Description
実施例1−プラスミド構築
野生型(WT)EKLF発現プラスミドpBabe puro HAII WT EKLFは、Belinda Singleton博士から親切に提供された。簡単には、全長EKLFを、以下のプライマーを用いて増幅し:5’−GATTACGCTGAATTCTCATGGCCCACAGCCGAGACC−3’(配列番号1)および5’−GATACTCGAGAATTCTCAAAGGTGGCGCTTCATG−3’(配列番号2)、pCR(登録商標)2.1−TOPOベクターにクローニングし、続いてpBabe puro HAII(プラスミド14738、Addgene Inc.、Cambridge、MA、US Dr.Adrienne Coxの研究室の好意)のEcoRI部位にサブクローニングした。
MiniMacsTM 磁気ビーズ分離システム(Miltenyi Biotech Ltd、Surrey、UK)を用いて臍帯血から単離したCD34+細胞を、以前に記載された3段階赤血球培養法[37];3%(v/v)のAB血清(Sigma)、2%(v/v)のウシ胎仔血清(Hyclone、Fisher Scientific UK Ltd)、10μg/mlのインスリン(Sigma)、3U/mlのヘパリン(Sigma)、200μg/mlのトランスフェリン(R&D Systems)を含むIMDM(Source BioScinece)を用いて、エキソビボで5〜8日間培養した。第1段階において(0〜8日目)、これに10ng/mlのSCF(幹細胞因子)、1ng/mlのIL−3、および3U/mlのEPO(エリスロポエチン)を;第2段階において(8〜11日目)、10ng/mlのSCF、3U/mlのEPOおよびさらに800μg/mlの鉄飽和トランスフェリンを、および最後の段階において(11〜20日目)、3U/mlのEPO、およびさらなる800μg/mlの鉄飽和トランスフェリンを補充した。
最低で5×105細胞を、1×ハンクス緩衝化食塩水溶液(HBSS、Sigma−Aldrich)で2回洗浄し、そしてRNA Later中で凍結した。全RNAを抽出し、そして収量を定量した。SuperScript II逆転写酵素(Invitrogen、Paisley)を用いて、400ngのRNAをcDNAへ逆転写した。BCL11AおよびEKLF発現を、標準的なポリメラーゼ連鎖反応(PCR)によって分析し、一方β−グロビンおよびγ−グロビンの遺伝子発現を、定量的(q)PCRによって分析した。全ての方法は、以前に記載された[38]。PCRおよびqPCRで使用する配列は、以下の通りである:
細胞ペレットを1×HBSSで2回洗浄し、次いで可溶化緩衝液(20mMのTris HCl pH7.5、150mMのNaCl、10%のグリセロール、1%のTriton、0.1%のSDS、1×完全プロテアーゼ阻害剤、および2mMのPMSF)中で1時間細胞全体を溶解し、続いて12.5UのBenzonase(登録番号)ヌクレアーゼ(Novagen、Damstadt、Germany)で1時間処置することによって、トランスフェクトしたK562細胞および臍帯血細胞を、ウェスタンブロット分析のために調製した。BioRad Protein Assay Dye試薬によって定量したタンパク質(3〜15μg)を、8%、12%、または18%のドデシル硫酸ナトリウム(SDS)−ポリアクリルアミドゲル電気泳動(PAGE)によって分離し、そしてPVDF膜に移した。以下の抗体と最低1時間インキュベートすることによって、EKLF、BCL11Aおよびβ−グロビンを検出した;HA.II(16B12 Covance、Crawley、UK)またはEKLF(H−210、Santa Cruz Biotechnology、Santa Cruz、CA)、Ctip1(14B5、Abcam、Cambridge、UK)、およびHemoglobinβ(37−8 Santa Cruz Biotechnology、Santa Cruz、CA)。タンパク質のバンドを、BIORAD Quantity Oneソフトウェアバージョン4.5.1を用いて定量した。
EKLFおよびBCL11A−XLコード領域をPCRによって増幅し、そしてIn−Fusionクローニングシステム(Clontech)を用いて、pXLG3−eGFPまたはpXLG3−mcherryレンチウイルスベクターに挿入した。HEK293T細胞を、ポリエチレンイミン(PEI)を用いて、構築物pMDG2(ウイルスコート)、pCMVR8.91(パッケージングタンパク質)およびpXLG3−eGFP−BCL11A−XLおよびpXLG3−mcherry−EKLFでトランスフェクトした。PEI/DNA溶液を、細胞と4時間インキュベートし、その後培地を交換した。48時間後、ウイルスを含む培地をろ過し、そしてアリコートに分けた。赤血球細胞を、8μg/mlのポリブレンを添加した1mlのウイルスとインキュベートした。細胞を48時間目に回収し、そして4%のパラホルムアルデヒドでポリ−L−リジンでコーティングしたカバーガラスに固定し、そして共焦点顕微鏡を用いて画像化した。
実施例6−K562細胞におけるBCL11Aの発現
BCL11A−XL、BCL11A−LおよびBCL11A−Sの転写物の発現を、K562細胞、HbEおよびHbFを発現するがHbAは発現しない赤血球生成(erythropoetic)細胞系、およびポジティブコントロールとして培養9日目の末梢血CD34+細胞から培養した赤芽球において、各BCL11Aバリアントに特異的なプライマーを用いたPCRによって決定した。3つのBCL11Aバリアント全ての転写物を、赤芽球において検出したが、K562細胞においては転写物を検出しなかった(図1A)。それに加えて、全長BCL11Aタンパク質を、赤血球において容易に検出したが、K562細胞においてはBCL11Aタンパク質の全長または短いバリアントのどちらも検出されなかった(図1B)。K562細胞においてKLF−1の転写物が検出されたが、9日目の赤芽球と比較して低いレベルであり、しかしKLF−1タンパク質は検出されなかった(図1C)。従ってK562細胞は、EKLFに関してヌルであると考えられる。従って、K562細胞におけるβ−グロビンの発現の欠如は、BCL11AおよびEKLFの欠如と関連している。
K562細胞を、pCDNA3−Flag−BCL11A−XL(BCL11A−XL;5μg)、pBp HA−EKLF(HA−EKLF;5μg)で一時的にトランスフェクトした、または両方のプラスミド(それぞれ5μg)でコトランスフェクトした。トランスフェクションの20時間後、細胞を回収し、そしてBCL11A、EKLF、およびβ−グロビンの発現に関して分析した。BCL11A−XLまたはHA−EKLFでトランスフェクトした細胞は、それぞれのタンパク質を発現し、コトランスフェクトした細胞は、両方のタンパク質を発現した(図2A)。次いで単独またはコトランスフェクトした細胞におけるβ−グロビン転写物のレベルを、qPCRによって分析した。BCL11Aでトランスフェクトした細胞において、わずかなβ−グロビンの転写物が検出され、一方HA−EKLFトランスフェクタントにおいて少しの増加(トランスフェクトしていないK562細胞の3倍)が検出された(図2B)。しかし、BCL11A−XLおよびHA−EKLFでコトランスフェクションすると、β−グロビンの転写物レベルは、トランスフェクトしていない細胞と比較して168倍増加し(図2B)、β−グロビンタンパク質のレベルが著しく増加した(チューブリンに対して標準化し、そしてトランスフェクトしていないK562細胞と比較した場合7.9倍;図2C)。
β−グロビンの発現レベルが、トランスフェクション後の時間と共に増加するかどうかを決定するために、BCL11A−XLおよびHA−EKLF(それぞれ5μg)でコトランスフェクトしたK562細胞を、トランスフェクションの24および48時間後に、BCL11A、EKLFおよびβ−グロビンに関して分析した。トランスフェクション後、BCL11AおよびEKLFの転写物およびタンパク質を容易に検出したが、そのレベルはトランスフェクション後24および48時間の間に減少した(図3AおよびB)。β−グロビンの発現は、再びトランスフェクトしていない細胞と比較してコトランスフェクトした細胞において著しく高かったが、転写レベルはトランスフェクション後24から48時間まで減少し、BCL11AおよびEKLFのレベルの減少とパラレルであった(図3AおよびC)。それに加えて、γ対β−グロビン発現の比は、トランスフェクション後減少したが、トランスフェクション後24時間と48時間との間にわずかに増加した(図3D)。
コトランスフェクトしたK562細胞において、β−グロビンの発現レベルを増加させる試みにおいて、我々は、トランスフェクションに使用するDNAの量を、5μgから10μgの各プラスミドに増加させた。我々はまた、トランスフェクション後異なる時間に転写レベルを比較し、そしてこの実施例において17時間が最適であることを見出した(データは示していない)。より高い濃度のDNAによるコトランスフェクションは、細胞におけるBCL11AおよびEKLFのレベルを増加させた(図4A)。K562細胞を、最初にBCL11A−XL(10μg)またはHA−EKLF(10μg)でトランスフェクトし、そしてトランスフェクションの17時間後に、qPCRによってβ−グロビンの発現を分析した。BCL11A−XLまたはHA−EKLFだけによるトランスフェクションは、トランスフェクトしていないK562細胞と比較して、β−グロビン転写物のレベルをそれぞれ5.9および7.5倍増加させた(図4B)。それぞれ5μgのBCL11A−XLおよびHA−EKLFによるコトランスフェクションは、β−グロビン転写物のレベルを305±156.9倍増加させた(n=2)(図4B)。しかし、それぞれ10μgのBCL11A−XLおよびHA−EKLFによるコトランスフェクションは、β−グロビン転写物のレベルを、トランスフェクトしていないK562細胞と比較して、887±143倍(n=2)増加させた(図4B)。両方の濃度のDNAによるトランスフェクション後、K562細胞においてベータグロビンタンパク質が検出された(図4A)。γ−グロビン対β−グロビン発現の比は、BCL11A−XLおよびEKLFによるコトランスフェクション時に再び減少したが、5μgの各プラスミドと比較して、10μgによるトランスフェクション時に、その比における非常に小さい減少のみが検出された(図4C)。
臍帯血CD34+前駆細胞から分化した赤芽球は、通常の成人赤血球細胞と同様に主にHbA(約94%)を発現する末梢血CD34+細胞から分化した赤芽球と対照的に、主にHbF(約65%)を発現する。図5Aは、臍帯RBCと比較して、成人RBCに存在するグロビンアイソフォームを示す。
臍帯血赤芽球においてBCL11AおよびEKLFの発現を増加させることは、成人β−グロビンの発現を増加させることが見出された。
EKLFおよびBCL11A両方の遺伝子を赤血球細胞へ伝達するための、より効率的なシステムを開発した。レンチウイルスpSEW−GFPプラスミドを用いた、BCL11AおよびEKLF遺伝子両方のためのレンチウイルス構築物を作成した(バックボーンプラスミド、ウイルスパッケージングプラスミド、導入ベクタープラスミドおよび293T細胞は、School of Biochemistryによって提供された)。そのアプローチの実行可能性を確認するために、赤血球生成(erythropoetic)細胞系、K562細胞を、レンチウイルス構築物pXLG3−mcherry−EKLF(図7A)およびpXLG3−eGFP−BCL11A−XL(図7B)で同時に形質導入した。形質導入効率は、慣用的に>80%であった。EKLFおよびBCL11A共焦点画像のオーバーレイは、>40%のその転写因子の二重発現を示した(図7C)。

使用される前記転写因子が、BCL11AとEKLFとの組合せである、いずれかの先行項目に記載の方法。
(項目10)
前記細胞が、臍帯血、人工多能性幹細胞、赤血球生成幹細胞または赤血球生成細胞系から得られる、いずれかの先行項目に記載の方法。
(項目11)
前記培養培地が、血清、胎仔ウシ血清、インスリン、ヘパリン、トランスフェリンのうちの少なくとも1つを含む、いずれかの先行項目に記載の方法。
(項目12)
前記培地に、SCF、EPOまたは鉄飽和トランスフェリンのうちの少なくとも1つが補充され得る、項目11に記載の方法。
(項目13)
項目1〜12のいずれか一項に記載の方法に従って調製された赤血球。
(項目14)
成人表現型を有し、除核され、かつ未改変細胞と比較してβ−グロビンの発現が増加するようにインビトロで改変された培養幹細胞から構成される、成人表現型の赤血球。
(項目15)
前記細胞が、前記未改変細胞と比較して減少したγ−グロビンの発現をさらに有する、項目14に記載の赤血球。
(項目16)
前記培養幹細胞が、転写因子BCL11AおよびEKLFまたはそれらの任意のバリアントでコトランスフェクトすることにより改変されている、項目14または15に記載の赤血球。
(項目17)
項目13〜16のいずれか一項に記載の赤血球および薬学的に受容可能なキャリア、希釈剤または賦形剤を含む、組成物。
(項目18)
項目13〜16のいずれか一項に記載の赤血球または項目17に記載の組成物を備える、輸血パック。
Claims (14)
- 培養中の成人赤血球を調製する方法であって、
エキソビボ供給源から幹細胞または細胞系を得る工程、
該細胞を規定培地中で培養する工程、
該細胞を転写因子BCL11AおよびEKLFで改変する工程、および
該細胞中の胎児グロビンを成人グロビンへと変換する工程、
を包含する、方法であって、該エキソビボ供給源は、臍帯血、人工多能性幹細胞、赤血球幹細胞、または赤血球生成細胞系から選択される、方法。 - 成人グロビンの発現が誘導される、請求項1に記載の方法。
- 成人β−グロビンの発現が増加し、および/または胎児γ−グロビンの発現が減少する、請求項1または2に記載の方法。
- 前記細胞が、改変前に胎児ヘモグロビンを発現する、請求項1〜3のいずれか一項に記載の方法。
- 前記細胞が、胎児表現型から成人表現型へと変換される、請求項1〜4のいずれか一項に記載の方法。
- 前記方法が、細胞を除核するさらなる工程を包含し、ここで該細胞が人工多能性幹細胞から誘導される、請求項1〜5のいずれか一項に記載の方法。
- 前記細胞が、表面抗原CD34を発現する、請求項1〜6のいずれか一項に記載の方法。
- 前記細胞をBCL11A、タギング形態のEKLF、GATA 1、FOG 1、SCL、SOX6のうちの1つまたはそれより多くで改変する工程をさらに包含する、請求項1〜7のいずれか一項に記載の方法。
- 前記培養培地が、血清、胎仔ウシ血清、インスリン、ヘパリン、トランスフェリンのうちの少なくとも1つを含む、請求項1〜8のいずれか一項に記載の方法。
- 前記培地に、SCF、EPOまたは鉄飽和トランスフェリンのうちの少なくとも1つが補充され得る、請求項9に記載の方法。
- 請求項1〜10のいずれか一項に記載の方法に従って調製された赤血球。
- 成人表現型を有し、除核され、かつ未改変細胞と比較してβ−グロビンの発現が増加するようにインビトロで改変された培養幹細胞から分化した、成人表現型の赤血球であって、該培養幹細胞は、臍帯血、人工多能性幹細胞、赤血球幹細胞、または赤血球生成細胞系に由来し、該成人表現型の赤血球が、該未改変細胞と比較して減少したγ−グロビンの発現を有し、該培養幹細胞が、転写因子BCL11AおよびEKLFでコトランスフェクトすることにより改変されている、赤血球。
- 請求項11または12のいずれか一項に記載の赤血球および薬学的に受容可能なキャリア、希釈剤または賦形剤を含む、組成物。
- 請求項11または12に記載の赤血球または請求項13に記載の組成物を備える、輸血パック。
