Description
本発明は、SCA5関連ポリヌクレオチド、当該ポリヌクレオチドによりコードされるポリペプチド、並びにかかるポリヌクレオチド、及びポリペプチドを同定する方法を含む。
疾患に関連するゲノム配列の同定は、当該疾患の改良された診断を可能とする。本発明は、SCA5ポリヌクレオチドにおける変異が脊髄小脳性運動失調5型(SCA5)に関連することを開示する。本発明は、SCA5ポリヌクレオチドにおいて変異を含むSCA5ポリヌクレオチドの如き本発明のポリヌクレオチドを、検出する方法、SCA5を発現する危険にさらされていない被験者を同定する方法、及びSCA5を発現する危険性を有する又は有している被験者を同定する方法を含む。本発明の方法は、典型的に、SCA5ポリヌクレオチド、通常はSCA5ポリヌクレオチドの一部分を分析すること、そして当該SCA5ポリヌクレオチドが変異を含むかどうか決定することを含む。
ヒト被験者。この試験に参加する全ての被験者、及び対照となる個人は、ミネソタ大学の被験者委員会又は参加機関により認可されたインフォームドコンセント用紙にサインした。関係のない対照DNAサンプルを、CEPHパネル、及び健常な北アメリカの住人から入手した(n=500)。DNAをPuregeneキット(Gentra Systems,Plymouth,MN)を使用して抹消静脈血から抽出した。
免疫ブロットを、ホースラディッシュ・ペルオキシダーゼ結合二次抗体で視覚化し、そして化学発光分析(Amersham Biosciences,Uppsala,Sweden)で強化した。
当該アメリカ系ファミリーは、エイブラハム・リンカーン大統領の父方の祖父母から降りる2つの主要分枝を有する(図1)。家族による「リンカーン病」といわれるSCA5は、リンカーン大統領の父方の伯父であるジョサイア、及び伯母であるメアリーの子孫に見られ、このことは、リンカーン大統領の父方の祖父母のいずれかがSCA5変異を有していることを示唆する。家族のこれらの2つの分鎖を、図1に示す。
Claims (9)
- ヒト対象が脊髄小脳性運動失調5型疾患(SCA5)の危険を有するか否かを決定する方法であって、前記ヒト対象由来の核酸試料について、配列番号1のヌクレオチド13823〜13861又は13827〜13865の欠失があるか否かを分析することを含み、ここで配列番号1のヌクレオチド13823〜13861又は13827〜13865の欠失が、SCA5の危険を有することを示す、方法。
- 前記核酸試料がSCA5ポリヌクレオチドを含む、請求項1に記載の方法。
- 前記分析がハイブリダイゼーションを含む、請求項2に記載の方法。
- 前記SCA5ポリヌクレオチドがゲノムSCA5ポリヌクレオチドである、請求項2に記載の方法。
- 前記SCA5ポリヌクレオチドが処理されたSCA5ポリヌクレオチドである、請求項2に記載の方法。
- 前記分析が前記SCA5ポリヌクレオチドの増幅を含む、請求項2に記載の方法。
- 前記分析が前記SCA5ポリヌクレオチドの部分配列の決定を含む、請求項2に記載の方法。
- 前記増幅がプライマーAGAGGCACTGTCCCTTGGT(配列番号19)及びGCTGGTTCACACTCCACAGA(配列番号20)の使用を含む、請求項6に記載の方法。
- 前記SCA5ポリヌクレオチドがエクソン7、エクソン14、又はその双方に変異を含むか否かを決定することを更に含むとともに、エクソン7の変異が、配列番号1のヌクレオチド7755のTからCへの変化を含み、エクソン14の変異が、配列番号1のヌクレオチド16010〜16024の欠失を含む、請求項2に記載の方法。
