Description
The present invention relates to a data processing apparatus for processing data collected by a chromatograph mass spectrometer using a mass spectrometer capable of MS n analysis (n is an integer of 2 or more) as a detector for a gas chromatograph or a liquid chromatograph. More specifically, the present invention relates to a data processing technique related to screen display processing for displaying analysis results.
In triple quadrupole mass spectrometers (TQMS) and ion trap time-of-flight mass spectrometers (IT-TOFMS), ions with a specific mass-to-charge ratio m / z are selected from various ions derived from the analyte. Select a precursor ion, dissociate the precursor ion by collision-induced dissociation (CID), and create a MS / MS (= MS 2 ) spectrum by mass-analyzing the product ion generated thereby. Is possible. In IT-TOFMS, it is also possible to create an MS n spectrum in which n is 3 or more by mass analysis of product ions finally generated by repeating ion selection and CID a plurality of times. . In the present specification, a mass spectrometer capable of MS n analysis in which n is 2 or more is referred to as an MS n type mass spectrometer.
In a chromatograph mass spectrometer that combines a liquid chromatograph (LC) or gas chromatograph (GC) with the above-described MS n- type mass spectrometer, if the component contained in the sample is known, the MS can be used for the retention time of that component. the object of n analysis preliminarily set the mass-to-charge ratio of the precursor ions can acquire the MS n spectrum of the target component. However, when the component contained in the sample is unknown, the precursor ion for performing the MS n analysis cannot be set in advance, and the MS n analysis result of the unknown component contained in the sample other than the target component can be obtained. I can't. In contrast, an apparatus having a function of automatically selecting an appropriate precursor ion from a result obtained by MS 1 analysis without CID and performing MS n analysis in real time (hereinafter referred to as auto MS n function). Conventionally known.
For example, in
FIG. 7 schematically illustrates the auto MS n function in a general chromatograph mass spectrometer. Here, as a selection criterion for precursor ions, a maximum of two of the peaks having a signal intensity equal to or greater than the threshold th in the MS spectrum are selected as precursor ions in descending order of signal intensity. However, an excluded ion list and a preferential ion list as shown in FIG. 2 are set separately, and ions having a mass-to-charge ratio registered in the excluded ion list are precursors even if they meet the above criteria. Do not select ions, and conversely, ions with a mass-to-charge ratio registered in the preferred ion list should be selected as precursor ions even if they do not meet the above criteria as long as there is a peak. To do. Usually, the excluded ion list is used to prevent a known contaminating component, an interfering component contained in a sample, or a component that is known not to be analyzed from being selected as a precursor ion. On the other hand, the preferred ion list is used to select a component to be analyzed as a precursor ion even if it is a trace amount. The reason why the number of precursor ions that can be selected for one MS spectrum is limited is that there is a limitation in the time for performing MS n analysis in real time.
When the waveform of the total ion current (total ion) chromatogram (TIC) is obtained as shown in FIG. 7A, it is assumed that the MS spectrum shown in A is obtained at time t1. In this MS spectrum, the peak f is listed as a precursor ion candidate in accordance with the signal intensity criterion described above. If the mass-to-charge ratio corresponding to this peak f is registered in the excluded ion list, it is not selected as the precursor ion. . On the other hand, the signal intensity of the peak g is lower than the threshold th. If the mass-to-charge ratio corresponding to the peak g is registered in the priority ion list, the ion corresponding to the peak g is automatically selected as the precursor ion, MS 2 analysis for this precursor ion is performed immediately. As a result, the MS 2 spectrum shown in B is obtained.
It is assumed that the MS spectrum shown in C is obtained at another time t2. In this MS spectrum, there are five peaks having a signal intensity equal to or greater than the threshold th, and two peaks are selected in descending order of intensity, but the mass-to-charge ratio corresponding to the peaks b and d is registered in the excluded ion list. Then, except for these, ions corresponding to the next highest intensity peaks a and c are automatically selected as precursor ions, and MS 2 analysis for the two precursor ions is immediately performed. As a result, two MS 2 spectra shown in D and E are obtained. In the analysis using the auto MS n function, while performing normal MS analysis without CID in this way, if there is an ion that satisfies the precursor selection condition based on the analysis result, it is converted to the precursor ion. Set up and run MS 2 analysis in real time. It is also possible to execute MS n analysis in which n is 3 or more by the same method.
When data collected using the auto MS n function as described above in a conventional chromatograph mass spectrometer is analyzed and displayed on the screen of the display unit, the MS obtained at the retention time specified by the analyst The spectrum and the MS 2 spectrum for the precursor ion automatically selected based on the MS spectrum are displayed side by side on the same screen (see Non-Patent Document 2). FIG. 8 shows a display example of this mass spectrum.
In FIG. 8, the MS spectrum at the holding time of 12.05 [min] is displayed in the
Of course, if the screen for setting the precursor selection condition is opened, it is possible to browse the excluded ion list and the preferential ion list as shown in FIG. It is possible to confirm the mass-to-charge ratio of excluded ions, the mass-to-charge ratio of preferred ions, and the like during the holding time. However, the operation and work for newly opening and referencing such another screen is cumbersome and troublesome, and the values listed in the excluded ion list and the preferential ion list are compared with the MS spectrum, and the peaks are excluded ions and It is troublesome to determine whether or not the ions are preferred ions. In particular, as shown in FIG. 2, since it is possible to set conditions with a high degree of freedom so that excluded ions and preferential ions can be arbitrarily set for each holding time range, the work is complicated and analysis is further performed. It is not easy to understand.
In addition, when performing MS 3 analysis using the auto MS n function, the MS 2 spectrum obtained by MS 2 analysis using a specific ion as a precursor ion or an automatically selected ion as a precursor ion is used. When the peak appearing in is a product ion due to a specific neutral loss, MS 3 analysis may be performed using the ion as a precursor ion. That is, this is a case where the generation of a specific neutral loss is set as a precursor ion selection condition for MS n analysis. When the precursor ion of MS 3 analysis is automatically found under such precursor ion selection conditions and the data collected by performing MS 3 analysis is analyzed in a conventional chromatograph mass spectrometer The MS spectrum obtained at the retention time specified by the analyst, the MS 2 spectrum for the precursor ion automatically selected based on the MS spectrum, and the precursor automatically selected based on the MS 2 spectrum. Three of the MS 3 spectra for ions are displayed side by side on the same screen as analysis results. In this case, if the number of peaks appearing in the spectrum is large, or if a relatively low intensity peak is a precursor ion, it is intuitive for the analyst which peak is selected as the precursor ion. There is a problem that it is difficult to understand.
The present invention has been made in view of the above problems, and the object of the present invention is to perform an operation by an analyst when analyzing data collected by chromatographic mass spectrometry and performing operations such as component identification. Another object of the present invention is to provide a mass spectrometry data processing apparatus capable of reducing the burden on the analyst by simplifying the work and improving the work efficiency and allowing the analyst to easily understand and grasp the result.

The present invention, which has been made to solve the above-mentioned problems, is characterized in that MS m over time is obtained by a chromatograph mass spectrometer that combines a chromatograph and a mass spectrometer capable of MS n analysis (n is an integer of 2 or more). -1 analysis (2 ≦ m ≦ n) obtained by performing MS m analysis by automatically selecting a precursor ion for each peak appearing in an MS m-1 spectrum obtained according to predetermined precursor selection conditions Is a mass spectrometry data processing device that processes the data collected in step (b) and displays it on a display screen, and includes excluded ions that are not selected as precursor ions regardless of other conditions and preferred ions that are preferentially selected as precursor ions. It is characterized by having.
The mass spectrometry data processing apparatus according to the present invention is typically m = 2, that is, the MS spectrum (mass spectrum) and the MS 2 spectrum can be displayed side by side by the spectrum display processing means. It may be 3 or more.
The mass spectrometry data processing apparatus according to the present invention executes a dedicated computer program for realizing the functions corresponding to each of the above means on a general-purpose computer including a display unit, an operation unit (keyboard, pointing device, etc.), etc. Can be embodied.
As an aspect of the mass spectrometry data processing apparatus according to the present invention, the additional display processing means may display the mass-to-charge ratio or the mass-to-charge ratio range of the excluded ions and the preferential ions in different display colors.
Further, in the mass spectrometry data processing apparatus according to the present invention, preferably, the additional display processing means has the mass to charge ratio or the mass to charge ratio on the same screen as the MS m-1 spectrum displayed by the spectrum display processing means. It is preferable that a selection operator for whether or not to display the range is arranged, and whether or not the mass-to-charge ratio or the mass-to-charge ratio range is displayed is switched according to the operation of the operator .
In the mass spectrometry data processing apparatus according to the present invention, for example, when an analyst clicks the selection operator with a pointing device such as a mouse and selects display, the additional display processing means automatically determines a precursor ion. The mass-to-charge ratio of the excluded ion and the preferential ion at that time or the range thereof are clearly shown on the original MS m-1 spectrum. Thus, for example, when the peak is not selected as a precursor ion even though it is a peak showing a large signal intensity on the MS m-1 spectrum, the peak is included in the mass-to-charge ratio range of the excluded ion. It is possible for an analyst to intuitively understand whether or not this is because. Whether or not the peak is included in the mass-to-charge ratio range of the preferential ion when it is selected as a precursor ion in spite of the peak showing a small signal intensity on the MS m-1 spectrum. Alternatively, the analyst can intuitively understand whether or not the reason is that a peak having a higher signal intensity is included in the mass-to-charge ratio range of the excluded ions.
According to the mass spectrometry data processing apparatus of the present invention, on the screen displaying the MS m-1 spectrum and the MS m spectrum side by side, the reason why the peak having a small signal intensity is selected as the precursor ion, and conversely, the signal intensity. The reason why a large peak is not selected as a precursor ion can be intuitively and easily understood by a very simple operation by an analyst.
Hereinafter, an LC-MS system including a mass spectrometry data processing apparatus according to the present invention will be described with reference to the accompanying drawings. FIG. 1 is a schematic configuration diagram of an LC-MS system according to the present embodiment.
This system is a liquid chromatograph (LC) 1 that temporally separates components contained in a liquid sample, and detects each separated component according to a mass-to-charge ratio m / z and An MS n type mass spectrometer (MS) 2 capable of performing MS n analysis, and a personal computer (PC) 3 that processes data acquired by the
MS2 is if MS 2 analysis or n is even are possible 3 or more MS n analysis, regardless its configuration. Therefore, a triple quadrupole mass spectrometer that mass-separates product ions with a quadrupole mass filter after dissociating the ions with CID in the collision chamber, and once the sample-derived ions are trapped in the ion trap, the precursor ions An ion trap mass spectrometer that performs sorting, CID, and mass separation of product ions in the ion trap, and once the ions from the sample are trapped in the ion trap, the precursor ions are sorted, and CID is performed in the ion trap to obtain the mass of the product ions. Separation may be performed using any ion trap time-of-flight mass spectrometer that is performed by a time-of-flight mass spectrometer outside the ion trap.
In the LC-MS system according to the present embodiment, the analyst performs a predetermined operation with the
When the analysis is started by an instruction to start the analysis and the liquid sample is introduced into the LC1, the components contained in the sample are temporally separated and eluted while passing through a column (not shown). In MS2, scan measurement involving mass scanning in a predetermined mass-to-charge ratio range is repeated at regular intervals. One scan measurement (mass scan) obtains data constituting one MS spectrum, for example, as indicated by A or C in FIG. In the
Therefore, by repeating scan measurement at a predetermined time interval, MS spectra are obtained at predetermined time intervals, and if a precursor ion that satisfies the precursor selection condition exists, one or a plurality of MSs during the time until the next scan measurement is present. Two spectra are obtained. A total ion current chromatogram (TIC) shown in FIG. 7A is obtained by adding all the ion intensities included in one MS spectrum and arranging them in the time direction.
In the LC-MS system of the present embodiment, from the time when one sample is introduced into LC1 (or when it is delayed by a predetermined time) from the time when the components in the sample are completely eluted until the time when it is appropriately delayed As described above, the MS spectrum data and the MS 2 spectrum data are repeatedly collected, and are collected into one data file and stored in the measurement
Next, a characteristic data processing operation at the time of reanalysis processing in the LC-MS system of the present embodiment will be described. FIG. 3 is a schematic diagram showing a part of a reanalysis screen displayed on the
The mass
On the other hand, when the analyst moves the cursor on the display screen to the “On” button in the precursor condition
Then, the
Further, since the MS spectrum displayed in the
In the above description, with respect to the background portion of the MS spectrum displayed in the
Next, a characteristic data processing operation when reanalysis processing is executed in the LC-MS system of the present embodiment will be described with another example.
Here, the data to be reanalyzed is data acquired up to the MS 3 spectrum using the auto MS n function as described above. Specifically, as the precursor selection conditions for MS 2 analysis, for example, a peak ion or the like showing the maximum intensity in the MS 1 spectrum is selected as the precursor ion as in the above example. On the other hand, as a precursor selection condition for MS 3 analysis, when a product ion generated by a neutral loss of a specific mass is detected in the MS 2 spectrum, the product ion is selected as a precursor ion for MS 3 analysis. . FIG. 5 is an example of a

As in the case of FIG. 3, the mass
In the mass
FIG. 6 is an example of a setting screen for setting whether or not the
In the LC-MS system of the present embodiment, when neutral loss or charge dross is set as a precursor selection condition, a specific peak selected as a precursor ion is displayed in color, so that the displayed mass spectrum Among them, the position of the precursor ion can be confirmed at a glance, and it can be immediately understood that the precursor selection condition is a neutral loss of mass 17.
In addition, although the said Example applies this invention to the process of the data collected by LC-MS, it is clear that it can apply to the process of the data collected by GC-MS. The MS m-1 spectrum and the MS m spectrum are displayed side by side. Of course, it can be extended to the screen to be displayed.
Moreover, the said Example is only an example of this invention, and even if it changes suitably in the range along the meaning of this invention, correction, and addition, it is clear that it is included by the claim of this application.
