Description
ヒトDCとマウスDCとの間には密接な関係がある(O’Keeffe et al.2003)。表現型CD11cloCD11b−CD45RAhiを伴うマウス血液細胞は、形態および機能が、ヒトpDCと極めて類似している。ヒト血液では、刺激に応じて大量のインターフェロンを産生できるCD11c−IL3R+CD45RA+pDCを見出すことができる。
[1]免疫療法剤を調製するための方法であって、(a)CD11b−/loおよびCD172+(Sirpα)である骨髄由来の非形質細胞様樹状細胞(非pDC)を提供するステップと、(b)1つ以上の抗原をステップ(a)の非pDCに負荷するように、該非pDCをエクスビボで修飾するステップと、を含む、方法。

したがって、本発明のmiDCは、抗原を負荷した、または抗原をパルスしたmiDCを包含する。故に、これらの抗原をパルスしたmiDCまたは抗原を負荷したmiDCは、抗原提示miDCである。本発明は、そのような抗原提示miDC(APmiDC)を提供する。本明細書で使用される抗原を負荷したまたは抗原をパルスしたmiDCは、抗原に曝露した本発明のmiDCを含む。本発明のmiDCは、当技術分野で知られている多数の手法によって抗原を負荷することができる。一実施形態において、miDCは、miDCが抗原を取り込むこと、および抗原の処理を開始することを可能にするのに十分な時間にわたって、培養物中で抗原およびmiDCをインキュベート(「パルス」)することによって、可溶性抗原を負荷することができる。例えば、miDCは、37℃で約1〜2時間、抗原をパルスしてもよい。例えば、Celluzi et al.,1996,J.Exp.Med.183:283−7、Hu et al.,1996,Cancer Res.56:2479−2483を参照されたい。概して、本発明によるAPmiDCは、例えば、抗原の存在下での培養中に、インビトロで抗原を負荷した状態になり得る。APmiDCはまた、抗原への曝露によってインビボで負荷してもよい。本発明による「抗原を負荷したAPmiDC」は、「抗原を負荷した抗原提示細胞(APC)」を調製する、以下の2つの既知の従来の方法に従って、調製されてもよい。すなわち、(1)抗原ペプチドとして知られるペプチド小断片を、APCの外側に直接「パルス」すること、または(2)APCを、APCによって次いで摂取されるタンパク質全体またはタンパク質粒子とともにインキュベートすること、である。これらのタンパク質は、APCによってペプチド小断片に消化され、APC表面に運搬されて、提示される。

1.マウス
メスのC57BL/6Jマウスを、Harlan Winkelmann GmbH,Borchen,Germanyから購入し、6〜10週齢で使用した。同種異系刺激性混合白血球応答(allo−MLR)で使用したT細胞は、Balb/cマウス由来のものであり、Harlan Winkelmann GmbHからも購入した。ユビキチンプロモータ(C57BL/6−Tg(UBC−GFP)30Scha/J、以下、UBC−GFPマウスと称する)の下で緑色蛍光タンパク質(GFP)を発現するマウスは、Charles River Laboratories(Sulzfeld,Germany)から購入し、ハウス内で飼育した。C57BL/6−Tg(TcraTcrb)1100Mjb/j(OTI)マウス、および破壊したNFkb1遺伝子(B6;129P−Nfkb1tm1Bal/J)を収容しているマウスは、最初にJackson Laboratoriesから購入し、ハウス内で飼育した。FLKOマウスは、McKenna et al.2000によって説明されており、Institute of Labortierkunde(University of Zurich)で飼育した。vavプロモータの下で発現したヒトBcl−2導入遺伝子を発現するマウスは、以前に説明されており(Ogilvy et al.1999)、G.Hacker,Institute for Medical Microbiology,Immunology and Hygiene,Technical University of Munichから入手した。
組み換え(rec)FLAGタグを付したマウス(mu)FLを、CHO細胞中で発現させて、前に説明したように精製した(O’Keeffe et al.2002b)。recmuGM−CSFおよびrecmuM−CSFは、Tebu−Bio(Frankfurt、Germany)から入手した。CpGモチーフ(CpG2216およびCpG1668)を含有するオリゴヌクレオチドは、公表された配列(Spies et al.2003)に従って、TIB MOLBIOL(Berlin、Germany)によって合成した。イミキモド(R837)およびパルミトイル−3−システイン−セリン−リシン−4(Pam−3−Cys)は、InvivoGen(San Diego,USA)から購入した。ポリ(シチジル−イノシン)酸(ポリI:C)およびリポ多糖類(LPS)は、Sigma−Aldrich(Taufkirchen,Germany)から購入した。
マウスの大腿骨および脛骨を取り除き、2%のFCSを含むRPMI−1640(Gibco/BRL(マウスの浸透性に調整する)を充填し、21gの針を取り付けた20mLの注射器で、細胞を骨からフラッシングした。細胞は、注射器によって繰り返してフラッシングすることによって再懸濁させた。低密度細胞の調製のために、細胞を、遠心分離によってペレット化し、Nycodenz(登録商標)1.077A(Progen Biotechnik,Heidelberg,Germany、マウスの浸透性308mOsmに調整する)中に再懸濁させた。抗CD49b mAbも枯渇カクテルに添加したこと、および抗Thy1.1を省略したことを除いて、密度分離、抗体枯渇カクテルとのインキュベーション、および磁気ビーズ枯渇は、本質的に、前に説明したもの(O’Keeffe et al.2002a)である。ハイブリドーマは、その上清を枯渇カクテルで使用し、Ken Shortman教授(WEHI,Australia)より快く提供していただいた。BM miDCおよびpDCを選別するために、枯渇ステップの後に、細胞を、CD11c(PE−Cy7またはFITC)、CD45RA−PE、およびCD11b(APCまたはPacific Blue)でルーチン的に染色した。CD24−FITCが染色剤に含まれる場合もあった。細胞は、pDC(CD11cintCD45RAhiCD11b−)またはmiDC(CD11cintCD45RA−CD11b−HSAint)として選別した。
CpGモチーフ(CpG2216およびCpG1668)を含有するオリゴヌクレオチドを、公表された配列に従って、TIB MOLBIOL(Berlin、Germany)によって合成した。Spies B,et al.(2003) Journal of Immunology 171(11):5908−5912。イミキモド(R837)およびパルミトイル−3−システイン−セリン−リシン−4(Pam−3−Cys)は、InvivoGen(San Diego,USA)から購入した。レシキモド(R848)は、Alexis Biochemicals(Axxora Deutschland GmbH,Lorrach,Germany)から購入した。ポリ(シチジル−イノシン)酸(ポリI:C)およびリポ多糖類(LPS)は、Sigma−Aldrich(Taufkirchen,Germany)から購入した。
樹状細胞の刺激は、50μL以下の容積のための丸底プレートまたはV底プレートにおいて0.25〜0.5×1.0×106個の細胞で行った。細胞は、添加した刺激物の有無に関わらず、完全培地(10%のFCS、50μMのベータメルカプトエタノール、100IU/mLのペニシリン/ストレプトマイシンを補充した、RPMI−1640倍地)中で、18〜48時間刺激した。使用した刺激物は、10ng/mLのGM−CSF、1μg/mLのPam−3−Cys、100μg/mLのポリ(I:C)、1μg/mLのLPS、1μg/mLのR837、0.5μMのCpG2216、または0.5μMのCpG1668であった。
T細胞またはmiDCを、製造業者の説明書に従って、(CellTrace CFSE Cell Proliferation Kit,Molecular Probes (Invitrogen GmbH,Karlsruhe,Germany)を使用して、最終濃度の2μMのCFSEで標識した。
DCサブセットを、BMおよび脾臓から精製して、FACS−ARIAを使用して95%を超える純度に選別した。Balb/cマウスからのT細胞を、皮下および腸間膜リンパ節から精製した。精製したT細胞を、CellTrace CFSE Cell Proliferation Kit(Molecular Probes,Invitrogen GmbH,Karlsruhe,Germany)を使用して、2μMのCFSEで標識した。3つの50,000個のT細胞を、媒体中のDCサブセットとともに、またはGM−CSF、LPS、またはCpG2216を含む刺激物の添加とともにインキュベートした。4日後、T細胞分裂(CFSE蛍光の消失)の程度を確認するために、FACSによって、37℃で、反復試料を分析した。分裂したT細胞の絶対数を数えることができるように、定量に使用されるビーズのアリコートも各ウエルに添加した。
前述のMLRに対して交差提示アッセイを同様に行い、それによって、OTI遺伝子導入マウス由来のT細胞をBalb/c T細胞の代わりに利用し、DCは、最初に、37℃で1時間、オバルブミンとともにインキュベートし、洗浄し、次いで、T細胞に添加した。
BMのpDCおよびmiDCを、UBC−GFPマウスから精製して選別し、MTPBS中で洗浄し、C57BL/6マウスの尾静脈に注射した。マウス1匹あたり100μLのMTPBS中の0.6×106個のpDCまたは0.3×106個のmiDCを、1つの細胞群あたり3匹のマウスに注射した。3匹のマウスには、MTPBSだけを受容させた。移入後約90時間に、レシピエントマウスを安楽死させ、BMおよび脾臓DCを調製し、細胞を受容した各マウスを、別々にしておいた。ドナー細胞は、GFP(FITC)hiおよびPIneg細胞上でゲートすることによって選択した。
BMにおけるpDCの除去をもたらす条件は、CpG−ODN刺激に応答して、全てのINF−αの産生を除去しないことが以前に示されている(Hochrein et al.2004)。Liuおよび同僚は、まだ完全なpDC表現型を有していないヒトの初期pre−PDCが、高レベルのIFN−Iを産生する能力も有することを以前に公表している(Blom et al.2000)。最初にpDCを除去した場合に、BM以外のいかなる他の器官もCpG−ODNに対していかなるINF−αを産生する能力も保持しないことが分かり、pDC前駆体は、マウスBM細胞のCD45R/CD45RAの枯渇後に残った細胞であり得ると考えられた。
低密度BM細胞を、次いで、精製して、CD45RAおよびCD11cで染色した(図2A)。粗製細胞集団を、pDC、CD11c+推定のBMのcDC、および「残り」の3つに選別した。これらの3つの集団のそれぞれを、CpG−2216で一晩刺激し、上清を、IFN−α活性について試験した。pDC以外に、CD11c+CD45RA−細胞集団も、IFN−αを産生した。CD11c+細胞集団は、異種のものであり、CD3、CD19、CD49b、およびNK1.1を発現する少数の細胞を含有していた。細胞選別は、ジャンクチャネルにおいて、これらの表面マーカーに対する抗体を含めて繰り返した。CD11c+CD3−CD19−CD49b−Ly6G−NK1.1−細胞は、CpG−2216に応答して、高レベルのIFN−αの発現を保持した。CD49b+CD11c+細胞を含む、残りのCD11c+細胞は、CpG−2216に応答して、IFN−αを産生しなかった。
CD11c+CD45RA−CD24intCD11b−細胞またはBMpDCを、CpG2216、GM−CSF、または培地で一晩刺激し、それらの形態を比較した。培地において、CD11c+CD45RA−CD24intCD11b−細胞およびpDCは、同様に振る舞い、いずれの集団においても、いかなる典型的なDC形態の徴候も示さなかった。しかしながら、驚くべきことに、CD11c+CD45RA−CD24intCD11b−細胞は、pDCとは異なって、GM−CSFにおいて、明らかに成熟していた(図2E)。さらに、この細胞は、CpG−2216に非常に良好に応答し、pDC培養物中に形成される小さい細胞塊とは異なり、典型的な細胞質突起を伴う大きい細胞塊を形成した。
IFN−αの産生および表面活性化を含む、CpG2216に対するmiDCの応答は、pDCに類似しており、これは、この細胞が、pDCの分化状態または異なるサブセットであった可能性を示唆した。しかしながら、LPSおよびGM−CSFに対するmiDCの応答は、miDCがpDCとは根本的に異なることを示唆した。種々のウイルスによるmiDCの活性化は、これらの2つの細胞型間の差をさらに明らかにした。HSVは、TLR9およびTLR9非依存性経路を介してBMのpDCを活性化することが知られており、miDCの表面活性化およびIFN−αの産生の強力な誘導原でもあった(図5)。miDCは、HSVに応答して、実質的なレベルのIFN−αを産生したが、それでもこれは、pDCによる産生の約50%であった。HSVは、CpG−2216のように、miDCからの高いIL−6の産生も誘導し、それは、pDCによる産生の約10倍であり、また、脾臓cDCによる産生の2倍以上でもあった。MVAは、miDC上でのCD86の上方調節、および、pDCによる産生の約1/3である、低レベルのIFN−αの産生を誘導した。IFN−αは、脾臓cDC由来の上清では検出されなかったことに留意されたい。
miDCは、CpG−2216に応答して、BMのpDCよりもはるかに優れた細胞表面活性化を明らかに示した(図4)。特に、CpG2216−ODNは、pDCからの検出可能なIL−6の産生をほとんど誘導しないが、全く対照的に、CpG2216−ODNは、miDCからの高レベルのIL−6の産生を誘導する(図3)。さらに、miDCは、pDCに対する極めて優れた生存を示し、これは、Bcl2遺伝子導入マウス由来のpDCの生存に類似している。miDCのpDCとの関係をさらに試験するために、NF−kbのp50サブユニットが不十分であるマウス(nfkb1−/−マウス)由来の精製したpDCおよびmiDCのTLR9応答を試験した。脾臓pDCおよびFLDC培養物由来のpDCについて以前に示したように、nfkb1−/−マウスのBMのpDCは、CpG1668−ODNによる活性化時に、実質的に減少したIL−6の産生(>80%の減少)を示す。しかしながら、nfkb1−/−マウス由来のmiDCは、IL−6産生の軽度の減少(約20%)しか示さず、これは、以前にnfkb1−/−マウスの従来の脾臓DCで見られた応答に類似している(O’Keeffe et al.2005)。
活性化時に、miDCは、CpG2216−ODN刺激時のCD45Rの発現および高レベルのIFN−αの産生を含む、pDCに類似する特徴を示した。同時に、miDCは、GM−CSFに応答する成熟、TLR4リガンドに対する反応性、およびnfkb1の非存在下での高レベルのIL−6の産生といった、cDCの特徴を示した。さらに、miDCの生存は、培地においても、刺激時においても極めて高かった(図6)。これらの因子を考慮すると、miDCが、実際は、pDCおよびcDC様のどちらの子孫も生じさせる分裂前駆体集団であったという可能性が考えられた。この仮説を試験するために、miDCを、CFSEで標識し、単独培地中で、またはTLR4、TLR7、もしくはTLR9、またはGM−CSFに対するリガンドの存在下で、40時間培養した。培養期間後の、miDCのFACS分析から、培養条件のいずれにおいても、いかなるCFSE標識の消失もなかったことが分かり、これは、miDCが、非分裂細胞であったことを示している(図7A)。CFSEで標識したおよび標識していないmiDCを、刺激後のIFN−α、IL−6、およびTNF−αを産生するそれらの能力について比較した。CFSEで標識した細胞は、正確にそれらの標識していない対照物として振舞った。
FLBM培養物は、FLによるDC前駆体からのcDCおよびpDCの発達を推進する。miDCの増殖能をさらに調査するために、培養物をCFSEで標識し、FLBM培養物中に「スパイク」した。4日目の後に、大部分のmiDCが1回の分裂を受け、5日目までに、細胞は、主に、1回または2回分裂した細胞であった(図7B)。培養物において、細胞は、CD45Rを上方制御したが、分裂時に、CD45Rは、細胞表面から消失した。細胞は、高レベルのSirpα(CD172)およびごくわずかなClec9Aを発現した。したがって、これらの細胞は、CD8−型FLDCにさらに類似していた。
miDCの刺激能を判定するために、miDCを、最初に、同種異系MLRで試験した。pDCと同様に、miDCは、事前の活性化を伴わない同種異系T細胞の不十分な刺激因子であった。活性化によって、miDCは、pDCよりも効率的にナイーブT細胞を刺激したが(図8A)、CD8−脾臓DCよりも小さい範囲であった。

Claims (20)
- 免疫療法剤を調製するための方法であって、
骨髄から単離され、CD11b−/loおよびCD172+(Sirpα)である、単離されたIFN−α産生非形質細胞様樹状細胞(非pDC)に、1つ以上の抗原を負荷するように、前記非pDCをエクスビボで修飾するステップ
を含み、前記抗原が、ウイルス抗原、細菌抗原、真菌抗原、寄生虫抗原または腫瘍抗原である方法。 - 前記非pDCをエクスビボで修飾する前記ステップは、(i)前記1つ以上の抗原および前記樹状細胞を培養物中でインキュベートすること、(ii)前記樹状細胞による前記1つ以上の抗原の免疫複合体に媒介される取り込み、(iii)前記1つ以上の抗原の存在下での前記細胞の電気穿孔、(iv)前記1つ以上の抗原のウイルス形質導入、ならびに(v)前記1つ以上の抗原をコードするmRNAまたはDNAのリポフェクションまたは形質移入、から成る群から選択される、請求項1に記載の方法。
