Description

[0017] 以下の詳細な説明は、特定の態様を記述し、そして限定する意味では利用されない。本明細書中の全ての重量、量、および比は、特に記載しない限り重量である。以下に示される用語は、以下の意味を有する。

[0066] チオール化キトサン溶液の粘度。それぞれ高分子量および低分子量キトサン塩を使用して作製した2種の非-チオール化キトサン溶液の粘性(図4中、曲線Aおよび曲線B)、そしてそれぞれ2.5%のHCS-TGA低および10%ULCS-TGA低を使用して作製した2種のチオール化キトサン溶液(図4中、データ点Cおよびデータ点D)を、脱イオン水中25℃で測定した。HCS-TGA低溶液は、約650 cPの相対的に高い粘性を有したが、一方ULCS-TGA低溶液は、約10 cPの非常に低い粘性を有した。これらの結果およびその他のキトサン溶液を使用して行ったスプレー実験から、ULCS-TGA低溶液は、10%の濃度そして10%を優に上回っても容易にスプレー可能であるようであった。
[0067] チオール化キトサンゲルの抗微生物特性。1.5%および5%のULCS-TBA高製剤および0.1%過酸化水素を用いて作製したゲルの抗微生物活性を、S. aureusに対して調べた。チオール化キトサンゲル製剤を、2重にして、無菌条件下にて、24ウェルポリスチレン組織培養プレート中に直接入れた。各ウェルを、1 mL(500,000コロニー形成ユニット)のS. aureus(ATCC 25923)の細菌懸濁物とともにインキュベーションした。陽性対照を、1 mlのトリプチケースソイブロス(TSB)とともにインキュベーションした。37℃で6時間のインキュベーションの後、培養液を新しいチューブに移し、連続10倍希釈を行った。適切な希釈物からの10μL分を、希釈追跡法(Jett B.D. et al., Biotechniques, 23, 648-650 (1997))を使用して、トリプチケースソイ寒天プレート上に3重にして置いた。プレートを37℃にて24時間インキュベーションし、そしてコロニー形成ユニット(CFU)を計測した。図5に示されるように、5%ゲル(バーB)は、トリプチケースソイブロス(TSB)対照(バーA)と比較して、完全なのバクテリアの殺傷能力(6 log減少以上)を示した。同様の結果(6 logの減少)が、1.5%ゲルを使用して得られた。
[0068] チオール化CSゲルの分解。1.5%(図6中の曲線Aを参照)および5%(図6中の曲線Bを参照)のULCS-TBA高および0.1%過酸化水素を使用して作製したゲルの分解の挙動を、リゾチーム(1 mg/mL)の存在下、pH 7.4でリン酸緩衝塩類溶液(PBS)中にゲルを設置することにより測定した。重量喪失を28日までの様々な時点で測定した。図6に示されるように、顕著な重量喪失が観察された。28日後、サンプルは完全には分解されていなかった。

Claims (13)
- 耳、鼻、または喉の粘膜組織を保護するための二成分を含む医薬組成物であって、第1の成分はチオール化キトサンを含み、そして第2の成分は十分な酸化剤または外部架橋剤を含み、それにより二成分が混合され流体の混合物が調製され、そして粘膜組織上にコーティングされる場合に、混合物が30分未満で保護ゲル層を形成する、前記二成分を含む医薬組成物。
- 第1の成分および第2の成分はそれぞれ、複数バレルシリンジのバレル中にパッケージングされる、請求項1に記載される医薬組成物。
- 第1の成分が自由流動性の乾燥粒子である、請求項1または2に記載される医薬組成物。
- 第1の成分および第2の成分がそれぞれ水溶液である、請求項1〜3のいずれかに記載される医薬組成物。
- 第2の成分が過酸化水素またはゲニピンを含有する、請求項1〜4のいずれかに記載される医薬組成物。
- 2つの成分を混合する場合、ゲル層が10分未満で形成される、請求項1〜5のいずれかに記載される医薬組成物。
- 2つの成分を混合する場合、ゲル層が5分未満で形成される、請求項1〜5のいずれかに記載される医薬組成物。
- 前記二成分の流体の混合物を、鼻腔、副鼻腔、中耳または内耳の上にスプレー塗布する、請求項1〜5のいずれかに記載される医薬組成物。
- 組成物を、スプレー塗布、洗浄(lavage)、ミスト、モッピング、ウィッキング、またはドリッピングにより適用する、請求項1〜5に記載される医薬組成物。
- チオール化キトサンと十分な酸化剤または外部架橋剤とを含み、それにより保護ゲル層がin situで30分未満で形成される、耳、鼻、または喉の粘膜組織上の流体層。
- 過酸化水素またはゲニピンを含有する、請求項10に記載される流体層。
- 1〜20 wt.%のチオール化キトサンと0.01〜1 wt.%の酸化剤を含有する、請求項10に記載される流体層。
- 1〜20 wt.%のチオール化キトサンと0.1〜10 wt.%の外部架橋剤を含有する、請求項10に記載される流体層。
