細菌によって産生され数々の手法により精製された市販の精製α2.8−ポリシアル酸(250mg、およそ70kDaの分子量を有するポリシアル酸、UK、Lipoxen、ここではPSA−180と命名)を、フラグメント化に使用した。分取工程において、サンプルを80℃で30分間加熱して自発的な加水分解を誘導した。次いでPSAを53mlのHRPセファロース陰イオン交換カラム(GE−Healthcare)で処理し、205/280nmでのUV光度検出器(Pharmacia Biotech)に連動させ、溶媒として流速4ml/分の2MのNH4HCO3緩衝液を利用する高速液体クロマトグラフィー(HPLC)システムによって分離した(表1)。それぞれ体積8mlを有する90本の試験管に流出液を収集した。連続3本の試験管をそれぞれプールして、30画分のPSA−180を得た。
大腸菌K1から調製されたおよそ70kDaの分子量を有する精製α2.8−ポリシアル酸(PSA−180)を使用して、Bice I.ら、Eng.Life Sci.2013、13、2号、140〜148で説明されている通りに、ポリシアル酸のフラグメント化および分離を繰り返した。分取用HPLCを使用した調製を、実施例1で説明されているようにして行った。上述したように、HPLC画分2から6をプールし、これをPSA−20と表示した。
異なる鎖長を有するシアル酸型の影響を研究するために、WO2010/125110で説明されているような人口多能性幹細胞由来ヒトミクログリア株を使用した。ヒトミクログリア細胞を、5μg/mlのポリ−L−リジン(lysin)(PLL、Sigma)でコーティングした培養皿で、1%N2(Invitrogen)、0.48mMのL−グルタミン(Gibco)および任意選択で100μg/mlのペニシリン/ストレプトマイシン(Gibco)が補充されたDMEM/F12培養培地を含有するN2−培地(Gibco)中で培養した。細胞を高密度で培養し、必要な場合に1:2に分けた。
ポリシアル酸がヒトミクログリアの炎症促進性の表現型に干渉するかどうかを分析するために、培養されたヒトミクログリアをリポ多糖(LPS)で活性化し、細菌毒素LPSにより誘導された炎症促進性サイトカインである腫瘍壊死因子−アルファの遺伝子転写に及ぼす、オリゴシアル酸画分やポリシアル酸画分を含む別個のシアル酸型の影響を決定した。様々な鎖長を有するシアル酸型の影響を研究するために、WO2010/125110で説明されているような人口多能性幹細胞由来ヒトミクログリア株を使用した。ヒトミクログリア細胞を、5μg/mlのポリ−L−リジン(PLL、Sigma)でコーティングした培養皿で、1%N2(Invitrogen)、0.48mMのL−グルタミン(Gibco)および任意選択で100μg/mlのペニシリン/ストレプトマイシン(Gibco)が補充されたDMEM/F12培養培地を含有するN2−培地(Gibco)中で培養した。細胞を高密度で培養し、必要な場合に1:2に分けた。ヒト人口多能性幹細胞由来ミクログリア株を、リポ多糖(LPS、1μg/ml、Invitrogen)および様々な鎖長を有するシアル酸型で24時間処置した。腫瘍壊死因子−α(TNF−アルファ)に関する遺伝子転写物を、定量RT−PCRによって決定して、GAPDHに正規化した。詳細には、RNeasyミニキット(Qiagen)を使用してRNAの単離を行い、スーパースクリプト第一鎖合成システム(Invitrogen)を用いて転写を行った。特異的なオリゴヌクレオチド(GAPDH:CTGCACCACCAACTGCTTAG(配列番号1)およびTTCAGCTCAGGGATGACCTT(配列番号2);TNFα:GACAAGCCTGTAGCCCATGT(配列番号3)およびAGGACCTGGGAGTAGATGAGG(配列番号4))を用いた定量RT−PCRを、マスターサイクラーepグラジェントS(Eppendorf)と共にSYBRグリーンPCRマスターミックスを使用して行った。結果をGAPDHに正規化した。デルタ−CT方法を使用した定量化を実行した。
アルツハイマー病は、凝集したアミロイドβ1−42ペプチドの細胞外蓄積から始まり、ミクログリアの活性化、活性酸素種の産生ならびにシナプスおよび軸索突起の喪失によって進行する神経変性疾患である。げっ歯類動物モデルは、好適なアルツハイマー病モデル系として部分的にしかヒト疾患をモデル化することができないため、共培養されたヒトニューロンとヒトミクログリアを使用し、そこに、ミクログリア細胞を刺激することが可能な線維性のアルツハイマー病関連アミロイドβ1−42ペプチドを添加した。実施例4で説明されているようにして人口多能性幹細胞由来ヒトミクログリア細胞株を使用した。
アルツハイマー病の脳組織の炎症性シグナル伝達を模擬するために、ヒトミクログリア細胞株を、ヒトアルツハイマー病関連の線維性アミロイド−ベータペプチド(Aβ)で処置した。Aβの線維性成分を得るために、合成アミロイド−ベータペプチド(アミロイド−ベータ1−42、Bachem/Brucker、10μM)を37℃で少なくとも3日間プレインキュベートした。ヒトミクログリア細胞を、様々な濃度の0.15μM、0.5μM、または1.5μMの低分子量ポリシアル酸(PSA−20)で60分間処置し、続いて線維性Aβと共に15分間インキュベートした。次いで、スーパーオキシドのアニオンラジカル産生を測定するために、30μMジヒドロエチジウム(DHE)を添加し、37℃で15分間インキュベートした。4%パラホルムアルデヒドと、0.25%グルタルアルデヒドで細胞を固定し、共焦点顕微鏡法によって分析した。DHE染色強度を定量化するために、実験ごとに6枚の図画を得て、ImageJソフトウェア(NIH)によって分析した。バックグラウンドを差し引いて、染色強度の平均値を比較した。細胞を固定し、DHEの強度を共焦点顕微鏡法によって定量化した。
次に、ヒトミクログリア株を、人口多能性幹細胞由来ニューロンと共に24時間共培養し、ミクログリアの神経毒性に及ぼす低分子量ポリシアル酸(PSA−20;1.5μM)の影響を評価した。ヒトニューロンをヒト人口多能性幹細胞(iPS細胞)から生成した。インビトロにおけるニューロンへの分化を短く改変したプロトコールを使用して実行し、これを使用して原始神経前駆体を誘導した。詳細に言えば、iPS細胞をフィーダー細胞上で培養して、小さいコロニーを形成した。次に培地を、LIFならびに3種の小分子CHIR99021(GSK−3βの阻害剤)およびSB431542(TGF−βおよびアクチビン受容体の阻害剤)、および化合物E(γ−セクレターゼの阻害剤)の存在下で、10日間、神経誘導培地に交換した。神経前駆体を拡大するために、アキュターゼによって細胞を単一の細胞に分離させて、白血病抑制因子(LIF)、CHIR99021およびSB431542の存在下で誘導培地を含むポリ−L−オルニチン/フィブロネクチンでコーティングした細胞培養皿でプレーティングした。ニューロンへの分化を誘導するために、神経前駆体細胞をアキュターゼによって分離させ、神経誘導培地中のポリ−L−オルニチン/ラミニンでコーティングした細胞培養皿に添加して、細胞を付着させて小さいコロニーを形成させた。次いで、培地を、脳由来神経栄養因子(BDNF)およびグリア細胞株由来神経栄養因子(GDNF)を含むニューロン分化培地に2週間交換した。2日に1回培地を交換した。共培養実験のために、ヒトミクログリア細胞を掻き取り、これを、1:4のミクログリア:ニューロンの比率で、1.5μMのPSA−20含有および非含有のニューロン分化培地中でニューロンに24時間添加した。細胞を洗浄し、4%パラホルムアルデヒド(PFA)中で15分間固定し、ブロッキングし、ウシ血清アルブミン10×(BSA)および5%正常ヤギ血清(nGS)および0.1%TritonX−100を含有する溶液で60分間透過処理した。次に、一次抗体(ニューロンに対してはβ−チューブリン−III、ミクログリア細胞に対してはIba−I)で4℃で一晩、続いて二次抗体で室温で90分間、細胞を免疫染色した。共焦点レーザー顕微鏡検査法(Fluoview 1000、オリンパス)によって条件ごとに10枚の無作為な写真を撮り、Neuron Jソフトウェア(NIH)によってニューロン分岐(neuronal branches)の長さを測定した。
ヒト人口多能性幹細胞由来ミクログリア(iPSdM)を、iPS細胞から得た。この研究には、iLB−C−35m−rlクローン(Bonn)から生成したiPSdM−1株を使用した。iPSdM−1(ここではミクログリアまたはミクログリア細胞と命名)を、1%N2(Invitrogen)、0.48mMのL−グルタミン(Gibco)および100μg/mlのペニシリン/ストレプトマイシン(Gibco)が補充されたDMEM/F12培養培地からなるN2−培地(Gibco)中で培養した。細胞を高密度で培養し、1:5に分けた。層剥離後、細胞を回収し、新しい培養皿に再度付着させた。
ヒト単球細胞株THP−1(ATCC TIB−202)を、75ml細胞培養フラスコ(Sarstedt)で、10%FCS、1%ピルビン酸ナトリウムおよび1%ペニシリン/ストレプトマイシン(100×)(全てGibco、Invitrogen)を含むRPMI培地中で培養した。組織マクロファージへの分化のために、この細胞株を、0.5μMの13−酢酸12−ミリスチン酸ホルボール(Sigma)を含有する正常細胞培養培地中で3時間培養し、その後13−酢酸12−ミリスチン酸ホルボール非含有培地で少なくとも24時間培養した。
敗血症は、酸化ストレスおよび補体活性化の増加に関連する食細胞機能不全を特徴とする過剰な炎症性の状況を伴うことが多い。敗血症の際の過剰な炎症性の状況は、細菌性リポ多糖(LPS)などの細菌から放出される生成物、加えて損傷を受けた細胞からの生成物によって引き起こされる。マクロファージなどの免疫細胞およびミクログリア細胞ならびに腫瘍壊死因子−a(TNF−a)などのそれらの炎症性メディエータは、敗血症性脳症に関与する。
これらの実験では、ヒト網膜、ヒトミクログリアおよびヒト化Siglec−11トランスジェニックマウス(Wang Yの2009年の学位論文、University Bonn、ユニバーサルリソースネーム:urn:nbn:de:hbz:5N−18095)および同腹子のコントロールマウスを使用した。
まず、ヒト網膜におけるSiglec−11の遺伝子転写を分析した。ヒト網膜のトータルRNAを生検から単離した。スーパースクリプトIII逆転写酵素(Invitrogen)およびランダム六量体プライマー(Roche Molecular Biochemicals)を使用してRNAの逆転写を行った。ポリメラーゼ連鎖反応(PCR)によってcDNAを35サイクルで増幅した。反応のために、1μgの各試験cDNAを使用した。使用されたPCRプログラムは、94℃で2分で最初の変性、94℃で90秒で変性、62.5℃で1分でアニーリング、68℃で1分で伸長、68℃で10分で最後の伸長であった。プライマー配列は、結果として352bpのPCR産物をもたらすフォワードACAGGACAGTCCTGGAAAACCT(配列番号9)およびリバースAGGCAGGAACAGAAAGCGAGCAG(配列番号10)であった。プライマーは、10pMの濃度で使用された。試験cDNAサンプルを、C57BL/6コントロールマウスおよびSiglec−11トランスジェニックマウスの脳、加えてヒト網膜から得た。網膜の非転写RNAを、PCR陰性コントロールとして利用した。図7Aからわかるように、ヒト網膜でSiglec−11に関する遺伝子転写物が検出された。
第二に、神経組織片で刺激されたミクログリアに及ぼすPSA−20の影響を分析した。ドルーゼンを含有する組織片は、老人性黄斑変性症の特徴の一つである。網膜のミクログリアに及ぼす変性物質の影響を研究するために、本発明者らは、人口多能性幹細胞由来ヒトニューロンを低張でかつ機械的に溶解させることによって神経組織片を調製した。次いで、本発明者らは、人口多能性幹細胞由来ヒトミクログリア株を処置して、PSA−20の影響を分析した。スーパーオキシド放出を、蛍光色素であるDHEによって決定した。神経組織片およびPSA−20またはコントロール媒体(PBS)を添加した後、スーパーオキシドのアニオンラジカル産生を測定するために、30μMジヒドロエチジウム(DHE)を添加して37℃で30分間インキュベートした。4%パラホルムアルデヒドと、0.25%グルタルアルデヒドで細胞を固定し、共焦点顕微鏡法によって分析した。DHE染色強度を定量化するために、実験ごとに6枚の図画を得て、ImageJソフトウェア(NIH)によって分析した。バックグラウンドを差し引いて、染色強度の平均値を比較した。細胞を固定し、DHEの強度を共焦点顕微鏡法によって定量化した。図7Bで示されるように、神経組織片の添加は、ミクログリアによるスーパーオキシド産生を増加させた。図7Bからわかるように、PSA−20は、神経組織片によって誘導されるミクログリアによるスーパーオキシド産生の増加を防止した。
レーザー技術により黄斑変性症の動物モデル症を誘発した。レーザーを使用したマウス網膜の実験的な傷害は、加齢性黄斑変性症に関して広く容認されている動物モデルである。ブルック膜のレーザー凝固と直接的な傷害により、急速にミクログリアおよび補体の活性化が起こり、血管漏出が誘発される。蛍光血管撮影法は、網膜中の血管漏出を検出するための確立された方法である。
オリゴ/ポリシアル酸の生理学的な発現に関して、長鎖ポリシアル酸(PSA−NCAM)、短鎖PSA(CD56)およびシアル酸三量体(A2B5)の免疫染色によってヒト網膜を分析した。網膜をスライスし、−80℃で凍結した。染色のために、スライドを室温で10〜15分間乾燥させ、PBSで2回洗浄した。切片を、10%ウシ血清アルブミン(BSA、Sigma)、5%ヤギ血清(Invitrogen)および0.1%Triton−X−100で20〜30分間ブロッキングした。切片を、以下の一次抗体:ウサギ抗Iba1(1:1000、和光)、マウス抗PSA−NCAM(1:500、ポリシアル酸、Millipore)、ラット抗CD56(1:200、オリゴシアル酸、BD Pharmingen)またはマウス抗A2B5(1:200、トリシアル酸、Invitrogen)の1種と共に4℃で一晩インキュベートした。切片をPBSで3回洗浄し、次いで対応するCy3−コンジュゲート二次抗体(Jackson)と共に室温で4時間インキュベートした。PBSで3回洗浄工程を行った後、切片を核色素TO−PRO(登録商標)ヨウ化物(1:2000、life technologies)と共に室温で15分間インキュベートし、ムービオーラにマウントした。細胞核をTO−PRO(登録商標)(核染色)で対比染色した。対応するアイソタイプ抗体を、コントロールとして使用した。少なくとも3回の独立した実験からの代表的な画像を分析した。網膜の外側では3種全てのシアル酸種が弱く発現されたが、内網状層(IPL)、ヒト網膜の神経節細胞層(GCL)および神経線維層中では強い発現が見出された。内網状層および神経節細胞層中ではオリゴシアル酸/ポリシアル酸が検出された。これから、ポリシアル酸は網膜の正常な成分であるため、PSA−20がこの機能を模擬することが可能になることが示される。
ヒト化Siglec−11トランスジェニックマウス(ヒト化Siglec−11マウス)および同腹子のコントロールマウス(正常なコントロールマウス)において、実験的自己免疫性脳脊髄炎を誘発した。したがって、6〜8週齢の成体雌ヒト化Siglec−11トランスジェニックマウス(Wangらの2009年の学位論文、University Bonn、ユニバーサルリソースネーム:urn:nbn:de:hbz:5N−18095)および同腹子のコントロールマウスを、不完全フロイントアジュバント中の100μgのミエリンオリゴデンドロサイト糖タンパク質MOG(アミノ酸35〜55;Seqlab)(両方ともDIFCO;BD GmbH、Heidelberg、Germany)で免疫化した。免疫化の0日目および2日目に、百日咳毒素(200ng;List Biological Laboratories、Epsom、UK)を注射した。臨床兆候を以下の通りにスコア付けした:0、臨床兆候なし;1、完全な尾の引きずり;2、完全な尾の引きずりおよび後脚の衰弱;3、少なくとも1本の後脚の対不全麻痺;4、完全な後脚の対不全麻痺および前脚の衰弱;ならびに5、前脚と後脚の麻痺または瀕死。20日目までに疾患が発病したマウス(≧1の臨床スコア)だけを実験に使用した。
パーキンソン病は、主としてドーパミン作動性ニューロンに影響を及ぼし、活性化ミクログリアおよび活性酸素種産生の増加が関与する神経変性疾患である。脳中の酸化ストレスの増加が、パーキンソン病の主要な特徴である。また、一次食作用(ファゴプトーシス(phagoptosis)と命名)による神経変性も説明されている。LPSなどの毒素の適用を、パーキンソン病に関する動物モデルとして使用した。げっ歯類動物モデルは、げっ歯類とヒトとの種間の差のためにヒト慢性神経変性疾患モデルに限定的な適性しか有さないため、近年、パーキンソン病のLSP毒素によって誘導された動物モデルの代用品として、ヒトニューロンおよびヒトミクログリアのヒト毒素および組織片によって誘導された培養モデル系が使用されている。
ヒト人口多能性幹細胞由来ミクログリア(iPSdM)を、人口多能性幹(iPS)細胞から誘導した。この研究には、iLB−C−35m−rlクローン(Bonn)から生成したミクログリアiPSdM株1(iPSdM−1)を使用した。iPSdM−1(ここではミクログリアまたはミクログリア細胞と命名)を、1%N2(Invitrogen)、0.48mMのL−グルタミン(Gibco)および100μg/mlのペニシリン/ストレプトマイシン(Gibco)が補充されたDMEM/F12培養培地からなるN2−培地(Gibco)中で培養した。ヒト胚性幹細胞から原始神経前駆体を得るために使用される改変されたプロトコールに従って、原始神経幹細胞(pNSC)の生成およびそれらのニューロンへの分化のために、ヒト由来多能性幹(iPS)細胞(Foreskin−1、WiCell)を使用した。簡単に言えば、iPS細胞をフィーダー細胞上で培養して、小さいコロニーを形成した。次に、培地を、白血病抑制因子(LIF;Millipore、10ng/ml)および3種の小分子CHIR99021(GSK−3βの阻害剤、Axon Medchem、4μM)およびSB431542(TGF−βおよびアクチビン受容体の阻害剤;Axon Medchem、3μM)、および化合物E(γ−セクレターゼの阻害剤;Axon Medchem、0.1μM)の存在下で10日間、神経幹細胞培地(DMEM/F12:Neurobasal;GIBCO)に交換した。ニューロンへの分化を誘導するために、pNSCをアキュターゼ(PAA)によって分離させ、ポリ−L−オルニチン(Sigma、0.15mg/ml)と、ラミニン(Sigma、1μg/ml)でコーティングした細胞培養皿で、神経幹細胞培地(DMEM/F12:Neurobasalと、LIF、CHIR99021およびSB431542;GIBCO)中で、細胞が付着して小さいコロニーが形成されるまで培養した。次いで、培地を、脳由来神経栄養因子(BDNF;Prospect、10ng/ml)およびグリア細胞株由来神経栄養因子(GDNF;10ng/ml)の存在下で2週間、ニューロン分化培地(DMEM/F12と、N2およびB27サプリメント、GIBCO)に交換した。2日に1回、神経栄養因子を含有する培地を交換した。
iPS細胞由来ヒトニューロンを、1.5μMのPSA−20の存在または非存在下で、ヒトの正常なリポ多糖(LPS)で事前に活性化されたミクログリア(活性化ミクログリア)と48時間共培養した。活性化ミクログリアにおいて、パーキンソン病で説明したような酸化ストレスが1μg/mlのLPS毒素によって誘導された。PSA−20の非存在または存在下で、ヒトニューロンにヒトミクログリア細胞を1:5(ミクログリア:ニューロン)の比率で48時間添加した。次いで、細胞を4%パラホルムアルデヒド(PFA、Sigma)で15分間固定し、ブロッキングし、10%ウシ血清アルブミン(BSA、Sigma)および5%正常ヤギ血清(nGS、Dianova、Hamburg)および0.1%TritonX−100(細胞核染色用、0.5%TritonX−100、Sigma)を含有する溶液で60分間透過処理した。次に、モノクローナル抗−β−チューブリン−III(Sigma)およびポリクローナルウサギ抗iba1(Dako)抗体を用いて4℃で一晩、続いてウサギIgGに対する二次Alexa488−コンジュゲート抗体(Molecular Probes)およびマウスIgGに対するCy3−コンジュゲートヤギ抗体(Dianova)を用いて室温で2時間、細胞を免疫染色した。
ヒトミクログリアを蛍光標識された神経組織片でチャレンジし、組織片の細胞への取り込みを、共焦点顕微鏡法および3D再構成によって決定した。
ミクログリア細胞によるスーパーオキシドの相対的な産生を測定するために、4つのチャンバーを有する培養皿でヒトミクログリア細胞をプレーティングした。24時間後、0.15μM、0.5μMまたは1.5μM濃度でPSA−20と1時間プレインキュベートして、またはしないで、細胞を5mg/mlの神経組織片で15分間処置した。次いで、細胞をクレブス−HEPES−緩衝液で2回洗浄した。スーパーオキシド放出を検出するために、その後細胞を、クレブス−HEPES−緩衝液で希釈した30μMのスーパーオキシド感受性蛍光色素であるジヒドロエチジウム(DHE)溶液と共に15分間インキュベートした。最後に、細胞をクレブス−HEPES−緩衝液で2回洗浄し、0.25%グルタルアルデヒドおよび4%PFAを用いて15分間固定した。共焦点レーザー顕微鏡検査法(Fluoview 1000、オリンパス)によって、合計で実験グループ当たり6つの画像を無作為に収集した。収集された画像の全ての細胞を、ImageJソフトウェア(NIH)によって分析した。
PSA−20の薬物動態および薬物毒性の決定
神経変性疾患の療法のためのほとんどの薬物が、神経毒性(neurotoxcity)または限られた脳実質への侵入のために失敗に終わっている。PSA−20のあらゆる神経毒作用を評価するために、ヒト人口多能性幹(iPS)細胞由来ニューロンを使用して、PSA−20のあらゆる毒作用を検出した。さらに、LPSによって誘導されたTNF−αの遺伝子転写物の抑制的調節に関するPSA−20の有効濃度を検出するために、ヒトiPS細胞由来ミクログリアを使用した。脳におけるPSA−20の生物学的利用率を決定するために、ビオチン−コンジュゲートPSA−20を腹腔内に適用し、様々な期間後の血清および脳組織中の濃度を分析した。

まず、PSA−20をビオチンとコンジュゲートさせて、ビオチン化PSA−20を検出するための高感度ELISAを確立した。PSA−20鎖の末端におけるシアル酸の第6の炭素原子で、PSA−20とビオチン分子とをカップリングした。そのため、過ヨウ素酸ナトリウムによってPSA−20の末端をアルデヒドに酸化した。その後、ヒドラジドがカップリングされたビオチンを室温でアルデヒド基にコンジュゲートして、ヒドラゾン結合を形成した。脱塩カラムを用いて(HiTrap脱塩カラム、GE Healthcare)精製を実行した。
Claims (6)
- 中枢神経系の変性疾患および脱髄疾患、ならびに変性性または炎症性の網膜疾患からなる群から選択される疾患の治療的および/または予防的処置で使用するための、以下に示される一般式(1):

に係る分岐状もしくは非分岐状のポリシアル酸および/もしくは該ポリシアル酸の医薬的に許容される塩または該ポリシアル酸(1)を含む多糖組成物であって、該ポリシアル酸のフラグメントは、4.9kDaから7.4kDaの間の平均分子量を有し、該フラグメントの90重量%以上100重量%以下は、4.3kDaから8kDaの間の分子量を有し、該フラグメントの0重量%以上5重量%以下は、3kDaから4.3kDaの間の分子量を有し、該フラグメントの0重量%以上5重量%以下は、8kDaから9.5kDaの間の分子量を有し、ここで該フラグメントの重量%はポリシアル酸フラグメントの総重量に基づく、ポリシアル酸および/もしくはポリシアル酸塩または多糖組成物。 - 神経変性疾患が、筋萎縮性側索硬化症、アルツハイマー病、軽度認知機能障害、レヴィ小体型認知症、パーキンソン病および睡眠時異常行動からなる群から選択される疾患であって、該神経変性疾患に使用するための、請求項1に記載のポリシアル酸および/もしくはポリシアル酸塩または多糖組成物。
- 変性性または炎症性の網膜疾患または眼疾患が、加齢性黄斑変性症、遺伝性網膜疾患を含む網膜変性、ブドウ膜炎、および糖尿病性網膜症からなる群から選択される疾患であって、該網膜疾患または眼疾患に使用するための、請求項1に記載のポリシアル酸および/もしくはポリシアル酸塩または多糖組成物。
- 中枢神経系の脱髄疾患が、多発性硬化症またはデビック病であって、該脱髄疾患に使用するための、請求項1に記載のポリシアル酸および/もしくはポリシアル酸塩または多糖組成物。
- 中枢神経系の変性疾患および脱髄疾患、ならびに変性性または炎症性の網膜疾患からなる群から選択される疾患の治療的および/または予防的処置で使用するための、活性成分として、以下に示される一般式(1):
