Description
それ故に、目的内での使用では、生体との結合性に富むことで所期する機能を果しえるものである。非特許文献3には、リン酸化合物の表面洗浄化方法として界面活性剤を用いている。リン酸化合物の表面に吸着する界面活性剤などはリン酸化合物の生体との結合性を低減するなどの機能阻害をしてしまう。また、タンパク質をはじめとする生体物質を表面から脱離させることができない。そのため新たな洗浄方法が求められていた。
しかし、目的使用後の再使用や、保管中に感染した生体適合性材料の浄化は不可能とされ、すべて廃棄することとされていた。
この発明により、従来は不可能とされていた生体センサーなどの再利用や、汚染されたインプラントなどの洗浄浄化が可能となり、従来は破棄されるべきものとされていた各種の生体適合性材料を再利用できる可能性を明らかにしたものである。
これにより、現存するそして今後も開発されるであろう各種の生体適合性材料に廃棄量を減少すると共に貴重な資源の再利用を生体材料の世界においても可能にした。
また、リン酸系生体親和性化合物を基材表面に配置する方法としては、従来より各種のものが知られている。
下記実施例に掲げた電気泳動堆積法以外の成膜法、例えばレーザーアブレーション法(上記文献3)やスパッター法により得られた薄膜あるいはその他の方法で得られた薄膜であっても、また、基材にリン酸系生体親和化合物を含有するものであっても本発明は適用可能である。
そして、タンパク質等の内部に存在するアミド結合などの弱い結合を切断する作用は、洗浄液の温度を0℃以上から100℃以下、好ましくは30℃以上から80℃以下、より好ましくは40℃以上から70℃以下にするのが効果的である。
この温度未満であるとラジカル発生が不十分でありタンパク質等の結合切断が進まず、これを超えるとリン酸系生体親和化合物薄膜のはく離などの欠点が生じる。
また、洗浄液としては、実施例に示すような、過酸化水素/アンモニア/水の混合液の他、生体物質の分子内に存在する弱い結合の切断作用を及ぼす液体であれば利用可能である。
当該処理において、洗浄物と接触する酸素の濃度は特に限定されることはないが、30Vol%以下から、好ましくは10Vol%から30Vol%、より好ましくは15Vol%から25Vol%とするのが適切である。
当該範囲よりも低い濃度では、酸素ラジカルの発生が生じなくなり、これを超える高い濃度では発火などの危険性が生じる。
また、紫外線の周波数として、一般に紫外線と認識されている170nmから270nmの範囲のものをさすが、特に周波数が185nmと254nmの併用が好ましく、これ以外の波長範囲であると十分な酸素ラジカルの発生が起きなくなる。
UVの185nmを照射することでチャンバー内にある大気中の酸素が分解され、さらに254nmのUVを照射することでオゾンが発生し、オキシラジカルを形成する。
なお、オゾン雰囲気中への暴露と、洗浄液への浸漬は、以下の実施例とは逆の順序とすることによっても、同様な効果が得られるものである。
図1(a)は、5μm×5μmの面積の表面高さ像であり、この範囲のRMS値 は3.5nmであった。また、図1 (b)、1μm×1μmの面積の表面高さ像であり、この範囲のRMS値 は4.0nmであった。
さらにUVオゾン((185nm、254nm)を10分間行った。この処理(表2No.1)を10回行った際のD−fプロットを図3に示す。
複合処理により吸着量(Δf:Δfn=3/3)・消散エネルギー(ΔD)の再現性があった。
また、洗浄処理及びフィブリノーゲン吸着表面に対する接触角測定結果を図6に示す。0回目と1回目では濡れ性が変化したものの、1回目以降は変化しなかった。(●) APM/UV法、(▲)APM法、(■) UV法
試料1のナノセンサーにフィブリノーゲン(1mg/ml、PBS)を流し、70℃に加熱した過酸化水素/アンモニア/水(5:1:1)およびUVオゾン(185nm、254nm)処理(表2No.1)を10分間行った、1回処理(a)のRMS値は7.3nm、5回処理(b)のRMS値は6.3nm、10回処理(c)のRMS値は6.6nmと処理回数により、表面粗さは殆ど変化しなかった。
複合処理により吸着量(Δf:Δfn=3/3)・消散エネルギー(ΔD)の再現性があった。

(10)は、液洗浄構造であって、アルカリ液洗浄槽(11)中の底部に、液攪拌機構(13)と液加熱ヒータ(14)を配置し、かご状の洗浄物入れ(12)を前記洗浄槽(11)内に出し入れして、前記洗浄槽内で加熱、攪拌されている洗浄液中に、前記洗浄物(S)を浸漬できるようにしてある。
なお、洗浄物入れ(12)の前記洗浄槽(11)への出し入れは、クレーンなどを用いて吊り下げて行う。
(20)は、UVオゾン処理装置で、前記液洗浄構造(10)にて、洗浄された洗浄物(S)にUVオゾン処理する装置である。
ベルトコンベア(22)により、入口から出口に至る間洗浄物(S)を移動するようにしてあるトンネルチャンバー(21)と、このチャンバーの天井に配置した紫外灯(23)と、酸素源(30)からの空気または酸素を前記チャンバー内に供給する酸素(空気)供給口(24)により構成してある。
つまり、酸素源(30)とは具体的には、供給量調整装置(レギュレータ)が付属した酸素ボンベ、空気ボンベもしくは空気供給ポンプのことである。
このようにして、酸素存在下の環境において紫外線を照射することで、オゾンを発生さるとともに、前記チャンバー(21)に入った洗浄物(S)にオゾン環境下で紫外線を照射するようにしてある。
洗浄物(S)への、紫外線照射時間は、前記コンベア(22)の移動速度により、また、チャンバー(21)内の酸素濃度は、酸素源(30)からの供給量により、また、紫外線の波長や強度は、前記紫外灯(23)の種類と同時に点灯する数量および照度により調整できるようにしてある。
なお、酸素源(30)に代わりオゾン発生装置を用いた場合は、チャンバー内への紫外線照射装置の設置は必ずしも必要ではない。
この結果、10%FBSを流した場合、周波数変化が−76.5Hzであった((b)領域)。PBSを流すこと((c)領域)で+22.5Hzと吸着したタンパク質の一部が脱着していることが分かった。さらにSDS溶液を流すこと((d)領域)で吸着したFBSは脱着しないことが明らかとなった。また、表面に吸着したFBS層の粘弾性がSDSを流すことで高くなっていることが分かった。

Claims (4)
- リン酸系生体適合化合物が表面に存在する生体適合性材料の表面浄化方法であって、前記リン酸系生体適合化合物の耐熱温度未満に加熱したアルカリ水溶液からなる洗浄液中に浸漬させる工程と、生体適合性材料をオゾン雰囲気中に暴露する工程とからない、この両工程のいずれかを他方に先行させて処理することを特徴とする生体適合性材料の表面浄化方法。
- 請求項1に記載の生体適合性材料の表面浄化方法において、前記洗浄液が、過酸化水素/アンモニア/水の混合溶液であることを特徴とする生体適合性材料の表面浄化方法。
- 請求項1または2に記載の生体適合性材料の表面浄化方法において、その生体適合性材料は、導電性基板上にリン酸系生体適合化合物を成膜した生体センサーであることを特徴とする生体適合性材料の表面浄化方法。
- 請求項1から3のいずれかに記載の表面浄化方法に用いる洗浄装置であって、前記洗浄液を貯留し、加熱する洗浄槽と、前記生体適合性材料をオゾン雰囲気中に暴露するオゾン暴露手段とからなることを特徴とする浄化装置。
