Description
本発明は、服用後約0.1〜約16時間、好ましくは約0.5〜約5時間、より好ましくは約1〜約4.5時間で、フェニレフリンのピーク血漿濃度を提供するように、それを必要としている被験体に、フェニレフリン又はその薬学的に許容し得る塩を送達するフェニレフリン粒子であって、前記フェニレフリンが、服用後少なくとも約6、約8、約12、約16、約20、及び/又は約24時間、約20、約40、約60、約80、約100、約120、約140、約160、約180、又は約200pg/mL超の濃度で維持される粒子を目的とする。
本明細書で使用するとき、フェニレフリンの薬学的に許容し得る塩としては、塩酸フェニレフリン、重酒石酸フェニレフリン、タンニン酸フェニレフリン等が挙げられるが、これらに限定されない。1つの好ましい実施形態では、フェニレフリンの薬学的に許容し得る塩は、塩酸フェニレフリンである。
ポリマーコーティングで被覆されたフェニレフリン粒子を含む製剤を調製した。長時間にわたってフェニレフリンを放出させる製剤は、25℃/60%RHで24ヶ月間、及び40℃/75%RHで3ヶ月間安定であることが証明された。フェニレフリンの多くの顆粒製剤は、経時的に安定ではなく、著しく劣化する。
2:固体重量。
3:エチルセルロース、セチルアルコール、及びラウリル硫酸ナトリウムを含む。
4:加工中に精製水、アセトン、及びイソプロピルアルコールを除去する。
1.精製水USPを、好適な大きさのステンレス鋼製容器に添加した。
2.エチルアクリレートNF及びメチルメタクリレートコポリマー分散液NF(Eudragit(登録商標)NE 30D)を、穏やかに撹拌しながら添加した。
3.フェニレフリンHCl USPを、撹拌しながら混合している間に添加し、混合した。
4.工程3の混合物を用いて、アルファ化加工デンプンNFを(層状に)被覆した。
5.工程4で得られた層状フェニレフリンHCl/アルファ化加工デンプンを乾燥させ、#20の篩で篩った。
6.以下を、記載する順序で、穏やかに撹拌しながら好適な大きさの容器に添加した:イソプロピルアルコールUSP、次いで、アセトンNF、次いで、アセチルトリブチルシトレートNF。
7.エチルセルロースNF(Ethocel(登録商標)Standard Premium 10)を撹拌しながら添加し、透明な溶液が形成されるまで混合した。
8.ステアリン酸マグネシウムを、撹拌しながら前記溶液に添加した。
9.工程5で得られた、篩い分けされた層状フェニレフリン/アルファ化加工デンプン粒子を、Wursterインサートを取り付けた好適な流動床コーティングユニットを用いて、工程8で得られた溶液で被覆した。
10.工程9で得られた粒子をオーブンで硬化させた。
11.Eudragit(登録商標)NE30D、次いで、精製水USP及びエチルセルロース水性分散液NF(Aquacoat ECD(登録商標))を、好適な大きさの容器に添加し、穏やかに撹拌しながら混合した。
12.工程10で得られたEthocel(登録商標)で被覆された層状粒子を、Wursterインサートを取り付けた好適な流動床コーティングユニットを用いて、コーティング溶液で被覆した。
13.工程12で得られたコーティング粒子を、コロイド状二酸化ケイ素NFと混合した。
14.工程13で得られた粒子をオーブンで硬化させた。
工程14で得られた被覆されたフェニレフリン粒子を、274nmにおけるUV検出を利用して、米国薬局方<General Chapter<711>Dissolution>に記載の装置、Apparatus II、回転パドルを用いて、0時間から14時間における溶解について分析した。溶解媒体は、最初の1時間については750mLの0.1N HClであり、次いで、2〜14時間については、1000mLの0.05Mリン酸ナトリウムバッファ(pH 6.8)であった。温度は、37℃であり、回転速度は、50rpmであった。溶解は、製剤中フェニレフリンの量100%で調製した標準に対して、放出されたフェニレフリンの割合が、1時間で50%以下、3時間で30%以下、8時間で50%以下であることを示した。使用した方法は、以下の通りであり、結果を以下の表2に示す。
1.溶解媒体の温度が標的値(37℃)に達していることを確認する。
2.45mgのフェニレフリンHClに等しいサンプルを量り分ける。(媒体溶液の表面上の)サンプルを、750mLの0.1N塩酸を収容している各容器に添加し、50rpmのパドル速度で溶解試験を開始する。0.1N塩酸中で1時間操作した後、1時間の時点での測定を完了する。250mLの0.20M第三リン酸ナトリウムを添加することによって、直ちに緩衝段階に進める。緩衝媒体のpHは、6.8±0.05である。
3.274nmでUV測定するためのラインプローブを備えるLEAPファイバー光学システムを用いることによって、媒体中に放出されたフェニレフリンHClのUV吸光度を測定する。
4.溶解しているフェニレフリンHClの量は、274nmの波長における標準溶液のUV吸光度と比較して、試験下のサンプル溶液のUV吸光度を用いて決定することができる。また、溶解しているフェニレフリンHClの量は、以下のアッセイ方法を用いて決定することができる。
サンプルの調製
1.約1600mgのフェニレフリンHCl粒子を正確に計量し、200mLのメスフラスコに移す。(固体の塊が形成されるのを避けるために、1mLの1%酢酸/水溶液を添加して粒子を濡らすことを推奨する。)
2.70mLの1%酢酸/アセトニトリル溶液を添加し、1時間低速でプラットフォームシェイカー上にてフラスコを振盪する。注記:フラスコを定期的に回して、溶媒レベルを超えて集まる粒子を除去する。
3.約50mLの1%酢酸/水溶液をフラスコに添加し、1時間低速でフラスコを連続的に振盪する。
4.1%酢酸/水溶液を用いて規定の体積に希釈し、十分混合する。
5.0.45μmのMillipore Millex PVDFフィルタを用いてアリコートを濾過する。最初の1〜2mLの濾液を廃棄した後、更に希釈するために濾液を回収する。
6.6mLの濾液を50mLのメスフラスコにピペットで移し、1%酢酸/水で規定の体積に希釈し、十分混合する。
標準(0.05mg/mLの、1%酢酸/水中フェニレフリンHCl)及びサンプルを、以下に推奨する条件と同様の条件下で、好適なHPLCシステムに注入する。パラメータは、クロマトグラフィーを最適化するために変更してもよい。標準溶液のピーク面積と比較して、試験下のサンプル溶液のピーク面積を用いて、フェニレフリンHClのアッセイを決定する。
サンプルの調製

標準(0.00025mg/mLの、1%酢酸/水中フェニレフリンHCl)及びサンプルを、以下に推奨する条件と同様の条件下で、好適なHPLCシステムに注入する。クロマトグラフィーを最適化するためにパラメータを変更してもよい。標準溶液のピーク面積と比較して、試験下のサンプル溶液のピーク面積を用いて、フェニレフリンHClの分解産物の量を決定する。
フェニレフリン及びカチオン交換樹脂を含む粒子を調製し、半透性膜で更に被覆した。コーティング成分の量の比(ある程度変動し得る)は、例えば、酢酸セルロース:ヒドロキシプロピルセルロース2:1、3:1、4:1、又は5:1であってよい。コーティング濃度(ある程度変動し得る)は、例えば、コーティング粒子の50重量%、45重量%、40重量%、35重量%、30重量%、25重量%、又は20重量%であってよい。出発カチオン交換樹脂中の粒子の大部分は、約74μm〜約177μm(マイクロメートル)の粒径の粒子を有していた。
2:量は、遊離塩基を表す(1mgのフェニレフリンHClは、0.821mgのフェニレフリン遊離塩基に等しい)。
3:加工中にアセトン及び精製水を除去する。
1.所望の粒径を有するポリスチレンスルホン酸ナトリウムUSPを、170メッシュの篩に通し、篩上に残った画分を回収した。
2.工程1で得られたポリスチレンスルホン酸ナトリウムUSPを精製水に分散させ、混合した。
3.混合しながら、工程2で得られたスラリーの一部を濾過し、精製水USPで洗浄した。水の大部分が除去されるまで濾過を続けた。
4.樹脂を容器に移した。
5.スラリーが全て除去されるまで工程3及び4を繰り返した。
6.精製水USPを、好適な大きさのステンレス鋼製容器に添加した。
7.混合しながら、フェニレフリンHCLを容器に添加し、溶解するまで混合した。
8.工程5で得られた洗浄した樹脂を、連続的に混合しながら添加し、スラリーに混合した。
9.混合しながら、工程8で得られたスラリーの一部を除去し、精製水USPで洗浄した。水の大部分が除去されるまで濾過を続けた。
10.工程9で得られた,洗浄され、濾過されたフェニレフリン樹脂酸を容器に移した。
11.スラリーが全て濾過されるまで工程9及び10を繰り返した。
12.フェニレフリン樹脂酸塩を乾燥させた。
13.精製水USP及びアセトンNFを、適切な大きさのステンレス鋼製容器に添加した。
14.ヒドロキシプロピルセルロースNFを容器にゆっくり添加し、溶解するまで混合した。酢酸セルロースNFをゆっくり添加し、溶解するまで混合した。
15.溶液が所望の重量になるまでアセトンNFを添加した。
16.工程12で得られたフェニレフリン樹脂酸塩を、Wursterカラムを取り付けた適切な流動床コーティング装置において、工程15で得られたコーティング溶液で被覆した。
17.被覆されたフェニレフリン樹脂酸塩を容器に排出した。
18.被覆されたフェニレフリン樹脂酸塩を乾燥させた。
19.乾燥した、被覆されたフェニレフリン樹脂酸塩を、米国標準#40メッシュの篩で篩い、篩を通過した画分を回収した。
工程19で得られた被覆されたフェニレフリン樹脂酸塩粒子を、274nmにおけるUV検出を利用して、米国薬局方<General Chapter<711>Dissolution>に記載の装置、Apparatus II、回転パドルを用いて、0時間から14時間における溶解について分析した。溶出溶媒は、最初の1時間については750mLの0.1N HClであり、2〜14時間については、1000mLの0.05Mリン酸ナトリウムバッファ(pH 6.8)であった。温度は、37℃であり、回転速度は、50rpmであった。溶解は、製剤中フェニレフリンの量100%で調製した標準に対して、放出されたフェニレフリンの割合が、1時間で50%以下、3時間で30%以下、8時間で50%以下であることを示した。使用した方法は、以下の通りであり、結果を以下の表5に示す。

3.1時間後、3時間後、6時間後(任意)、及び8時間後に、各容器から溶解サンプル溶液10mLを注ぐ。Varian Full Flowフィルタ(10μm)を通してサンプル溶液を濾過する。
4.溶解しているフェニレフリンの量は、274nmの波長における標準溶液のUV吸光度と比較したUV吸光度から決定することができる。また、溶解しているフェニレフリンの量は、フェニレフリンアッセイ方法を用いて決定することができる。
5.より早い時点で注いだ量を加えることによって、3、6、及び8時間で溶解した量を補正する。DISSLプログラム(又は等価物)を用いて又は手動で、中間体サンプリングについて補正する。
数ロットの樹脂及び樹脂系粒子を、粒径分布について分析した。サンプルは、(1)Amberlite(商標)IRP69樹脂(DOW Chemical Companyから市販)、(2)選択された粒径を有する無負荷樹脂(それぞれ、プロセスA又はプロセスBによって調製)、及び(3)負荷樹脂酸塩粒子(すなわち、フェニレフリンを含有)を含んでいた。粒径分布は、11分間90ボルトに設定したFMC Syntron Sieve analyzer(FMC Technologies,Houston,TX)において、1サンプル当たり約75グラムを用いて分析した。篩を、ほんの少量のステアリン酸マグネシウムで処理して、操作中の粘着を予防した。結果を表6及び7に示す。

サンプルの調製
1.好適な量の被覆されたフェニレフリン樹脂酸塩サンプル(25mgのフェニレフリンHClに相当する量を含有)を正確に計量し、計量したサンプルを500mLのメスフラスコに移す。
2.希釈液(1N HCl)400mLを添加し、2時間以上低速でプラットフォームシェイカー上にてフラスコを振盪する。
3.粒子が溶媒レベルを超えて集まらないことを確実にするために、定期的に希釈液で粒子を溶液にすすぐ。
4.希釈液で規定の体積に希釈し、十分混合する。
5.0.45μmのMillipore Millex PVDFシリンジフィルタ又は等価物を用いてアリコートを濾過する。μおよそ最初の5mLの濾液を廃棄した後、分析のためにHPLCバイアルに残りを回収する。
標準(0.05mg/mLの、1N HCl中フェニレフリンHCl)及びサンプルを、以下に推奨する条件と同様の条件下で、好適なHPLCシステムに注入する。パラメータは、クロマトグラフィーを最適化するために変更してもよい。システムの適合性規格を満たしている場合、分析結果は有効である。
実施例5の第1のPK試験、第2のPK試験、及びPD試験で使用した被覆されたフェニレフリン樹脂酸塩粒子を、実施例2に記載の方法を用いて0〜8時間の溶解について分析した。結果を以下の表10Aに示す。
1.溶解媒体の温度が標的値に達していることを確認する。
2.(好適なチューブを用いて媒体溶液に直接)サンプルを、750mLの0.1N塩酸を収容している各容器に添加し、75rpmのパドル速度で溶解試験を開始する。0.1N塩酸中で1時間操作した後、1時間後のサンプルを注ぎ、250mLの0.20M第三リン酸ナトリウムを添加することによって、直ちに緩衝段階に進めた。媒体のpHは、6.8±0.05でなければならない。
3.1時間後、3時間後、6時間後(任意)、及び8時間後に、各容器から溶解サンプル溶液10mLを注ぐ。Varian Full Flowフィルタ(10μm)を通してサンプル溶液を濾過する。
4.274nmの波長における標準溶液のUV吸光度と比較したUV吸光度から溶解しているフェニレフリンの量を決定する。

実施例5の第1のPK試験及び第2のPK試験において使用した被覆されたフェニレフリン樹脂酸塩粒子を、25℃及び相対湿度60%で1ヶ月間、並びに40℃及び相対湿度75%で1ヶ月間保存した後の安定性について分析した。全てのサンプルについて、3−ヒドロキシベンズアルデヒドのレベルは、0.5%以下であり、フェニレフリン4,6異性体(N−メチル−4,6−ジヒドロキシ−1,2,3,4−テトラヒドロキシイソキノロンHCL)及びフェニレフリン4,8異性体(N−メチル−4,8−ジヒドロキシ−1,2,3,4−テトラヒドロキシイソキノロンHCL)のレベルは、2.0%以下であった。フェニレフリンに関して定量された合計分解産物は、各環境において1ヶ月で2.0%以下であった。
分解産物法のためのサンプル調製
1.好適な量の被覆されたフェニレフリン樹脂酸塩サンプル(25mgのフェニレフリンHClに相当する量を含有)を正確に計量し、計量したサンプルを500mLのメスフラスコに移す。
2.希釈液(1N HCl)400mLを添加し、2時間以上低速でプラットフォームシェイカー上にてフラスコを振盪する。
3.粒子が溶媒レベルを超えて集まらないことを確実にするために、定期的に希釈液で粒子を溶液にすすぐ。
4.希釈液で規定の体積に希釈し、十分混合する。
5.0.45μmのMillipore Millex PVDFシリンジフィルタ又は等価物を用いてアリコートを濾過する。μおよそ最初の5mLの濾液を廃棄した後、分析のためにHPLCバイアルに残りを回収する。
標準(0.00025mg/mLの、1N HCl中フェニレフリンHCl)及びサンプルを、以下に推奨する条件と同様の条件下で、好適なHPLCシステムに注入する。パラメータは、クロマトグラフィーを最適化するために変更してもよい。システムの適合性規格を満たしている場合、分析結果は有効である。
2つの薬物動態(PK)試験、及び薬力学(PD)試験を実施した。
実施例1の被覆された徐放フェニレフリン粒子及び実施例2の被覆された徐放フェニレフリン樹脂酸塩粒子の薬物動態プロファイル、バイオアベイラビリティ、及び代謝を調べるために、16人の被験者に対してパイロット試験を実施した。一晩の絶食後、被験者を4つの処理群に割り付けた。4つの期間の間に7日間の休薬期間を設けた。両方の場合において、20mgのフェニレフリンHCl用量に相当するコーティング粒子をアップルソースに入れて健常被験者に投与した。更に、実施例2の徐放フェニレフリン樹脂酸塩粒子と市販の即時放出液体との組み合わせを評価した。組み合わせ処理において、15mgのフェニレフリンHClに等しい被覆されたフェニレフリン樹脂酸塩粒子をアップルソースに入れて投与し、5mgのフェニレフリンHClに等しい10mLの液体を経口注射器によって投与した。
以下の目的で第2のパイロット試験を実施した:(i)30mgのフェニレフリンが、4時間間隔で投与される2つの10mg用量の即時放出フェニレフリンと同様の最高薬物濃度を得ることができるかどうかを決定する、及び(ii)20mgのフェニレフリン及び1300mgのアセトアミノフェンのER PKプロファイル及びバイオアベイラビリティを評価する。
PK試験の結果を、以下の図1〜11及び表13に示す。
図は、概数にされている。
20mgのフェニレフリンを含むER−IRブレンドは、10mgのIR用量の50%であるCmax、及び2つの10mgのIR用量(20mg)よりも15%大きいAUCinfを有していた。
上気道感染症に起因する鬱血及び疼痛の症状を有する被験者におけるフェニレフリン及びフェニレフリン−アセトアミノフェン徐放製剤の効果を調べるために、無作為化二重盲検プラセボ対照試験を実施した。30mgのER用量、45mgのER用量、及び1300mgのアセトアミノフェンと同時投与した30mgのER用量を評価し、プラセボと比較した。各例において、本発明の被覆されたERフェニレフリン樹脂酸塩粒子を使用した。30mgのER用量、45mgのER用量、及び1300mgのアセトアミノフェンと同時投与した30mgのER用量は、それぞれ、1日目の0〜12時間における(1)鼻詰まり/鼻閉;(2)副鼻腔の圧迫感/圧痛;及び(3)頭部鬱血についての重篤度スコアにおいてプラセボに対して良好な性能を示した。

[1] フェニレフリン及びカチオン性ポリスチレンスルホン酸を含む薬物−樹脂複合体であって、前記カチオン性ポリスチレンスルホン酸が、前記フェニレフリンと組み合わされる前に約74μm〜約177μmの粒径の粒子を含む、複合体。
[2] 前記カチオンが、ナトリウム、銅、亜鉛、鉄、カルシウム、ストロンチウム、マグネシウム、及びリチウムからなる群から選択される、[1]に記載の薬物−樹脂複合体。
[3] 前記カチオンが、ナトリウムである、[2]に記載の薬物−樹脂複合体。
[4] コーティングで被覆された[3]に記載の薬物−樹脂複合体を含む、徐放性粒子。
[5] 前記コーティングが、セルロース材料を含む、[4]に記載の徐放性粒子。
[6] 前記セルロース材料が、酢酸セルロース及びヒドロキシプロピルセルロースからなる群から選択される、[5]に記載の徐放性粒子。
[7] [6]に記載の徐放性粒子を含む、医薬製剤。
[8] 即時放出型のフェニレフリンを更に含む、[7]に記載の医薬製剤。
[9] [1]に記載の薬物−樹脂複合体をコーティングすることを含む、コーティングされた薬物−樹脂複合体を形成する方法。
[10] 前記粒子の少なくとも約50%が、約74μm〜約177μmの粒径を有する、[1]に記載の薬物−樹脂複合体。
[11] 前記粒子の少なくとも約80%が、約74μm〜約177μmの粒径を有する、[10]に記載の薬物−樹脂複合体。
[12] 前記粒子の少なくとも約90%が、約74μm〜約177μmの粒径を有する、[11]に記載の薬物−樹脂複合体。
[13] 前記粒子の15%未満が、約44μm未満の粒径を有する、[1]に記載の薬物−樹脂複合体。
Claims (19)
- フェニレフリン及びカチオン性ポリスチレンスルホン酸の複合体を含む徐放性粒子であって、
前記カチオン性ポリスチレンスルホン酸の少なくとも90%が、前記フェニレフリンと組み合わされる前に74μm〜177μmの粒径を有しており、
前記フェニレフリン及びカチオン性ポリスチレンスルホン酸の複合体はコーティングで被覆されており、
前記コーティングの量は、前記徐放性粒子の30〜45重量%であり、
前記コーティングは、酢酸セルロースおよびヒドロキシプロピルセルロースを含み、
前記コーティングは、67〜83重量%の酢酸セルロースを含み、
前記徐放性粒子は、投与後8時間以上にわたってフェニレフリンを放出する
徐放性粒子。 - 前記カチオンが、ナトリウム、銅、亜鉛、鉄、カルシウム、ストロンチウム、マグネシウム、及びリチウムからなる群から選択される、請求項1に記載の徐放性粒子。
- 前記カチオンが、ナトリウムである、請求項2に記載の徐放性粒子。
- 請求項1に記載の徐放性粒子を含む、医薬製剤。
- 即時放出型のフェニレフリンを更に含む、請求項4に記載の医薬製剤。
- フェニレフリン及びカチオン性ポリスチレンスルホン酸を含む薬物−樹脂複合体をコーティングすることを含む、請求項1に記載の徐放性粒子を形成する方法。
- 前記カチオン性ポリスチレンスルホン酸の15%未満が、前記フェニレフリンと組み合わされる前に44μm未満の粒径を有する、請求項1に記載の徐放性粒子。
- 酢酸セルロースとヒドロキシプロピルセルロースの重量比(酢酸セルロース:ヒドロキシプロピルセルロース)が、2:1、3:1、4:1および5:1からなる群より選択される請求項1に記載の徐放性粒子。
- 前記酢酸セルロースとヒドロキシプロピルセルロースの重量比(酢酸セルロース:ヒドロキシプロピルセルロース)が3:1である、請求項8に記載の徐放性粒子。
- 前記コーティングの量は、前記徐放性粒子の35重量%である請求項1に記載の徐放性粒子。

- 前記徐放性粒子は、投与後12時間以上にわたってフェニレフリンを放出する請求項1に記載の徐放性粒子。
- 請求項4に記載の医薬製剤と1つ以上の追加の治療剤とを含む組合せ物であって、前記追加の治療剤は、抗ヒスタミン剤、鬱血除去剤、鎮痛剤、抗炎症剤、解熱剤、鎮咳剤、および去痰剤からなる群より選択される組合せ物。
- 前記追加の治療剤がアセトアミノフェンである、請求項13に記載の組合せ物。
- 前記追加の治療剤が、ブロモフェニラミン、クロルシクリジン、デクスブロムフェニラミン、フェニンダミン、フェニラミン、ピリラミン、トンジルアミン、プリポリジン、エフェドリン、プソイドエフェドリン、フェニルプロパノールアミン、クロルフェニラミン、デキストロメトルファン、ジフェンヒドラミン、ドキシルアミン、アステミゾール、テルフェナジン、フェキソフェナジン、ナファゾリン、オキシメタゾリン、モンテルカスト、プロピルヘキサドリン、トリプロリジン、クレマスチン、アクリバスチン、プロメタジン、オキソメマジン、メキタジン、ブクリジン、ブロムヘキシン、ケトチフェン、テルフェナジン、エバスチン、オキサタミド、キシロメタゾリン、ロラタジン、デスロラタジン、及びセチリジン;これらの異性体、並びにこれらの薬学的に許容し得る塩類及びエステル類からなる群より選択される、請求項13に記載の組合せ物。
- 前記追加の治療剤が、イブプロフェン、ナプロキセン、ケトプロフェン、フルルビプロフェン、フェンブフェン、フェノプロフェン、インドプロフェン、ケトプロフェン、フルプロフェン、ピルプロフェン、カルプロフェン、オキサプロジン、プラノプロフェン、スプロフェン、セレコキシブ、アセトアミノフェン、アセチルサリチル酸、インドメタシン、ジクロフェナク、スリンダク、トルメチン、メフェナム酸、メクロフェナム酸、フルフェナム酸、ジフルニサル、フルフェニサール、ピロキシカム、スドキシカム、イソキシカム、及びメロキシカム;これらの異性体、並びにこれらの薬学的に許容し得る塩類及びプロドラッグ類からなる群より選択される、請求項13に記載の組合せ物。
- 前記追加の治療剤が、ジフェンヒドラミン、デキストロメトルファン、ノスカピン、クロフェジアノール、メントール、ベンゾナテート、エチルモルヒネ、コデイン、アセチルシステイン、カルボシステイン、アンブロキソール、ベラドンナアルカノイド、ソブレロール、グアヤコール、及びグアイフェネシン;これらの異性体、並びにこれらの薬学的に許容し得る塩類及びプロドラッグ類からなる群より選択される、請求項13に記載の組合せ物。

徐放性粒子。 - 19.5重量%のフェニレフリン及び45.5重量%のカチオン性ポリスチレンスルホン酸の複合体を含む徐放性粒子であって、
前記カチオン性ポリスチレンスルホン酸の少なくとも90%が、前記フェニレフリンと組み合わされる前に74μm〜177μmの粒径を有し、
前記フェニレフリン及びカチオン性ポリスチレンスルホン酸の複合体はコーティングで被覆されており、
前記コーティングの量は、前記徐放性粒子の35重量%であり、
前記コーティングは、75重量%の酢酸セルロースおよび25重量%のヒドロキシプロピルセルロースを含み、
前記徐放性粒子は、投与後8時間以上にわたってフェニレフリンを放出する
