Description
従って、苦痛なくカロリー制限と同様の効果を期待できるカロリー制限模倣物(CR mimetics)を開発することは、病気を未然に防ぎ、進行を抑制する可能性があることから産業的に非常に大きなインパクトを有していると考えられる。しかしながら、これまでカロリー制限模倣物の効率的なスクリーニング方法は確立されていなかった。
カロリー制限による寿命延長効果に寄与する遺伝子に関しては、多くの研究がなされており、いくつかの遺伝子がその本態の候補として挙げられている。それらの多くはGH(growth hormone)系もしくはInsulin系に属するシグナル遺伝子に関連するものである。本発明者も、GH−IGF−I(下垂体成長ホルモン−インスリン様成長因子−I)シグナル伝達系にその本態がある可能性を研究し、GHのアンチセンス遺伝子を導入したトランスジェニックラットを作成した。該トランスジェニックラットでは、確かに寿命延長効果が認められ、カロリー制限による効果の一部がGH−IGF−Iシグナル伝達系によるものであることを証明した。しかしながら、該トランスジェニックラットでは、カロリー制限をした正常マウスほど大きな寿命延長効果は得られず、また該トランスジェニックラットをカロリー制限するとさらに寿命が延長することから、GH−IGF−Iシグナル伝達系の他にもメカニズムがあることが考えられた(非特許文献1)。
これまでのように、カロリー制限による寿命延長効果に寄与する複数の遺伝子のうち1つの遺伝子を用いてその効果を追跡すると、その遺伝子が関わった効果しか得ることができず、その遺伝子に影響するカロリー制限模倣物しかスクリーニングできないため、カロリー制限模倣物のスクリーニング能力は不十分である。
また、PGC−1αが酸化ストレスによる神経変性の防止に重要であることがノックアウトマウスを用いた研究から明らかになっている(非特許文献2)。HNF−4の過剰発現等によって一部カロリー制限と似た遺伝子発現パターンを示すことも報告されている(非特許文献3)。しかしながら、恒常的なPGC−1αとHNF−4の活性化は、糖新生が亢進して血糖値が上昇するなどの糖尿病と似たような表現型を示すことが知られている。

該プロモーターが、該レポーター遺伝子の転写を駆動し得るように、該レポーター遺伝子の5’側に連結しており;且つ
(A)のヌクレオチド配列が、該プロモーターによる該レポーター遺伝子の転写を調節し得るように、該プロモーターの5’側に連結している、レポーターベクター。
(2) (A)のヌクレオチド配列が、以下の(a’)〜(d’)から選択されるいずれか1つであり、且つHNF−4αへ結合し得るヌクレオチド配列である、(1)記載のレポーターベクター:
(a’)配列番号1で表されるヌクレオチド配列、
(b’)配列番号1で表されるヌクレオチド配列において、1若しくは複数のヌクレオチドが欠失、置換、挿入または付加されたヌクレオチド配列、
(c’)配列番号1で表されるヌクレオチド配列に少なくとも70%の同一性を有するヌクレオチド配列、及び
(d’)上記(a’)〜(c’)に記載の配列に相補的なヌクレオチド配列。
(3) (1)記載のレポーターベクターが導入された形質転換体。
(4) (1)記載のレポーターベクターが導入された非ヒトトランスジェニック哺乳動物。
(5) 以下の工程を含む、カロリー制限模倣物質のスクリーニング方法:
1)被検物質の存在下で、(1)記載のレポーターベクターを用いてレポーターアッセイを行うこと;
2)被検物質の存在下でのレポーター遺伝子の発現レベルを、被検物質の不在下でのそれと比較すること;及び
3)比較の結果、レポーター遺伝子の発現レベルを上昇させた被検物質を、カロリー制限模倣物質の候補として選択すること。
(6) (1)記載のレポーターベクター、該レポーターベクターが導入された形質転換体、又は該レポーターベクターが導入された非ヒトトランスジェニック哺乳動物を含む、カロリー制限模倣物質をスクリーニングするためのキット。
(7) 以下の工程を含む、寿命を延長する物質のスクリーニング方法:
1)被検物質の存在下で、(1)記載のレポーターベクターを用いてレポーターアッセイを行うこと;
2)被検物質の存在下でのレポーター遺伝子の発現レベルを、被検物質の不在下でのそれと比較すること;及び
3)比較の結果、レポーター遺伝子の発現レベルを上昇させた被検物質を、寿命を延長する物質の候補として選択すること。
(8) (1)記載のレポーターベクター、該レポーターベクターが導入された形質転換体、又は該レポーターベクターが導入された非ヒトトランスジェニック哺乳動物を含む、寿命を延長する物質をスクリーニングするためのキット。
(9) (2)記載のレポーターベクター、該レポーターベクターが導入された形質転換体、又は該レポーターベクターが導入された非ヒトトランスジェニック哺乳動物、及び
HNF−4α又はHNF−4αを発現し得る発現ベクター
を含む組み合わせ物。
(10) 更に、PGC−1α又はPGC−1αを発現し得る発現ベクターを含む、(9)記載の組み合わせ物。

ここで「相補的」とは、ヌクレオチド配列間で約70%以上、好ましくは約80%以上、より好ましくは約90%以上、更に好ましくは約95%以上、最も好ましくは100%の相補性を有することをいう。相同性計算アルゴリズムNCBI BLAST(NationalCenter for Biotechnology Information Basic Local Alignment Search Tool)を用い、以下の条件(期待値=10;ギャップを許す;フィルタリング=ON;マッチスコア=1;ミスマッチスコア=−3)にて計算することができる。
(a’)配列番号1で表されるヌクレオチド配列、
(b’)配列番号1で表されるヌクレオチド配列において、1若しくは複数(好ましくは1〜10個、より好ましくは1〜5個、さらに好ましくは1又は2個)のヌクレオチドが欠失、置換、挿入又は付加されたヌクレオチド配列、
(c’)配列番号1で表されるヌクレオチド配列に少なくとも70%(好ましくは80%以上、より好ましくは90%以上、さらに好ましくは95%以上)の同一性を有するヌクレオチド配列、及び
(d’)上記(a’)〜(c’)に記載の配列に相補的なヌクレオチド配列。
本発明は、上記本発明のレポーターベクターが導入された形質転換体を提供するものである。宿主としては、例えば、エシェリヒア属菌(エシェリヒア コリ(Escherichia coli)等)、バチルス属菌(バチルス サブチルス(Bacillus subtilis)等)、酵母(サッカロマイセス セレビシエ(Saccharomyces cerevisiae)等)、昆虫細胞(夜盗蛾の幼虫由来株化細胞(Spodoptera frugiperda cell;Sf細胞)等)、哺乳動物細胞(肝細胞、脂肪細胞、腎臓細胞(293細胞等)、線維芽細胞等)などが用いられる。宿主は好ましくは哺乳動物細胞である。
本発明は、上記した本発明のレポーターベクターが導入された非ヒトトランスジェニック哺乳動物を提供するものである。本発明のトランスジェニック哺乳動物を用いることにより、下記のカロリー制限模倣物質のスクリーニング方法をインビボで実施することが可能となる。
本発明は、以下の工程を含む、カロリー制限模倣物質(又は寿命を延長する物質)のスクリーニング方法を提供するものである:
1)被検物質の存在下で、上記した本発明のレポーターベクターを用いてレポーターアッセイを行うこと;
2)被検物質の存在下でのレポーター遺伝子の発現レベルを、被検物質の不在下でのそれと比較すること;及び
3)比較の結果、レポーター遺伝子の発現レベルを上昇させた被検物質を、カロリー制限模倣物質(又は寿命を延長する物質)の候補として選択すること。
1)非ヒト哺乳動物に候補物質を投与し、飼育すること;
2)候補物質の投与を受けた非ヒト哺乳動物の寿命を、候補物質の投与を受けていない非ヒト哺乳動物のそれと比較すること;及び
3)比較の結果、非ヒト哺乳動物の寿命を延長した候補物質を選択すること。
HNF−4α又はHNF−4αを発現し得る発現ベクター
とを含む、組み合わせ物を提供するものである。
長寿命を示すAmes dwarf miceとコントロールマウスとを、自由摂食(ad libitum:AL)、および自由摂食群の70%の食餌量を与えたカロリー制限下で4ヶ月間飼育し屠殺後、肝臓からRNAを抽出した。cDNAを合成後、Affmetrix mouse U74Av2 arrayを用いて遺伝子発現の変化を解析した。その結果Dwarfismにより312の遺伝子発現が変化し、CRにより177の遺伝子発現が変化した。(Tsuchiya et. al., Physiol Genomics. 2004)。その中でdwarf(DF)効果とCR効果が相加的であると思われるものは95遺伝子であった(Tsuchiya et. al., Physiol Genomics. 2004)。さらに、その中からいずれの効果も遺伝子の発現を亢進させる56遺伝子について以下の解析を行った。56遺伝子のうちインターネット上に公開されているEZ retrieve(http://siriusb.umdnj.edu:18080/EZRetrieve/index.jsp)によってデータベース上から転写開始点から上流3000bpを取得できた43遺伝子を抽出した。さらにその43遺伝子の上流の配列を用いて、インターネット上で利用可能なMEME(http://meme.sdsc.edu/meme/)を利用して、高頻度に見られるモチーフ配列を4種類同定した(表1。モチーフ1〜4はそれぞれ、本明細書中の配列番号1〜4に記載の塩基配列と同一)。また、モチーフ2と相同性のある配列を上流にもつ遺伝子群を表2に示した。表中で番号が大きいものほどDF効果、CR効果による遺伝子の発現亢進が高い。
表1で示した配列のうち、モチーフ2について文献検索を行ったところ、この配列にはHNF−4α(hepatocyte nuclear factor−4α)が結合する可能性を見いだした(例えば、Hirota K et. al., Int J Mol Med. 2005 Mar;15(3):487-90.)。そこで、モチーフ2の末端をビオチン化したDNAプローブを用いて、ゲルシフトアッセイをEMSA:electrophoretic mobility shift assay kit(ピアス社)を用いて行った。この実験ではアッセイ系に添加するタンパク質を、in vitro転写・翻訳反応を行い作成した。In vitro転写・翻訳は、Flag−tagをN末端にもつHNF−4αプラスミドに対して、TNT(登録商標) System(プロメガ社)を用いて行った。
その結果、モチーフ2にはHNF−4αが結合する可能性を見いだした(図1)。矢印がモチーフ2とFlag−HNF−4αとの複合体である(レーン2)。抗Flag抗体添加により結合が阻害されバンドが薄くなり(レーン3)、非ビオチン化DNAとの競合反応によりバンドが消失した(レーン4)。Flag−HNF−4αタンパク質なしではこのバンドは見られなかった(レーン1)。
ApoC−IIおよびGstm遺伝子について、モチーフ2と相同性のある転写開始点より上流の配列を含む、約400bpを増幅するようにPCRプライマーを設計し、クロマチン免疫沈降法によるHNF−4αとの結合解析を行った。クロマチン免疫沈降はUpstate社のEZ−Chipキットを用いて行った。反応に用いたゲノムDNAはマウス肝細胞由来培養細胞より抽出した。
結果を図2に示す。矢印が増幅された400bpの塩基とHNF−4の結合物である。ApoC−IIおよびGstm遺伝子の上流にはHNF−4αと結合する可能性のある配列があることが示唆された。
実施例1〜3よりモチーフ2へのHNF−4αの結合が強く示唆された。
分泌型アルカリフォスファターゼ発現用ベクターである、pSEAP2−Controlベクター(クロンテック社)のNheI/BglII部位にモチーフ2を5個直列に並べて導入した。また遺伝子導入効率を測定するためのコントロール用ベクターとして、分泌型ルシフェラーゼ発現ベクター、pMetLuc−Controlベクター(クロンテック社)を用いた。293細胞(ヒト胎児腎臓由来)にFuGENE(登録商標) HDトランスフェクション試薬(ロシュ アプライド サイエンス社)を用いて、これらのレポーター遺伝子と、HNF−4αおよびその転写のコアクチベーターであるPGC−1αとを組み合わせてトランスフェクションした。24時間および48時間後に培養上清を集め、Ready−To−Glow Dual Secreted Reporter System(クロンテック社)により分泌型アルカリフォスファターゼ(SEAP)およびルシフェラーゼの活性を測定した。活性測定には、ルミネッセンスリーダー(アロカ社製:AccuFLEX Lumi400)をもちいて、発光量を15秒間測定した。
その結果、24時間後においてcontrol(タンパク発現ベクター無し)と比較して、HNF−4α発現ベクター(1μg)、HNF−4α発現ベクター(1μg)+PGC−1α発現ベクター(1μg)、HNF−4α発現ベクター(2μg)+PGC−1α発現ベクター(2μg)の順に、レポーターが活性化されることが明らかとなった(図3)。この結果はトランスフェクション後、48時間後でも同様の結果であった(図4)。したがって、モチーフ2は細胞内においてもHNF−4α/PGC−1αが結合し、転写を活性化させる可能性が示唆された。
なお、PGC−1α等のグルコースや脂質代謝関連遺伝子がカロリー制限によりアップレギュレートされること、カロリー制限によってHNF−4αとPGC−1αとが相互作用することが既に報告されている(T. Chiba et al., Molecular and Cellular Endocrinology, Volume 309, Issues 1-2, Pages 17-25)。
実施例4のレポーター遺伝子を恒常的に発現する293T細胞株をG418を用いた選択培養法により樹立した。その細胞を用いて、HNF−4α/PGC−1αの活性を制御することが知られている、インスリン/デキサメタゾン添加によるレポーターの活性化を検討した。活性測定は実施例4と同様に行った。
その結果、インスリン/デキサメタゾン添加後、24時間後の培養上清を用いてアッセイを行ったところ、100nM、1000nMのインスリン/デキサメタゾン添加したものでは、このレポーターが活性化されていることが示唆された(図5)。
したがって、医薬品、および食品成分等の候補分子(被験物質)を本実施例において使用したインスリン/デキサメタゾンの代わりに添加することによって、この細胞のレポーターが活性化されれば、モチーフ2を持つ遺伝子の活性化、つまり表2で示した長寿命や代謝疾患の改善と関連すると考えられる、一連の遺伝子群の転写が活性化されると予測される。そのような分子は、寿命を延長させる効果を有しており、抗老化、老化に伴う疾患または代謝疾患の予防または治療剤等の候補分子として有用であることが示唆される。
実際にマウスの生体内においてもこのレポーターアッセイ系が利用可能かを、約8週間、30%のカロリー制限下で飼育したICRマウス(20週齢)を用いて解析した。マウスの尾静脈よりハイドロダイナミック法に基づく遺伝子導入試薬であるTransIT(登録商標)−EE Hydrodynamic Delivery Solution(Mirus Bio社)を用いてレポーター遺伝子(5μg)を導入し、48時間後に血中に分泌されたSEAPを測定した。活性測定には、ルミネッセンスリーダー(アロカ社製:AccuFLEX Lumi400)をもちいて、発光量を15秒間測定した。
その結果、図6に示すように、自由摂食マウスにおいてはモチーフ2をもつベクターを導入しても、モチーフ2をもたないベクターを導入した場合と同程度のSEAP活性を示した。しかし、カロリー制限マウスにモチーフ2をもつベクターを導入した際には、有意にSEAPの活性が上昇した。このことはマウスの生体内においてもこのアッセイ系が機能し、かつ予測されたようにカロリー制限下においては、レポーター活性が高いことが示唆された。従って、カロリー制限はおそらくHNF−4α/PGC−1αなどの転写因子を活性化させ、モチーフ2と相同性のある遺伝子群の発現を上昇させることが示唆された。
つまり、このことから、一過性および構成的トランスジェニックマウスに、医薬品や機能性食品の候補分子(被験物質)を餌や飲水中に添加して飼育後、血中に分泌されたSEAP活性を定量することにより、生きたまま、その候補分子の抗老化、抗老化疾患作用をスクリーニングすることが可能であると考えられた。

- レポーターベクターであって、
(A)以下の(a)及び(b)から選択されるいずれか1つのヌクレオチド配列であり、且つHNF−4αへ結合し得るヌクレオチド配列、
(a)配列番号1で表されるヌクレオチド配列、及び
(b)配列番号1で表されるヌクレオチド配列において、1又は2個のヌクレオチドが置換されたヌクレオチド配列、
(B)プロモーター、並びに
(C)レポーター遺伝子
を含み;
該プロモーターが、該レポーター遺伝子の転写を駆動し得るように、該レポーター遺伝子の5’側に連結しており;且つ
(A)のヌクレオチド配列が、該プロモーターによる該レポーター遺伝子の転写を調節し得るように、該プロモーターの5’側に連結している、レポーターベクター。 - 請求項1記載のレポーターベクターが導入された形質転換体。
- 請求項1記載のレポーターベクターが導入された非ヒトトランスジェニック哺乳動物。
- 以下の工程を含む、カロリー制限模倣物質のスクリーニング方法:
