Description
1抗原賦活化は、多くの資料で「エピトープ賦活化」とも称されている。さらに、抗原賦活化中に試料の加熱が含まれる場合、それは、熱誘導抗原賦活化または熱による抗原賦活化と称されることもある。
既存の組織処理装置は、技術の進歩というよりむしろ、ある一定の程度までスループットおよび再現性を向上させるために手動の方法を自動化および並列処理しているに過ぎないと言える。目下の問題の1つは、処理サイクルの所要時間が長いことである。一般的に、この方法は終夜行われ、処理、染色された組織は翌日にならないと得ることができない。このように所要時間が長く、外科的介入中に分析を完了することができないため、これは現在のIHC法の大きな障害となっている。しかし、介入中にIHCを完了することができれば、IHCの結果を直ちに使用することにより外科的処置プロトコルを微調整することができる。非常に重要な例の1つは、癌診断およびその後の治療工程である。患者が癌と診断された場合、通常の処置では、一般的に、疑わしい組織の組織生検を実施する。次いで、これらの試料のIHC分析を行い、癌の疑いが真であるかどうか調べる。答えがイエスの場合、2回目の手術を行い、身体から全ての腫瘍を除去するが、生きた癌細胞が1個でもあると増殖して再度腫瘍になり得るため、これは重要な工程である。残念ながら、現在まで、腫瘍を完全に除去することが立証された方法はない。従って、時折、患者は、残存し得るこれらの細胞を除去するために、追加の手術または化学療法を必要とすることがある。数えると、手術の回数は1回〜3回となり、それにより患者のリスク、費用および不安、ならびに、医師の時間および手術室の利用可能性などの医療資源の著しい損失が増加する。
術中の態様の他に、特定のアッセイを行う時に必要な免疫組織化学により得られるシグナルの程度を使用する定量的なバイオマーカー発現分析を必要とする事例を扱う場合、現在までどの技術を用いても、得られる結果の正確度は限られている。従来の方法では、このような半定量分析を必要とする事例の最大20%で不明瞭な結果が得られることがあり、免疫組織化学だけを使用して最終的な診断結果を得ることはできない。従って、現在の標準では、最終的な診断結果を得るために、その後にこれらの事例の遺伝分析(in situハイブリダイゼーション)を行うため、診断法にかなりの費用と時間(数日)が嵩む。
最新式の自動装置には、工程所要時間が長く正確度が限られているという本質的な問題に加えて、他の幾つかの欠点がある。最近の市販の自動IHCは、嵩張り、実験台に組み込まれて供給されるかまたは実験台上に載置される。従って、それらは持ち運び可能で手持ち式であるのとは程遠い。持ち運び可能であることは、例えば、術中動作の要件ではないが、これにより、検査室環境または電気がない遠隔地でも利用し易くなる。
現在まで、拡散時間の短縮および流体交換制御の改善により有利となり得る、従来のIHCの特定の態様を改良するためのマイクロ流体IHC法が幾つかあった。これらには全分析時間の短縮を目的とするものもあれば、比較的高い抗体希釈度で、空間的に離れた位置にある異なる標的バイオマーカーを探索するために複数の平行な小さいチャネルを使用して多重IHCを行うことを目的とするものもある。しかし、これらの研究のいずれにおいも、マイクロ流体法により、定量分析の正確度が向上し、このような分析によって得られる不明瞭な診断結果が減少することは示唆されなかった。
前述したように、上市されている処理装置でIHC工程時間が短いのは、「波動」システム(PCT/US2006/015020号明細書および国際公開第2006/116037号)だけである。この技術は、「波動」機構と称される現象に基づいており、隣接する2枚のスライドの「波状の」ヒンジ運動を採用し、その一方が組織薄片を有する(Celerus Diagnostics)。それは、凍結固定されたスライドの染色時間を15分に短縮することができ、従って、術中と称することができる。他方、これらの15分には、固定、観察および画像化時間は含まれず、決定を行うために、これらはさらに少なくとも約15分を要し得、それは術中の条件を超える。
標本が固定化されているスライドを受け入れる、縦穴をベースにするデバイスが文献に記載されている。Mcneelyら(PCT/US2002/07113号明細書および国際公開第2002/072264号)は、DNAマイクロアレイ処理用に製造されたこのようなデバイスを紹介した。IHCに必要とされるような面積の広いチャンバではなく、このDNAマイクロアレイ処理デバイスは、DNAマイクロアレイの要素が存在する小さい場所それぞれに試薬を送達するための複数の縦穴と網目状のマイクロ流体チャネルとを有する。組織または細胞単層の特定の箇所を染色するために非常に小さい場所(対角線で約100um)に縦方向のマイクロ流体穴が配置されている、「マイクロ流体プローブ」と称される類似のデバイスも開示され(A.Queval et.al.Lab Chip,vol:10,pp.326−334,2010ならびに特許文献PCT/IB2010/052018および国際公開第2010/128483号)、このプローブヘッドは空間的に移動させることができる。Delamarcheらによる別の特許(PCT/IB2003/005350および国際公開第2004/050246号)では、表面上で液体を流動させるデバイスが紹介されたが、ここでは縦穴およびスペーサを使用して表面にチャンバが形成されている。
組織切片が固定化されている単なる標準的な顕微鏡スライドである組織スライド(TS)をマイクロ流体チャネルに迅速に組み立てることは極めて重要であるが、その理由は全アッセイ時間が重要なパラメータとなるからである。迅速な組み立ての他に、TSしか変える必要がなく、他の態様(マイクロ流体回路)は変えないシステムが考えられる。図2は、(a)流入および流出用に組織チャンバの縁部に沿って、ならびに(b)流入用にチャンバの中心に、および流出用にチャンバの縁部に配置されているマイクロ流体アクセス穴を有するデバイスの断面図を示す。デバイスは、マイクロ流体最上層を使用して、TSに送達される流体の案内および事前調整を行うための横方向のマイクロ流体チャネルと、O−リングを使用してマイクロ流体チャネルデバイス部分にTSを封止することにより形成される薄いチャンバに到達するための縦方向のマイクロ流体アクセス穴とを有する。
本発明では、マイクロ流体デバイスは、より高い精密さおよび破壊圧力を達成するために、図6に示されるように、マイクロ流体溝の多段階深掘り反応性イオンエッチング(DRIE)を行った後、Parylene−Cが塗工されたPyrexウエハと結合させることにより製造される。1)2.5μmの湿潤酸化膜(wet oxide)を有する4インチのシリコンウエハを出発点とした。2)次いで、5μmのAZ9260フォトレジストをスピンコートし、DNCマスクを被せて露光し、現像してチャネルを形成した。下にある酸化物をRIE(Alcatel 601E)でエッチングした。3)レジストを剥離し、5μmのAZ9260フォトレジスト(MicroChemicals GmbH,Germany)およびチャンバマスクを使用して追加のリソグラフィー工程を実施した。リソグラフィー後、前面を2段階でDRIEエッチングする(Alcatel 601E)。これは、チャネルおよびチャンバを異なる高さに形成するために行う。まず、チャンバを深さ100μmにエッチングし、レジストを剥離した。4)レジストを剥離した後、第一段階でパターニングされた硬質マスクを介してチャネルをエッチングした。エッチング深さは、設計に応じて、50から200μmまで様々であった。5)後で、このウエハを、2μmのparyleneが塗工されたPyrexウエハに低応力parylene−C結合法で結合させる。この工程にアノード結合も使用できるが、これにより比較的高い応力および亀裂が生じ得ることに留意されたい。6)結合後、結合した積層体に、シリコン側から厚み8μmのAZ9260レジストを用いて追加のリソグラフィー工程を適用した。後で、チャンバに達するまでDRIEをもう一段階行うが、それは、O−リングを取り付けるための切り欠きを形成するのにも使用された。最後に、レジストを剥離する。ウエハをダイシングすることにより加工は完了する。図7(a)は、加工およびダイシング後のデバイスの最上層のマイクロ流体チャネルを示す。図7(b)は、デバイスの底部側にある、マイクロ流体アクセス穴とO−リングが配置されている切り欠きとを示す。
濃度応答時間および均一性のキャラクタリゼーション
オープンソースのImageJソフトウェアを使用して得られる画像から組織チャンバの強度プロファイルを解析することにより、濃度のキャラクタリゼーションを行った。分析を行うため、1秒当たりのフレーム数が5のビデオをグレースケールに変換する。まず、40μL/sの流量で長時間(数100分間)にわたりチャンバ全体での平均強度を算出することにより、最大濃度での強度(MxCI)を実験的に求めた。同様に、PBS緩衝液を流した後、最小濃度での強度(MnCI)を実験的に求めた。これらの2つの値を使用して、実験で記録された強度を正規化し、正規化された値を試薬濃度、Cとして使用するが、それは0〜1の範囲となる。後で、共に16秒の周期と50%のデューティーサイクルを有するが、180°位相シフトさせた矩形波の形態で適用される試薬および緩衝液を適用した。得られたビデオから、組織チャンバの異なる領域における濃度変化の応答時間、および平均挙動を調べた。生成した応答時間曲線を使用して、本デバイスの可能な時間性能を評価する。そのため、Cおよび1−Cの対数プロットに直線フィッティングを使用した。これらのフィッティングにより、所与の充填/洗浄純度に必要な洗浄および充填時間を算出するための指標である、1秒当たりのディケード(dec/s)に関する充填および洗浄性能が得られる。他方、組織染色面積のピクセルヒストグラムの標準偏差を完全な充填および洗浄の時点でのカメラピクセル深度で除することにより、組織チャンバ全体での濃度均一性を算出した。
マイクロ流体チャンバの時間応答について示したが、次にヒト乳癌組織上の診断バイオマーカーの分析について記載する。以前の分析に基づき、逐次的プロトコルを開発し、陽性の場合に、再現性よく高いシグナル対バックグラウンド(SBR)比を有する蛍光画像を生成する最適なインキュベーション時間を求める必要がある。この最適化試験では、同じ腫瘍摘出術試料から隣接して切除された強い発現を有する7つのHER2/neu陽性組織薄片を一次抗体および二次抗体の様々なインキュベーション時間(tinc)について使用して、インキュベーション条件を確立し、インキュベーション時間は対数的に変え、2n分(式中、n=−2、−1、0、1、2、3、4である)のインキュベーションを行って、得られるシグナルに対するインキュベーション時間の影響を15秒から16分まで評価した。これは、充填後、中心から組織薄片表面まで1分の拡散所要時間を示すデバイス設計に関連しており、上記の組のインキュベーション時間は、理論値を中心に対数的に分布した事例となる。
開発されたMTPシステムを実際の場で試験するために、病理学研究所のアーカイブから取り出された76の浸潤性乳管癌1組について一連の免疫組織化学的反応を行った。一次および二次抗体のインキュベーション時間を2分(n=1)にして、MTPでのHER2/neu免疫組織化学に最適な実験条件を確立した後、本プロトコルを76の浸潤性乳管癌に適用した。従来のIHCとMTP−IHCとの診断結果の比較を図11に示す。MTP−IHCでは、従来のIHCと比較して、不明瞭な結果が90%少なくなり、ISH増幅結果が正確に予測される。挿入された表は、試験した76例に関する、従来のIHCのスコアとMTP−IHCのスコアとの相互相関を示す。MTP−IHC法を使用した場合、それぞれスコア(0)および(+)の事例ならびにスコア(+++)の事例に関して、偽陽性または偽陰性結果は1つも得られなかった。さらに重要なことには、スコア(++)の不明瞭な事例の数は、27例から3例に顕著に減少した(ほぼ90%の減少)。従来のIHCで診断されたスコア(++)の不明瞭な事例24例は、MTP−IHCにより(0)/+または(+++)のいずれかに評価され、これらの24例ではそれぞれ、割り当ては遺伝子増幅状態に対応している。最初に(+++)の事例と診断された一例には、(++)のスコアが再割り当てされた。従って、従来のIHCの代わりに前述のMTP−IHCを使用すると、最終的な診断HER2/neu結果がさらにずっと正確なに予測される。
本発明の流体動作は、圧力駆動流の発生だけに限定されるものではない。本デバイスの進歩性(インキュベーション時間の同時短縮、プロトコル反応が速度論的状態に維持されること、得られる読み取りシグナルと標的抗原量が示された比例関係になること、および流体の迅速かつ均一な交換による高い均一性)はまた、流体の流れが、動電流または熱誘起流れを含むがこれに限定されるものではない他の作動機構により生じる場合にも適用される。例えば、動電流または熱誘起流れを採用する他の発明も存在する(国際公開第2011/102801号および欧州特許第1974814号明細書)が、これらの文献では、進歩性は、流れを生じさせる特定の電極の特定の配置(または設計、形状等)自体にある。このような以前の請求項は、従って、前記マイクロチャネルおよび前記組織チャンバ内での流体の流れを生じさせる他の方法と組み合わせても、本発明の使用を限定することはできない。

Claims (12)
- 生物および化学試料処理デバイスであって、
固定化された成分、生物試料または分子などの試料を含有する着脱可能なスライドを受け取るように構成されたマイクロ流体デバイスを具え、
前記マイクロ流体デバイスが、前記マイクロ流体デバイスの上部側と底部側との間に延在する複数のマイクロ流体アクセス穴のうちの第1の複数のマイクロ流体アクセス穴の配置および第2の複数のマイクロ流体アクセス穴の配置とを具え、前記マイクロ流体デバイスの前記底部側には封止リングが配置されており、前記第1の複数のマイクロ流体アクセス穴の配置は、試料処理の間、前記マイクロ流体デバイス及び前記スライドが前記封止リングと接触することにより形成される単一のマイクロ流体チャンバへ流体を注入するために構成され、前記第2の複数のマイクロ流体アクセス穴の配置は前記マイクロ流体チャンバからの流体の回収を行うために構成され、前記底部側が前記マイクロ流体チャンバに面し、前記第1の複数のマイクロ流体アクセス穴の配置および第2の複数のマイクロ流体アクセス穴の配置は前記マイクロ流体チャンバの縁部に沿って分布され、かつ前記マイクロ流体チャンバに均一に流体物質および試薬を移流輸送するように構成されており、
前記マイクロ流体デバイスがさらに、前記マイクロ流体チャンバの外部に形成されている入口ポート、出口ポート、およびマイクロ流体チャネルを具え、当該マイクロ流体チャネルは前記第1および第2の配置の複数のマイクロ流体アクセス穴に連結されて前記マイクロ流体デバイスの上部側に分布した網状チャネルを構成する、
ことを特徴とするデバイス。 - 請求項1に記載の生物および化学試料処理デバイスにおいて、前記マイクロ流体チャンバ内に流体の流れを生じさせるために入口と出口との間に圧力差を生じさせる、圧力駆動流発生手段を備えることを特徴とするデバイス。
- 請求項1に記載の生物および化学試料処理デバイスにおいて、前記マイクロ流体チャンバ、前記マイクロ流体チャネルおよび前記マイクロ流体アクセス穴の任意の2箇所以上の間に電位差を印加することにより、前記マイクロ流体チャンバ内に流体の流れを生じさせるための動電流発生手段を備えることを特徴とするデバイス。
- 請求項1に記載の生物および化学試料処理デバイスにおいて、前記マイクロ流体チャンバ内に流体の流れを生じさせるための、前記マイクロ流体チャンバ、前記マイクロ流体チャネルおよび前記マイクロ流体アクセス穴の任意の2箇所以上の間に温度差を発生させることにより流体の流れを生じさせることによる、熱誘起流れ発生手段を備えることを特徴とするデバイス。
- 請求項1乃至4の何れか1項に記載の生物および化学試料処理デバイスにおいて、前記マイクロ流体チャンバ、前記マイクロ流体チャネルおよび前記マイクロ流体アクセス穴に閉じ込められた前記流体の温度を調節および制御するための加熱素子および温度センサを備えることを特徴とするデバイス。
- 請求項1乃至5の何れか1項に記載の生物および化学試料処理デバイスにおいて、明視野を得るためにまたは蛍光検出を行うためにスライドガラス上に固定化された成分をデジタル画像化するため、シリコンマイクロエレクトロニクス技術で製造された光検出器および光源または光検出器および光源のアレイを組み込むことを特徴とするデバイス。
- 請求項1乃至6の何れか1項に記載の生物および化学試料処理デバイスにおいて、光検出器および光源または光検出器および光源のアレイの前に配置される、マイクロレンズアレイ、偏光フィルタおよび/または蛍光フィルタを備えることを特徴とするデバイス。
- 請求項1乃至7の何れか1項に記載の生物および化学試料処理デバイスにおいて、前記チャンバの高さが100μm未満であることを特徴とするデバイス。
- 請求項1乃至8の何れか1項に記載の生物および化学試料処理デバイスにおいて、前記第1のマイクロ流体アクセス穴の配置が前記マイクロ流体チャンバの一方の縁部に沿って配置され、前記第2のマイクロ流体アクセス穴の配置が前記マイクロ流体チャンバの反対側の縁部に沿って配置されていることを特徴とするデバイス。
- 請求項1に記載の生物および化学試料処理デバイスにおいて、前記第1のマイクロ流体アクセス穴の配置が前記マイクロ流体チャンバの一方の縁部に沿って分布され、前記第2のマイクロ流体アクセス穴の配置が前記マイクロ流体チャンバの反対側の縁部に沿って分布されていることを特徴とするデバイス。
- 請求項1乃至10の何れか1項に記載の生物および化学試料処理デバイスを使用して行われる生物および化学試料処理方法において、
−1つ以上の側に成分を有する前記スライドを手動でまたは自動的に得る工程と、
−前記スライドを前記マイクロ流体デバイスに手動でまたは自動的に組み込むことにより前記形成される前記マイクロ流体チャンバを確立する工程と、
−所定の時間中に、所定の温度で前記スライド上の成分を順次適切な流体物質または反応性物質に曝す工程と、
−検出を表示するため、所定の時間中に、所定の温度で前記スライド上の成分を順次適切な流体物質または反応性物質に曝す工程と、
を含むことを特徴とする方法。 - 所定の時間中に前記スライドの表面上の成分を適切な流体物質もしくは反応性物質または任意の一連の適切な流体反応性物質に曝すための、請求項1乃至10の何れか1項に記載のデバイスの使用において、次の操作:
i.組織化学法
ii.細胞化学法
iii.免疫組織化学法
iv.免疫細胞化学法
v.免疫組織蛍光法
vi.免疫細胞蛍光法
