本特許出願は、2010年12月2日に出願された米国仮特許出願第61/419,218号の利益を主張し、当該出願は、その全体において本明細書において特に参考として組み込まれる。
分子6−シクロヘキシル−1−ヒドロキシ−4−メチルピリジン−2(1H)−オンは、シクロピロクス(Ciclopirox)としても公知であり、市販の局所用抗真菌剤である。シクロピロクスは、表在性真菌症および癜風(Tinea versicolor)を処置するために用いられてきた。しかし、シクロピロクスは、水性の液体中で非常に低い可溶性を有し、これが、水性の媒体中での使用および対象への投与を限定する。ヒトのシステムは高度に水性であるため、シクロピロクスは、水溶性の欠如に起因してバイオアベイラビリティーが不十分である。しばしば、シクロピロクスは、シクロピロクスオラミンとして塩形態において用いられるが、なお低い水溶性を有する。したがって、バイオアベイラビリティーを改善するために、改善された水溶性のためにシクロピロクスを設計することは有益であろう。

一態様において、シクロピロクス−POMプロドラッグは、医薬組成物中に含まれていてもよい。医薬組成物は、静脈内、経口または経皮などの投与の経路に好適となるように処方することができる。
一般的に、シクロピロクスは、シクロピロクス−POMプロドラッグを精製するための本明細書において記載される反応プロトコルを通して得ることができ、次いで反応させることができる。また、本明細書において記載される合成プロトコルに従って、シクロピロクス誘導体を得、反応させることができ、ここで、シクロピロクス誘導体試薬は、対応するシクロピロクス−POM誘導体プロドラッグをもたらす。一部の場合において、シクロピロクスまたはその誘導体は、オラミン塩などの塩において得ることができる。したがって、反応スキームは、シクロピロクスを、POMプロドラッグ部分との抱合の前に脱塩することを含んでもよい。
本明細書において記載されるシクロピロクス−POMプロドラッグ、(((6−シクロヘキシル−4−メチルピリジン−1(2H)−イル)オキシ)メチルホスファートの2ナトリウム塩、MW361)を研究し、シクロピロクス遊離塩(MW207)およびシクロピロクスオラミン(MW268)と比較した。これらの3つの実体についてシクロピロクスの等用量を、以下のように投与した:シクロピロクスオラミンは2.0mg/mLで、シクロピロクス−POMプロドラッグは2.69mg/mL。(ng/mL)における血漿シクロピロクス濃度を、マウスモデルにおいて、10mg/kgのIVボーラスでのシクロピロクス−POMプロドラッグのボーラス投与の後で決定した。データを収集した時点を、表1において、5、15、30、60、90、120、180および240分として、データと共に示す。さらなる試料を、360、480、720および1440分において収集したが、濃度は、10ng/mLのBQLより下に低下し、PKデータ分析において含めない。プロドラッグのためのIVボーラス処方は、25mMのリン酸緩衝液中のものであった。
Claims (19)
- 式1の構造:
R1は、メチルであり;
R2〜R12は、各々独立して、水素、ハロゲン、ヒドロキシル、1〜6個の炭素原子を含むアルコキシ、1〜6個の炭素原子を含む直鎖脂肪族、1〜6個の炭素原子を含む分枝状脂肪族、1〜6個の炭素原子を含む不飽和脂肪族、1〜6個の炭素原子を含む脂肪族アミン、の1または2以上であり;
R13〜R14は、各々独立して、陽イオン、ナトリウムイオン、水素、1〜6個の炭素原子を含む直鎖脂肪族、1〜6個の炭素原子を含む分枝状脂肪族、1〜6個の炭素原子を含む飽和脂肪族、1〜6個の炭素原子を含む不飽和脂肪族、アリール置換基を有するアルキル基(アルキル基が、1〜6個の炭素原子を含む分岐状または非分岐状の飽和炭化水素基であり、アリール置換基が、単一の芳香族環、または共に融合しているか、直接的に架橋されているか、または間接的に架橋されている複数の芳香族環を含む芳香族置換基である)、アミン、1〜6個の炭素原子を含む脂肪族アミン、の1または2以上であり;
nは、1であり;ならびに
mは、1である、

6−シクロヘキシル−1−ヒドロキシ−4−メチルピリジン−2(1H)−オンを提供すること;
ジ−tert−ブチル(クロロメチル)ホスファートまたはジベンジル(クロロメチル)ホスファートから選択されるホスホリルオキシメチル(POM)試薬を提供すること;および
請求項1の式1を有する化合物を提供するために、前記6−シクロヘキシル−1−ヒドロキシ−4−メチルピリジン−2(1H)−オンを、前記ホスホリルオキシメチル(POM)試薬と反応させること
を含む、前記方法。 - 式10の構造
ホスホリルオキシメチル(POM)の保護基を除去することによって、リン上のOH基を残すこと;および
塩を形成させるためにNa + を導入すること
を含む、請求項16の方法。 - ホスホリルオキシメチル(POM)試薬を、ジ−tert−ブチル(クロロメチル)ホスファートとなるよう合成することを含み、前記合成することが、テトラ−メチルアンモニウムジ−tert−ホスファートをヨードクロロメタンと反応させることを含む、請求項16の方法。
- ジベンジル(クロロメチル)ホスファートを、ホスホリルオキシメチル(POM)試薬として合成することを含み、前記合成することが、銀ジベンジルホスファートをヨードクロロメタンと反応させることを含む、請求項16の方法。
