を含む、クロマチン構造を評価する方法に関する。1つ又は複数の細胞は、1つ又は複数の生細胞であり得るか、又は、1つ又は複数の真核細胞であり得るか、又は、1つ又は複数の哺乳動物細胞、及び/又は他の種類の細胞、若しくはその任意の組合せであり得る。複数の画像を、任意の期間にわたりキャプチャすることができる。1ピクセル当たりの染色強度の平均値及び/又はSD値を、各画像に関して算出することができる。その期間にわたるSD値の変化は、例えば任意の期間にわたり、クロマチン構造の変化と相関し得る。核酸染色剤は、蛍光染色剤、又は他の種類の染色剤であり得る。核酸染色剤は、例えばヨウ化プロピジウムであり得る。本発明の方法は、核酸染色剤で処理する前にRNaseで細胞を処理することをさらに含み得る。任意選択として、画像を、共焦点顕微鏡又は他の種類の顕微鏡若しくは検出機器を利用して、キャプチャすることができる。1つ又は複数の細胞の試料は、平切断切片(planar section slice)を含み得る。画像は、約512×512ピクセルの解像度に代えて、任意の解像度で、例えば、1ピクセル当たりの染色強度を理解することを可能にするような解像度で、キャプチャすることができる。平均値及び/又はSD値を、各核の少なくとも2つのキャプチャした画像の平均から算出することができる。核の画像は、任意の量のピクセル、例えば500ピクセル以上、又は1000ピクセル以上、又は約6000ピクセル以上を含み得る。本発明の方法は、1ピクセル当たりの平均染色強度に基づき、クロマチン構造を低凝縮度の、中程度の凝縮度の、又は高凝縮度のものと判定することをさらに含み得る。核酸染色剤はヨウ化プロピジウムを含むことができ、及び/又は1ピクセル当たりの染色強度の平均値は、約2400〜約3000の範囲であり得る。この範囲外の他の範囲を使用することができる。
1つ又は複数の細胞を化合物と接触させること、
1つ又は複数の細胞におけるクロマチン構造を上述したように評価すること、及び
化合物と接触させた1つ又は複数の細胞のSD値を1つ又は複数の対照細胞のSD値と比較すること、
を含み得る。該化合物は、1つ又は複数の細胞の有糸分裂の進行を刺激することができる。該化合物は、1つ又は複数の細胞におけるアポトーシスを誘発することができる。該化合物は、成長因子を含み得る。
核酸染色剤で処理した1つ又は複数の細胞の核の、少なくとも1つの画像をキャプチャする光学機器(又は他の機器)、
画像の各ピクセルで染色強度を定量する分光計機器(又は他の機器)であって、光学機器に光学的に接続されており、計算機器と動作可能に接続されている分光計機器、並びに
任意に、1ピクセル当たりの染色強度の平均値及び標準偏差(SD)値を算出する計算機器、及び
任意に、算出した平均値及びSD値を表示する表示機器、
を含み得る。該光学機器は、共焦点顕微鏡を含み得る。該分光計機器は、ヨウ化プロピジウム又は他の化合物により放射される光を検出することができる。
プラスミド。ヒト野生型c−Src(c−Src−wt、1−536、開始メチオニンを1と定める)((非特許文献24)、D. J. Fujitaにより提供された)をコードするcDNAを、従前(非特許文献7)のようにpcDNA4/TOベクター(Invitrogen)中にサブクローニングした。C−末端にHAエピトープタグを付したSrc−HA(1−532)は、c−Src−wt cDNAをpMHベクター(Roche)中にサブクローニングすることにより構築した。ヒト野生型Lyn(Lyn−wt)をコードするcDNAは、T. Yamamotoにより提供された(1−512、(非特許文献25))。C−末端の負の調節テールを欠くLynにHA−タグを付した構築物、Lyn−HA(1−506、キナーゼ活性がある)及びLyn(K275A)−HA(1−506、キナーゼ活性がない)は、以前に説明した(非特許文献5)。ヒト野生型Fyn(1−537、(非特許文献26))及びヒト野生型c−Yes(1−543、(非特許文献27))(T. Yamamotoにより提供された)をコードするcDNAは、C−末端にHAエピトープタグを付した。C−末端の負の調節テールを欠くHA−タグを付した構築物、Fyn−HA(1−524、キナーゼ活性がある)及びYes−HA(1−530、キナーゼ活性がある)は、pMH中にサブクローニングすることにより構築した。ヒト野生型Syk(1−635、(非特許文献28))(E.A. Clarkにより提供された)をコードするcDNAは、pcDNA4/TO中にサブクローニングした。構築物のN−末端に核局在シグナル(NLS)(非特許文献29)を導入するために、pcDNA4/TO−NLS−mcsベクターを、NLS−Chk(PTK)−FLAG(非特許文献30)をコードするpcDNA4/TOのBalI−BssHII断片と、pcDNA4/TOベクターのEcoRV−BssHII断片とを置換することにより構築した。pcDNA4/TO−NLS−mcs中のSrc−HA、Lyn−HA、Lyn(K275A)−HA、Fyn−HA、Yes−HA及びSykのN−末端にNLSを添加した。NLS融合緑色蛍光タンパク質(NLS−GFP)を、以前に説明したように構築し、pcDNA4/TO中にサブクローニングした(非特許文献30、31)。pcDNA3/Flag−Suv39h1及びpcDNA3/Flag−G9aは、M. Nakao及びY. Shinkaiにより提供された(非特許文献32)。ヒトCD25をコードするcDNAは、A. Iwamaにより提供された(非特許文献33)。CD25−Lyn−HAは、Lyn−HAのN末端でのCD25(1−270)との融合により構築し、配列番号1の配列(I−P−E−F−P−R−D−P−L−C−W−T−R−P−A−A−P−K−L−S−P−R−A−G−N)をCD25及びLyn−HAの間に挿入した。
クロマチン構造変化の定量。図1A、図2A、図3A、図4A、図5A及び図6Aは、ヨウ化プロピジウム(PI)染色で得られるDNA蛍光画像を示す。「材料及び方法」で説明したように、画像を多数のピクセルに等しく分割した。各ピクセルの蛍光強度を測定し、平均強度及びS.D.値を各細胞に関して算出した。スケールバーは10μmを示す。図1B、図2B、図3B、図4B、図5B及び図6Bに示すヒストグラムは、低凝縮度のDNA(700≦1ピクセル当たりの蛍光強度<2200、緑色)、中程度の凝縮度のDNA(2200≦1ピクセル当たりの蛍光強度≦3200、黄色)、高凝縮度のDNA(1ピクセル当たりの蛍光強度>3200、赤色)及びバックグラウンド(1ピクセル当たりの蛍光強度<700、青色)の領域を表す。2D等角投影法による強度プロファイルを、図1C、図2C、図3C、図4C、図5C及び図6Cに示す。右に示すPI蛍光強度のスペクトル(0〜4095)は、高凝縮度のDNA(赤色)、中程度の凝縮度のDNA(黄色)、低凝縮度のDNA(緑色)及びバックグラウンド(青色)を表す。図7A〜図7C及び図8は、「材料及び方法」で説明したように、S期又はG2期で同期化したCOS−1細胞に関する。DNA含有量は、非同期化した細胞と、S期及びG2期が豊富な細胞集団とに関してフローサイトメトリーにより分析し、結果はそれぞれ図7A〜図7Cにヒストグラムとして示す。G2期が豊富な細胞集団は、G2期に同期化した細胞から得た。ピークは、2N(G1期)及び4N(G2期)のDNA含有量を表す。図8に示すプロットは、各細胞に関する1ピクセル当たりのPI強度の各S.D.値を表し、バー及び値は3回の独立の実験における代表的な実験(細胞数81〜94)に基づく平均±S.D.を表す。アスタリスクは、スチューデントt検定により算出した有意差を示す(***p<0.001、NS:有意でない)。図9及び図10は、3mMのチミジンに18時間曝露することによりS期で停止させ、洗浄してチミジンを含まないものとし、さらに9時間培養したCOS−1細胞に関する。細胞を、DNAに関して抗リン酸化ヒストンH3S10抗体及びPIで染色し、有糸分裂の染色体を検出した。PI強度のS.D.値を図9に示す。スケールバーは10μmを示す。図10におけるプロットは、各細胞に関する1ピクセル当たりのPI強度の各S.D.値を表し、バー及び値は代表的な実験(細胞数35〜74)に基づく平均±S.D.を表す。アスタリスクは、スチューデントt検定により算出した有意差を示す(***p<0.001)。図11A〜図11D及び図12は、表示した濃度のH2O2で1時間処理し、DNAに関してPIで染色したCOS−1細胞に関する。DNA含有量をフローサイトメトリーにより分析し、結果を図11A〜図11Dにヒストグラムとして示す。図12におけるプロットは、各細胞に関する1ピクセル当たりのPI強度の各S.D.値を表し、バー及び値は代表的な実験(細胞数61〜78)に基づく平均±S.D.を表す。アスタリスクは、スチューデントt検定により算出した有意差を示す(***p<0.001)。図13は、ヒストンH2B−GFPを安定的に発現している、DNAに関してPIで染色したCOS−1細胞に関する。DNA及びヒストンH2B−GFPの染色の蛍光画像を図13に示す。PI強度のS.D.値を、図13に示した画像の上部に示す。スケールバーは10μmを示す。
Claims (33)
- 細胞におけるクロマチン構造を評価する方法であって、
1つ又は複数の細胞の試料を用意すること、
該1つ又は複数の細胞を核酸蛍光染色剤で処理すること、
該核酸蛍光染色剤で処理した細胞のうち1つ又は複数の細胞の核の画像(複数のピクセルを含む)をキャプチャすること、
該複数のピクセルの各ピクセルで染色強度を定量すること、
1ピクセル当たりの染色強度の平均値及び標準偏差(SD)値を算出すること、
該クロマチン構造を判定すること、ここで該SD値がクロマチン凝縮のレベルの指標である、
を含むクロマチン構造を評価する方法。 - 核酸蛍光染色剤がヨウ化プロピジウムである、請求項1に記載の方法。
- 核酸蛍光染色剤で処理する前に、該1つ又は複数の細胞をRNaseで処理することをさらに含む、請求項1または2に記載の方法。
- 該1つ又は複数の細胞が1つ又は複数の生細胞である、請求項1に記載の方法。
- 該1つ又は複数の細胞が1つ又は複数の真核細胞である、請求項1に記載の方法。
- 該1つ又は複数の細胞が1つ又は複数の哺乳動物細胞である、請求項1に記載の方法。
- 複数の画像を或る期間にわたりキャプチャし、1ピクセル当たりの染色強度の該平均値及び該SD値を各画像に関して算出する、請求項1に記載の方法。
- 該期間にわたるSD値の変化が、該期間にわたる該クロマチン構造の変化と相関する、請求項7に記載の方法。
- 該画像を、共焦点顕微鏡を利用してキャプチャする、請求項1に記載の方法。
- 該1つ又は複数の細胞の試料が平切断切片を含む、請求項1に記載の方法。
- 該画像を512×512ピクセルの解像度でキャプチャする、請求項1に記載の方法。
- 該平均値及び該SD値を、各核の少なくとも2つのキャプチャした画像の平均から算出する、請求項1に記載の方法。
- 該核の画像が6000ピクセル以上を含む、請求項1に記載の方法。
- 1ピクセル当たりの該平均染色強度に基づき、該クロマチン構造を、低凝縮度の、中程度の凝縮度の、又は高凝縮度のものと判定することをさらに含む、請求項1に記載の方法。
- 該核酸蛍光染色剤がヨウ化プロピジウムを含み、1ピクセル当たりの染色強度の該平均値が2400〜3000の範囲である、請求項1に記載の方法。
- 被験体における病変の診断、予後診断又はモニタリングを補助する方法であって、該被験体由来の1つ又は複数の病変細胞の試料における該クロマチン構造を請求項1から3のいずれか1項に記載の方法に従って評価することを含み、ここで該SD値が疾患状態の指標である、病変の診断、予後診断又はモニタリングを補助する方法。
- 該被験体由来の1つ又は複数の非病変細胞の試料における該クロマチン構造を請求項1から3のいずれか1項に記載の方法に従って評価すること、及び該1つ又は複数の病変細胞の該算出したSD値を、該1つ又は複数の非病変細胞の該算出したSD値と比較することをさらに含む、請求項16に記載の方法。
- 該1つ又は複数の病変細胞の該算出したSD値を、正常な組織から疾患状態までの進行曲線に沿う範囲である同じ種類の病変の予め算出したSD値の範囲と比較することをさらに含む、請求項16に記載の方法。
- 該工程を、所定の時間の経過後に繰り返し、該病変が疾患状態に進行しているのか、又は疾患状態から退行しているのかを判定する、請求項16に記載の方法。
- 該所定の時間の間の該病変の進行を、該病変を治療する方法の有効性を測定するために利用する、請求項19に記載の方法。
- 被験体における癌の診断、予後診断及び/又はモニタリングを補助する方法であって、該被験体由来の1つ又は複数の癌性細胞の試料における該クロマチン構造を請求項1から3のいずれか1項に記載の方法に従って評価することを含み、ここで該SD値が悪性疾患の指標である、癌の診断、予後診断及び/又はモニタリングを補助する方法。
- 該被験体由来の1つ又は複数の非癌性細胞の試料における該クロマチン構造を請求項1から3のいずれか1項に記載の方法に従って評価すること、及び該1つ又は複数の癌性細胞の該算出したSD値を、該1つ又は複数の非癌性細胞の該算出したSD値と比較することをさらに含む、請求項21に記載の方法。
- 該1つ又は複数の癌性細胞の該算出したSD値を、正常な組織から悪性疾患までの進行曲線に沿う範囲である同じ種類の癌の予め算出したSD値の範囲と比較することをさらに含む、請求項21に記載の方法。
- 該工程を、所定の時間の経過後に繰り返し、該癌が悪性疾患に進行しているのか、又は悪性疾患から退行しているのかを判定する、請求項21に記載の方法。
- 1つ又は複数の細胞の該クロマチン構造を調節する化合物をスクリーニングする方法であって、
該1つ又は複数の細胞を該化合物と接触させること、
該1つ又は複数の細胞における該クロマチン構造を請求項1から3のいずれか1項に記載の方法に従って評価すること、及び
該化合物と接触させた該1つ又は複数の細胞の該SD値を、1つ又は複数の対照細胞の該SD値と比較すること、
を含む、化合物をスクリーニングする方法。 - 該化合物が該1つ又は複数の細胞の有糸分裂の進行を刺激する、請求項25に記載の方法。
- 該化合物が該1つ又は複数の細胞におけるアポトーシスを誘発する、請求項25に記載の方法。
- 該化合物が成長因子を含む、請求項25に記載の方法。
- 1つ又は複数の細胞におけるクロマチン構造を評価するシステムであって、
核酸蛍光染色剤で処理した1つ又は複数の細胞の核の、少なくとも1つの画像をキャプチャする光学機器と、
該画像の各ピクセルで染色強度を定量する分光計機器であって、該光学機器と光学的に接続されており、計算機器と動作可能に接続されている分光計機器と、
1ピクセル当たりの染色強度の平均値及び標準偏差(SD)値を算出する計算機器と、
該算出した平均値及びSD値を表示する表示機器と、
を含む、評価するシステム。 - 該核酸蛍光染色剤がヨウ化プロピジウムである、請求項29に記載のシステム。
- 核酸蛍光染色剤で処理する前に、該1つ又は複数の細胞をRNaseで処理することをさらに含む、請求項29または30に記載のシステム。
- 該光学機器が共焦点顕微鏡を含む、請求項29から31のいずれか1項に記載のシステム。
- 該分光計機器がヨウ化プロピジウムにより放射される光を検出する、請求項29から32のいずれか1項に記載のシステム。
