Description
IMS Healthによると、中枢神経系(CNS、脳及び脊髄)病態を処置するための薬物の世界市場は、2007年にはおおよそ700億ドルであり、この総額のうちのほぼ90億ドルが薬物送達技術から生まれた製品に相当した(Jain, 2008, Jain PharmaBiotech Report, Drug Delivery in CNS disorders)。このように、今日、神経学は、心臓血管内科及び腫瘍学と共に、三大治療領域の1つである。世界中のCNS疾患及び病態の罹患患者数は、心血管疾病又は癌患者数より多いが、神経学は、依然として未開拓市場である。これは、CNS病態を治療するための可能性のある薬物の98%が、血液脳関門すなわちBBBを通過しないという事実によって一部には説明される(Pardridge, 2003, Mol. Interv., 3, 90-105)。現在市場に出回っている薬物の僅か5%が神経系に関する。
侵襲的アプローチは、活性物質の脳への直接脳室内注射、脳内注射もしくは髄腔内注入によって、又はBBB/BSCBの崩壊(これらのバリアの完全性の一次的破壊)によって遂行され得る。
分子を輸送するための薬理学的戦略は、活性物質に脂質又は親油性基を付加することによってより疎水性にした分子の経細胞拡散(細胞透過性親油性拡散、すなわちTLD)、又はリポソームの使用(Zhou et al., 1992, J. Control. Release, 19, 459-486)、及び正に荷電したベクター分子を介したイオン性吸着による又は活性分子のカチオン化による輸送(吸着媒介輸送すなわちAMT)を含む。
ベクター化への生理的アプローチに基づく戦略は、バリアの様々な天然輸送メカニズムの活用に存する。BBBを通過する分子の能動輸送のこれらのメカニズムは、特異的受容体基質とのカップリングを介して、又は特異的受容体基質との分子的擬態(担体媒介輸送すなわちCMT)によって、又は受容体を特異的にターゲティングするリガンドとのカップリングもしくは融合(RMT、受容体媒介輸送、又は受容体媒介経細胞輸送)を介して作用する。

VxSzDy (III)

Vは本発明のペプチド又は疑似ペプチド又は多量体を示し、Sはスペーサーを示し、Dは活性物質又は対象の物質を示し、xは1〜5の整数であり、y及びzは1〜10の整数である)
で示されるコンジュゲート化合物に関する。特定の態様において、x=z=y=1又はx=z>y又はy=z>x又はz>x>yである。
実施例I
ペプチドの合成及びトレーサー分子(酵素活性を有するビオチン、フルオレセイン、ローダミン、シアニン又はStagペプチド)とのカップリング
ペプチドを、アドバンスケムテックApex396(AAPPTec)シンセサイザー又はLiberty(登録商標)(CEM)マイクロ波シンセサイザーで、固相ペプチド合成(SPPS)法によって、ポリスチレン−1%DVB上のリンクアミドAMレジン、ポリスチレン−1%DVB上のワンレジン、ポリスチレン−1%DVB上のBarlosレジン(2−クロロトリチルクロリド)、又はポリスチレン−1%DVB上のSieberアミドレジンでのFmoc/tBu戦略を使用して合成した。ロード(又は置換)は使用されるレジンに応じて、0.25mmol/gから1.6mmol/gである。
− Chromolith Flash C18カラム(4.6mm×25mm)、
− 3ml/分の流速、
− 2.5分間で0〜100%のBの線形勾配(A:0.1%のH2O/HCO2H;B:0.1%のACN/HCO2H)。
ペプチドベクターの設計
ペプチドがhLDLRに結合する能力を決定した。この目的を達成するために、接着及びコンフルエントなCHO−hLDLR−RFP細胞を、6ウェルプレート中で培養した。1つの条件につき3つの細胞のウェルを使用する。
(i)HamF12−1%BSA培地。
(ii)HamF12−1%BSA培地+10μMの対照ペプチドCTRL−STag(STagを含む任意のペプチドの非特異的結合の評価)。
(iii)HamF12−1%BSA培地+10μMのペプチド配列番号17−STag+10μMの対照ペプチドCTRL(対象のペプチドと対照ペプチドCTRLの間の「非特異的」競合の評価)。
(i)100nMの範囲のhLDLRに対する見かけの親和性(Km)及び10nMの範囲の親和性(KD)、これは以前のペプチドより非常に優れており、かつ、ペプチドの結合、エンドサイトーシス、細胞内放出又は経細胞輸送/排出後の放出に適合している(Yu et al., 2011, Sci Transl Med., May 25;3(84):84ra44);
(ii)hLDLRに対するその特異性を支持する物理化学的な特徴(ジスルフィド架橋の環化を介した拘束されたコンフォメーション);8アミノ酸までに低減されたサイズ;この受容体に対するその特異性を促進するために非天然アミノ酸は4つ以下;
(iii)酵素的タンパク質分解に対するより大きな抵抗性:内因性ペプチド及び一般的には天然アミノ酸のみを含む小さな鎖状ペプチドは、血液中で非常に短いインビトロにおける半減期(t1/2)を有し、典型的には分単位の範囲を有するが(Foltz et al., 2010, J Nutr., Jan;140(1):117-8)、本発明のペプチドのt1/2は、1.5時間から9.2時間までの範囲である。
(iv)小さなサイズ及び1kDaに近い分子量により、合成費用及び工業規模での将来的な生産量を減少させることが可能となる。
CHO−LDLR−GFP細胞系のhLDLRに対して親和性を有する合成ペプチドの結合及びエンドサイトーシス
本発明のペプチドは、C末又はN末の位置において、3つのGly残基から一般的に構成されるスペーサーによって隔てられて、様々なトレーサー分子である、ローダミンRed−X、Cy3.5、FITC又はSTagのいずれかとカップリング/コンジュゲートしている(図2)。STag(ウシ膵リボヌクレアーゼAの配列1〜15に由来する15アミノ酸ペプチド)は、一方で、免疫細胞化学又はFACSアプローチのために抗STag抗体によって認識され得、他方で、FRETWorks STagアッセイキット(Novagen 70724-3)を使用したインビトロ活性試験においてリボヌクレアーゼS−タンパク質(C末部分、アミノ酸21〜124)と結合させることによって酵素活性を復元することができる。このようにして活性化されたリボヌクレアーゼは、RNA基質を消化して、遮蔽された蛍光物質を放出し、これはFRET(蛍光共鳴エネルギー移動)によって可視化され、96ウェルプレート中、Beckmann分光蛍光光度計で定量される。これらのFRET実験のために、対照CHO細胞を使用し、hLDLR及びmLDLRとC末の位置において融合させたGFP(これは、FRETに使用された波長で強力なバックグラウンドノイズを発生する)を、赤色蛍光タンパク質(RFP)によって置換した。FRET実験のために生成された安定な細胞系は、従って、CHO−RFP及びCHO−hLDLR−RFPである。
BBBのインビトロモデルにおける内皮細胞上のLDLRに対して親和性を有する合成ペプチドの毒性、エンドサイトーシス及び経細胞輸送
内皮細胞に対するペプチドの可能性ある毒性作用、これらの細胞におけるペプチドの結合/蓄積、及びペプチドの経細胞輸送による通過を、インビトロのBBBモデルにおいて評価する。共培養モデルを組み立てるのに必要とされる細胞は、ラット又はマウス脳毛細血管内皮細胞(BCEC)及びラット又はマウス星状膠細胞である。このタイプのインビトロのBBBモデルを使用して、数多くの分子、特に薬理学的薬剤の、BCECを通過する受動通過又は能動輸送を、及び従って、インビボにおいてCNS組織に到達するその能力を推定することによって評価する。現在までに開発された種々のモデル(ウシ、ブタ、マウス、ヒト)は、脳内皮、特にタイトジャンクション、開窓がないこと、経内皮チャネルがないこと、疎水性分子に対する低い透過性、及び高い電気抵抗を特徴とする、超微細構造的な特性を有する。さらに、これらのモデルは、BBBを通過するその特性についてインビトロ及びインビボで評価された様々な分子について行なわれた測定の結果間に確かな相関を示した。現在までに得られた全てのデータは、これらのインビトロのBBBモデルが、細胞培養実験に伴う利点を保持しつつ、インビボで存在する細胞環境の幾分の複雑さを再現することによって、インビボの状況を厳密に擬態することを示す。
参照ペプチド2の生体分子融合、並びに、参照ペプチド1及び配列番号1の、ヒトIgG1抗体の定常フラグメント(Fcフラグメント)への生化学的カップリング、並びに、CHO hLDLR細胞系及びインビトロラットBBBモデルにおける結合、エンドサイトーシス及びRMTの評価
hLDLRに対する親和性を有するいくつかのペプチドを、細胞系又は一次内皮細胞において発現されるhLDLRへの、共有結合的に連結されたFcフラグメントの結合及び/又はエンドサイトーシス及び/又は経細胞輸送を促進するその能力について試験した。この目的のために、本発明らは、Fcフラグメントと融合したペプチド配列番号43をコードする配列、又は、同じFcフラグメントに化学的にカップリングさせたペプチド配列番号17及び配列番号1をクローニングした。

対象の治療分子又はイメージング剤又は診断剤又は任意の他の分子、例えば分子プローブを、細胞膜を、より特定するとBBBを通過して輸送及び通過させた後、例えばベクターと活性物質との間の化学結合の加水分解又は酵素的切断によるプロドラッグ戦略を通して、ベクターから切断/遊離/塩析させることができる。
− 直列型合成(すなわち、2つの実体間の中間体を全く伴わない直接的なカップリング)、
− リンカーを介した合成(Temsamani et al., 2004, Drug Discov. Today, 23, 1012-1019)。
ペプチド、又は治療上関心のある薬物、イメージング(又は診断)剤、もしくはトレーサーとして使用される任意の他の物質にコンジュゲートさせたペプチドの組織分布
LDLRをターゲティングするペプチド及びペプチドコンジュゲートの分布を、マウスにおいて評価して、高レベルのターゲット受容体を発現している組織及び器官に優先的に蓄積するその能力を示す。
ベクター単独、及び対象の治療分子又はイメージング(又は診断)剤又は任意の他の分子、例えば分子プローブとコンジュゲートさせたベクターのインサイツにおける脳灌流、並びに、BBBを通過するその輸送動態及びマウス脳におけるその蓄積の研究
インサイツにおける脳灌流技術(成体雄OF1マウス)は、BBBを通過する脳内の通路を示す。
多量体の合成及び特性
hLDLR−GFP/RFPに対して親和性を有する本発明のペプチドを、単量体として使用し、二量体を合成した。単量体を、アミン、酸、チオール、アジド、アルキン、カルボニル、ヒドラジンの中の少なくとも3つの反応性官能基を含む分子プラットフォーム上で、溶液中又は固相支持体上のいずれかでカップリングさせた。単量体は、スペーサー(一般的に1つのグリシンから構成されているが、それは、PEG分子、アミノヘキサン酸又は一連のグリシンであってもよい)によってプラットフォームから隔離されている。プラットフォームは、市販の有機化合物であっても、又は臨機応変にペプチド化学反応を使用して設計されてもよい。
Claims (16)
- 配列番号1、配列番号2及び配列番号5から選択されたペプチド。

を有する多量体物質。 - h及びiが、互いに独立して、1又は2であり、好ましくはh=i=1である、請求項2の多量体物質。
- Mがジリジンである、請求項2又は3の多量体物質。
- ホモ二量体である、請求項2〜4のいずれか一項の多量体物質。
- 前記ペプチドが、メチレン(−CH2−)又はリン酸(−PO2−)基、第2級アミン(−NH−)又は酸素(−O−)、アルファ−アザペプチド、アルファ−アルキルペプチド、N−アルキルペプチド、ホスホンアミデート、デプシペプチド、ヒドロキシメチレン、ヒドロキシエチレン、ジヒドロキシエチレン、ヒドロキシエチルアミン、レトロインベルソ型ペプチド、メチレンオキシ、セトメチレン、エステル、ホスフィネート、ホスフィン酸、ホスホンアミド又はカルバ基の挿入から選択された少なくとも1つのペプチド模倣結合を含む、請求項1〜5のいずれか一項のペプチド又は多量体物質。
- 前記ペプチドのN末端(N末)官能基がアシル化によって保護され、及び/又は前記ペプチドのC末端(C末)がアミド化もしくはエステル化によって保護される、請求項1〜6のいずれか一項のペプチド又は多量体物質。
- 以下の式(III):

- 活性物質又は対象の物質が、治療上関心のある分子、診断剤又は医療用イメージング剤、分子プローブ、化学的低分子、ペプチド又はポリペプチド、タンパク質、抗原、抗体又は抗体の一部、核酸又はオリゴヌクレオチド、siRNA、miRNA、リボザイム、マーカー又はトレーサーであることを特徴とする、請求項9のコンジュゲート化合物。
- V、D及びSの間のカップリングが、生理的培地中で又は細胞内で切断可能な又は切断不能な1つ以上の共有結合、イオン結合、水素結合、疎水性結合又はファンデルワールス結合によって行なわれることを特徴とする、請求項9のコンジュゲート化合物。
- Dが、Vと、場合によりSを介して、VのN末端及び/もしくはC末端の一方もしくは両方において、並びに/又はVを構成する天然もしくは非天然アミノ酸側鎖によって担持されている1つ以上の反応性基においてカップリングしている、請求項9のコンジュゲート化合物。

- 請求項8又は13の少なくとも1つのコンジュゲート化合物及び1つ又はいくつかの薬学的に許容される賦形剤を含む薬学的組成物。
- 請求項8又は13のコンジュゲート化合物から構成される診断剤又は医療用イメージング剤を含む診断用組成物。
