Description
本発明は、動物の毛の表面と選択的に結合する抗体を提供する。
マウスの免疫化と続くハイブリドーマ技術の使用によって、動物毛特異的抗体を作製した。ビーグルから得た切断毛を、ホモジナイザーを使用して細かく刻んだ。細かく刻んだ毛は、約200μmの長さであった。
モノクローナル抗体B7.3およびB18−4.3を、プロテインGによって、あるいは、限外濾過の沈殿によってハイブリドーマ培養上清から精製した。精製されたタンパク質の収率は、0.1〜1.1mg/mLの範囲であった。精製された抗体を、その後のアッセイに使用した。
抗体結合−エッペンドルフベースのELISA
精製されたモノクローナル抗体B7.3およびB18−4.3の、毛表面との結合を定性的に確認するために、エッペンドルフベースのELISAを使用した。抗体B7.3およびB18−4.3は、特異性を有してヨークシャテリアの毛と結合する。重要なことに、ヒト毛との結合は検出されなかった。
2種の抗体B7.3およびB18−4.3の、毛との結合を定量的に比較できるよう、別の種類のELISAを確立した。イヌ毛を2〜5mm長の断片に切断し、PBS(5mg/ml)に溶解した。この懸濁液200μlを、96ウェルマイクロタイター濾過プレート(Multiscreen HTS、メンブレンポアサイズ1.2μm、Millipore)の各ウェルにピペットで入れた。マイクロタイター濾過プレート中の毛がこれらの手順で減少しないよう、遠心分離によってバッファーおよび基質交換を行う。
抗体重鎖および軽鎖の完全性を確認するために、SDSポリアクリルアミドゲル電気泳動によって精製されたタンパク質を分析した。抗体B7.3は、予測される大きさのインタクトな重鎖および軽鎖を有するが、抗体B18−4.3は、両方とも全長重鎖よりも小さい2種の重鎖バンドを有する(55kDaの代わりに約44および47kDa)(図2)。これらの突然変異体重鎖の一方は、中途停止コドンをもたらすC2ドメインの末端でのフレームシフト突然変異の結果である。もう一方の突然変異重鎖の起源は、まだわかっていない。B7.3およびB18−4.3重鎖の分解産物は観察されず、このことは、精製された抗体の完全性を実証する。
最大結合能を決定するために、規定数のヨークシャテリアの毛(20本の毛、各20mm長)を使用してエッペンドルフベースのELISAにおいてB18−4.3の希釈物を試験した。各エッペンドルフ管中の毛の表面は、3.15mm2に相当していた。ELISAによれば、2.5〜5.0ngの抗体B18−4.3が、3.15mm2の毛表面と結合する。
抗体の機能性に対する結合パートナーの影響を分析するために、精製されたモノクローナル抗体を、ビオチン化し、その後、アビジンビーズと結合させ、これをFACS分析によって確認した(図3)。イヌ毛との結合をエッペンドルフベースのELISAを使用して分析した。ビオチン化抗体B7.3およびB18−4.3の結合が、HRPOにコンジュゲートしているアビジンによって検出され、このことは、ビオチン化および結合パートナーが抗体の機能性に影響を及ぼさないことを実証した。
ビーズとカップリングしているモノクローナル抗体の結合を分析するために、0.5%(v/v)Tween20を含有する1×PBSを用いてウェストハイランドテリアから得た毛を前処理して汚染を除去し、抗原構造に堅くなく固定した。続いて、ビオチン化抗体B18−4.3(0.8mg/mL)を適用し、ストレプトアビジンコーティングしたFITC蛍光ビーズ(Kisker)を使用して結合している抗体を検出した。ビーズの大きさは、0.8μmであった。さらに、ビオチン化抗体B18−4.3を、ストレプトアビジンコーティングしたFITC蛍光ビーズにカップリングし、複合体を、毛サンプルに適用して結合を分析した(図4a)。
表1に示される結果は、表面抗原の少なくとも一部は、毛サンプルを穏やかな洗浄剤で洗浄することによって除去され得るということを示す。抗原構造の除去は、抗体結合の減少につながった。しかし、抗体結合は依然として検出可能であり、このことは、抗原の相当な部分が、毛表面と結合したままであるということを実証した。
モノクローナル抗体の遺伝情報を手に入れるために、抗体結合特異性を決定する可変抗体ドメインをクローニングした。以下の工程を実施した:
・ハイブリドーマクローンからのRNA単離
・cDNAへの逆転写
・PCRによる抗体重鎖および軽鎖に由来する可変領域の増幅
・スプライシングオーバーラップ伸長(splice overlap extension)(SOE)PCRによる短いリンカー断片による両可変ドメインの融合
・一本鎖抗体(scFv)断片の細菌ベクターへのクローニング
・細菌の形質転換
・プラスミド単離および配列決定
各毛サンプルをELISAによって3回測定した。図表は、サンプルの平均および標準偏差を示す。2回のELISAを並行して実施し、各図表は、1回のELISAの結果である。両図表中に同一サンプルが出ている場合には(ウェストハイランドテリア、ヨーロピアンハウスキャット)、1つの毛サンプルを使用して、両ELISAプレートにピペットで入れた。
マウスの免疫化と、それに続くハイブリドーマ技術の使用によってウマ毛特異的抗体を作製した。ウマから得た毛を、PBS−T(1×PBS、Tween20 0.05%)を用いて洗浄し、ホモジナイザーを使用して細かく刻んだ。細かく刻んだ毛は、約200μmの長さであった。
マウスの免疫化と、それに続くハイブリドーマ技術の使用によってネコ毛特異的抗体を作製した。ヨーロピアンショートヘアのネコの切断毛を、ホモジナイザーを使用して細かく刻んだ。細かく刻んだ毛は、約200μmの長さであった。
粒子は、ポリマーからなる。それらは、その大きさが1〜10μmの間であるので毛皮上では目に見えない。有効成分は、被包され、拡散によって継続的に放出される。放出は、粒子設計(ポリマーの種類、分子量、添加剤)によって幅広く制御できる。粒子表面は、抗体の連結に必要である官能基、例えば、アミンまたはカルボキシルで修飾する。
毛特異的抗体の、薬物デリバリー粒子とのカップリングは、以下の方法によって達成した。
抗体を備えた微粒子が、イヌ毛皮と特異的に結合するという証拠は、有効成分の分光学的検出によって与えられる。マルチウェルプレートに、等量(mg)のイヌおよびヒトの毛を部分的に入れる。ウェルの底は、30〜40μmのポアサイズを有するフィルターである。次いで、それらの毛を含有するウェルに、抗体保持微粒子7c_6+7_EEを含有する懸濁液を入れる。次いで、マルチウェルプレートを遠心分離し、上清分散物を30〜40μmポアを通して圧迫することによって、液体および結合していない粒子のすべてを除去する。粒子の大きさは、わずか1〜10μmであるので、毛と結合していない粒子は、フィルターを通過する。毛での粒子結合は、ウェルの内側の毛と接着しているままである。毛を、0.05% Tween20を含有するバッファーで5回洗浄する。各洗浄の後に、遠心分離によってバッファーを再度除去する。この後、薬物を溶解するが、毛またはマルチウェルプレートの材料を溶解しないアセトニトリルをウェルに加える。アセトニトリルは、被包された全薬物を微粒子から抽出する。有効成分の定量的回収は、HPLCによってよりも早く証明された。次いで、ウェルの底で、ポアのない透明なマルチウェルプレートを、毛を含有するマルチウェルプレートの下に入れる。それは、極めてわずかなUV吸収度しか示さないポリマーからなる。アセトニトリルを遠心分離して、透明なマルチウェルプレートのウェルに入れる。アセトニトリル溶液中の有効成分は、268nmのUV光によって検出できる。較正後、UVリーダーは、ウェル中に存在する薬物の量を検出でき、したがって、間接的に、微粒子の、イヌまたはヒトの毛との接着を証明する。多くの微粒子が毛に連結されるほど、UVシグナルはより強い。この相関は、直線的依存型である(図8)。
製剤の1つの有用な実施形態では、抗体を備えた薬物含有微粒子を、動物の毛皮に噴霧製剤として適用する。
本研究の目的は、実験的にクリイロコイタマダニを寄生させたイヌ上で、抗体を介してイヌ毛皮に結合している、フルメトリンを負荷した微粒子を含有する2種の異なる噴霧製剤のマダニに対する有効性評価することであった(表4)。

記載された実施例は、本発明の1つの高度に有用な実施形態を実証する。広い範囲にわたって薬物の放出を制御するのに適している薬物含有微粒子の、動物毛(この場合には、イヌ毛)と特異的に結合する抗体を用いた備え(equipment)。本発明のこの適用には、複数の利点がある:
−寄生虫駆除剤を特定の哺乳類種にターゲッティングさせ、したがって、製剤が適用される場合に、または種々の種が、例えば、遊ぶまたはなめることによって相互作用する場合に、交差適用または、例えば、ヒトもしくはその他の哺乳類種への薬物移動の危険を大幅に低減することを可能にする。
−薬物が動物の毛皮に方向付けられ、それによって、皮膚と大きく接触するのを避けるのを可能にし、これが皮膚刺激作用または重篤な皮膚損傷の可能性を低減する。
−COOH、NH2またはその他の基を介して、単一種特異的抗体と、種々の微粒子表面を連結し、微粒子を毛皮に配置するという原則の普遍性によって、単一製剤のために、種々の異なる微粒子の種類(種々の放出プロフィールを有する種々の薬物を含有する種々のポリマー)を選択することが可能となる。分散物中の種々の種類の微粒子の組み合わせ、それによって、さもなければ化学的に適合しない場合もある薬物を単一製剤中に組み合わせることを許可する。
−適当な薬物放出プロフィールを有する、適した微粒子の使用によって、粒子および薬物の種類に応じて薬物の有効性を数週間または数カ月に延長し(extent)、それによって、よくある局所適用される液体駆除剤およびその他の薬物製剤の有効性期間をかなり延長する可能性を提供することが可能になる。
−微粒子の、動物毛との接着は、例えば、動物が雨天において歩き回る場合、または池でしばらく泳いでいる場合の、水との強い接触またはさらには穏やかな洗浄であっても、打ち勝つのに十分強力である。この場合には、通常の製剤は、新たな適用を頻繁に必要とする。

- イヌ、ネコ、ウシ、ヒツジ、ヤギ、ラクダ、ラマおよびウマの群から選択される1種の動物種の毛の表面と選択的に結合する抗体。
- ポリクローナル抗体、モノクローナル抗体または組換え抗体およびそれらの断片を含む群から選択される、請求項1に記載の抗体。
- 請求項1または2に記載の抗体を含む薬物デリバリーシステム。
