本明細書において用いているように、以下の用語は、特に指定しない限り、それらについて記載する意味を有する。
本発明のT1Rsあるいはその断片又は変異体の分離及び発現は、下記のように実施することができる。味覚受容体リガンド結合領域をコードする核酸の増幅にPCRプライマーを用いることができ、そして、これらの核酸のライブラリーを場合によって発生させることができる。次に、これらの核酸又はライブラリーの機能的発現のために、個々の発現ベクター又は発現ベクターのライブラリーを用いて宿主細胞を感染又はトランスフェクトすることができる。これらの遺伝子及びベクターは、in vitro又はin vivoで造り、発現させることができる。当業者は、核酸の発現を変化させ、制御するための所望の表現型は、本発明のベクター内で遺伝子及び核酸(例えば、プロモーター、エンハンサー等)の発現又は活性を調節することにより得ることできることを認識するであろう。発現又は活性を増加又は減少させるための記載された既知の方法のいずれかを用いることができる。本発明は、科学及び特許文献に十分に記載されている当技術分野で知られているいずれかの方法又はプロトコールと組み合わせて実施することができる。
核酸ハイブリダイゼーション法を用いるT1R遺伝子及び遺伝子発現の検出に加えて、T1Rsを検出するために、例えば、味覚受容体細胞並びにT1Rファミリーメンバーの変異体を同定するためにイムノアッセイも用いることができる。イムノアッセイを用いて、T1Rsを定性的又は定量的に分析することができる。適用可能な技術の一般的な概要は、Harlow&Lane、Antibodies:A Laboratory Manual(1988年)に見いだすことができる。
T1Rファミリーメンバーと特異的に反応するポリクローナル及びモノクローナル抗体を生産する方法は、当業者に知られている(例えば、Coligan、Current Protocols in Immunology(1991年)、Harlow&Lane、前出、Goding、Monoclonal Antibodies:Principles and Practice(第2版、1986年)及びKohler&Milstein、Nature、第256巻、495〜497頁(1975年)を参照)。そのような技術としては、ファージ又は同様のベクターにおける組換え抗体のライブラリーからの抗体の選択による抗体の調製並びに免疫化ウサギ又はマウスによるポリクローナル及びモノクローナル抗体の調製などがある(例えば、Huse et al.、Science、第246巻、1275〜1281頁(1989年)、Ward et al.、Nature、第341巻、544〜546頁(1989年)を参照)。
T1Rタンパク質、断片及び変異体は、多くのよく認識されている免疫学的結合アッセイのいずれかを用いて検出し、かつ/又は定量することができる(例えば、米国特許第4,366,241号、第4,376,110号、第4,517,288号及び第4,837,168号を参照)。一般的イムノアッセイの総説については、Methods in Cell Biology:Antibodies in Cell Biology、第37巻(Asai編、1993年)、Basic and Clinical Immunology(Stites&Terr編、第7版、1991年)も参照。免疫学的結合アッセイ(又はイムノアッセイ)では、一般的に最適のタンパク質又は抗原(この場合、T1Rファミリーメンバー又はその抗原性サブ配列)に対して特異的に結合する抗体を用いる。抗体(例えば、抗T1R)は、当業者によく知られている多くの手段のいずれかにより、及び上記のように生産することができる。
サンプル中のT1Rポリペプチドを検出するためのイムノアッセイは、競合的又は非競合的であってよい。非競合的イムノアッセイは、抗原の量を直接測定するアッセイである。1つの好ましい「サンドイッチ」アッセイにおいて、例えば、抗T1R抗体を、それらが固定化されている固体基体に直接結合させることができる。これらの固定化抗体が試験サンプル中に存在するT1Rポリペプチドを捕捉する。T1Rポリペプチドは、そのように固定化され、次いで、標識を帯びている第2T1R抗体のような標識物質により結合される。あるいは、第2抗体は、標識を欠いてもよいが、次に、第2抗体が得られる動物種の抗体に対して特異的な標識第3抗体により結合される。第2又は第3抗体は一般的に、検出可能な部分を得るために、例えば、ストレプトアビジンのような他の分子が特異的に結合することができるビオチンのような検出可能な部分で修飾されている。
競合的アッセイでは、サンプル中に存在するT1Rポリペプチドの量を、サンプル中に存在する未知のT1Rポリペプチドにより抗T1R抗体から置換された(競合脱離した)既知の添加(外因性)T1Rポリペプチドの量を測定することにより間接的に測定する。1つの競合的アッセイにおいて、既知量のT1Rポリペプチドをサンプルに加え、次いで、サンプルをT1Rに特異的に結合する抗体と接触させる。抗体に結合する外因性T1Rポリペプチドの量は、サンプル中に存在するT1Rポリペプチドの濃度に逆比例する。特に好ましい実施形態において、抗体が固体基体に固定化されている。抗体に結合したT1Rポリペプチドの量は、T1R/抗体複合体に存在するT1Rポリペプチドの量を測定するか、あるいは、残りの複合体化していないタンパク質の量を測定して決定する。T1Rポリペプチドの量は、標識されたT1R分子を供給することによって検出することができる。
競合的結合方式のイムノアッセイは、交差反応性の測定にも用いることができる。例えば、本明細書に開示する核酸配列により少なくとも部分的にコードされたタンパク質を固体担体に固定化することができる。固定化抗原への抗血清の結合に対して競合するタンパク質(例えば、T1Rポリペプチド及び同族体)をアッセイに加える。固定化タンパク質への抗血清の結合に対して競合する添加タンパク質の能力を、それ自体と競合する本明細書に開示する核酸配列によりコードされたT1Rポリペプチドの能力と比較する。上記のタンパク質の公差反応性の割合を標準計算法により計算する。上記の添加タンパク質のそれぞれと10%未満の交差反応性を示す抗血清を選択し、プールする。交差反応性抗体は場合により、考慮された添加タンパク質、例えば、関連性が小さい同族体を用いる免疫吸収によりプールした抗血清から除去する。さらに、T1Rファミリーのメンバーを同定するのに用いられる保存モチーフを表すアミノ酸配列を含むペプチドは、交差反応性の判定に用いることができる。
ウエスタンブロット(イムノブロット)分析を用いて、サンプル中のT1Rポリペプチドの存在を検出し、定量する。この手法は一般的に、分子量に基づきゲル電気泳動によりサンプルタンパク質を分離し、分離したタンパク質を適切な固体担体(ニトロセルロースフィルター、ナイロンフィルター又は誘導体化ナイロンフィルター)に移し、サンプルをT1Rポリペプチドに特異的に結合する抗体と共にインキュベートすることを含む。抗T1Rポリペプチド抗体は、固体担体上のT1Rポリペプチドに特異的に結合する。これらの抗体は、抗T1R抗体に特異的に結合する標識抗体(例えば、ヒツジ抗マウス抗体)を用いて直接標識することができるか、あるいは後に検出することができる。
当業者は、イムノアッセイにおける特異的結合を最小限にすることがしばしば望ましいことを認識しているであろう。特に、アッセイが固体基体に固定化されている抗原又は抗体を用いるものである場合、基体への非特異的結合の量を最小限にすることが望ましい。そのような非特異的結合を低減する手段は、当業者によく知られている。一般的に、この手法は、タンパク質性組成物で基体をコーティングすることを含む。特に、ウシ血清アルブミン(BSA)、脱脂粉乳及びゼラチンのようなタンパク質組成物が広く使用されており、粉乳が最も好ましい。
アッセイに用いられる特定の標識又は検出可能な基は、アッセイに用いる抗体の特異的結合を有意に妨げない限り、本発明の重要な態様ではない。検出可能な基は、検出可能な物理的又は化学的特性を有する物質であってよい。そのような検出可能な標識は、イムノアッセイの分野において十分に発達しており、一般的に、そのような方法において有用なほとんどの標識は、本発明に適用することができる。したがって、標識は、分光学的、光化学的、生化学的、免疫化学的、電気的、光学的又は化学的手段により検出可能なあらゆる組成物である。本発明における有用な標識としては、磁気ビーズ(例えば、DYNABEADSTM)、蛍光色素(例えば、フルオレセインイソチオシアネート、テキサスレッド、ローダミン等)、放射性標識(例えば、3H、125I、14C、35S)、酵素(例えば、西洋ワサビ、リン酸アルカリ及びELISAに一般的に用いられている他のもの)コロイド金又は着色ガラス又はプラスチックビーズ(例えば、ポリスチレン、ポリプロピレン、ラテックス等)のような測色標識などがある。
試験化合物が本発明のT1R受容体にin vitro及びin vivoで特異的に結合するかどうかを検討するための組成物及び方法を以下に記載する。本発明のT1Rポリペプチドへのリガンド結合の影響を評価するために、細胞生理学の多くの側面をモニターすることができる。これらのアッセイは、化学感覚受容体を発現する完全な細胞について、透過性化細胞について、あるいは、標準的方法により生成させた膜断片又はin vitroでの新規合成タンパク質について実施することができる。
味覚変換も本発明のT1Rポリペプチドを用いて、可溶性又は固体状態反応によりin vitroで検討することができる。特定の実施形態において、リガンド結合を検定するために、in vitro可溶性又は固体状態反応にT1Rリガンド結合ドメインを用いることができる。
他の実施形態において、蛍光偏光(「FP」)に基づくアッセイを用いてリガンド結合を検出し、モニターすることができる。蛍光偏光は、平衡結合、核酸ハイブリダイゼーション及び酵素活性を測定するための汎用性のある実験室技術である。蛍光偏光アッセイは、遠心分離、ろ過、クロマトグラフィー、沈殿又は電気泳動のような分離段階を必要としない点で均一である。これらのアッセイは、溶液中で実時間で、直接的に行われ、固定化相を必要としない。偏光の測定は迅速で、サンプルを破壊しないので、偏光値は繰り返して、また、試薬の添加後に測定することができる。一般的に、この手法を用いて、低いピコモルからマイクロモルまでの偏光値を測定することができる。この項では、本発明のT1Rポリペプチドへのリガンドの結合を単純かつ定量的方法で測定するのに、どのようにして蛍光偏光を用いるのかを述べる。
他の実施形態において、本発明は、ヘテロオリゴマーT1Rポリペプチド複合体あるいは細胞又は組織同時発現T1Rポリペプチドを用いる可溶性アッセイを提供する。好ましくは、細胞は、機能的T1R1/T1R3(うま味)味覚受容体又はT1R2/T1R3(甘味)味覚受容体を安定に同時発現する。他の実施形態において、本発明は、T1RポリペプチドあるいはT1Rポリペプチドを発現する細胞又は組織を固相基体又は味覚刺激物質に結合させ、T1R受容体と接触させ、適切な標識又はT1R受容体に対して産生させた抗体を用いて結合を検出する、高処理能力方式での固相に基づくin vitroアッセイを提供する。
処理の好ましい実施形態において、T1Rタンパク質又はポリペプチドの組合せを真核細胞中で、非修飾形で、あるいは、分泌経路によりその突然変異及びターゲッティングを促進する異種シャペロン配列を含む、又は好ましくは含まないキメラ、変異体又は切断型受容体として一時的又は安定に同時発現させる。そのようなT1Rポリペプチドは、HEK−293細胞のようなあらゆる真核細胞中で発現させることができる。細胞は、機能的Gタンパク質、例えば、Gα15又は以前に同定されたキメラGタンパク質、あるいは、キメラ受容体を細胞内シグナル伝達経路又はホスホリパーゼCのようなシグナル伝達タンパク質に結合させることができる他のGタンパク質を含むことが好ましい。また、そのような細胞は味覚刺激物質に対する強い応答(同じT1Rの組合せを一時的に発現する細胞に関連)を示すことが見いだされた(実験的実施例に示すように)ので、T1R1/T1R3又はT1R2/T1R3を安定に同時発現する細胞を発生させることが好ましい。そのような細胞中のT1R受容体の活性化は、細胞中のFluo−4依存性蛍光を検出して細胞内カルシウムの変化を検出することによるなどの標準的な方法を用いて検出することができる。そのようなアッセイは、この適用例において示す実験所見の基礎である。
本発明のT1R味覚受容体配列の組合せを発現するヒト以外の動物も受容体アッセイに用いることができる。そのような発現は、化学感覚受容体又はそのリガンド結合領域をコードする核酸を安定又は一時的にトランスフェクトしたヒト以外の動物を試験化合物と接触させることにより、試験化合物が哺乳類味覚膜貫通受容体複合体にin vivoで特異的に結合するかどうかを検討し、受容体ポリペプチド複合体に特異的に結合することにより、動物が試験化合物に対して反応するかどうかを検討するために用いることができる。
T1Rファミリーメンバーの調節物質として試験される化合物は、小化合物あるいはタンパク質、核酸又は脂質のような生物学的物質であってよい。それらの例として、51IMP及び51GMPなどがある。水溶液に溶ける化合物が試験されることが非常に多いが、本質的にあらゆる化合物を本発明のアッセイにおける可能性のある調節物質又はリガンドとして用いることができる。アッセイ段階を自動化し、化合物を都合のよい源から供給することにより、大きい化学ライブラリーをスクリーニングするためのアッセイを設計することができる。これらのアッセイは一般的に並行して実施される(例えば、ロボットアッセイにおけるマイクロタイタープレート上のマイクロタイターフォーマットで)。化学ライブラリーは多くの化学反応のうちの1つ(例えば、Senomyx独占の化学)により合成することができることは認識されよう。さらに、Sigma(ミズーリ州セントルイス)、Aldrich(ミズーリ州セントルイス)、Sigma−Aldrich(ミズーリ州セントルイス)、Fluka Chemika−Biochemica Analytika(スイス、ブークス)等の化合物の多くの供給業者が存在する。
T1R遺伝子及びそれらの相同体は、化学感覚受容体細胞の同定に、法医学及び父性決定に、味覚変換の検討に有用なツールである。T1Rプローブ及びプライマーのようなT1R核酸と特異的にハイブリダイズするT1Rファミリーメンバー特異試薬並びにT1Rポリペプチドに特異的に結合するT1R特異試薬、例えば、T1R抗体は、味覚細胞発現及び味覚変換制御を検討するために用いられる。
本発明を上文で詳細に記述したが、好ましい実施形態を例示するために、以下の実施例を記載する。これらの実施例は、例示を目的とするものであって、本発明の範囲を限定するものではない。
イントロンを含まないhT1R発現構成体をcDNA法及びゲノムDNA法の組合せによるクローンした。全長hT1R1発現構成体を得るために、クローンhT1R1区分(受入番号AL159177)において同定された2つの5’コード化エキソンをPCRオーバーラップにより結合させ、次いで、5’−切断型精巣cDNAクローンに結合させた。hT1R2発現構成体を部分的に配列決定したhT1R2ゲノム区分から得た。2つの欠落hT1R2 5’エキソンを、対応するラットコード配列から得られたプローブを用いてクローンゲノム区分のショットガンライブラリーをスクリーニングすることにより同定した。次いで、コード化エキソンをPCRオーバーラップにより結合させて、全長発現構成体を生成させた。hT1R3発現構成体は、配列決定済みhT1R3ゲノム区分(受入番号AL139287)からPCRオーバーラップにより得た。ラットT1R3を、hT1R3系縮重PCRにより得られたrT1R3エキソン断片を用いてラット味覚組織由来のcDNAライブラリーから分離した。部分hT1R1 cDNA、rT1R2 cDNA及び部分hT1R2ゲノム配列は、Dr.Charles Zuker(カリフォルニア大学、サンディエゴ)から入手した。

上記から選択したクローンT1R配列の対応するラットT1Rsに対するアライメントを行った。図1に示すように、ヒトT1R1、ヒトT1R2及びヒトT1R3並びにラットT1R3の以前に記載したT1Rs(受入番号AAD18069を有するrT1R1及び受入番号AAD18070を有するrT1R2)、ラットmGluR1メタボトロピックグルタミン酸受容体(受入番号P23385)及びヒトカルシウム検出受容体(受入番号P41180)に対するアライメントを行った。比較を明確にするために、mGluR1及びカルシウム検出受容体のC末端を切り離す。7種の可能な膜貫通セグメントを青色の枠で囲む。mGluR1結晶構造中のグルタミン酸塩側鎖カルブチレートに接触する残基は赤色の枠で囲み、グルタミン酸塩αアミノ酸部分に接触する残基は緑色の枠で囲む。サブユニット間ジスルフィドに基づく構成体に含まれているmGluR1及びカルシウム検出受容体システイン残基は、紫色で囲む。これらのシステインは、T1R1及びT1R2には保存されていないが、潜在的に類似のT1R3システイン残基を含むアライメントの分解領域にあり、これも紫色で囲む。
図2に示すように、hT1R2及びhT1R3は味覚組織において発現し、両遺伝子の発現は、切除されたヒト有郭乳頭のRT−PCRにより検出することができる。
Gα15を安定に発現するHEK−293誘導体(Chandradhekar et al.、Cell第100巻、第6号、703〜11頁(2000年))を、10%FBS、MEM非必須アミノ酸(Gibco BRL)及び3μg/mlブラスチシジンを補足したダルベッコ修正イーグル培地(DMEM、Gibco RRL)中で37℃で成長させ、維持した。カルシウム画像化実験のために、細胞を最初に24ウエル組織培養プレートに播種し(約0.1×106細胞/ウエル)、Mirus Translt−293(PanVera)を用いてリポフェクションによりトランスフェクトした。グルタミン酸及びグルコース誘発性脱感作を最小限にするために、トランスフェクションの約24時間後に補足DMEMを低グルコースDMEM/GlutaMAX(Gibco BRL)と交換した。24時間後に細胞にダルベッコのPBS緩衝液(DPBS、Gibco BRL)中3μMのカルシウム色素Fluo−4(Molecular Probes)を室温で1.5時間負荷した。250μlDPBSで置換した後、味覚刺激物質を補足した200μlDPBSを室温で添加して、刺激を行った。カルシウム動員をImaging Workbench 4.0ソフトウエア(Axon)を用いてAxiovert S100TV顕微鏡(Zeiss)でモニターした。T1R1/T1R3及びT1R2/T1R3の応答は、著しく一過性であり、カルシウムの増加が15秒以上持続することはまれであり、非同時性であった。したがって、応答細胞の数の時間的推移は比較的一定であり、したがって、一定の時点、一般的に刺激物質を添加してから30秒後に応答細胞の数を手動で数えて、細胞応答を定量した。
Gα15を安定に発現するHEK細胞をヒトT1R2、T1R3及びT1R2/T1R3で一時的にトランスフェクトし、ショ糖の濃度の増加に応答しての細胞内カルシウムの増加についての測定を行った(図3a)。また、数種の甘味刺激物質についてT1R2/T1R3の用量応答を測定した(図3b)。応答細胞の最大パーセンテージは、異なる甘味料で10〜30%の範囲の差があった。明確にするために、用量応答を応答細胞の最大パーセンテージに対して正規化した。図3の値は、4回の独立した応答の平均値±s.e.である。味覚試験により決定した精神物理学的味覚検出閾値をX軸に丸印で示す。グルマリン(ろ過済み10g/l Gymnema sylvestre水性抽出物の50倍希釈溶液)は、250mMショ糖に対するT1R2/T1R3の応答を阻害したが、20μMイソプロテレノールに対する内因性β2−アドレナリン受容体の応答は阻害しなかった(図3(b))。図3(c)に種々の甘味料(ショ糖、アスパルテーム、D−トリプトファン及びサッカリン)に対するT1R2/T1R3同時発現細胞系の正規化応答を示す。
Gα15を安定に発現するHEK細胞をhT1R2/hT1R3、rT1R2/rT1R3、hT1R2/rT1R3及びrT1R2/hT1R3で一時的にトランスフェクトした。次いで、これらのトランスフェクトした細胞の、350mMショ糖、25mMトリプトファン、15mMアスパルテーム及び0.05%のモネリンに応答しての細胞内カルシウムの増加についての測定を行った。ショ糖及びアスパルテームに関する結果は、図4に示すが、rT1R2/rT1R3も甘味受容体として機能することを示している。また、これらの結果は、T1R2がT1R2/T1R3リガンド特異性を制御している可能性があることを示唆している。
Gα15を安定に発現するHEK細胞をhT1R2及びhT1R3で一時的にトランスフェクトした。これらの細胞にカルシウム色素Fluo−4を負荷し、甘味料に対するそれらの応答を蛍光プレートリーダーを用いて測定した。図5にhT1R2/hT1R3を発現した細胞及びhT1R3のみを発現した細胞(J19−22)のシクラメート(12.5mM)応答を示す。得られた蛍光結果は、これらの味覚刺激物質に対する応答がhT1R2/hT1R3を発現した細胞においてのみ起こることを示している。図6に正規化用量応答曲線を示す。その結果は、甘味刺激物質との用量特異的相互作用があることから、hT1R2とhT1R3が一緒にヒト味覚受容体として機能していることを示している。特に、図6にショ糖、トルプトファン及び他の種々の市販甘味料に対する用量応答を示す。これらの結果は、アッセイにおいて得られた順位序列および閾値がヒト甘味味覚の値を密接に反映しているので、T1R2/T1R3がヒト甘味受容体であることを示している。
図6に示すように、mGluR1の重要なリガンド結合残基がT1R1に保存されている。グルタミン酸塩とmGluR1との相互作用が示され、いくつかの重要な残基が図1と同じ色スキームに従って強調されている。
Gα15を安定に発現するHEK細胞をヒトT1R1、T1R3及びT1R1/T1R3で一時的にトランスフェクトし、グルタミン酸塩の濃度の増加(図8(a))並びに0.5mMグルタミン酸塩、0.2mM IMP及び0.5mMグルタミン酸塩+0.2mM IMP(図8(b))に応答しての細胞内カルシウムの増加についての測定を行った。ヒトT1R1/T1R3の用量応答を0.2mM IMPの存在下及び非存在下でグルタミン酸塩について測定した(図8(c))。応答細胞の最大パーセンテージは、グルタミン酸塩に対して約5%であり、グルタミン酸塩+IMPに対して約10%であった。明確にするために、用量応答を応答細胞の最大パーセンテージに対して正規化した。値は、4回の独立した応答の平均値±s.e.である。味覚試験により決定した味覚検出閾値をX軸に丸印で示す。
6つの残基PDZIP配列(SVSTW(配列番号1))をhT1R2のC末端に融合し、次いで、キメラ受容体(すなわち、hT1R2−PDZIP)を免疫蛍光及びFACS走査データを用いてモニターした。図9A及び図9Bに示すように、PDZIP配列を含めることにより、hT1R2−PDZIPの表面発現がhT1R2と比べて増加した。より具体的には、図9Aに、myc標識hT1R2の免疫蛍光染色により、PDZIPが形質膜上のhT1R2の量を有意に増加させたことが実証されたことを示す。図9Bに同じ結果を示しているFACS分析データを示す。myc標識hT1R2を発現した細胞を点線で、myc標識hT1R2−PDZIPを発現した細胞を実線で示す。特に、図10AにHBS緩衝液中の未トランスフェクトGα15安定宿主細胞を示し、図10Bに甘味料no.5プール(HBS緩衝液中で、サッカリン、シクラミン酸ナトリウムアセスルフェームK及びアスパルテーム−各20mM)中のhT1R2−PDZIPトランスフェクトGα15安定宿主細胞を示し、図10Cに甘味料no.5プール中のhT1R3−PDZIPトランスフェクトGα15安定宿主細胞を示し、図10Dに甘味料no.5プール中のhT1R2−PDZIP/hT1R3−PDZIPコトランスフェクトGα15安定宿主細胞を示す。さらに、図10E〜10Hに、ショ糖−E:HBS緩衝液中0mM;F:30mM;G:60mM及びH:250mMに対するhT1R2/hT1R3コトランスフェクトGα15安定宿主細胞の用量依存的応答を示す。図10I〜10Lに、個々の甘味料−I:アスパルテーム(1.5mM);J:アセスルフェームK(1mM);K:ネオテーム(20mM);L:シクラミン酸ナトリウム(20mM)に対するhT1R2/hT1R3コトランスフェクトGα15安定宿主細胞の応答を示す。図10のカルシウム像により示されているように、hT1R2及びhT1R3は両者とも甘味刺激物質による活性の刺激を必要とする。
それぞれhT1R1又はhT1R2発現構成体(T1R1の場合はプラスミドSAV2485、T1R2の場合はプラスミドSAV2486)及びhT1R3(T1R3の場合はプラスミドSXV550)をそれぞれ含む線状化PEAK10由来(Edge Biosystems)ベクター及びpCDNA3.1/ZEO由来(Invitrogen)ベクターをGα15発現細胞系にトランスフェクトすることにより、ヒトT1R1/T1R3又はヒトT1R2/T1R3を安定に同時発現する細胞系を発生させた。特に、線状化SAV2486及びSXV550を、Gα15を安定に発現するAurora BioscienceのHEK−293細胞系に同時トランスフェクトすることにより、T1R2/T1R3安定細胞系を発生させた。線状化SAV2485及びSXV550を、Gα15を安定に発現する同じHEK−293細胞系に同時トランスフェクトすることにより、T1R1/T1R3安定細胞系を発生させた。SAV22485/SXV550及びSAV2486/SXV550トランスフェクションの後に、プロマイシン耐性及びゼオシン耐性コロニーを選択し、拡大し、カルシウム画像化法により、甘味又はうま味刺激物質に対する応答を試験した。細胞を、GlutaMAX、10%透析済みFBS及び0.003mg/mlブラスチシジンを補足した低グルコースDMEM中37℃で0.0005mg/mlプロマイシン(CALBIOCHEM)及び0.1mg/mlゼオシン(Invitrogen)で選択した。耐性コロニーを拡大し、甘味刺激物質に対するそれらの応答を蛍光顕微鏡により評価した。VIPR−II機器(Aurora Biosciences)を用いた自動蛍光分析画像化法のために、T1R2/T1R3安定細胞を最初に96ウエルプレート(約100,000細胞/ウエル)に播種した。24時間後に、細胞にPBS中0.005mMのカルシウム色素fluo−3−AM(Molecular Probes)を室温で1時間負荷した。70μlPBSで置換した後、味覚刺激物質を添加した70μlPBSを室温で添加して、刺激を行った。化合物の添加の20〜30秒後の蛍光(480nm励起、535nm発光)応答を平均し、化合物添加前に測定したバックグラウンド蛍光で補正し、0.001mMイオノマイシン(CALBIOCHEM)(カルシウムイオノフォア)に対する応答に対して正規化した。
うま味受容体を安定に発現したT1R1/T1R3 HEK293細胞系は、一時的にトランスフェクトした細胞と比較して、活性の頑強な向上を示す。しかし、短所は、細胞増殖中に急速に活性を失うことがあり得ることである。
アルキルカルボン酸であるラクティソールは選択的な甘味阻害物質であると考えられていた(例えば、Lindley(1986年)米国特許第4,567,053号及びSchiffman et al.、Chem Senses第24巻、439〜447頁(1999年)を参照)。T1R2/T1R3をトランスフェクトしたHEK−Gα15細胞の種々の濃度のラクティソールの存在下での150mMショ糖に対する応答を測定した。ラクティソールは、ヒトT1R2/T1R3の活性を阻害し、IC50は24μMである。
Claims (57)
- 少なくとも1種のT1R2ポリペプチドと、少なくとも1種のT1R3ポリペプチドを含む組換え又は単離されたT1R2/T1R3受容体であって、
(1)配列番号6によって表されるか、又は配列番号10によってコードされるヒトT1R2アミノ酸配列に対して、少なくとも90%の同一性を有する少なくとも1種のT1R2ポリペプチドと、
それぞれ配列番号4、配列番号7、配列番号9及び配列番号11によって表されるか又はコードされるヒト、及びラットT1R3アミノ酸配列の何れかに対して、少なくとも90%の同一性を有する少なくとも1種のT1R3ポリペプチドとを含み、甘味刺激物質に特異的に結合及び/又は活性化される受容体。 - 配列番号6によって表されるか、又は配列番号10によってコードされるヒトT1R2アミノ酸配列に対して、少なくとも95%の同一性を有する少なくとも1種のT1R2ポリペプチドと、
それぞれ配列番号4、配列番号7、配列番号9及び配列番号11によって表されるか又はコードされるヒト、及びラットT1R3アミノ酸配列の何れかに対して、少なくとも95%の同一性を有する少なくとも1種のT1R3ポリペプチドとを含み、甘味刺激物質に特異的に結合及び/又は活性化される受容体とを含む、請求項1に記載の受容体。 - 前記T1R2ポリペプチド及び前記T1R3ポリペプチドが同じ動物種に由来する請求項1記載の受容体。
