【発明の属する技術分野】
本発明は抗菌活性を有するカルバペネム化合物合成のためのアゼチジノン誘導体および、その製造方法に関する。

特開昭64-79180号公報において、カルバペネム化合物を構築する際の中間体の製造法が参考例として記載されている。

【発明が解決しようとする課題】
しかし、これらの方法では、収率、用いる試剤の入手容易性と価格、取り扱いやすさ、操作性といった点から工業的製法としてそれほど満足すべきものではなかった。具体的には、例えば、前記式(9)の方法は、2成分の溶媒系で反応を行っており、また、3フッ化ホウ素は、必ずしも取り扱い性が良いわけではない。
また、前記式(11)あるいは(12)の方法は、反応途中に溶媒置換が必要であり、前記式(13)の方法においては、(3S,4S)−3−〔(1R)−1−tert−ブチルジメチルシリルオキシエチル〕−4−〔(1R)−1−フェニルチオカルボニルエチル〕−1−tert−ブトキシカルボニルメチル−2−アゼチジノンをカラムクロマトグラフィー精製しおり、また、4塩化チタンを用いる脱保護反応は2成分の溶媒系で行われており、工業的製法としてはさらに優れた方法が望まれている状況にある。

【課題を解決するための手段】
本発明者らは、上記課題を解決するため鋭意研究を行った結果、後記一般式(2)で表される化合物から、後記一般式(5)で表される化合物が高収率で得られること、そして後記一般式(5)で表される化合物から、温和で簡便な反応条件下、高収率で後記一般式(6)で表される化合物が得られることを見いだし、さらに、後記一般式(8)で表される化合物をチタン化合物で処理し、次いで水または塩酸水で処理することにより、工業的製法に適した次式(1)

で表されるアゼチジノン誘導体一水和物が得られることを見いだし、本発明を完成したものである。

以下、本発明で用いられる種々の定義について詳細に述べる。
Yで表されるシリル基としては、tert−ブチルジメチルシリル基、トリイソプロピルシリル基、トリエチルシリル基、トリメチルシリル基のようなトリアルキルシリル基;フェニルジメチルシリル基のようなジアルキルアリールシリル基;ジフェニルメチルシリル基のようなアルキルジアリールシリル基;トリフェニルシリル基のようなトリアリールシリル基;などが挙げられる。好ましくはtert−ブチルジメチルシリル基、トリイソプロピルシリル基、トリエチルシリル基、トリメチルシリル基のようなトリアルキルシリル基が挙げられる。
Arで表されるアリール基またはヘテロアリール基としては、フェニル基、o−クロロフェニル基、p−クロロフェニル基、2,4−ジクロロフェニル基、2,4,6−トリクロロフェニル基、p−ニトロフェニル基、p−メチルフェニル基、p−メトキシフェニル基などの無置換またはハロゲン原子、ニトロ基、炭素原子数1〜6個のアルキル基、もしくは炭素原子数1〜6個のアルコキシ基で置換されたフェニル基;2−ピリジル基、3−ピリジル基、4−ピリジル基、2−ピリミジル基、2−(4,6−ジメチル)ピリミジル基などのヘテロアリール基;などが挙げられる。好ましくは、フェニル基、p−クロロフェニル基、2,4,6−トリクロロフェニル基などの無置換またはハロゲン原子で置換されたフェニル基が挙げられる。

Zで表される保護基としては、tert−ブチル基、2−メチル−2−ブチル基、ジフェニルメチル基、トリフェニルメチル基などの無置換または置換低級アルキル基が挙げられる。好ましくはtert−ブチル基が挙げられる。
チタン化合物としては、4塩化チタン、4ヨウ化チタンなどが挙げられる。好ましくは4塩化チタンが挙げられる。
Xで表されるハロゲン原子としては、塩素原子、臭素原子、ヨウ素原子などが挙げられる。好ましくは塩素原子が挙げられる。
Rで表される低級アルキル基又はアリール基としては、メチル基、エチル基、イソプロピル基、secブチル基などの、低級アルキル基;フェニル基、p−メチルフェニル基などのアリール基;が挙げられる。

以下、本発明の、化合物(2)から化合物(5)の製造方法、および化合物(5)から化合物(6)の製造方法を以下に詳細に説明する。

(1)工程1
化合物(5)は、化合物(2)を一般式(3)

で表される化合物と反応させ、さらに、一般式(4)

で表される化合物と反応させることにより製造することができる。

本工程は、好ましくは塩基の存在下で行う。この反応に使用される塩基としては、例えば、トリエチルアミン、ジイソプロピルエチルアミン等のトリアルキルアミン、例えば、ピリジン、ルチジン、ピコリン等のピリジン化合物、キノリン、イミダゾール、N−メチルピロリジン、1,5−ジアザビシクロ〔4.3.0〕ノン−5−エン、1,4−ジアザビシクロ〔2.2.2〕オクタン、1,8−ジアザビシクロ〔5.4.0〕ウンデシ−7−エンを挙げることができる。好ましくは、トリエチルアミン等のトリアルキルアミンが挙げられる。
塩基の当量数としては、化合物(2)に対して1〜5当量が使用される。好ましくは1〜2当量が、より好ましくは1〜1.5当量が挙げられる。
反応試剤を加える順序としては、好ましくは、一般式(2)で表される化合物を、上記の塩基と処理し、得られた混合物もしくはその溶液を、一般式(3)で表される化合物もしくはその溶液に加えるか、または、一般式(2)で表される化合物および塩基を、一般式(3)で表される化合物もしくはその溶液に同時に加える。

反応は例えば、ジオキサン、ジエチルエーテル、テトラヒドロフラン等のエーテル類、塩化メチレン、クロロホルム、1,2−ジクロロエタン等のハロゲン化炭化水素類、ベンゼン、トルエン、モノクロロベンゼン等の芳香族炭化水素類、などのような反応に悪影響を及ぼさない常用の溶媒を使用して行う。好ましくは塩化メチレン、クロロホルム、1,2−ジクロロエタン等のハロゲン化炭化水素類が挙げられる。
反応温度は特に限定されないが、通常は冷却下または加温下で行う。好ましくは−10〜5℃が挙げられる。
一般式(3)で表される化合物および一般式(4)で表される化合物の当量数としては、各々化合物(2)に対して0.8〜1.5当量が使用される。好ましくは0.9当量から1.3当量が挙げられる。
一般式(2)で表される化合物を一般式(3)で表される化合物と反応させ、一般式(4)で表される化合物と反応させたあと、さらに引き続いて、一般式(3)で表される化合物と反応させ、一般式(4)で表される化合物と反応させる操作を1度づつもしくは、2度づつ以上繰り返すことにより、収率が向上する。これらの操作の4回以上とは、4回から10回を意味する。この場合の、一般式(3)で表される化合物の当量数は、0.01〜0.5当量が挙げられ、好ましくは0.05〜0.3当量が挙げられ、また、一般式(4)で表される化合物の当量数は0.01〜0.5当量が挙げられ、好ましくは0.05〜0.3当量が挙げられる。また、この場合、塩基を追加してもよく、この場合の当量数は、0.01〜1当量が挙げられ、好ましくは0.05〜0.5当量が挙げられる。2度目以降に加える一般式(3)で表される化合物および塩基は、それぞれ、1回目に用いた一般式(3)で表される化合物および塩基と同じでも異なってもよい。

(2)工程2
化合物(6)は、化合物(5)を反応補助剤の存在下または非存在下、単一溶媒中または混合溶媒中チタン化合物と処理することにより得られる。この工程は、工程1の終了後、引き続いて行うことも可能である。
反応は例えば、ジオキサン、ジエチルエーテル、テトラヒドロフラン等のエーテル類、塩化メチレン、クロロホルム、1,2−ジクロロエタン等のハロゲン化炭化水素類、ベンゼン、トルエン、モノクロロベンゼン等の芳香族炭化水素類、などのような反応に悪影響を及ぼさない常用の溶媒を使用して行う。好ましくは塩化メチレン、クロロホルム、1,2−ジクロロエタン等のハロゲン化炭化水素類が挙げられる。
反応補助剤としては、例えばアニソール、o−ジメトキシベンゼン、m−ジメトキシベンゼン、p−ジメトキシベンゼンなどの芳香族エーテル類があげられる。
反応温度は特に限定されないが、通常は冷却下または加温下で行う。好ましくは0〜20℃が挙げられる。
チタン化合物としては、好ましくは4塩化チタンが挙げられる。用いる当量数としては、化合物(5)に対して1〜10当量が挙げられる。好ましくは、2〜5当量が挙げられる。
後処理としては、水または、塩酸水で処理する方法が挙げられる。好ましくは水または10重量%以下の塩酸水が挙げられる。

上記の工程1、工程2で得られる化合物(5)および(6)は、精製せずに次工程に使用することが可能であるが、必要な場合には蒸留、結晶化、クロマトグラフィー等の単離、精製を行うこともできる。
上記の工程2により得られる化合物(6)から、公知の方法、例えば、特開昭62-103084号公報に開示されている方法、あるいは、Chem. Pharm. Bull. 42(7) 1381-1387, (1994)に記載された方法により、優れた抗菌活性を有することが知られているカルバペネム化合物
を製造することが可能である。

【発明の効果】
本発明により、優れた抗菌活性を有するカルバペネム化合物製造時の重要中間体が、工業的に一層有利な方法で提供されることが可能になった。すなわち、前記一般式(2)で表される化合物から、前記一般式(5)で表される化合物が高収率で製造できること、そして前記一般式(5)で表される化合物から、温和で簡便な反応条件下、高収率で前記一般式(6)で表される化合物を製造することが可能になった。
さらに本発明により、重要な中間体である化合物(1)が濾過操作や取り扱いに便利な態様で得られ優れた工業的製法の提供が可能になった。に適したものとなった。

【実施例】
次に実施例を挙げて、本発明をさらに具体的に説明するが、本発明はこれらによって何ら限定されるものではない。なお、略号の意味は次のとおりである。

(3S,4S)−3−〔(1R)−1−tert−ブチルジメチルシリルオキシエチル〕−4−〔(1R)−1−カルボキシエチル〕−1−tert−ブトキシカルボニルメチル−2−アゼチジノン(235g)、トリエチルアミン(65.8g)、を塩化メチレン(820g)に溶かした液を、クロロ炭酸イソプロピル(76.2g)の塩化メチレン(982g)溶液に−10〜0℃で滴下する。1時間、−10〜0℃で攪拌の後、同温度で、p−クロロチオフェノール(83.4g)を塩化メチレン(250g)に溶かした液を加える。1時間、−10〜0℃で攪拌の後、同温度で、クロロ炭酸イソプロピル(6.9g)とトリエチルアミン(5.7g)を加える。1時間、−10〜0℃で攪拌の後、p−クロロチオフェノール(8.2g)を塩化メチレン(25g)に溶かした液を加え、同温度で1時間、攪拌する。こうして得られた(3S,4S)−3−〔(1R)−1−tert−ブチルジメチルシリルオキシエチル〕−4−〔(1R)−1−p−クロロフェニルチオカルボニルエチル〕−1−tert−ブトキシカルボニルメチル−2−アゼチジノンを含む塩化メチレン溶液に、5〜15℃で、4塩化チタン(375g)を加え、5〜15℃で4時間、攪拌する。この反応液を、3%塩酸水(1200g)と塩化メチレン(1000g)の混合溶液に、0〜20℃で滴下する。滴下終了後、0〜10℃で攪拌し、析出した結晶を濾過し、水洗(2000g)、洗浄(ヘキサン:アセトン=2:1混液(w/w)、700g)の後、乾燥により、(3S,4S)−3−〔(1R)−1−ヒドロキシエチル〕−4−〔(1R)−1−p−クロロフェニルチオカルボニルエチル〕−1−カルボキシメチル−2−アゼチジノンを一水和物の結晶として得た(収量200g、収率90.5%、融点98〜107℃)。

[Problems to be solved by the invention]
However, these methods are not very satisfactory as an industrial production method in terms of yield, availability and price of reagents to be used, ease of handling, and operability. Specifically, for example, in the method of the above formula (9), the reaction is performed in a two-component solvent system, and boron trifluoride is not always easy to handle.
The method of formula (11) or (12) requires solvent substitution during the reaction. In the method of formula (13), (3S, 4S) -3-[(1R) -1- tert-Butyldimethylsilyloxyethyl] -4-[(1R) -1-phenylthiocarbonylethyl] -1-tert-butoxycarbonylmethyl-2-azetidinone is purified by column chromatography, and is removed using titanium tetrachloride. The protection reaction is carried out in a two-component solvent system, and a more excellent method is desired as an industrial production method.

[Means for Solving the Problems]
As a result of intensive studies to solve the above problems, the present inventors can obtain a compound represented by the following general formula (5) in a high yield from a compound represented by the following general formula (2).
