神経伝達物質は、あるニューロンまたは神経細胞と別のものとの間の間隔またはシナプス間隙の電気信号を中継する機能を有する化学的「メッセンジャー」である。神経伝達物質は、神経末端に局在する小胞と称される小さな嚢に貯蔵される。電気信号がニューロン末端に到達すると、小胞は神経細胞膜に移動し、その神経伝達物質分子をシナプス間隙に放出する。シナプス前(または送信)ニューロン中に形成される神経伝達物質は間隔を横切って拡散し、隣接したシナプス後(または受信)ニューロンの細胞膜上の結合部位またはレセプター上に閉じ込められる。イオン輸送およびいくつかの酵素の放出または阻害などの種々の生化学的過程は、シナプス後ニューロンにて、神経伝達物質がその表面上のレセプターを占める場合に開始する。その結果、新たな電気信号がシナプス後ニューロンにて生成し、その信号が継続する。

本発明の実践にて用いることができる具体的なシドノンイミン誘導体の製造のための化学反応は、以下にさらに詳細に記載する。この反応の出発物質は市販されている。米国特許3,277,108も参照のこと。
N-フェニルカルバモイル-3-(ベンジル)シドノンイミンの製造
7.5N水性HCl 1.2 mlをベンジルアミン0.94 gおよびシアン化カリウム0.58 gの水2 ml中の混合物中にて撹拌(0℃にて)した。次いでホルムアルデヒド0.7 gを混合物に滴加した。得られた混合物を室温にて2時間撹拌した後、0℃まで冷却した。硝酸ナトリウム0.62 gの水1 ml溶液を混合物に滴加し、次いで7.5N HCl水溶液1.2 mlを冷却しながら加えた。混合物を室温にて1時間撹拌した。エーテルを用いて得られた混合物を三回抽出した。集めたエーテル溶液を無水硫酸ナトリウムで乾燥した。溶媒を減圧留去した。N-ニトロソ-ベンジルアミノアセトニトリル(反応式を参照のこと)を黄色油状物として得た。次いでニトロソ中間体を500 ml HCl エーテル溶液(2.0 M)で処理し、室温にて30分間撹拌した。白色析出物を得、2-プロパノールで再結晶し、白色結晶を得た。このように得られた3-ベンジル-シドノンイミン塩酸塩2.84 gを水25 mlに溶解した。その溶液に炭酸水素ナトリウム1.34 gを0℃にて加えた。次いでフェニルイソシアネート2.45 mlを混合物に滴加し、0℃にて4時間撹拌し、得られた混合物にエーテル10 mlを加え、黄色結晶析出物を得た。メタノールを再結晶用溶媒として用いた。所望の生成物を黄色結晶として得た。
化合物N-フェニルカルバモイル-3-(p-カルボキシルベンジルシドノンイミン (2);N-フェニルカルバモイル-3-(p-メチル-ベンジル)シドノンイミン (3);N-(3',4'-ジクロロフェニル)カルバモイル-3-フェネチル シドノンイミン (4);N-(3',4'-ジニトロフェニル)カルバモイル-3-p-ニトロフェネチル シドノンイミン (5);N-フェニルカルバモイル-3-(フェニルプロピル)シドノンイミン (6);およびN-フェニルカルバモイル-3-(p-フルオロ-ベンジル)シドノンイミン (7)を上記実施例1に記載のものと実質的に同じ手順に従い等量の異なる出発物質に適当に置き換えて合成した。
I. ドーパミン輸送体結合アッセイ[3H]WIN 35,428
本アッセイは、Javitch, J. J. et al., Mol. Pharmacol. 26: 35-44; 1984に記載の手順に従い行った。
A. 組織調製(すべての溶液は氷上にて調製)
1. 雄モルモットからの凍結脳を解凍し(アッセイ緩衝液にて)、50 mM TRIS-HCl中に置く(25℃にて120 mM NaC1でpH 7.4)。線条体を単離する。
2. ポリトロン(Polytron)を用いて組織を20 vols. (w/v)の50 mM Tris-HClに均質化する(25℃にて120 mM NaC1でpH 7.4)。
3. ホモジネートを48,400 x g、4℃にて約10分間遠心分離する。上清を廃棄する。
4. 工程2および3に記載のようにペレットをさらに洗浄する。
5. 結合アッセイに必要となるまで氷上にてペレットを貯蔵する。
6. ポリトロン(設定5;約10秒)を用い、50 mM Tris-HCl(25℃にて120 mM NaC1でpH 7.4)中にてペレットを再懸濁し、最終濃度が8 mg/mlまたは組織4.0 mg/チューブとなるように、初期濃度10 mg/ml(元の湿重量)とする。

2. 組織を加えながら結合反応を開始し、0℃(氷上)にて120分間インキュベートする。
3. 未処理Whatman GF/Cフィルター上でのチューブ内容物の急速なろ過により結合反応を終了する。
4. アッセイチューブを50 mM氷冷TRIS HC1(25℃にて120 mM NaC1でpH 7.4, BSA)で1回すすいだ後、同じ洗浄緩衝液6 x 1 ml/チューブでフィルターを素早くすすぐ。
5. LSM器具を用いてフィルター上にトラップされた放射能を分析する。シンチカクテル中に3時間置く。
物質および試薬
1. [3H]-WIN 35-428を50 mM TRIS HC1(25℃にて120 mM NaC1でpH 7.4)中60 nMの濃度に希釈する。従って、最終リガンド濃度は6 nMである。
2. 非特異的結合を1 x 10-6 M GBR12909の存在下にて残存するものとして定義する(室温キャビネット)。GBR12909 MW=523.5g/mol. 4% DMSOにて可溶化する(超音波分解15分)。アリコートを用いることができる(1E-3)。
3. アッセイの参照化合物はGBR12909であり、4% DMSO中に希釈した後、以下の最終濃度にて行う:1x10-10、5x10-10、2x10-9、5x10-9、1x10-8、2x10-8、5x10-8、1x10-7、2x10-7、5x10-7、1x10-6、5x10-6。
4. 陽性対照GBR12909を最終濃度:2x10-8、1x10-7、5x10-7Mにて行う。
5. レセプターについてのKdは28.0 nMである。
本アッセイはRaisman, R, et al. Eur. Jrnl. Pharmacol. 78: 345-351 (1982)記載の手順を改変して行った。文献(Langer, S., et al., Eur. Jrnl. Pharmacol, 72: 423-4 (1981))も参照のこと。
組織調製
1. hrNET(レセプター生物学;RBNET)を各チューブが0.5マイクロ単位または2マイクロ単位/mlを含むように、アッセイ緩衝液中にて2.5マイクロ単位/mlまで希釈する。
注:100マイクロアッセイ、緩衝液20 mlを加える。
1. 各チューブは以下の成分を含む:
薬物またはビヒクル25 ul
[3H]-ニソキセチン25 ul
組織上清200 ul
2. 組織を加えながら結合反応を開始し、室温にて1時間インキュベートする。
3. 0.1% PEI処理GF/Cフィルター(TopCount)上でのチューブ内容物の素早いろ過により結合反応を終了する。
4. 氷冷50 mM NaCl上にてアッセイチューブをすすいだ後、同じ洗浄緩衝液6 x 1 ml/チューブでフィルターを素早くすすぐ。
5. シンチレーションカクテルに少なくとも1時間浸した後、液体シンチレーション分光光度計を用いてフィルター上にトラップされた放射能を分析する。
1. レセプター源はヒト組換え/CHOである。
2. アッセイ緩衝液中10 nMの濃度に希釈した[3H]-ニソキセチンを放射性リガンドとして用いる。従って、最終リガンド濃度は1 nMである。
3. 非特異的結合は1 x 10-6Mデシプラミン(MW = 302.8)の存在下にて残存するものとして定義される(新鮮なものとする:バッグ内のrocks 1をデシケートしながら、水1E-3に溶解する)。
4. アッセイの参照化合物はデシプラミンである。デシプラミンを以下の最終濃度にて行う:

自発運動(オープン・フィールド研究)
NIDA医薬開発課(MDD)自発運動研究標準プロトコールによれば、実施例3の化合物により誘発される運動低下について用量反応研究を行った。2セットにて音波減衰チャンバー内に収容された40 Digiscan自発運動試験チャンバー(40.5 X 40.5 X 30.5 cm)を用いて本研究を行った。赤外線(16ビーム)および対応する光検出器のパネルを各活動チャンバーの側に沿って水平方向に置いた。各チャンバー上に7.5 W白色光を薄暗い照明で供した。ファンは、チャンバー内で80dB周囲ノイズレベルを提供した。慣らされていない8の雄Swiss Websterマウス(Hsd:ND4, 2-3月齢)の別の群に自発運動試験の20分前にビヒクル(2%メチルセルロース)または上記実施例3の化合物(3、10、30または100 mg/kg)を腹腔内(IP)経路により注射した。器具に置いた直後、すべてのマウスに食塩水を腹腔内注射した。全研究にて、水平活動(光細胞ビームの遮断)は10分以内で1時間測定した。1のマウス/活動チャンバーで試験を行った。
実施例6の化合物により誘発される運動低下の研究と同じプロトコールを続けた。
経時変化(8時間)マウス自発運動試験
実施例3の化合物の運動低下誘発効果の経時変化/用量応答研究は、自発運動試験直前にビヒクル(2%メチルセルロース)または実施例3の化合物(3、10、30または100 mg/kg)を8のマウスの群に注射したことを除いて、同じMDD自発運動研究経時変化プロトコールに従って行った。100 mg/kgの試験化合物投与後30、120および480分にて各マウスの行動観察を記録した。本研究にて用いたビヒクルは2%メチルセルロースであった。
本発明者らは、コカイン乱用および異常ドーパミン再摂取と関連する他の障害の治療および予防における使用のためのDAT選択的阻害剤を認識した。そのような二の阻害剤、すなわち上記実施例3および6の化合物は、少なくとも200 mg/kgまでよく許容される。観察可能な行動変化が数分で起こり、約2-3時間続いた。実施例3の化合物で処理した対象は125 kg/mgを超えた用量にて顕著に不活発となったが、この効果はより高濃度の実施例6の化合物にて減少した。
Figures 2A-2Dに示されるように、さらなる薬物動態研究を行った。同用量における実施例3の化合物と比較した場合に、実施例6の化合物は血漿および脳にてわずかに良いバイオアベイラビリティを示すようだ。実施例6の化合物はまた、異なる用量にて投与される場合に比較的高い経口バイオアベイラビリティを示す。Figure 3を参照のこと。

各マウスについて、5分間隔にて60分間の全路程および路程を自発運動の尺度として測定した。さらに、試験中の各実験動物により生じた糞をカウントした。各チャンバーを個々のマウス試験間に掃除した。
Figure 8に示したデータは、実施例3の化合物がオープン・フィールド試験における自発運動を減少するのに有効であることを示す。さらに別のアッセイにて、同じ化合物が明らかな抗不安効果を生じた。Figures 9A-9Dを参照のこと。

- 請求項1〜4のいずれかに記載の化合物を有効成分として含む、医薬組成物。
- 治療を必要とする患者におけるドーパミン再摂取を阻害することにより緩和された障害を治療し、またはその進行を遅延させるための医薬組成物であって、当該障害が肺疾患;虚血再かん流傷害;心疾患;高プロラクチン血症およびミクロプロラクチノーマ;疼痛;運動障害;ストレス、慢性心的外傷後ストレス障害、不安障害、強迫性障害、分娩後うつ病;統合失調症、躁病、双極性障害および情動障害;実行機能障害;コカイン、アンフェタミン、アルコール依存症、および常習行為;神経内分泌調節障害;炎症性疾患、自己免疫疾患およびリウマチ;腫瘍性疾患;視覚障害、色覚異常;および射精機能不全および関連性機能障害から選択される、請求項5の医薬組成物。
- さらに少なくとも一つの選択的セロトニン再摂取阻害剤を含む、請求項6記載の医薬組成物。
- コカイン依存症、注意力欠如障害、うつ病、統合失調症、ナルコレプシー、肥満症またはパーキンソン病の治療のために、任意で他の少なくとも一つの治療剤とともに投与されるべく構成された、請求項5記載の組成物。
- パーキンソン病の治療用L-dopa;うつ病および/またはコカイン乱用および中毒の治療用選択的セロトニン再摂取阻害剤;統合失調症の治療用ドーパミンD2アンタゴニスト;およびアルツハイマー病または患者が認知障害を有する他の疾患もしくは病態の治療用コリン作動性モジュレータの群から選択される、少なくとも一つのさらなる治療剤と同時にまたは連続して投与されるべく構成された、請求項5に記載の組成物。
- 任意の他の治療剤とともにコカイン依存症の治療のために投与されるべく構成された、請求項8または9記載の組成物。
- 任意の他の治療剤とともに注意力欠如障害の治療のために投与されるべく構成された、請求項8または9記載の組成物。
- 化合物を0.01〜200 mg/患者体重キログラム/日の単位剤形で投与すべく構成された、請求項5または6記載の組成物。
- 医薬的に許容される賦形剤を含む、固形の請求項5〜12のいずれかに記載の組成物。
- 医薬的に許容される希釈剤を含む、液体の請求項5〜12のいずれかに記載の組成物。
- 化合物が経口投与されるべく構成された、請求項5または6記載の組成物。
- 化合物が非経口投与されるべく構成された、請求項5または6記載の組成物。
