルスに対するこのタンパク質の特異性を示す。
・Sp2CBMTDの投与は、抗HA抗体の発生に影響を及ぼさず、免疫応答のレベルは、H7N9ウイルスの再感染に対して保護するには十分であった。

例3に示されたデータによって示されるように、インフルエンザウイルス複製に必須の細胞因子を標的とする化合物は、抗ウイルス療法に対する魅力的なアプローチを表す。Sp2CBMTDは、インフルエンザウイルスによって認識される気道上皮上のシアル酸を含有する細胞受容体をマスクする遺伝子操作された多価タンパク質である。出現ヒトA/Anhui/1/2013(H7N9)インフルエンザウイルスを用いたマウスの致死的感染に対するSp2CBMTDの抗ウイルス能力は、インビボでのその活性のメカニズムの基礎があったように調査された。Sp2CBMTDは、H7N9ウイルス接種の前(7日、3日もしくは1日)または後(6時間もしくは24時間)に単回投薬量または反復投薬量(0.1、1、10もしくは100μg)として鼻腔内にマウスに投与された。H7N9の致死的チャレンジ前の7日に投与された場合、単回Sp2CBMTD投薬量(10または100μg)は80%から100%のマウスを保護した。反復Sp2CBMTD投与は最大保護を与え、試験された最低投薬量(0.1μg)でさえ、マウスの100%生存をもたらした。Sp2CBMTDの投与は、炎症誘発性メディエーター(IL−6、RANTES、MCP−1、Il−1β、TNFα、MIP−1α、IP−10)の肺発現を誘導し、H7N9ウイルス感染前に気道に好中球を動員した。これは、より少ない顕著な炎症およびマウス肺からの迅速なウイルスクリアランスをもたらした。Sp2CBMTD投与は、ウイルス特異的な適応免疫応答に影響を及ぼさず、相同H7N9ウイルスまたは異種H5N1ウイルスのより高い投与量を用いた再感染に対して保護するのに十分であった。したがって、Sp2CBMTDは、致死的マウスモデルにおけるH7N9感染の予防に効果的であり、人畜共通のインフルエンザウイルスに対する予防オプションとして期待される。
ウイルス、細胞および生物製剤
インフルエンザA/Anhui/1/2013(H7N9)およびA/トルコ/15/2006(H5N1)ウイルスは、世界保健機関のネットワークを介して取得され、35℃で48時間、発育鶏卵で増殖させた。以前に記載されるように(22)、Madin−Darbyイヌ腎臓(MDCK)細胞をアメリカン・タイプ・カルチャー・コレクションから取得され、維持した。ストレプトコッカス・ニューモニエNanAシアリダーゼ由来の炭水化物結合モジュール40(CBM40)ドメインをコードする遺伝子とシュードモナス・エルギノーサのシューダミニダーゼ由来の三量体化ドメインをコードする遺伝子を用いて、Sp2CBMTDをPCRに基づくクローニング技術により生成させた(21)。H7N9とH5N1インフルエンザウイルスを用いた実験は、米国の農務省によって承認された動物バイオセーフティーレベル3+封じ込め施設で実施された。
6週齢の雌性BALB/cマウス(体重、18〜20g;Jackson Laboratories,Bar Harbor,ME)は、イソフルラン吸入により軽く麻酔し、50μLのPBS中の50%マウス致死的投薬量(MLD50)のA/Anhui/1/2013(H7N9)インフルエンザウイルスを5回用いて鼻腔内接種された。第一の研究について、グループあたり5匹のBALB/cマウスは、H7N9ウイルス接種前の7日、3日もしくは1日、またはH7N9ウイルス接種の6時間もしくは24時間後に鼻腔内に単回投薬量のSp2CBMTD(0.1、1、10もしくは100μg/マウス)を与えられた。Sp2CBMTDの反復投薬は、H7N9接種前に2回(3日および1日)投薬量または3回(7日、3日および1日)投薬量のいずれかとして与えられた。マウスの生存は、感染後(p.i.)の21日間、毎日モニターされた;重症疾患の兆候および25%の体重減少を示した動物を屠殺した。対照マウスは、7日、3日および1日に滅菌PBSを受けた。
A/Anhui/1/2013(H7N9)インフルエンザウイルスを用いた接種から3週間後、生存マウスは、25MLD50の相同なウイルスを用いて再感染された。Sp2CBMTDおよびH7N9ウイルス接種による初期処理で生存した他のマウスは、3週間後にSp2CBMTDの第二の投与を受け、次に、20MLD50のA/トルコ/15/2016(H5N1)ウイルスを感染された。
4つのサイトカイン[ガンマインターフェロン(IFN−γ)、インターロイキン−6(IL−6)、活性化調節された発現および分泌正常T細胞(RNTES)、単球走化性タンパク質−1(MCP−1)]の濃度、および4つのケモカイン[インターロイキン−1β(Il−1β)、腫瘍壊死因子アルファ(TNF−α)、マクロファージ炎症性タンパク質−1α(MIP−1α)、誘導タンパク質(IP−10)]の濃度は、製造業者の指示に従ってマウスMYCTOMAG−70K−PMX MILLIPLEX(登録商標)予備混合キット(Millipore)を用いて測定された。各サイトカインについて、標準曲線は、3.2〜10,000pg/mLの範囲であった。サイトカインは、感染後の0日、3日、6日および9日に25μLの肺ホモジネート試料において測定された。マルチプレックスプレートは、xPonentデータ収集および分析ソフトウェアを用いて、ルミネックス100/200アナライザーで読み取った。
第二の研究の各実験グループ(n=3)におけるマウスの肺を10%中性緩衝ホルマリン(NBF)を用いて全身灌流後に回収した。マウス肺を気管注入を介して膨張させ、包埋、切片化、インフルエンザAウイルス[核タンパク質(NP);US Biological]および好中球[ミエロペルオキシダーゼ(MPO);Thermo Shandon]についてヘマトキシリンおよびエオシンまたは免疫組織化学的(IHC)染色を用いた従来の組織病理学について染色する前の少なくとも7日間、10%NBF中に維持する。インフルエンザAウイルスNP染色およびMPO染色した切片を病理評価のために盲検化した。抗原の存在は、Aperio ScanScope XT Slide Scanner(Aperio Technologies)を用いて全肺切片のデジタル画像を捕捉し、次に、著しく減少または不存在のNPおよびMPO染色の場所と一緒に領域全体を手動で輪郭を描くことにより定量された。染色範囲が減少した肺領域の割合は、Aperio’s ImageScopeソフトウェアを用いて算出された。
血清試料をH7N9またはH5N1ウイルス感染の21日後に得、受容体−破壊酵素(Denka Seiken Co)を用いて処理し、56℃で1時間、熱不活性化し、0.5%シチメンチョウ赤血球(Rockland Immunochemicals)を用いた赤血球凝集素(HA)阻害アッセイにより抗HA抗体の存在について試験した。血清試料中の抗SpCBM抗体の存在は、96ウェルプレート(Corning)上に固定化された精製タンパク質(1μg/ウェル)を用いて、ELISAにおいて測定された(21)。
統計分析
ウイルス肺力価は、サイトカインおよびケモカインの濃度、ならびに抗SpCBM抗体力価は、対応のないスチューデントt検定によって分析された。NP染色およびMPO染色された肺細胞数は、GraphPad Prism 5.0ソフトウェアを用いて分散分析(ANOVA)により比較された。累積生存率は、カプラン・マイヤーログランク検定により算出された。
H7N9インフルエンザウイルスを用いて致死的にチャレンジされたマウスの生存におけるSp2CBMTDの効力。Sp2CBMTDが、A/Anhui/1/2013(H7N9)インフルエンザウイルスを用いて致死的にチャレンジされたマウスの生存率を改善したかどうかを決定するために、生物製剤が、ウイルスチャレンジ前の7日、3日または1日に0.1、1、10または100μgの投薬量で1回投与された。ウイルス接種され、PBS処理された対照動物は、進行性の体重減少を示し、感染後の8日と10日の間に全て死亡した(図17)。H7N9ウイルスチャレンジ前のSp2CBMTDの単回の鼻腔内投与は、マウスの用量依存的な保護をもたらした。Sp2CBMTDの最大単回投薬量(100μg)は最大保護を提供し、生物製剤をウイルス接種前の7日、3日または1日に投与した場合に、100%のマウスが感染に対して生存した(図17A)。10μgの単回投薬量は、ウイルス接種前の1日または7日もしくは3日に投与した場合に、それぞれ100%、80%および80%のマウスを保護した。しかしながら、ウイルスチャレンジ前の3日または1日に1μg投薬量を投与した場合に、たった60%および40%のマウスが生存した(図17A)。試験した最低投薬量(0.1μg)は、少なくとも効果的であり、たった20%のマウスが保護された。
A/Anhui/1/2013(H7N9)インフルエンザウイルスを用いて感染させたマウスの致死的チャレンジ前に2回または3回処方計画として与えられたSp2CBMTDの反復投与は、試験された生物製剤の全ての投薬量で完全な保護を与えた(図17C)。これらの結果は、H7N9ウイルス接種前のSp2CBMTDの反復投与がマウスの生存および体重減少の最小化に最も効果的であることを示した(表1b)。
H7N9インフルエンザウイルスを用いたマウスの再感染。Sp2CBMTD処理が適応免疫の誘導と干渉するかどうかを調べるために、A/Anhui/1/2013(H7N9)インフルエンザウイルスに対する血清抗HA抗体の力価を決定した。全ての生存マウスは、使用された処方計画に関係なく、抗HA抗体の中程度の力価(1:40〜1:160)を有した(表2b)。抗HA抗体力価は、25MLD50投薬量の相同H7N9ウイルス再感染に対して、生存動物を100%を保護するのに十分であった(データ示さず)。
本発明者らのデータは、Sp2CBMTDの抗ウイルス作用のメカニズムは複雑であり、2つの主要な要因:1)SAを含有する細胞受容体へのウイルス結合を妨げ(21)、2)宿主免疫応答を調節することによって駆動されることを示唆する。インフルエンザウイルス感染に対する保護におけるSp2CBMTDの多機能的役割は、先天性免疫応答の刺激、およびインフルエンザウイルス感染部位への免疫細胞の動員によって実証され、したがって、H7N9ウイルスによって誘導される免疫病理の重症度を低減する。Sp2CBMTDは、SAに加えて、まだ未知の受容体(単数または複数)に結合し、したがって、免疫応答を調節する能力を獲得することが可能である。
