Description
図を参照しながら、本発明の第1実施形態に係る細胞培養装置10を備えた細胞培養システム100について説明する。本実施形態に係る細胞培養システム100は、主に多能性細胞を培養するものであるが、これに限らず、他の細胞を培養する装置として用いてもよい。なお、ここでいう多能性細胞とは、複数の種類の細胞に分化する能力を有する細胞を意味する。例えば、多能性細胞は、胚性幹細胞(ES細胞)、精子幹細胞(GS細胞)、胚性生殖細胞(EG細胞)、人工多能性幹細胞(iPS細胞)、培養線維芽細胞や骨髄幹細胞由来の多能性細胞(Muse細胞)、及び体性幹細胞を含むがこれらに限定されない。また、多能性細胞は、種々の生物に由来するものであってよい。好ましくは、ヒトを含む哺乳類動物由来の多能性細胞であり、より好ましくは、マウス由来の多能性細胞や霊長類由来の多能性細胞である。特に好ましくは、ヒト由来の多能性細胞である。
ここで、本実施形態に係る細胞培養装置10の効果を確かめるため、以下の試験を行った。
試験1:本実施形態に係る細胞培養装置10を用いて、細胞培養部12へ送液される培養液の流量を微小流量計で計測し、図10に示した。また、細胞培養装置10から圧力均一化機構18及び加圧機構22を取り外した比較例の細胞培養装置を用いて、細胞培養部12へ送液される培養液の流量を微量流量計で計測し、図9に示した。
試験2:本実施形態に係る細胞培養装置10を用いて、細胞培養部12へ送液される培養液の流量を0.3μl/min、0.5μl/min、0.75μl/minに設定し、それぞれの場合において、培養開始後1日目と3日目の培養室12Bに生着しているiPS細胞の個数を顕微鏡で数えて表1に記載した。また、上述した比較例の細胞培養装置を用いて同様の試験を実施した。なお、比較例の細胞培養装置における培養液の平均流量は0.5μl/minとした。また、何れも培養はCO2インキュベーター内で行い、培養液は、1.0×105細胞/mlのものを用いた。
次に、本発明の第2実施形態に係る細胞培養装置70について説明する。なお、第1実施形態と同様の構成については、同じ符号を付し、説明を省略する。

Claims (7)
- 細胞を培養する細胞培養部と、
培養液を貯蔵する貯蔵部と、
前記細胞培養部と前記貯蔵部とを接続する流路と、
前記流路に設けられ、前記貯蔵部から前記細胞培養部へ前記培養液を送液する送液手段と、
前記流路に設けられ、前記細胞培養部へ送液される前記培養液にかかる圧力の変動を抑制する圧力均一化機構と、
前記細胞培養部の流出口側の前記流路に設けられ、前記培養液へ所定の圧力を加える加圧機構と、
を有し、
前記圧力均一化機構は、
前記培養液が充填される流体室と、
前記流体室に形成され、前記流路と接続される流入口及び流出口と、
前記流体室の内壁の一部を構成すると共に、前記流体室の圧力の変動に応じて撓む可撓性膜と、を有し、
前記加圧機構は、
前記流路に接続され、前記流路より断面積が小さい加圧部と、
前記加圧部の壁面の一部を構成し、前記加圧部へ流入した前記培養液の圧力により弾性変形する弾性膜と、
を有する細胞培養装置。 - 前記圧力均一化機構が有する前記流入口及び前記流出口は、前記流体室の対向する端部に形成され、
前記流体室は、前記端部から中央部に向かって徐々に広幅に形成されている請求項1に記載の細胞培養装置。 - 前記流体室は、平面視で、角が丸められたひし形状である請求項2に記載の細胞培養装置。
- 前記細胞培養部の流入口側の前記流路には、気泡を除去する気泡除去手段が設けられて
いる請求項1〜3の何れか1項に記載の細胞培養装置。 - 前記圧力均一化機構を複数備えた請求項1〜4の何れか1項に記載の細胞培養装置。
- 請求項1〜5の何れか1項に記載の細胞培養装置と、
前記細胞培養装置を格納する恒温器と、
を有する細胞培養システム。 - 請求項1〜5の何れか1項に記載の細胞培養装置を用いて細胞を培養する細胞培養方法であって、
前記細胞培養部、前記貯蔵部、及び前記圧力均一化機構の少なくとも1つに細胞を添加する工程と、
前記加圧機構で前記流路中の前記培養液を加圧しつつ、前記送液手段で前記流路中の前記培養液を循環させる工程と、
を有する細胞培養方法。
