Description
本発明は組換え融合タンパク質の分野、より具体的にはクモ糸タンパク質(スピドロイン)に由来する部分を含む新規融合タンパク質に関する。本発明は、スピドロインに由来する部分を含む組換え融合タンパク質を含むポリマーであるタンパク質構造を提供する方法を与える。また、有機ターゲットに結合するための新規なタンパク質構造を与える。
応用タンパク質化学において、活性、典型的には生物活性ペプチドを、活性に関連する部位、典型的には有機ターゲット、例えば、核酸、タンパク質、タンパク質の複合体、タンパク質及び/又は脂質及び/又は糖質及び/又は核酸などに対していかに調製又は提示するかは一般的な問題である。最も簡単な解決策は、生物活性ペプチド又はタンパク質の水溶液を提供することである。しかしながら、多くの適用が所望の目標を達成するために幾つかの更なる手段を要求する。例えば、ペプチド/タンパク質は、脂質混合物と会合するか又は支持構造体に化学的に固定化されてもよい。
本発明は、有機ターゲットとの選択的相互作用が可能な固体タンパク質構造が、反復構造単位として組換え融合タンパク質のポリマーの形態で調製できるとする見識に一般的に基づいている。融合タンパク質は、有機ターゲット(抗原/エピトープ)との選択的相互作用が可能である少なくとも一の免疫グロブリン(Ig)断片及びクモ糸タンパク質の少なくともC末端断片に対応する部分を含んでいる。驚くべきことに、クモ糸タンパク質に由来する部分は、構造的に再配置するために誘導することができ、その結果、ポリマー性の固体構造を形成するが、一方、例えばパラトープを含む免疫グロブリン(Ig)断片は、構造的に再配置されないが、その所望の構造と機能、即ち、有機ターゲットと選択的相互作用する能力を維持している。タンパク質構造は、化学的結合工程又は変性法による工程無しで得ることができ、これは、特に機能がその部分の2次構造に依存しているときに、その部分の維持された機能性を持つ融合タンパク質を得る手順を促進し、及びその可能性を向上する。これらの融合タンパク質ポリマーの形成は厳しく制御され、この見識は、更なる新規タンパク質構造、タンパク質構造を作り出す方法、及び様々な応用と方法におけるタンパク質構造の用途へと発展している。

アラニンリッチセグメント又はグリシンリッチセグメントは、N末端又はC末端のリンカーセグメントに隣接しているかどうかは重要ではないということになる。nは2から10の整数、好ましくは2から8、好ましくは4から8、より好まれるのは4から6であり、即ち、n=4、n=5、又はn=6である。
融合タンパク質の概念を証明するために、Rep4CTタンパク質(4回の内部反復とCT部分を持つREP部分)が単一鎖断片変異体(scFv)(B部分)と融合して産生された。scFvは、柔軟なポリペプチドリンカーを介して遺伝子的に一緒に連結されたVH及びVLからなる。これは、インタクトな抗原結合能力を持つ最小(27kDa)の実体である目標は、Rep4CTに融合したScFvからなる融合タンパク質から、繊維、薄膜及び膜などの構造体を生成し、及びScFvの抗原結合能力、並びにRep4CTの構造形成特性をなお保持することが可能であるかどうかを探索することであった。そのために、Rep4CTのN末端のScFv及びC末端のScFvからなる融合タンパク質をクローニングした。
His6ScFvRep4CT及びHis6Rep4CTScFv融合タンパク質(配列番号61−62)をコードする遺伝子(配列番号59−60)が構築された。ベクターは、カナマイシン(70μg/ml)を補充した寒天プレート上で増殖させたケモコンピテント(chemocompetent)大腸菌(E. coli)BL21(DE3)細胞に形質転換した。コロニーをその後採取し、正しい挿入のためにPCR検査をし、その後またDNA配列を確認するために配列決定した。
pT7His6ScFvRep4CT又はpT7His6ScFvRep4CTベクターを有する大腸菌BL21(DE3)細胞を、カナマイシン(70μg/ml)を補充したルリア−ベルターニ(Luria-Bertani)培地(全6リットル)で、30℃で1−1.5のOD600値まで増殖させ、次に、300μMのIPTG(イソプロピルβ−D−1−チオガラクトピラノシド)で発現を誘導し、約2時間、20℃でさらにインキュベートした。次に、細胞を4700rpmで20分間の遠心分離により回収し、得られた細胞ペレットを、20mMのトリス(pH8.0)中に溶解した。
20mMトリス(pH8.0)中に溶解させた細胞ペレットは、細菌細胞を溶解するために、リゾチーム及びDNアーゼIが補充され、細胞溶解物を30分間15000回転の遠心後に回収した。次に、回収した細胞溶解物を分割し、合計4つのキレートセファロースファーストフローZn2+カラムにロードし、タンパク質がHis6タグを介してカラムマトリックスに結合するように保った。洗浄後、結合したタンパク質を20mMのトリス/300mMのイミダゾール(pH8.0)で溶出した。次に、プールされた溶出液を一晩、20mMトリス(pH8.0)の5リットルに対して透析し、1mg/mlに濃縮し、最終的に繊維又は薄膜を形成させた。
二つの異なる方法が、ScFv単独あるいはRep4CTと融合して結合する抗原を検出するために使用された。A)Alexa647標識抗原の直接添加、洗浄、及びその後の蛍光測定。B)ビオチン化血清サンプルの添加、洗浄、Alexa647標識ストレプトアビジンの添加、洗浄、及びその後の蛍光測定。
ScFvRep4CT及びRep4CTScFvの両方とも発現、精製され、薄膜又は繊維に構築することができた。以下のすべての実験は、薄膜上で行われた。Alex647標識抗原の直接添加を用いた抗原結合の分析は、ScFv4repCT及び4RepCTScFv両方ともより強いスポットを与え、従ってScFv単独の場合よりも多くの抗原が結合した(図3A)。スポットの強度は、異なる検出強度で測定され、図3Bにプロットした。

Rep4CTタンパク質とのScFv融合体からスポットされた薄膜は、ScFvと比較して純粋な抗原よりも10倍超結合する。しかし、ビオチン化血清サンプルをフルオロフォア標識されたストレプトアビジンを用いて分析した場合、ScFvを含まないRep4CT薄膜に対して若干の非特異的な結合(約5倍低い)がある。

His6ScFvCT及びHis6CTScFv融合タンパク質(配列番号63−64)をコードする遺伝子が構築される。ベクターは、カナマイシン(70μg/ml)を補充した寒天プレート上で増殖させたケモコンピテント(chemocompetent)大腸菌(E. coli)BL21(DE3)細胞に形質転換する。コロニーをその後採取し、正しい挿入のためにPCR検査をし、その後またDNA配列を確認するために配列決定する。
pT7His6ScFvCT又はpT7His6ScFvCTベクターを有する大腸菌BL21(DE3)細胞を、カナマイシン(70μg/ml)を補充したルリア−ベルターニ(Luria-Bertani)培地(全6リットル)で、30℃で1−1.5のOD600値まで増殖させ、次に、300μMのIPTGで発現を誘導し、約2時間、20℃でさらにインキュベートする。次に、細胞を4700rpmで20分間の遠心分離により回収し、得られた細胞ペレットを、20mMトリス(pH8.0)中に溶解する。
20mMトリス(pH8.0)中に溶解させた細胞ペレットは、細菌細胞を溶解するために、リゾチーム及びDNアーゼIが補充され、細胞溶解物を30分間15000回転の遠心後に回収する。次に、回収した細胞溶解物を分割し、合計4つのキレートセファロースファーストフローZn2+カラムにロードし、タンパク質がHis6タグを介してカラムマトリックスに結合するように保つ。洗浄後、結合したタンパク質を20mMのトリス/300mMのイミダゾール(pH8.0)で溶出する。次に、プールされた溶出液を一晩、20mMトリス(pH8.0)の5リットルに対して透析し、1mg/mlに濃縮し、最終的に繊維又は薄膜を形成させる。

NTCT及びCTは、単一鎖断片変異体1(scFv1)と命名される操作された抗体断片との融合体として産生された。scFv1は、自己免疫疾患、全身性エリテマトーデス(SLE)に特異的な抗原を認識する、27kDaの一価、操作された抗体断片である。我々の目標は、NTCTに融合したscFv1タンパク質ドメイン(His6−scFv1−NTCTと表記;配列番号72)及びCTに融合したscFv1タンパク質ドメイン(His6−scFv1−CTと表記;配列番号74)からそれぞれなる融合タンパク質から、例えば、繊維及び薄膜などの構造体を生成し、及びscFv1タンパク質ドメインの抗原結合能力、並びにNTCT及びCTの構造形成特性をなお保持することが可能であるかどうかを探索することであった。そのために、NTCT及びCTのN末端に融合したscFv1ドメインからなる2つの融合タンパク質がクローニングされた。
His6−scFv1−CT融合タンパク質(配列番号74)をコードする遺伝子(配列番号73)は以下のように構築した。プライマーは、そのようなscFv1配列を含むベクターからドメインscFv1のPCR断片を生成するために設計された。また、プライマーは、制限エンドヌクレアーゼNdeI及びEcoRIの認識部位を含有していた。得られたPCR生成物は、その後、標的ベクター(カナマイシン耐性遺伝子を有するpAff8His6TrxHis6CTを示す)のように、制限エンドヌクレアーゼNdeI及びEcoRIで処理した。ターゲットベクターの制限切断に際し、His6TrxHis6部分が切断された。切断されたPCR断片と目標ベクターを、T4 DNAリガーゼを用いて互いに連結し、そして、得られた正しく連結されたベクター(pT7His6−scFv1−CT)は、カナマイシン(50μg/ml)を補充した寒天プレート上に増殖させたケモコンピテント(chemocompetent)大腸菌(E. coli)BL21(DE3)細胞に形質転換された。コロニーをその後採取し、正しい挿入のためにスクリーニングし、続いてまた、標的ベクター中に挿入されたscFv1のDNA配列を確認するために配列決定した。
pT7His6−scFv1−CTベクターを有する大腸菌BL21(DE3)細胞を、カナマイシン(50μg/ml)を補充したルリア−ベルターニ(Luria-Bertani)培地(全3リットル)で、30℃で1−1.5のOD600値まで増殖させ、次に、300μMのIPTGでpT7His6−scFv1−CTの発現を誘導し、約17時間、14℃でさらにインキュベートした。次に、細胞を4700rpmで20分間の遠心分離により回収し、得られた細胞ペレットを、20mMトリス(pH8.0)中に溶解した。
20mMのトリス(pH8.0)中に溶解した細胞ペレットは、細菌細胞を溶解するために、リゾチーム及びDNアーゼIを補充され、その後、NaClとイミダゾールをそれぞれ200mM及び10mMの最終濃度まで添加した。30分間の遠心分離(15000rpm)後、細胞溶解物を回収した。次に、回収した細胞溶解物をキレートセファロースファストフローZN2+カラムにロードし、His6タグを介してHis6−scFv1−CT(配列番号74)タンパク質がマトリックスに結合するように保った。洗浄後、結合したタンパク質を20mMのトリス/200mMのイミダゾール(pH8.0)/300mMのNaClで溶出した。溶出物は、A280の測定によると0.93mgのHis6−scFv1−CTタンパク質を含んでいた。次に、溶出したタンパク質を、一晩、20mMのトリス(pH8.0)を3リットルに対して透析し、その後、0.87mg/mlに濃縮し、0.348mgのHis6−scFv1−CT融合タンパク質(配列番号74)の最終量を得た。
His6−scFv1−CTの薄膜は、薄膜あたり5μMの可溶性融合タンパク質の1μlからマイクロアレイスライド(プラスチックMaxiSorp、Nunc)上にスポットされた。次いで、温度と湿度が調節された部屋で、薄膜を一晩凝固させた。5μMのタンパク質溶液の1μlからHis6−scFv1−NTCTの薄膜を成型するために同じ手順が続けられた。
純粋な抗体(scFv1、コントロール)及び糸が融合した抗体(scFv1−NTCT)は、透明及び黒のポリマーマキシソープマイクロアレイスライド(NUNC、25x76mm)上に5μMのタンパク質溶液の1μlを手動で添加することでマイクロアレイ形式にスポットされ、135ピコモルの純粋な抗体(scFv1)及が274ピコモルの融合した抗体(scFv1−NTCT)をそのスポットされた薄膜中に得た。薄膜形態中にタンパク質をスポットした後、薄膜を、温度と湿度が調節された部屋で一晩乾燥させた。次いで、アレイは、200μlのサンプルバッファー(PBS中に1%(w/v)の無脂肪粉乳及び1%(v/v)のTween−20を含む)を90分間適用することによりブロックし、その後、200〜300μlの洗浄緩衝液(PBS中に0.05%(v/v)のTween−20を含む)を適用することによって3回洗浄した。全てのインキュベーションは、室温で穏やかに撹拌して行った。次に、サンプルバッファー中で希釈されたビオチン化抗原サンプル(10nM)の100−200μlが適用され、1時間インキュベートした。アレイは、その後、200〜300μlの洗浄緩衝液を適用することによって3回洗浄し、結合した抗原を検出するために、試料緩衝液中に希釈されたアレクサ647標識ストレプトアビジン(1μg/ml)の100〜200μlを、アレイ上に適用し、1時間インキュベートした。最後に、アレイを200〜300μlの洗浄緩衝液で3回洗浄し、窒素気流下で乾燥した。アレイは、共焦点マイクロアレイ蛍光スキャナー(ScanArray Express、Perkin-Elmer Life & Analytical Sciences)を用いてスキャンした。スキャンアレイエクスプレス(ScanArray Express)ソフトウェアV2.0(Perkin-Elmer Life & Analytical Sciences)を用いて各スポットの強度を定量した。同じ分析手順を、His6−scFv1−CT融合タンパク質を分析するために行った。
Claims (16)
- B及びCT及び任意でREPの部分を含む組換え融合タンパク質であって、該組換え融合タンパク質はポリマーを形成することが可能であり;
Bは有機ターゲットとの選択的相互作用の能力を提供する、少なくとも一の免疫グロブリン断片を含み;
CTは70から120アミノ酸残基からなる部分で、クモ糸タンパク質のC末端断片に由来し、該部分が、配列番号7及び配列番号14〜44の何れか一のアミノ酸配列に少なくとも90%同一性を有し;及び
REPは70から300アミノ酸残基からなる部分で、クモ糸タンパク質の反復断片に由来し、REP部分は、L(AG) n L、L(AG) n AL、L(GA) n L、L(GA) n GLの群から選択され、
nは2から10の整数であり;
各個々のAセグメントは8から18アミノ酸残基のアミノ酸配列であり、アミノ酸残基の0から3はAlaでなく、残りのアミノ酸残基はAlaであり;
各個々のGセグメントは12から30アミノ酸残基のアミノ酸配列であり、アミノ酸残基の少なくとも40%はGlyであり;及び
各個々のLセグメントは、0から20アミノ酸残基のリンカーアミノ酸配列であり;及び
クモ糸タンパク質に由来した部分の一又は複数が、ポリマー形成の能力を提供する、組換え融合タンパク質。 - B部分の各免疫グロブリン断片は、免疫グロブリン可変領域から選択される、請求項1に記載の組換え融合タンパク質。
- B部分は、少なくとも一の重鎖可変領域(VH)及び少なくとも一の軽鎖可変領域(VL)を含む、請求項1又は2に記載の組換え融合タンパク質。
- B部分は、一本鎖可変断片(scFv断片)、抗原結合断片(Fab断片)、F(ab’)2断片、ドメイン抗体(dAb)及び単一ドメイン抗体(sdAB)からなる群から選択される、請求項1から3の何れか一項に記載の組換え融合タンパク質。
- B部分が、一本鎖可変断片(scFv)である、請求項4に記載の組換え融合タンパク質。
- B部分は、30−1000アミノ酸残基を含む、請求項1から5の何れか一項に記載の組換え融合タンパク質。
- B部分は、150−400アミノ酸残基を含む、請求項1から6の何れか一項に記載の組換え融合タンパク質。
- CT部分は、配列番号7に対して少なくとも90%、又は少なくとも95%同一性を有する、請求項1から7の何れか一項に記載の組換え融合タンパク質。
- 請求項1から8の何れか一項に記載の融合タンパク質をコードする単離された核酸。
- 有機ターゲットと選択的相互作用が可能なタンパク質構造であって、前記タンパク質構造が、請求項1から8の何れか一項に記載の組換え融合タンパク質を反復構造単位として含有するポリマーであり、B部分が有機ターゲットとの選択的相互作用の能力を提供する、タンパク質構造。
- 前記タンパク質構造が、繊維、薄膜、発泡体、網、メッシュ、球及びカプセルからなる群から選択される物理的形態である、請求項10に記載のタンパク質構造。
- サンプルからの有機ターゲットの分離における、請求項10又は11に記載のタンパク質構造の使用。
- 細胞の培養における、請求項10又は11に記載のタンパク質構造の使用。
- 有機ターゲットを含むサンプルを提供し;
親和性培地は有機ターゲットとの選択的相互作用が可能である、親和性培地を提供し;
前記親和性培地を、親和性培地と有機ターゲット間の結合を達成するために適した条件下で前記サンプルと接触させ;及び
非結合サンプルを除去する工程を含む、
サンプルからの有機ターゲットの分離のための方法であって、
親和性培地が、(i)請求項1から8の何れか一項に記載の融合タンパク質を含む有機ターゲットの固定化のための親和性培地であって、B部分が有機ターゲットとの選択的相互作用が可能である親和性培地、(ii)請求項10又は11に記載のタンパク質構造を含み、該タンパク質構造が組換え融合タンパク質を含むポリマーである(i)の親和性培地、(iii)有機ターゲットを更に含み、B部分が前記有機ターゲットとの選択的相互作用が可能であり該有機ターゲットに結合される(i)又は(ii)の親和性培地、及び(iv)有機ターゲットが第二有機ターゲットとの選択的相互作用が可能である(iii)の親和性培地、からなる群から選択される、方法。 - 前記親和性培地上で固定化ターゲットの存在を検出し、及び任意で定量化する工程を更に含む、請求項14に記載の方法。
- 前記親和性培地から有機ターゲットを放出し、及び回収する工程を更に含む、請求項14又は15に記載の方法。
