Description
[1]外来性の導入遺伝子の発現が抑制された人工多能性幹細胞を選別する方法であって、以下の工程を含む方法
(a)人工多能性幹細胞において外来性の導入遺伝子の発現量を測定する工程、および
(b)当該外来性の導入遺伝子の発現量が、対照値と同じまたはそれ以下である人工多能性幹細胞を選択する工程。
[2]測定する導入遺伝子がOct3/4、Sox2、KLF4、c-Mycから成る群より選択された少なくとも一つを含む、[1]に記載の方法。
[3]測定する導入遺伝子がOct3/4、Sox2、KLF4、c-Mycから成る群より選択された少なくとも二つを含む、[2]に記載の方法。

4F2, 4F3, 4F4, 4F5, 4F6、4F7, 4F8, 4F10, 4F12,4F16、4F19, 4F20, 4F21、4F24、4F25、4F26および4F31は、作製したサンプルiPS細胞であり、201B7および201B2は、外来性の遺伝子の発現が抑制されている対照iPS細胞である。87E6は、外来性の遺伝子の発現が抑制されていない対照iPS細胞である。
(A) 体細胞ソース
iPS細胞作製のための出発材料として用いることのできる体細胞は、哺乳動物(例えば、ヒト、マウス、サル、ブタ、ラット等)由来の生殖細胞以外のいかなる細胞であってもよく、例えば、角質化する上皮細胞(例、角質化表皮細胞)、粘膜上皮細胞(例、舌表層の上皮細胞)、外分泌腺上皮細胞(例、乳腺細胞)、ホルモン分泌細胞(例、副腎髄質細胞)、代謝・貯蔵用の細胞(例、肝細胞)、境界面を構成する内腔上皮細胞(例、I型肺胞細胞)、内鎖管の内腔上皮細胞(例、血管内皮細胞)、運搬能をもつ繊毛のある細胞(例、気道上皮細胞)、細胞外マトリックス分泌用細胞(例、線維芽細胞)、収縮性細胞(例、平滑筋細胞)、血液と免疫系の細胞(例、Tリンパ球)、感覚に関する細胞(例、桿細胞)、自律神経系ニューロン(例、コリン作動性ニューロン)、感覚器と末梢ニューロンの支持細胞(例、随伴細胞)、中枢神経系の神経細胞とグリア細胞(例、星状グリア細胞)、色素細胞(例、網膜色素上皮細胞)、およびそれらの前駆細胞(組織前駆細胞)等が挙げられる。細胞の分化の程度や細胞を採取する動物の齢などに特に制限はなく、未分化な前駆細胞(体性幹細胞も含む)であっても、最終分化した成熟細胞であっても、同様に本発明における体細胞の起源として使用することができる。ここで未分化な前駆細胞としては、たとえば神経幹細胞、造血幹細胞、間葉系幹細胞、歯髄幹細胞等の組織幹細胞(体性幹細胞)が挙げられる。
てその培養に適した自体公知の培地で前培養することができる。そのような培地としては、例えば、約5〜20%の胎仔ウシ血清を含む最小必須培地(MEM)、ダルベッコ改変イーグル培地(DMEM)、RPMI1640培地、199培地、F12培地などが挙げられるが、それらに限定されない。核初期化遺伝子(さらに必要に応じて、他のiPS細胞の樹立効率改善物質)との接触に際し、例えば、カチオニックリポソームなど導入試薬を用いる場合には、導入効率の低下を防ぐため、無血清培地に交換しておくことが好ましい場合がある。
本発明に用いられる導入遺伝子とは、体細胞の核を初期化してiPS細胞を樹立するために体細胞において外来的に発現させた核初期化機能を有する遺伝子(核初期化遺伝子)であり、WO 2007/069666に記載の遺伝子であってもよい。より詳細には、Oct3/4, Klf4, Klf1, Klf2, Klf5, Sox2, Sox1, Sox3, Sox15, Sox17, Sox18, c-Myc, L-Myc, N-Myc, TERT, SV40 Large T antigen, HPV16 E6, HPV16 E7, Bmil, Lin28, Lin28b, Nanog, EsrrbまたはEsrrgが例示される。また、本導入遺伝子は、必ずしもタンパク質をコードする必要がなく、例えば、Science, 296, 550-553 (2002)に記載のp53に対するRNA干渉機能を有するRNAを発現するDNAであってもよい。

核初期化遺伝子のcDNAは、宿主となる体細胞で機能し得るプロモーターを含む適当な発現ベクターに挿入される。発現ベクターとしては、例えば、レトロウイルス、レンチウイルス、ヘルペスウイルスなどのウイルスベクター、動物細胞発現プラスミド(例、pA1-11,pXT1,pRc/CMV,pRc/RSV,pcDNAI/Neo)などが用いられ得る。
切な方法により、該ベクターを細胞に感染させる。例えば、ベクターとしてレトロウイルスベクターを用いる具体的手段が WO2007/69666、Cell, 126, 663-676 (2006) 及び Cell, 131, 861-872 (2007) に開示されており、ベクターとしてレンチウイルスベクターを用いる場合については、Science, 318, 1917-1920 (2007) に開示がある。
iPS細胞は、樹立効率を高めるために、例えば、ヒストンデアセチラーゼ(HDAC)阻害剤[例えば、バルプロ酸 (VPA)(Nat. Biotechnol., 26(7): 795-797 (2008))、トリコスタチンA、酪酸ナトリウム、MC 1293、M344等の低分子阻害剤]、G9aヒストンメチルトランスフェラーゼ阻害剤[例えば、BIX-01294 (Cell Stem Cell, 2: 525-528 (2008))等の低分子阻害剤]、L-calcium channel agonist [例えばBayk8644 (Cell Stem Cell, 3, 568-574 (2008))、UTF1(Cell Stem Cell, 3, 475-479 (2008))、Wnt Signaling(例えばsoluble Wnt3a)(Cell Stem Cell, 3, 132-135 (2008))、2i/LIF (2iはmitogen-activated protein kinase signallingおよびglycogen synthase kinase-3の阻害剤)、(PLoS Biology, 6(10), 2237-2247 (2008))]等が挙げられるが、それらに限定されない。
iPS細胞の樹立のため、核初期化遺伝子が導入された細胞は例えばES細胞の培養に適した条件下で培養することができる。マウス細胞の場合、通常の培地に分化抑制因子としてLeukemia Inhibitory Factor(LIF)を添加して培養を行う。一方、ヒト細胞の場合には、LIFの代わりに塩基性線維芽細胞増殖因子(bFGF)および/または幹細胞因子(SCF)を添加することが望ましい。また通常、細胞は、フィーダー細胞として、放射線や抗生物質で処理して細胞分裂を停止させたマウス胎仔由来の線維芽細胞の共存下で培養される。マウス胎仔由来の線維芽細胞としては、通常STO細胞株(ATCC CRL-1503)等がフィーダーとしてよく使われるが、iPS細胞の誘導には、STO細胞にネオマイシン耐性遺伝子とLIF遺伝子を安定に組み込んだSNL細胞(SNL76/7 STO細胞; ECACC 07032801)(McMahon, A. P. &
Bradley, A. Cell 62, 1073-1085 (1990))等がよく使われている。しかしながら、本発明においては、マウス胎仔由来の初代線維芽細胞(MEF)を用いた方がヒトiPS細胞の樹立効率がより改善されるので、MEFの使用がより好ましい。マイトマイシンC処理済のMEFは、ミリポア社やリプロセル社から市販されている。これらのフィーダー細胞との共培養は、核初期化物質の接触より前から開始してもよいし、該接触時から、あるいは該接触より後(例えば1-10日後)から開始してもよい。
の酸素濃度よりも低い酸素濃度の条件が挙げられ、例えば雰囲気中の酸素濃度が18%以下の条件が該当する。好ましくは、雰囲気中の酸素濃度は15%以下(例、14%以下、13%以下、12%以下、11%以下など)、10%以下(例、9%以下、8%以下、7%以下、6%以下など)、または5%以下(例、4%以下、3%以下、2%以下など)である。また、雰囲気中の酸素濃度は、好ましくは0.1%以上(例、0.2%以上、0.3%以上、0.4%以上など)、0.5%以上(例、0.6%以上、0.7%以上、0.8%以上、0.95%以上など)、または1%以上(例、1.1%以上、1.2%以上、1.3%以上、1.4%以上など)である。
上記のように樹立されたiPS細胞における外来性の導入遺伝子の発現量と、外来性の導入遺伝子の発現量とそれに対応する内在性遺伝子の発現量を合わせた発現量を区別して測定する方法以下に示す。ここで、それ(外来性の導入遺伝子)に対応する内在性遺伝子とは、例えば、外来性の導入遺伝子がOct3/4の場合に、細胞が内在的に発現しているOct3/4遺伝子を意味する。

外来性の導入遺伝子の発現量とそれに対応する内在性遺伝子の発現量を合わせた発現量は、当業者に周知の方法により、外来性と内在性の共通する部分を測定すればよい。
の他にも、ノザンハイブリダイゼーション法を用いる場合も、プローブに付加した蛍光や放射線同位体の量を測定することで、発現量を数値化することができる。
(A)絶対的評価による選択方法
上記のように測定した導入遺伝子の発現量に対応する数値は、測定に用いた機器等の特性又は能力に依存するため、標準溶液を用いて同様の測定を行った結果の値に対する相対値として換算されることが望ましい。ここで用いる標準溶液としては、分子量が既知であるiPS細胞の樹立に用いた発現ベクターを溶解させた標準溶液が挙げられる。換算される単位としては、1つの細胞に含まれるmRNAの個数(以下、コピー数)が例示される。この他にも、細胞量に比例する物、例えば、細胞内全RNA量または全DNA量に対するコピー数も換算されるべき単位として挙げられる。
5000, 1000、900、850、840、830、820、810または800コピーと同じかそれ以下である。より好ましくは、導入遺伝子ごとの対照値を用いることであり、例えば、導入遺伝子がOct3/4である場合の対照値は、細胞内RNA 50ngあたり18700, 853, 800、700、600、500、400または345コピーであり、導入遺伝子がSox2である場合の対照値は、細胞内RNA 50ngあたり7950, 5000, 1000、900、810又は379コピーであり、導入遺伝子がKLF4である場合の対照値が、細胞内RNA 50ngあたり2990, 1000, 685,500、400、300、200、100、50または23コピーであり、導入遺伝子がc-Mycである場合の対照値は、細胞内RNA 50ngあたり4020, 1890, 500、400、300または270コピーである。iPS細胞の選択に際しては、導入遺伝子全てにおいて対照値と同じかそれ以下であるiPS細胞を選択してもよく、導入遺伝子の中のいずれか、3遺伝子、2遺伝子もしくは1遺伝子のみが対照値以下であるiPS細胞を選択してもよい。
測定した外来性の導入遺伝子の発現量とそれに対応する内在性遺伝子の発現量を合わせた発現量に対する外来性の導入遺伝子の発現量の割合を算出して、外来性の導入遺伝子の発現が抑制されているiPS細胞の選択を行うことができる。例えば、事実上外来性の導入遺伝子の発現が抑制されている対照iPS細胞における割合(以下、対照割合)以下のiPS細胞を選択することが好ましい。ここで、対照割合は、外来性の導入遺伝子の発現の状態が知られている入手可能な任意のiPS細胞株の外来性の導入遺伝子の発現の換算値を調べて表2を作成し、表2に示す感度および/または特異性の値が共に0.9以上、0.95以上または0.99以上になるように予め設定した値を用いるてもよい。より好ましくは、感度および特異性の値は、共に1である。ここで、感度および特異性が共に1を示すということは、偽陽性および偽陰性が全くない理想的な対照割合である事を意味する。前記入手可能な任意の細胞株は、例えば、Takahashi et al., Cell, 131, 861-872 (2007)に記載の201B1、201B2、201B3、201B6または201B7が例示される。。
Takahashi, K. et al., Cell, 131: 861-872 (2007)に記載の方法でヒトiPS細胞を樹立した。201B7および201B2細胞株は、この文献で樹立したものを使用した。同様の方法により、ヒトOCT3/4、ヒトSOX2、ヒトKLF4、ヒトc-MYCを発現するレトロウイルスを用いて、Chronic Infantile Neurologic, Cutaneous, Articular syndrome (CINCA 症候群)の患者から同意を得て生検した皮膚から樹立した線維芽細胞よりサンプルiPS細胞株を樹立した。このサンプルiPS細胞は、4F2, 4F3, 4F4, 4F5, 4F6、4F7, 4F8, 4F10, 4F12, 4F16、4F19, 4F20, 4F21、4F24、4F25、4F26、4F31と命名した。
各iPS細胞から、全RNAを抽出し、このうち1μgをプライマーdT(20)および逆転写酵素Rever Tra Ace(TOYOBO)を用いてmRNAを相補鎖DNAへと逆転写した。反応物の最終容量は、20μlであった。この逆転写物1μl(50ngのRNA量に相当)を鋳型として、表3に記載の外来性遺伝子の発現量と内在性遺伝子の発現量を合わせた発現量測定用および外来性遺伝子発現量測定用プライマーセットを用いて、SYBR Green IIを指標として導入遺伝子ごとに定量PCRを行った。増幅のための条件は以下の通りであった。すなわち、95℃/30秒で1サイクル、次に、表3におけるKlf4(total)#2を除くプライマーセットの場合には(95℃/5秒、60℃/30秒)を50サイクル、Klf4(total)#2のプライマーセットの場合には(95℃/10秒、60℃/10秒、72℃/40秒)を50サイクルであった。
図1〜4には、外来性遺伝子の陰性対照として、iPS細胞の作成の由来となった線維芽細胞4F6, 4F16, 4F21, 4F24, 4F25, 4F26, 4F31, 201B7、フィーダー細胞として用いたSN
L細胞、胚性幹細胞(H9)およびサイレンシングされていない陽性対照iPS細胞として87E6の結果を示した。図3及び4では、外来性とこれに対応する内在性のKlf4遺伝子の発現量を合わせた発現量、及びc-Myc遺伝子の場合における発現量を測定するために、Klf4(total)#1及びc-Myc(total)#1のプライマーセットをそれぞれ用いた。これらの線維芽細胞、SNL細胞および陰性対照H9細胞では、外来性の導入遺伝子の発現は全く見られなかったが、内在性の遺伝子の発現は確認された。一方、サイレンシングされている対照iPS細胞である201B7の結果を表4にも示した。
4F12, 4F16 および 4F20 では外因性遺伝子がサイレンシングされていることが確認された(表7)。
さらに、対照値として全RNA 50ngあたり10000 コピーを用いた場合、4F3, 4F4, 4F5, 4F6, 4F10, 4F20, 4F24 および 4F26では、外因性遺伝子が抑制されることが見い出された(表7)。この場合において、表1におけるOct3/4, Sox2, Klf4, およびc-Mycの外因性発現に対する「感度」は、それぞれ88%, 100%, 100%, および100%であった。また表1におけるOct3/4, Sox2, Klf4, およびc-Mycの外因性発現に対する「特異性」は、それぞれ100%, 100%, 60%, および14%であった。
Claims (6)
- 外来性の導入遺伝子の発現が抑制された人工多能性幹細胞を選別する方法であって、
(a)人工多能性幹細胞において外来性の導入遺伝子の発現量を測定する工程、および
(b)当該外来性の導入遺伝子の発現量が、対照値と同じかそれ以下である人工多能性幹細胞を選択する工程、を含み、
当該対照値が、人工多能性幹細胞内の全RNA 50ngあたり10000コピー又はそれ以下である、方法。 - 測定する導入遺伝子がOct3/4、Sox2、およびKlf4を含む、請求項1に記載の方法。
- 測定する導入遺伝子がOct3/4、Sox2、Klf4、およびc-Mycを含む、請求項1に記載の方法。
- 外来性の導入遺伝子の発現が抑制された人工多能性幹細胞を選別する方法であって、
(a)人工多能性幹細胞において外来性の導入遺伝子の発現量を測定する工程、および
(b)当該外来性の導入遺伝子の発現量が、対照値と同じかそれ以下である人工多能性幹細胞を選択する工程、を含み、
当該外来性の導入遺伝子がOct3/4、Sox2、およびKlf4であり、
当該対照値が、以下の(i)から(iii)である、方法;
(i)測定する導入遺伝子がOct3/4である場合の対照値が、人工多能性幹細胞内の全RNA 50ngあたり18700コピー又はそれ以下、
(ii)測定する導入遺伝子がSox2である場合の対照値が、人工多能性幹細胞内の全RNA 50ngあたり7950コピー又はそれ以下、および
(iii)測定する導入遺伝子がKlf4である場合の対照値が、人工多能性幹細胞内の全RNA 50ngあたり2990コピー又はそれ以下。 - 当該外来性の導入遺伝子としてさらにc-Mycを含み、
当該対照値が、以下の値である、請求項4に記載の方法
(iv)測定する導入遺伝子がc-Mycである場合の対照値が、人工多能性幹細胞内の全RNA 50ngあたり4020コピー又はそれ以下。 - 導入遺伝子の発現量を、配列番号9及び10、配列番号11及び12、配列番号13及び14、又は、配列番号15及び16のプライマーセットを用いて測定する工程を含む、請求項4または5に記載の方法。
