Description
The present invention relates to a device such as a detect.
近年、低濃度の試料分子を検出する高感度分光技術の1つとして、SPR(Surface Plasmon Resonance:表面プラズモン共鳴)、特にLSPR(Localized Surface Plasmon Resonance:局在表面プラズモン共鳴)の利用したSERS(Surface Enhanced Raman Scattering:表面増強ラマン散乱)分光が注目されている(特許文献1,2)。
In recent years, as one of highly sensitive spectroscopic techniques for detecting a low concentration of sample molecules, SPR (Surface Plasmon Resonance), particularly SERS (Surface) using LSPR (Localized Surface Plasmon Resonance) is used. Enhanced Raman Scattering (surface enhanced Raman scattering) spectroscopy has attracted attention (
This type of surface plasmon resonance sensor is obtained by fixing metal fine particles such as gold and silver on a substrate. A detection apparatus equipped with this sensor irradiates sample molecules adsorbed on metal nanoparticles of a surface plasmon resonance sensor with light, and detects enhanced Raman scattered light.
Here, as one of uses of the surface plasmon resonance sensor, for example, monitoring of environmental pollutants is cited. In order to monitor contaminants, they must be detected in real time.
However, the above-described detection apparatus can detect whether or not the sample molecule adsorbed on the metal nanoparticles of the surface plasmon resonance sensor is a contaminant, but it can be detected only once. In order to continuously detect with the same apparatus, the sample molecules adsorbed on the metal nanoparticles must be desorbed before the next detection.
An object of some aspects of the present invention is to provide an optical device capable of reliably desorbing sample molecules adsorbed on metal nanoparticles and capable of real-time detection, and a detection apparatus using the same. To do.

(1) One aspect of the present invention is
A base material having at least one surface as a conductor surface;
A metal nanostructure having a convex part of 1 to 1000 nm formed on the conductor surface;
A power supply terminal connected to the conductor surface;
It is related with the optical device which has.
According to one embodiment of the present invention, since the metal nanostructure having a convex portion of 1 to 1000 nm is provided, an enhanced electric field is formed around the convex portion of the metal nanostructure. Accordingly, this optical device can be used as a sensor that increases the signal intensity of light reflecting the detection target substance that is enhanced by the enhanced electric field, for example, Raman scattered light. Since the optical device includes a conductor surface connected to the metal nanostructure and the power supply terminal, the metal nanostructure can be energized from the external power supply through the power supply terminal and the conductor surface. Thereby, Joule heat is generated from the metal nanostructure. By this Joule heat, energy can be given to the fluid sample adsorbed by the optical device to desorb the fluid sample. At times other than the time of detachment, the power supply from the power source can be cut off.
(2) In one mode of the present invention, it can further have a measurement terminal connected to the conductor surface.
Here, when molecules of the fluid sample are adsorbed on the optical device, the movement of electrons moving on the surface of the conductor to which power is supplied is hindered, and the electrical resistance of the optical device increases. On the contrary, when the adsorbed molecule is detached from the optical device, the factor that obstructs the movement of electrons moving on the surface of the conductor to be supplied with power decreases, and the electrical resistance of the optical device decreases. Using this phenomenon, the electrical resistance measured through the measurement terminal can be used to detect the adsorption or desorption of molecules in the fluid sample. The power supply terminal may be used as a measurement terminal. The measurement terminal may be any terminal as long as the electrical resistance can be detected as a result, and measuring the current flowing through the optical device under a constant voltage is synonymous with measuring the electrical resistance.

It is related with the detection apparatus which has.
According to another aspect of the present invention, a detection apparatus can be configured using the optical device according to one aspect of the present invention. In particular, energy can be applied to and desorbed from the fluid sample adsorbed by the optical device, so that termination control of the desorption mode can be performed accurately.
(4) In another aspect of the invention, the optical device further includes a measurement terminal connected to the conductor surface, and the detection device is connected to the measurement terminal and controls one of current and voltage. And a measuring unit that measures the electrical resistance value of the optical device. In this detection device, the measured electrical resistance value can be used to detect the adsorption or desorption of molecules in the fluid sample. In addition, when the measurement unit controls one of the current and voltage, in the desorption mode, one of the current and voltage is increased to generate Joule heat, and in the adsorption mode, one of the current and voltage is decreased to generate Joule heat. The resistance value can be measured while suppressing.
(5) In another aspect of the present invention, based on the output from the measurement unit, the first mode including a period detected by the light detection unit, and the fluid sample adsorbed on the metal nanostructure are removed. It is possible to further include a control unit that controls execution of the second mode. Since the electrical resistance value of the optical device reflects the adsorption or desorption of molecules in the fluid sample, the control unit performs each of the first mode involving the adsorption of molecules and the second mode involving the desorption of molecules. The situation can be acquired based on the output from the measurement unit.
(6) In another aspect of the invention, the control unit can switch from the second mode to the first mode based on a decrease in the electrical resistance value of the optical device. This is because an increase in the electrical resistance value of the optical device reflects the desorption of molecules from the optical device.
(7) In another aspect of the invention, the control unit can start and control light irradiation from the light source in the first mode based on an increase in the electrical resistance value of the optical device. Since the increase in the electrical resistance value of the optical device reflects the adsorption of molecules to the optical device, light irradiation can be started at the timing when the molecules are adsorbed to the optical device.
(8) In another aspect of the present invention, the optical device allows light from the light source to be incident on the metal nanostructure side and reflect the light reflecting the fluid sample on the surface of the conductor. Since the surface of the conductor is difficult to transmit light, light from the light source is incident on the metal nanostructure side, and the surface of the conductor can also be used as a light reflecting surface reflecting the fluid sample.
Hereinafter, preferred embodiments of the present invention will be described in detail. The present embodiment described below does not unduly limit the contents of the present invention described in the claims, and all the configurations described in the present embodiment are indispensable as means for solving the present invention. Not necessarily.
1. Basic Configuration of Detection Device FIG. 1 shows a configuration example of a detection device according to this embodiment. In FIG. 1, the
In the chamber 120, the
The
The
In the present embodiment, in the first mode, the fluid sample sucked at the flow velocity V1 can be adsorbed to the
In the second mode, the flow velocity V2 is set higher than the flow velocity V1 in the first mode (adsorption mode or detection mode). Therefore, in the second mode, desorption of the fluid sample adsorbed on the
As described above, when the first mode and the second mode are alternately performed, the fluid sample once adsorbed to the
Switching between the first and second modes can be performed based on the output of the

3. Principle of optical detection and optical device structure FIGS. 5A to 5C are explanatory diagrams of the principle of detection of Raman scattered light as an example of a light detection principle reflecting a fluid sample. As shown in FIG. 5A, incident light (frequency ν) is irradiated to the
FIG. 5B is an enlarged view of the
As the second structure on the plurality of
As shown in FIGS. 5B and 5C, in the
5A to 5C, surface enhanced Raman scattering (SERS) occurs when the
The phenomenon of “adsorption” of the

The energy required to desorb the DMS molecule from the Ag surface is, for example, 21.5 kcal / mol (= 90 kJ / mol) although it depends on the Ag state (measured value). The kinetic desorption characteristics of a molecule are represented by the following formula.
Consider that power is applied to the
And the fluctuation | variation of resistance value is monitored by making it constant with the current value 0.206A. Due to the detachment, the resistance value becomes small as indicated by an arrow A1 in FIG. In FIG. 9, the horizontal axis represents the coverage of the
4.2. Use in the first mode (adsorption / detection mode) In the first mode, power from the
The reason will be described qualitatively with reference to FIGS. FIG. 10A shows electrons (current) 2 </ b> A flowing through the conductor surface (Ag thin film) 221 in a state where the fluid sample is not adsorbed on the
On the other hand, FIG. 10B shows electrons (current) 2A flowing through the conductor surface (Ag thin film) 221 in a state where the fluid sample, for example, pyridine molecules as the
Next, the resistance value fluctuation amount ΔR is quantitatively expressed by Equation (6) (reference: quoted from the lecture book by Toshiyuki Totani, the National Institute of Informatics Electronic Library, Hokkaido University).
In the period in which ΔR / R is less than, for example, 3.5% (threshold value) in FIG. 9, the
When ΔR / R exceeds 3.5% (threshold value) in FIG. 9, the
5. Control Method of Light Source and Measuring Unit FIGS. 13 and 14 show laser intensity (light intensity) control from the

The
