試験した個体がADである可能性を診断する方法であって、
(i)上記個体から得た末梢血試料中のγδT細胞のレベルと、少なくとも1種のMDSCのレベルとを測定すること;ならびに
(ii)(i)で測定したレベルと、年齢を一致させた対照群から得た血液試料中のγδT細胞のレベル範囲および上記少なくとも1種のMDSCのレベル範囲により示される基準レベルとを比較して、(i)の測定レベルを基準レベルに対する相対レベルとして表した分析結果を得ること
を含み、
上記個体のγδT細胞のレベルに上昇が見られ、かつ上記少なくとも1種のMDSCのレベルのいずれにも変化が見られない場合、上記個体がADである可能性が、年齢を一致させた対照群よりも高いことが示されることを特徴とする方法
に関する。
上記工程(i)において、上記血液試料中の少なくとも1種の炎症性単球のレベルを測定すること;および
上記工程(ii)において、上記少なくとも1種の炎症性単球のレベルを、年齢を一致させた対照群から得た血液試料中の上記少なくとも1種の炎症性単球のレベル範囲で示される基準レベルと比較すること
をさらに含み、
上記個体のγδT細胞のレベルに上昇が見られ、上記少なくとも1種のMDSCのレベルのいずれにも変化が見られず、かつ上記少なくとも1種の炎症性単球のうちの少なくとも1種においてレベルの上昇が見られる場合、上記個体がADである可能性が、年齢を一致させた対照群よりも高いことが示されることを特徴とする。
ADに罹患していると診断された患者におけるADの治療の有効性を測定する方法であって、
(i)2つの時点、すなわち上記治療の実施前または実施中である先の時点と、上記治療の実施中である後の時点とにおいて、上記患者から得た末梢血試料中のγδT細胞のレベルを測定すること;および
(ii)2つの時点で測定したγδT細胞レベルを比較すること
を含み、
それぞれの時点の測定レベルを、年齢を一致させた対照群から得た血液試料中のγδT細胞のレベル範囲で示される基準レベルに対する相対レベルとし、後の時点を先の時点と比較したときのγδT細胞レベルの低下が、上記治療の有効性と相関していることを特徴とする方法
に関する。
上記工程(i)において、上記2つの時点で上記血液試料中の少なくとも1種の炎症性単球のレベルを測定すること;および
上記工程(ii)において、上記2つの時点で測定した上記少なくとも1種の炎症性単球のレベルを比較すること

試験した個体がADである可能性を診断するため、またはADに罹患していると診断された患者におけるADの治療の有効性を測定するためのキットであって、
(i)γδT細胞と少なくとも1種のMDSCとを含む複数種の細胞;
(ii)上記各細胞種に対する抗体;
(iii)上記抗体を検出するための試薬;
(iv)年齢を一致させた対照群から得た血液試料中の上記各細胞種のレベル範囲で示される基準レベル;および
(v)使用説明書
を含むキット

患者:
患者群には、ADに罹患していることが臨床的に診断されており、インフォームドコンセントへの署名に合意した患者(男性および女性)が含まれていた。対照群には、ADの臨床症状を示さず、インフォームドコンセントへの署名に合意した男女のボランティアが含まれていた。研究を実施する前に、参加したアルツハイマー病患者群および対照群に簡易知能検査を行い、認知能力を検査した。20mL以下の血液試料を採取して実験室に送付し、HCV、HBSAG、HIV、HTLVまたはTPHAのウイルス反応が存在しないことを確認した後、様々な細胞成分について分析した。血液分析は採取から18〜24時間後に行った。
全血試料50μLを指定の各モノクローナル抗体5μLとともに4℃で45分間インキュベートした。FACSlyse(ベクトン・ディッキンソン社、カリフォルニア州サンノゼ)2mLを各チューブに加え、チューブを室温で12分間インキュベートし、PBS2mLで洗浄した。FACSCalibur(ベクトン・ディッキンソン社、カリフォルニア州サンノゼ)によって各試料から105のイベントを記録し、FCS Express V3ソフトウェアで分析した。

この研究では、生きている末梢血単核細胞(PBMC)全体における単一マーカーの分布を試験した。まず若年健常ボランティアから得た血液を使用して測定の正確性および頑強性を調査し、次いでアルツハイマー病患者と年齢を一致させた対照群とを比較した対照研究を行った。

(i)末梢血試料中のガンマ・デルタ(γδ)T細胞のレベルと、CD11b+/CD14-/CD15+細胞のレベルと、CD14+/CD16+細胞のレベルとを測定すること;ならびに
(ii)(i)で測定したレベルと、年齢を一致させた対照群から得た血液試料中のγδT細胞のレベル範囲、CD11b+/CD14-/CD15+細胞のレベル範囲およびCD14+/CD16+細胞のレベル範囲で示される基準レベルとを比較して、(i)の測定レベルを基準レベルに対する相対レベルとして表した分析結果を得ること
を含み、
γδT細胞レベルに上昇が見られ、CD11b+/CD14-/CD15+細胞レベルに変化が見られず、かつCD14+/CD16+細胞レベルに上昇が見られる場合、ADである可能性が、年齢を一致させた対照群よりも高いことが示されることを特徴とする方法。 - アルツハイマー病(AD)の治療の有効性を測定する方法であって、
(i)2つの時点、すなわち前記治療の実施前または実施中である先の時点と、前記治療の実施中である後の時点とにおいて、末梢血試料中のガンマ・デルタ(γδ)T細胞のレベルを測定すること;および
(ii)2つの時点で測定したγδT細胞レベルを比較すること
を含み、
それぞれの時点の測定レベルを、年齢を一致させた対照群から得た血液試料中のγδT細胞のレベル範囲で示される基準レベルに対する相対レベルとし、後の時点を先の時点と比較したときのγδT細胞レベルの低下が、前記治療の有効性と相関していることを特徴とする方法。 - 前記工程(i)において、前記2つの時点で前記血液試料中の少なくとも1種の炎症性単球のレベルを測定すること;および
前記工程(ii)において、前記2つの時点で測定した前記少なくとも1種の炎症性単球のレベルを比較すること
をさらに含み、
それぞれの時点の測定レベルを、年齢を一致させた対照群から得た血液試料中のγδT細胞のレベル範囲および前記少なくとも1種の炎症性単球のうちの少なくとも1種のレベル範囲で示される基準レベルに対する相対レベルとし、後の時点を先の時点と比較したときのγδT細胞のレベルおよび/または前記少なくとも1種の炎症性単球のうちの少なくとも1種のレベルの低下が、前記治療の有効性と相関していることを特徴とする請求項9に記載の方法。 - 前記少なくとも1種の炎症性単球がCD14+/CD16+細胞であることを特徴とする請求項10に記載の方法。
- 前記先の時点が前記治療の実施前または実施中であり、前記後の時点が、前記先の時点から1ヶ月後またはそれ以降であることを特徴とする請求項9〜11のいずれか一項に記載の方法。
- アルツハイマー病(AD)である可能性を評価するため、またはADの治療の有効性を測定するためのキットであって、
(i)ガンマ・デルタ(γδ)T細胞と少なくとも1種の骨髄由来抑制細胞(MDSC)とを含む複数種の細胞;
(ii)前記各細胞種に対する抗体;
(iii)前記抗体を検出するための試薬;
(iv)年齢を一致させた対照群から得た血液試料中の前記各細胞種のレベル範囲で示される基準レベル;および
(v)使用説明書
を含むキット。 - 前記複数種の細胞が少なくとも1種の炎症性単球をさらに含むことを特徴とする請求項13に記載のキット。
- 前記少なくとも1種の炎症性単球がCD14+/CD16+細胞であることを特徴とする請求項14に記載のキット。
