Description
本願は、米国仮特許出願第61/466,188号(出願日:2011年3月22日)に基づき優先権を主張する。当該仮特許出願の内容は全て、参照により本願に組み込まれる。
1D CSIについての基本的な数式を考慮する。
CSI実験の目的は、M個の異なる位相エンコードで収集したM個の公知の信号(S)から、M個の未知のスペクトルを数4のマトリクスρで再構築することである。しかし、スカウトMRIから、ρが含むのはC(<M)個のMRS対象コンパートメント、および、各コンパートメントの空間的位置のみであることが分かる。理論上、ρの解を一義的に求め、C個のスペクトルを再構築するために必要なのは、C個の位相エンコードステップで得られるC個の測定結果のみである。
要約すると、SLAMの実施形態は、以下に説明するステップ1からステップ5で実行される。
1.MRIを取得して、コンパートメント数に関する先行情報を抽出(C<<M)、SLAM再構築のために各コンパートメントの空間的位置を取得
2.M´(≧C)個の位相エンコードステップを選択する。理論的には、任意に選択することができるが、選択結果によっては、
3.選択したM´個のエンコード傾斜を適用して、M´個の信号を収集
4.MRIで特定される各コンパートメントの空間的位置に基づいてbマトリクスを決定
5.M個からC個へと次元を減らして、
M´個の位相エンコードステップの選択は、数4において整数であるkに対応する元々の基本セットのM個のCSIステップに限定する必要はない。M個の位相エンコード傾斜は、再構築の所望の特性を最適化するように選択され得る。例えば、以下で説明するように、傾斜は、SNRを最大限まで大きくするように、および/または、コンパートメント間の信号の汚染を最小限に抑えるように、および/または、不均一な信号ソースによるコンパートメント内のエラーを最小限に抑えるように、最適化される。これは実質的に、その他の全ての実験パラメータを変えることなく、数4のCSIにおいて分数のkを可能にすることを含む。SLAMとは違い、この分数SLAM方法は、fSLAMと呼ばれるが、スキャナ側での傾斜の最適化および事前設定が必要になる。
SNRを最大限まで大きくするべく、数19を変形して、ノイズ項である
これまでのところ、各コンパートメントが均一であると仮定している。しかし、コンパートメントの平均から逸脱するCSI基本セットにおけるスペクトルは、再構築後、各コンパートメントの間、および、各コンパートメント内を伝搬する信号を発生させる可能性がある。漏れを抑制するべくM´個の位相エンコードステップのfSLAM実験を最適化することを目的として、数8を書き直して、元々のρマトリクスを平均部分および不均一部分に分割する。
二番目の部分は以下の式で表される。
fSLAM実験においてi番目のコンパートメント内の不均一性に起因するエラーを最小限に抑えるべく、i番目のコンパートメント自体の内側から発生する係数の二乗和を最小限に抑える。
要約すると、fSLAM実験は、ステップ2の位相エンコード傾斜が数27のSNRコスト関数または数47のエラーコスト関数を最小限に抑えることによって取得されることを除いて、SLAMプロトコル(図2)と同じステップ1−ステップ5を用いて実施される。概して、最適化方法が異なると、得られる位相エンコード傾斜のセットも異なる。SNRおよび最小エラーの両方について最適化された傾斜のセットが求められている場合、数27および数47のコスト関数の合計の最小化は、スケーリングが異なるので、利用できない。これに代えて、fSLAMのコスト関数のSLAMのコスト関数に対する比率の加重和を最小限に抑えれば、十分である。重み付けは、用途およびエラー許容範囲に応じて選択される。ステップ2の位相エンコード傾斜は通常、分数である。
<コンピュータシミュレーション>
SLAMを人間の心臓の31P MRSに適用した場合の精度について調べるべく、コンピュータシミュレーションを実行した。われわれの研究室では[17−21]1D CSIが主であった。3つのコンパートメントを仮定した。心臓、胸部の骨角筋、および、その他である。実際には、「その他」コンパートメントが必要なのは、コンパートメントが割り当てられていない指定コンパートメントの外部で生成した任意の信号は胸部および心臓で止まり、大きさによってはエラーを発生させるためである。胸部スペクトルおよび心臓スペクトルは、図3の(A)および(C)に示す。16ボクセルで1cmの分解能の1D CSIモデルを用いて、所定のコンパートメント分布を持つこれらのスペクトルから信号が生成される。
(i)胸部および心臓は静止している(部分体積エラーなし)
(ii)胸部および心臓は共に±2mm移動(部分体積エラーあり)胸部は、絶対に心臓と重ならないように、制限された。SLAMおよびSLIMの両方について、中央のk空間から4つのCSI位相エンコードを用いて、シミュレーションを行った。SLIM再構築は、先述したように[3]、位相エンコード係数を、心臓、胸部およびその他の3つのコンパートメントのモデルにわたって積分して、4×3のGマトリクスを生成する[3]することによって、実行された。再構築された信号と、真の結果またはCSI結果との平均(±SD)パーセントエラーは、1000回実行した場合について算出された。
31P 1D CSI、SLAMおよびfSLAMを、フィリップス(Philips)社の3T Achieva MRI/MRSシステムにおいて、ファントム、人間の脚、および、人間の心臓に対して実施した。ファントムの検査は、直径が14cmの1つのループ状の送受信コイルで実行され、人間に対する検査は、上述したように[22]、直径が17cm/11cmの二重ループ送信コイルおよび直径が8cmの単一ループ受信31Pコイルのセットを利用した。人間に対する検査は全て、ジョンズ・ホプキンス・メディシン・インスティシューショナル・レビュー・ボード(ジョンズ・ホプキンス医療機関審査委員会)によって承認されており、全ての被験者はインフォームドコンセントを得た。各コンパートメントを構成している体積要素の全てからのそれぞれのCSIスペクトルは、SLAMおよびfSLAMを用いて再構築された均等な体積からのスペクトルと全て比較するべく、収集後に加算された。
<コンピュータシミュレーション>
図3の(A)から(D)は、元々の16個の位相エンコードステップのうち中央の(k空間)3つの位相エンコードステップのみを用いて再構成された胸部および心臓のSLAMスペクトルが、不均一性またはノイズが無い場合には、元々のシミュレーションされたスペクトルと見分けがつかないことを示す。ノイズおよび不均一性を加えた場合に多岐にわたる胸筋および心臓のコンパートメント分布について再構成されたSLAMスペクトルへの影響は、図4の(A)から(E)に示す濃度のばらつきおよび感度のばらつきの両方のモデルについてのモンテカルロシミュレーションによって図示している。これらによると、再構成の精度は、エラーの平均によって示されているように、全ての胸部/心臓の人体構造上の組み合わせについて10%未満であることが分かる。予想されるかもしれないが、濃度が高くなるほど、または、コンパートメントサイズが大きくなるほど、エラーSDは小さくなる。心臓について、シミュレーションからは、心臓コンパートメントの実効的な範囲が最も小さくなる場合にエラーが最も高くなると予想される。
b:胸部/心臓信号比は4で胸部の不均一性は±15%(合計30%)の場合について算出
CSIおよびSLAMを用いて再構築された2つのディスク状の無機リン酸塩のファントムからのスペクトルを図7に示す。H3PO4は、2.9ppmにおいて31Pの単一ピークがあり、H3PO2の共鳴は、水素との異核結合であるので、13.5ppmを中心とした三重項である(結合定数、545Hz)。スキャン時間を4分の1に縮小できるが、2つのディスクからのSLAMスペクトルは、コンパートメントの体積が同じ場合の加算CSIスペクトルと非常に類似しており、シミュレーションに見られる漏れは無視してよい(図4)。
PRESS[26]、STEAM[27]またはISIS[28]等の単一ボクセル方法は、単一のコンパートメントのMRSを実行する場合の局所化方法としては良い選択であるが、複数のコンパートメントのMRSについてスキャン時間を一定とした場合にはSNRが最適な値とならない。また、緩和効果(T1およびT2の両方)および動きに対する感度は、特に31PのMRSにおいて[1、26]、定量化に関して大きな問題を呈する。CSIは、全サンプル、全タイミングからの全信号を収集する単純なパルス−収集実験であって、現時点において定量的MRSに対して最もクリーンな方式であり、SNR効率も最も高い可能性がある。しかし、感度を持つ全体積または検出コイルの全FOVをエンコードするために必要な最小スキャン時間によって制限がある。これによって、例えばB0磁界強度が高くなることによって得られるSNRゲインがスキャン時間の短縮に直接つながる可能性が制限され得る。

2Dおよび3DのSLAM実験はそれぞれ、フィリップス(Philips)社の3T MRIシステムで人間の脳(1H)およびリン酸のファントム(31P)に対して実施された。コンパートメントは、頭皮、脳、側脳室およびバックグラウンド(2D)、および、H3PO4、H3PO2のディスク状のファントムおよびバックグラウンド(3D)であった。追加で、胸部、心臓およびバックグラウンド(1D)のコンパートメントを含む31Pの心臓の検査も図示している。SLAMスペクトルは、中央の4個(1D)、7×7個(2D)、2×4×2個(3D)の位相エンコードで再構築され、16個(1D)、32×25個(2D)、および、10×20×8個(3D)の位相エンコードを含む全データセットから取得されたコンパートメントの平均CSIスペクトルと比較した。心臓の検査は、ECGゲーティングされ(TR=15.7s)、脳の検査は、脂質/水分が抑制され(TE/TR=0.144/3s)、ファントムの検査は、TR=0.72sであった。

Claims (22)
- 空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法であって、
被験体の磁気共鳴画像を受信する段階と、
少なくとも1つの対象コンパートメントを含む前記被験体において磁気共鳴スペクトロスコピー信号を生成する複数のC個のコンパートメントを特定する段階と、
少なくとも1つの空間的次元において、前記被験体の前記磁気共鳴画像を前記複数のC個のコンパートメントに分割する段階と、
前記少なくとも1つの空間的次元において適用される、M´≧Cである複数のM´個の位相エンコードを適用することによって、前記複数のC個のコンパートメントから磁気共鳴スペクトロスコピー信号を収集する段階と、
前記少なくとも1つの対象コンパートメントから、空間的に局所化された磁気共鳴化学シフトスペクトルを算出する段階と、
前記少なくとも1つの対象コンパートメントから、磁気共鳴化学シフトスペクトルの空間的平均に略等しい空間的に局所化された磁気共鳴スペクトルをレンダリングする段階と
を備え、
前記少なくとも1つの対象コンパートメントから、前記空間的に局所化された磁気共鳴化学シフトスペクトルを前記算出する段階は、線形代数法を利用する、空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。 - 前記被験体の前記磁気共鳴画像を受信する段階、前記複数のC個のコンパートメントを特定する段階、および、前記磁気共鳴画像を前記複数のC個のコンパートメントに分割する段階はそれぞれ、前記被験体から前記磁気共鳴スペクトロスコピー信号を収集する段階の前に実行され、
前記複数のM´個の位相エンコードはさらに、前記少なくとも1つの対象コンパートメントの空間的選択または信号ノイズ比の少なくとも一方を最適化するように選択される、請求項1に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。 - 前記複数のM´個の位相エンコードは、前記被験体の前記磁気共鳴画像のk空間の中央部分から選択される、請求項1又は2に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。
- 前記複数のM´個の位相エンコードのうち少なくとも1つは、ゼロの位相エンコードであり、前記複数のM´個の位相エンコードのうち別の少なくとも1つは、ゼロでない最小位相エンコードの整数倍である、請求項3に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。
- 前記複数のM´個の位相エンコードのうち少なくとも1つは、ゼロでない最小位相エンコードの非整数倍である、請求項2から4のいずれか一項に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。
- 前記複数のM´個の位相エンコードの選択はさらに、分割後の前記磁気共鳴画像に基づいて行われ、
前記少なくとも1つの対象コンパートメントにおける信号ノイズ比の最適化は、前記被験体の前記磁気共鳴画像のk空間の中央部分から位相エンコードを選択することを含み、
前記少なくとも1つの対象コンパートメントの空間的選択の最適化は、前記対象コンパートメントの外部から発生する磁気共鳴スペクトロスコピー信号、または、前記対象コンパートメント内で発生する非均一な磁気共鳴スペクトロスコピー信号分布から発生するエラー含有信号のうち少なくとも一方を最小限に抑えることを含む、請求項5に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。 - 前記複数のM´個の位相エンコードの選択は、前記少なくとも1つの対象コンパートメントにおいて前記空間的選択および前記信号ノイズ比の両方を最適化するメトリックに基づいて行われる、請求項6に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。
- 前記少なくとも1つの空間的次元は、2つの空間的次元または3つの空間的次元のうち一方であり、
前記複数のM´個の位相エンコードは、前記2つの空間的次元または前記3つの空間的次元のそれぞれにおいて適用される2つまたは3つのサブセットの位相エンコードから構成される、請求項1から7のいずれか一項に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。 - 前記2つまたは3つのサブセットの位相エンコードの各サブセットにおける位相エンコードの数は、対応する前記空間的次元において分割されている、磁気共鳴スペクトロスコピー信号を生成する前記被験体のコンパートメントの数以上である、請求項8に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。
- 前記少なくとも1つの対象コンパートメントは、複数の対象コンパートメントである、請求項1から9のいずれか一項に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。
- 前記方法によると、少なくとも1つの対象コンパートメントにおいて、前記少なくとも1つの対象コンパートメントに寄与する全ての体積要素からの信号が加算される従来の化学シフトイメージングプロセスに比べて、信号ノイズ比において改善が得られ、
前記改善は、前記対象コンパートメントの体積と、前記化学シフトイメージングプロセスの体積分解能との比率の二乗根に略等しい、請求項1から10のいずれか一項に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。 - 前記方法は、M個の位相エンコードステップを用いて実行される従来の化学シフトイメージングプロセスよりも、M/M´に略等しい倍数で算出される短い収集時間で、少なくとも1つの対象コンパートメントから空間的に局所化されたスペクトルを提供する、請求項1から11のいずれか一項に記載の空間的に局所化された磁気共鳴スペクトロスコピーを実行する方法。
- 磁気共鳴イメージングスキャナと、
前記磁気共鳴イメージングスキャナと通信して、被験体の磁気共鳴スペクトロスコピー信号を受信するデータ処理システムと
を備え、
前記データ処理システムは、
前記被験体の磁気共鳴画像を受信し、
前記磁気共鳴画像を表示して、少なくとも1つの対象コンパートメントを含む、前記被験体において磁気共鳴スペクトロスコピー信号を生成する複数のC個のコンパートメントを特定できるようにし、
少なくとも1つの空間的次元において、前記被験体の前記磁気共鳴画像を前記複数のC個のコンパートメントに分割し、
前記少なくとも1つの空間的次元において複数のM´個の位相エンコードを適用することによって、前記磁気共鳴画像に対応する前記被験体から磁気共鳴スペクトロスコピー信号を受信し、
前記少なくとも1つの対象コンパートメントから、空間的に局所化された磁気共鳴化学シフトスペクトルを算出し、
前記少なくとも1つの対象コンパートメントから、前記磁気共鳴化学シフトスペクトルの空間的平均に略等しい空間的に局所化された磁気共鳴スペクトルを提供し、
M´≧Cであり、
前記少なくとも1つの対象コンパートメントから、前記空間的に局所化された磁気共鳴化学シフトスペクトルを前記算出することは、線形代数方法を利用する、磁気共鳴局所化スペクトロスコピーイメージングシステム。 - 前記磁気共鳴イメージングスキャナはさらに、
前記被験体から前記磁気共鳴スペクトロスコピー信号を受信する前に、前記複数のC個のコンパートメントの分割および特定を可能とし、
前記磁気共鳴イメージングスキャナはさらに、
前記少なくとも1つの対象コンパートメントにおいて、空間的選択または信号ノイズ比の少なくとも一方について、前記複数のM´個の位相エンコードを最適化する、請求項13に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。 - 前記複数のM´の位相エンコードは、前記被験体の前記磁気共鳴画像の前記少なくとも1つの空間的次元に対応するk空間の中央部分から得られる、請求項13又は14に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。
- 前記複数のM´個の位相エンコードの少なくとも1つは、ゼロでない最小位相エンコードの整数倍である、請求項15に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。
- 前記複数のM´個の位相エンコードの少なくとも1つは、ゼロでない最小位相エンコードの非整数倍である、請求項14から16のいずれか一項に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。
- 分割後の前記磁気共鳴画像から決定される複数のM´個の位相エンコードが提供され、
前記データ処理システムはさらに、
前記被験体の前記磁気共鳴画像のk空間の中央部分からの位相エンコードの選択を含む、前記少なくとも1つの対象コンパートメントにおける信号ノイズ比の最適化、および、
前記対象コンパートメントの外部から発生する磁気共鳴スペクトロスコピー信号の少なくとも1つを最小限に抑えることによる、または、前記対象コンパートメント内で発生する非均一な磁気共鳴スペクトロスコピー信号分布から発生するエラー含有信号を最小限に抑えることによる、前記少なくとも1つの対象コンパートメントの空間的選択の最適化
のうち少なくとも1つを実行する、請求項17に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。 - 前記複数のM´個の位相エンコードは、前記信号ノイズ比および前記空間的選択の両方を最適化するメトリックを前記少なくとも1つの対象コンパートメントに含めることによって提供される、請求項18に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。
- 前記少なくとも1つの空間的次元は、2つの空間的次元または3つの空間的次元のうち一方であり、
前記複数のM´個の位相エンコードは、前記2つの空間的次元または前記3つの空間的次元においてそれぞれ適用される2つまたは3つのサブセットの位相エンコードを含む、請求項13から19のいずれか一項に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。 - 前記2つまたは3つのサブセットの位相エンコードの各サブセットにおける位相エンコードの数は、対応する前記空間的次元において分割されている、磁気共鳴スペクトロスコピー信号を生成する前記被験体のコンパートメントの数以上である、請求項20に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。
- 前記少なくとも1つの対象コンパートメントは、複数の対象コンパートメントである、請求項13から21のいずれか一項に記載の磁気共鳴局所化スペクトロスコピーイメージングシステム。
