Description
本発明は、ちしゃとうの全草または一部を有効成分とする抗アレルギー剤に関する。また前記抗アレルギー剤を含んでなる、食品または医薬組成物に関する。
近年、アレルギー疾患は日本のみならず、世界的にも急増している。特に、アレルギー性鼻炎、花粉症、アレルギー性皮膚炎および気管支喘息などのアレルギーは、I型アレルギー(即時型アレルギー)と呼ばれており、その罹患率は年々増加し、大きな社会問題となっている。
本発明の抗アレルギー剤は、ちしゃとうの全草または一部を有効成分とする。
ちしゃとう(Lactuca sativa var.asparagina)は、中国原産の植物であり、その葉および肥大している茎の部分を食用としている。
本発明のちしゃとうの全草または一部を有効成分とする抗アレルギー剤は、後述する実施例に記載の通り、抗アレルギー作用を有しており、特に、アレルギー性鼻炎、花粉症、およびアレルギー性皮膚炎などのI型アレルギー疾患の発症を予防し、症状の改善および治療効果が期待できる。したがって、本発明の抗アレルギー剤は、アレルギー疾患の予防、改善および治療に用いることができる。
本発明の抗アレルギー剤が効果を有するアレルギー疾患は、好ましくはI型アレルギー疾患である。具体的には、アレルギー性鼻炎、花粉症、アレルギー性皮膚炎、アレルギー性結膜炎、アナフィラキシー反応、および気管支喘息などが挙げられ、即時型アレルギー反応を示すものをいう。
本発明によれば、本発明による抗アレルギー剤を含んでなる食品組成物が提供される。このような食品組成物は、例えば、本発明による血圧上昇抑制剤を、食品の材料成分に添加することを含んでなる製造方法によって製造することができる。
本発明によれば、本発明による抗アレルギー剤を含んでなる医薬が提供される。
ちしゃとうの抗アレルギー作用について、アレルギー性鼻炎モデルマウス(アレルゲン感作マウス)を用いて評価した。
ちしゃとう凍結乾燥物の製造
生のちしゃとうを、水で洗浄し、土などの異物を取り除いた。茎部と葉部とに分け、茎部は外皮をピーラー(厚み5〜8mm)で剥き翡翠色の可食部分を原料とし、葉部はそのまま原料とした。茎部、葉部ともに1〜5%の食塩を加えた80〜100℃のお湯で、1〜5分ブランチングを行った。ブランチングは、ちしゃとうに含まれる酵素活性を抑え、菌数を減少させるために行った。ブランチング後、直ちに茎部、葉部ともに冷水に入れ、30℃以下に急冷した。遠心脱水機(NE3−26、株式会社アサヒ製作所製)を用いて1400rpmで3〜5分遠心し、またはザルにあげ静置状態にして、茎部、葉部の水気を除去した。ちしゃとうを、トレーに並べた(トレー張り)。緩慢に凍結、または急速凍結を行い、中心温度−15〜−30℃にした。TFD−LF5S(東洋技研株式会社製)を用いて凍結乾燥した。
ちしゃとう茎部の凍結乾燥物500mgを、乳鉢を用いて磨砕し、粉末化した。得られた粉末を、蒸留水で0.5%に調整したカルボキシメチルセルロース(和光純薬工業株式会社より入手)水溶液10mlに懸濁し、ちしゃとう茎部懸濁液(50mg/ml)を得た。
さらに、ちしゃとう葉部の凍結乾燥物についても、同様に処理し、ちしゃとう葉部懸濁液(50mg/ml)を得た。
アレルギー性鼻炎モデルの作成
5週齢の雌性BALB/c系マウス(日本エスエルシー株式会社より入手)を、室温24±2℃、湿度55±15%、明暗サイクル12時間(明期8:00〜20:00)の条件下にある動物室で飼育した。マウスは、飼育用ケージ(幅22cm、奥行き32cm、高さ14cm)で、1ケージ7匹飼いとし、1群7匹で下記の試験に供した。標準餌(MF飼料、オリエンタル酵母工業株式会社より入手)と水は自由に摂取させた。マウスの体重は、平均17〜18gであった。
さらに、初回感作14日目以降は、局所感作として、1日1回、卵白アルブミン1.2mgを生理食塩液4μlに溶解した卵白アルブミン生理食塩液(300mg/ml)を、マウスの両側鼻腔内に、マイクロピペットを用いて、4μlずつ点鼻投与した。
ちしゃとう懸濁液を投与した試験群(ちしゃとう茎部投与群、ちしゃとう葉部投与群)と、対照群とで、アレルギー性鼻炎症状を測定した。
ちしゃとう投与群は、感作0日目から、1日1回、初回感作14日目以降の場合は卵白アルブミン生理食塩溶液の点鼻1時間前に、それぞれ、ちしゃとう懸濁液10ml/kg(ちしゃとう乾燥物500mg/kg)の用量で、マウスに、それぞれの懸濁液を、経口投与した(図1参照)。
卵白アルブミン生理食塩溶液を、マウスの両側鼻腔内にマイクロピペットで4μlずつ点鼻投与した直後から、マウスのアレルギー性鼻炎症状を測定した。マウスのアレルギー性鼻炎症状として、抗原溶液の点鼻直後から誘発されるくしゃみ反応と、鼻掻き行動の回数とを、20分間測定した。
ちしゃとうの抗アレルギー作用として、ヒスタミンを接種したマウス、すなわちヒスタミン誘発マウスを用いて、ヒスタミン誘発鼻炎症状抑制作用、ヒスタミン誘発皮膚掻痒行動抑制作用、およびヒスタミン誘発皮膚血管透過性亢進反応抑制作用について評価した。
ヒスタミン誘発鼻炎マウスを用いて、ちしゃとう懸濁液を投与した試験群(ちしゃとう茎部投与群、ちしゃとう葉部投与群)、および抗ヒスタミン薬であるセチリジン投与群と、対照群とで、ヒスタミン誘発鼻炎症状を測定した。
にいれて、10分間馴化させた。1群9匹で下記の試験に供した。
ヒスタミン誘発皮膚掻痒行動マウスを用いて、ちしゃとう懸濁液を投与した試験群(ちしゃとう茎部投与群、ちしゃとう葉部投与群)、および抗ヒスタミン薬であるセチリジン投与群と、対照群とで、ヒスタミン誘発皮膚掻痒行動を測定した。
ヒスタミン誘発皮膚血管透過性亢進反応マウスを用いて、ちしゃとう懸濁液を投与した試験群(ちしゃとう茎部投与群、ちしゃとう葉部投与群)、および抗ヒスタミン薬であるセチリジン投与群と、対照群とで、ヒスタミン誘発皮膚血管透過性亢進反応を測定した。
Claims (5)
- ちしゃとうの全草または一部を有効成分とする、抗アレルギー剤(飲食品を除く)。
- I型アレルギーを予防、改善または治療するためのものである、請求項1に記載の抗アレルギー剤。
- I型アレルギーが、アレルギー性鼻炎、花粉症またはアレルギー性皮膚炎である、請求項2に記載のアレルギー剤。
- 請求項1〜3のいずれか一項に記載の抗アレルギー剤を含んでなる、医薬組成物。
- ちしゃとうの全草または一部を有効成分として摂取させることを含んでなる、アレルギー反応を抑える方法(ヒトへの医療行為を除く)。
