Description
a.1つの特定の種に属する各バキュロウイルスゲノムが他のものよりも同一または異なるODVであることができる、少なくとも1つの製造される閉塞由来ビリオンにおいて共に包まれるバキュロウイルスゲノムを有する昆虫種の幼虫または培養細胞を重感染する;
b.多角体病により死亡するまで感染される幼虫を育てるまたは細胞を培養する;
c.それらの死亡後幼虫または培養される細胞から製造されるOBを単離する;
細胞または幼虫は、製造される閉塞由来ビリオンの少なくとも1つにおいて共に包まれるものであるゲノムのバキュロウイルスのいずれかによる感染に感受性がある。
−バキュロウイルスゲノムを含むウイルス粒子が注射される場合、BVまたは遊離ODVを使用することができる。血体腔注射が好ましい。
−ヌクレオカプシドを経口的に昆虫の幼虫に接種する場合、BV、遊離ODVまたはOBを使用することができる。BVはこの経路による感染の最少限に効果的な粒子である。遊離ODVを使用することができるが、封入体は自然に昆虫の幼虫によって取り込まれ、それらは自然感染性粒子であるので、これらは封入体に閉じ込められることが好ましい。経口接種が使用される場合、封入体を昆虫に供給される水性懸濁液で投与することができ、またはそれらは、食事または昆虫の幼虫により摂取される他の物質と混合される、固体または半固体の形態で投与されることができる。
a)同時接種または最初の感染の徴候の発現のために必要な時間に劣る時間の遅延による逐次接種により、2つの異なるバキュロウイルス種で昆虫の幼虫に重感染する、そして
b)多角体病による明らかな病気または死まで必要な条件で接種される幼虫を飼育する。
PCRアッセイを下記の表1に示されるプライマーを使用して実施した。
ビリオンは最終容量500μlで100μlの0.5MのNa2CO3、50μlの10% (w/v)ドデシル硫酸ナトリウムを用いて109のOBs/mlを含む100μlのOB懸濁液を混合および10分間、60℃でインキュベートすることによりOBから放出された。未溶解のOBと他の破片を低速遠心分離(3,800 x g、5分)により取り除いた。ビリオンを含む上清を1時間、50℃で25μlのプロテイナーゼK(20mg/ml)で処理した。ウイルスDNAを2回飽和フェノールと1回クロロホルムで抽出し、アルコール沈殿により水相から分離した。ペレットを10分間、60℃で50〜100μlの0.1xTE緩衝液(トリスEDTA、pH8)中に懸濁させた。DNA濃度を260nmで光吸収を読み取ることにより評価した。
この例では、バキュロウイルスの2種は同じ細胞で複製することができ、ODV中に位置する2つのタンパク質(PIF1およびPIF2)を共有することができ、バキュロウイルスの経口伝達に不可欠であるという証拠を示す。
−シロイチモジヨトウMNPV(SeMNPV)、SeUS2−A変異体
SeMNPVのSeUS2−A変異体は、核多角体病ウイルスSeMNPVの少なくとも4つの遺伝子型変異体(Munozら、1999年)のヘテロ遺伝子混合物を含む殺虫剤Spod-X(登録商標)(Certis USA, LLC)生物殺虫剤に基づくフロリダからのSeUS−2分離株から生じ、それは感染し、シロイチモンジョトウ、シロイチモジウヨトウの幼虫を殺す。SeUS2−Aは、処理される幼虫の約10%を殺したOBの低用量(LD10)を摂取したシロイチモジヨトウの幼虫においてインビボクローニングにより得られた。SeUS2−Aは、SeUS2分離株に存在する最も豊富な変異体であり、シロイチモジヨトウに唯一感染性である。この変異体はまたSe301細胞株においてインビトロで複製すことができる。
SfNIC−C変異体は、少なくとも9つの遺伝子型変異体(Simonら、2004年b)のヘテロ遺伝子混合物を含むSfMNPVニカラグア分離株から生じ、それは遺伝的および表現型的特徴を有している(Simonら、2005年a,b;2011年;2012年)。SfNIC−CをSf9細胞においてインビトロクローニングによって単離した(Simonら、2004年b)。感受性宿主昆虫への摂取材料の注射後に生産的感染を製造することができるが、この変異体は、pif1およびpif2遺伝子を取り除く〜16kbのゲノム欠失を有し、この変異体は摂取により感染性でないことを意味する。この変異体はまた細胞株Sf21およびSf9において複製することができる。
SeUS2−AおよびSfNIC−Cは、制限エンドヌクレアーゼPstI(たから)でゲノムDNAの処理により分化されることができ、アガロースゲル中の電気泳動に続いた。各ゲノムDNAから取得されるこの制限プロファイルは、明確に区別され、サンプル中において前記ゲノムの1つ(または両方)の存在の識別を許す(図2参照)。
シロイチモジヨトウの第4齢は、SeUS2−AとSfNIC−CのODVの混合物(各ウイルスのOBの等しい数を混合することにより得られる)の血体腔内注射により接種された。死亡および得られたOBが前述の方法(Caballeroら、1992年)により回収されるまで、注射された幼虫は、Greeneら(1976年)により記載されるように、小麦胚芽、酵母および大豆に基づく半合成飼料でそれぞれ飼育された。簡単に言えば、滅菌水中で死体の均質化およびろ過と分画遠心法による得られた懸濁液の精製後、それらを死んだ幼虫から抽出した。
これらの結果から、それは以下に結論付けることができる:
(a)シロイチモジヨトウの細胞は、2つのウイルスにより同時に感染することができ、それらの両方は複製できる。
(b)PIF1およびPIF2タンパク質は、ツマジロクサヨトウの幼虫において(これらのpif遺伝子を欠く)SfNIC−Cの経口感染性を救うことができる。
SeUS2−AおよびSfNIC−Cの混合物を注射した幼虫において製造される混合ウイルスOBは(セクションC参照)、シロイチモジヨトウの幼虫の経口で6連続継代(P1〜P6)を含む実験において接種材料(継代0、P0:SeUS2−AのOBとSfNIC−CのOBの1:1の比率から放出されるODVを使用するシロイチモジヨトウの重感染に得られる)として使用された。
これらの実験で使用された2つのウイルスは以下の通りであった:
−オートグラファカリフォルニカ多発性核多角体病ウイルス(AcMNPV)
AcMNPVは、アルファバキュロウイルス族のタイプ種である。それは、BV膜において糖タンパク質(GP64)の存在により特徴付けられる、グループIのNPVに属するマルチカプシドウイルスである。本研究では、クローン化された遺伝子型AcC6が使用され、それは完全に配列決定された最初のバキュロウイルスゲノムであった(GenBank信託番号L22858:Ayresら、1994年)。このウイルスは、Sf21およびSf9においてよく複製し、ツマジロクサヨトウの幼虫に生産的致死感染を生じさせることができる。
これは、BVでFタンパク質の存在により特徴付けられるグループIIのNPVであり(Herniouら、2004年)、したがって、AcMNPVからの系統発生学的に離れている。本研究で、SfIC−Bと命名されたインビトロクローンが使用され、ニカラグア(SfNIC)からの野生型分離株の存在する最も豊富な遺伝子型であった(Simonら、2004年b)。SfNIC−Bは、完全なゲノムであり(GenBank信託番号HM595733.1、Simonら、2012年)、ツマジロクサヨトウの幼虫に経口で感染性がある。
混合ウイルスODVとOBを製造するために、ツマジロクサヨトウ第4齢は、液滴供給方法を使用して、各ウイルスの5×107OB/mlの濃度で1:1の割合でSfNIC−BのOBとAcMNPVのOBの混合物を接種した(Hughes & Wood、1986年)。ツマジロクサヨトウ幼虫は両方のウイルスに感受性がある。24の幼虫のグループを接種し、実験を3回行った。接種された幼虫を死亡まで制御された温度(25℃)で例1でまた用いた半合成実験用飼料で飼育した。OBは感染した死体から採取され、前述のろ過および遠心分離により精製された(Munozら、1998年)。
実施された繰り返しのそれぞれにおいて、nはAcMNPV、SfMNPVまたは両方のウイルスのいずれかの分析された細胞培養プラークの数である。S.D.は標準偏差を示す。
陽性ウェルは、バキュロウイルス感染(OB)の病理学的徴候を有する少なくとも1つの細胞に含まれたものである。P(0)は、感染の徴候なしにウェルを有する各繰り返しで計算された確率である。P(1)、P(2)およびP(3)は、ポアゾン分布により推定され、それぞれ1つ、2つまたは3つのODVにより感染するために特定のウェル中で細胞のための確立を指す。
A)これらの実験で使用された2つのウイルスは以下の通りであった:

昆虫の死亡率上の2つの異なるウイルス種による感染の間隔の影響を決定するために、最近、脱皮ツマジロクサヨトウ第4齢は、液滴供給方法を使用して最初のウイルスの5 x 107OB/mlの懸濁液を経口的に接種された(Hughes & Wood、1986年)。接種された昆虫は、最初の接種後12、24、48または72時間で25℃で半合成実験食で別個に飼育し、これらの幼虫は第2のウイルスのOBの同じ濃度で供給するようにされた。全ての場合において、24の幼虫のグループは、各処理で使用され、実験は3回実施され、第1のウイルス接種材料としてAcC6のOB、第2の接種材料としてSfNIC−BのOB、逆もまた同様に使用した。1回または2回接種さていた幼虫は、個別にふ化または死亡まで半合成実験食で飼育された。

−(i)単独でAcC6のOBで処理された幼虫:この処理は、ウイルス死亡率の最も低い有病率をもたらした。
−(ii)単独でSfNIC−BのOB、混合物中で両ウイルスのOBの同時投与、またはSfNIC−BのOBの後に続いてAcMNPVのOBで処理された幼虫:これらの処理の全ては、死亡率の同様に高い有病率を得た。
−(iii)AcC6のOBで最初に処理され、その後SfMNPVのOBで処理された幼虫:死亡率の有病率は、わずかだが単独でAcC6のOBで処理された幼虫で観察されたよりも有意に高かった。
前記のように、DNA抽出物により決定されることに続きウイルス特異的プライマーを用いる各ウイルスDNAのqPCR定量のように(表1参照)、第1と第2の接種処理の間の時間的間隔は、子孫のOBで各ウイルスの割合に影響を与えた。これらの結果の統計的分析は、5つの区別できるグループの存在を同定した(分散分析:F10,75 =187.57; p<0.001):
−(i)AcC6単独
−(ii)SfNIC−B単独
−(iii)同時に両方のウイルスで処理された幼虫とおよびSfNICのOBで初期処理を含む全ての処理
−(iv)AcC6のOBで最初におよび48または72時間後にSfNIC−BのOBで処理された幼虫
−(v)AcC6のOBで最初におよび48または72時間後にSfNIC−BのOBで処理された幼虫。
ODVのウイルス組成物は、例2で記載されるように、プラークアッセイにより分析された。第1と第2の接種の間の時間が増加すると、単一種のゲノム(ビリオン)または両方の種の割合は、接種の間隔および第1または第2のウイルスの同一性に従って一緒に変化した(図11)。例えば、AcC6のOBの接種に続き24時間後のSfNIC−Bの接種は、混合ウイルスビリオン内に両方のウイルスを含むODVの約50%で、約50%のAcC6ゲノムと50%のSfNIC−Cゲノムを含む子孫のOBをもたらした。

Claims (28)
- a.昆虫の幼虫または昆虫細胞を、少なくとも1つは多発性核多角体病ウイルスである、2つ以上の異なるバキュロウイルス種で重感染し、
b.前記感染した昆虫の幼虫または昆虫細胞からウイルス封入体(OB)を単離し、または場合によっては更に前記ウイルス封入体(OB)からビリオンを遊離し、
c.同じビリオンに、異なるバキュロウイルスゲノムが存在することを確認することを含む、
同じビリオンに少なくとも2つ以上の異なるバキュロウイルス種からのゲノムを含む混合ウイルス封入体由来ビリオン(ODV)、または少なくとも1つの前記混合ウイルス封入体由来ビリオンが閉じ込められている混合ウイルス封入体(OB)を製造する方法。 - a.同じ封入体由来ビリオンに共に包まれる2以上の異なるバキュロウイルスゲノムで昆虫種の幼虫または昆虫種の培養細胞を重感染し;
b.多角体病による明らかな病気または死亡まで、前記感染した幼虫を飼育、または前記感染した細胞を培養し;
c.前記幼虫または前記培養された細胞から、ウイルス封入体(OB)を単離し、
d.同じビリオンに、異なるゲノムが存在することを確認することを含む、
同じビリオンに少なくとも2つ以上の異なるバキュロウイルス種からのゲノムを含む混合ウイルス封入体由来ビリオン(ODV)、または少なくとも1つの前記混合ウイルス封入体由来ビリオンが閉じ込められている混合ウイルス封入体(OB)を製造する方法。 - 前記バキュロウイルス種の全てが、多発性核多角体病ウイルスである、請求項1又は2に記載の方法。
- 前記同じビリオンに含まれる前記異なるゲノムの少なくとも2つが、それぞれ感染性バキュロウイルス粒子を生じることができ、細胞内で完全なウイルスサイクルを形成することができるゲノムである、請求項1〜3のいずれか1項に記載の方法。
- 前記バキュロウイルスゲノムが、封入体に閉じ込められている封入体由来ビリオン(ODV)を形成しながら包まれ、前記封入体の少なくとも1つは、1つより多い種のゲノムが共に包まれる少なくとも1つの混合ウイルス封入体由来ビリオン(ODV)を含む、請求項1〜4のいずれか1項に記載の方法。
- 前記重感染は、前記バキュロウイルス種、前記ウイルス種のゲノムまたは前記ゲノムを有するヌクレオカプシドを、昆虫細胞または昆虫の幼虫に、
a. 他の前記バキュロウイルス種、前記ウイルス種のゲノムまたは前記ゲノムを有するヌクレオカプシドと同時に、或いは
b. 他の前記バキュロウイルス種、前記ウイルス種のゲノムまたは前記ゲノムを有するヌクレオカプシドに対して別々の時間で
接種することで実施される、請求項1〜5のいずれか1項に記載の方法。 - 前記バキュロウイルス種、前記ウイルス種のゲノムまたは前記ゲノムを有するヌクレオカプシドが、他の前記バキュロウイルス種、前記ウイルス種のゲノムまたは前記ゲノムを有するヌクレオカプシドの摂取に対して48時間より短い時間間隔内で前記昆虫細胞または昆虫の幼虫に接種される、請求項6に記載の方法。
- i)前記バキュロウイルス種のヌクレオカプシドが、発芽ビリオン(BV)または遊離ODVの形態で、注射により昆虫の幼虫に接種され、或いは
ii)前記ヌクレオカプシドが、発芽ビリオン(BV)、遊離ODVまたはウイルス封入体(OB)の形態で、経口で昆虫の幼虫に接種され、それらは、封入体に閉じ込められているODVを形成しながら包まれる、請求項1〜7のいずれか一項に記載の方法。 - a.2つ以上の異なるバキュロウイルス種で昆虫の幼虫を、同時接種、または最初の感染の徴候の発現に必要な時間より短い時間の遅延での順次接種により、重感染し、
b.多角体病による明らかな病気または死亡まで必要な条件で、前記接種された幼虫を飼育し、
c.前記幼虫から、ウイルス封入体(OB)を単離し、
d.同じビリオンに、異なるゲノムが存在することを確認することを含む、
請求項1〜8のいずれか一項に記載の混合ウイルスウイルス封入体(OB)の製造方法。 - シロイチモジヨトウ(Spodoptera exigua)多発性核多角体病ウイルス(SeMNPV)のバキュロウイルス変異体SeUS2−Aおよびツマジロクサヨトウ(Spodoptera frugiperda)多発性核多角体病ウイルス(SfMNPV)の変異体SfNIC−Cの混合ウイルス封入体(OB)の製造方法であって、
a.前記変異体の両方で、シロイチモジヨトウの幼虫を重感染し、
b.多角体病による明らかな病気または死亡まで、前記シロイチモジヨトウの幼虫を飼育し、
c.前記幼虫から、ウイルス封入体(OB)を単離し、
d.同じビリオンに、異なるゲノムが存在することを確認することを含む、方法。 - オートグラファカリフォルニカ(Autographa californica)多発性核多角体病ウイルス(AcMNPV)の変異体AcC6およびツマジロクサヨトウ多発性核多角体ウイルス(SfMNPV)の変異体SfNIC−Bの混合ウイルス封入体由来ビリオン(ODV)または混合ウイルス封入体(OB)の製造方法であって、
a.ツマジロクサヨトウの幼虫を前記変異体の両方で重感染し、
b.前記幼虫を、多角体病による明らかな病気または死亡まで飼育し、
c.前記幼虫から、ウイルス封入体(OB)を単離し、
d.同じビリオンに、異なるゲノムが存在することを確認することを含む、方法。 - 前記製造される混合ウイルス封入体(OB)は、水中で前記病気または死んだ幼虫をつぶし、得られた懸濁液をろ過し、ウイルス封入体(OB)を懸濁液から分離することを可能とし若しくは分離を引き起こし、沈殿、遠心分離または関連する技術により沈殿することを可能とし若しくは沈殿を引き起こし、上澄みからOBのペレットを分離することにより単離される、請求項1〜11のいずれか一項に係る方法。
- 遊離の前記封入体由来ビリオン(ODV)は、前記単離したウイルス封入体(OB)にアルカリ溶解を施すことにより得られる、請求項1〜12のいずれか一項に係る方法。
- 同じビリオンに共に包まれた少なくとも2つの異なるバキュロウイルス種からのゲノムを含む、遊離の混合ウイルス封入体由来ビリオン(ODV)。
- 前記ゲノムの少なくとも1つは、多発性核多角体病ウイルスのゲノムである、請求項14に記載の遊離の混合ウイルス封入体由来ビリオン(ODV)。
- 少なくとも、AcMNPVの変異体AcC6のゲノムコピーとSfMNPVの変異体SfNIC−Bのゲノムコピーとを含む、請求項15に記載の遊離の混合ウイルス封入体由来ビリオン(ODV)。
- 少なくともSeMNPVの変異体SeUS2−AのゲノムコピーとSfMNPVの変異体SfNIC−Cのゲノムコピーとを含む、請求項15に記載の遊離の混合ウイルス封入体由来ビリオン(ODV)。
- 請求項1〜12のいずれか一項に記載の方法によって得られた単離した混合ウイルス封入体(OB)にアルカリ溶解を施すことを含む、同じビリオンに少なくとも2つの異なるバキュロウイルス種からのゲノムを含む、遊離の混合ウイルス封入体由来ビリオン(ODV)の製造方法。
- 前記遊離の混合ウイルス封入体由来ビリオン(ODV)は、少なくともAcMNPVの変異体AcC6のゲノムコピーとSfMNPVの変異体SfNIC−Bのゲノムコピーとを含む、請求項18に記載の方法。
- 前記遊離の混合ウイルス封入体由来ビリオン(ODV)は、少なくともSeMNPVの変異体SeUS2−AのゲノムコピーとSfMNPVの変異体SfNIC−Cのゲノムコピーとを含む、請求項18に記載の方法。
- 同じビリオンに共に包まれた少なくとも2つの異なるバキュロウイルス種からのゲノムを含む、混合ウイルス封入体由来ビリオン(ODV)が少なくとも1つ閉じ込められている、単離された混合ウイルス封入体(OB)。
- 前記混合ウイルス封入体由来ビリオン(ODV)は、少なくとも、AcMNPVの変異体AcC6のゲノムコピーとSfMNPVの変異体SfNIC−Bのゲノムコピーとを含む、請求項21に記載の単離された混合ウイルス封入体(OB)。
- 前記混合ウイルス封入体由来ビリオン(ODV)は、少なくとも、SeMNPVの変異体SeUS2−AのゲノムコピーとSfMNPVの変異体SfNIC−Cのゲノムコピーとを含む、請求項21に記載の単離された混合ウイルス封入体(OB)。
- 請求項21〜23のいずれか一項に記載の混合ウイルス封入体(OB)、及び/又は
請求項14〜17のいずれか一項に記載の混合ウイルス封入体由来ビリオン(ODV)を含む、害虫の制御のための組成物。 - それぞれが単一ウイルス種のゲノムを含む複数のウイルス封入体(OB)を更に含む、請求項24に係る組成物。
- 2つ以上の害虫種の駆除に用いる、請求項24または25に記載の組成物。
- 害虫の制御のための、請求項21〜23のいずれか一項に記載の混合ウイルス封入体(OB)、請求項14〜17のいずれか一項に記載の混合ウイルス封入体由来ビリオン(ODV)および/または請求項24又は25に記載の組成物の使用。
- 2つ以上の害虫種の駆除に用いる、請求項27に記載の使用。
