基本手順
クロロ−2−ピリジル−t−ブチルホスフィンの合成のためのグリニャール試薬は、塩化イソプロピルマグネシウムにより「Knochel法」によって調製した(Angew.Chem.2004,43,2222−2226)。後処理は、Budzelaarの方法に従って行った(Organometallics 1990,9,1222−1227)。
1mlのグリニャール溶液を、NH4Clの飽和水溶液中においてクエンチし、エーテルで抽出する。Na2SO4で乾燥させた後、当該エーテル溶液のGCを記録する。定性的には、排他的にo−キシレンが形成されていることが観察される。
1mlのグリニャール溶液を、2mlの0.1MのHClでクエンチし、過剰な酸を0.1MのNaOHで滴定する。好適な指示薬は、0.04%ブロモクレゾール水溶液である。色は、黄色から青色へと変わる。0.74mlの0.1MのNaOHが消費されていた。2ml−0.74ml=1.26mlであり、0.126mmolのグリニャール化合物に対応する。ジグリニャールが存在するので、当該グリニャール溶液は0.063Mである。これは、90%を超える収率である。
