実施例1 バナナの葉とバナナの茎の前立腺肥大抑制作用
バナナの葉のメタノール抽出物とバナナの茎メタノール抽出物による前立腺肥大抑制効果を調べるために以下の実験を行った。バナナの葉と茎粗切物をそれぞれメタノールに一日浸漬し、減圧乾固しバナナの葉とバナナの茎それぞれの抽出物を得た。7週齢雄ddYマウス(n=5)の精巣を摘出し、その翌日からバナナの葉抽出物(100mg/kg)、バナナの茎抽出物(100mg/kg)を0.2mlの蒸留水に溶かしたものを経口投与した。同時に男性ホルモンとしてプロピオン酸テストステロン(2.0mg)を0.1mlのコーンオイルに溶かしたものを腹腔内注射した。これを一日一回、10日間行い最終投与日の翌日に解剖をし、前立腺の短径と長径の大きさを測定した。その結果を表1に示す。また同時に精嚢腺を摘出して重量を測定した。その結果を表2に示す。ここでA群(コントロール)は精巣摘出したものにプロピオン酸テストステロンのみ投与、B群は精巣摘出したものにプロピオン酸テストステロンとバナナの葉抽出物を投与、C群は精巣摘出したものにプロピオン酸テストステロンとバナナの茎抽出物を投与したものである。

実施例2 バナナの葉とバナナの茎の前立腺肥大抑制作用
バナナの葉のメタノール抽出物とバナナの茎メタノール抽出物が男性ホルモン抑制作用すなわち抗アンドロゲン作用を有することを調べるために以下の実験を行った。バナナサンプル抽出物の調製は実施例1と同様に行なった。アンドロゲン依存的に増殖することが知られているヒト前立腺ガン由来LNCaP細胞にアンドロゲンを添加し、一定期間培養することで増殖した細胞数の割合を調べた。アンドロゲンの非存在下で7日間培養した場合、細胞数は1.5倍程度に増加するが、アンドロゲンとしてテストステロン(T)2mg/ml添加では約3倍程度にまで増加する。生細胞数はMTT処理により細胞内に生じたホルマザン色素を吸光度計で測定(550nm)することにより算出した。サンプルをアンドロゲンと同時に添加した際の細胞増殖率の変化により、サンプルの抗アンドロゲン作用を評価した。結果を図1に示したとおり、サンプルとしてバナナサンプル(25μg/ml)を添加した場合には、アンドロゲンによる細胞増殖を濃度に依存して顕著に抑制したことから、バナナサンプルには抗アンドロゲン様作用があることが示された。男性ホルモン依存性脱毛は男性ホルモン抑制作用すなわち抗アンドロゲン作用により抑制できることが知られていることから、抗アンドロゲン作用を有するバナナ葉・茎抽出物が男性ホルモン依存性脱毛の抑制剤としても使用可能であることが明らかである。
