従って、L2蛋白質のこの領域はHPVが細胞に結合・侵入する際に重要な役割を担っており、抗体が結合することでHPVの感染が阻害される機構を想定できる(図8)。
1)Katharina Slupetzkyらは、HIVのgp41蛋白質のB細胞エピトープとされている8アミノ酸の領域を、HPV16型L1蛋白質のアミノ酸286と287の間に挿入、アミノ酸281-287領域を置換、及びC末端に付加の3つの構造を作り、粒子の形成を確認するとともに、アミノ酸282-286領域が抗L1中和抗体のエピトープに関わっていることを報告した(Katharina Slupetzky, et al. : Chimeric papillomavirus-like particles expressing a foreign epitope on capsid surface loops. [3]に記載のキメラ蛋白質を集合させることでキャプシドを形成させることを含む、[1]に記載のキャプシドの製造方法。
本発明のL1-キャプシドは、ヒトパピローマウイルス(HPV)16型L1蛋白質のループ部位に、ヒトパピローマウイルスのL2エピトープ群のうちの少なくとも1つのL2エピトープを挿入したキメラ蛋白質の集合体であるキャプシドである。
本発明者らは、HPV16型のL2蛋白質のアミノ酸108-120領域にL2-エピトープが含まれることを先に報告した(Kawana. K, et al.: Common neutralization epitope in minor capsid protein L2 of human papillomavirus types 16 and 6. J. Virology., 73, 6188-6190, 1999 )。そして、16型のL2-エピトープは、16型だけでなく6、11、52型の感染を阻害することを示した。尚、本発明においては、蛋白質のアミノ末端から順にアミノ酸残基に番号を付け、例えば50番目のアミノ酸をアミノ酸50と記載した。
同様に、L2エピトープ群は、VEESGIVDV で表されるアミノ酸配列において、1または数個のアミノ酸が、欠失、置換、または付加されたアミノ酸からなり、かつ本発明のキャプシドにHPV31型の抗原性を付与するアミノ酸配列を有するL2エピトープの変異体を含むことができる。
同様に、L2エピトープ群は、IEETTFIES、で表されるアミノ酸配列において、1または数個のアミノ酸が、欠失、置換、または付加されたアミノ酸からなり、かつ本発明のキャプシドにHPV52型の抗原性を付与するアミノ酸配列を有するL2エピトープの変異体を含むことができる。
同様に、L2エピトープ群は、IEDSSVVTSで表されるアミノ酸配列において、1または数個のアミノ酸が、欠失、置換、または付加されたアミノ酸からなり、かつ本発明のキャプシドにHPV18型の抗原性を付与するアミノ酸配列を有するL2エピトープの変異体を含むことができる。
同様に、L2エピトープ群は、IEESAIINAで表されるアミノ酸配列において、1または数個のアミノ酸が、欠失、置換、または付加されたアミノ酸からなり、かつ本発明のキャプシドにHPV6/11型の抗原性を付与するアミノ酸配列を有するL2エピトープの変異体を含むことができる。
部位2と部位3に同時にL2エピトープを挿入したキメラL1蛋白質(16L1-140/266VA)は、キャプシドを形成した。
部位1と部位2と部位3に同時にL2エピトープを挿入したキメラL1蛋白質(16L1-56/140/266VA)は、キャプシドを形成した。
L2ペプチド(アミノ酸107-122)の作製は、常法により、96カラム自動ペプチド合成装置でのFmoc固相法により合成した。なお、このペプチドは抗体作製を目的としたため、KLH接合されており、このKLHによるペプチド領域のマスキングを防ぐためにN末にシステイン残基を付加してある。
キメラL1遺伝子を作製し、これをバキュロウイルスベクターを用いて発現させて、キメラL1蛋白質、キメラキャプシドを作製した。キメラL1遺伝子の作製は、PCRによった。詳細は以下の通りである(図23参照)。
(B)挿入部分を持つプライマーの一方とプライマー1またはプライマー2を組み合わせてPCRを行い、
(C)片側に挿入断片を持つDNAを2つ合成した。それぞれの相補部分をアニールさせ、伸長反応を行い、
(D)挿入DNAを持つキメラ遺伝子を作成した。
(E)このDNAを鋳型にプライマー1とプライマー2を用いてPCRを行い、
(F)挿入遺伝子を持つDNAを増幅した。
尚、リン酸緩衝液に0.14モルの食塩を溶かしたものをPBS(phosphate buffered serine)とした。
HPV16L1キメラ遺伝子は、L2-エピトープ領域の塩基配列を持つプライマーを用いたPCRによって作製した。プロトタイプHPV16DNAの6240番目の塩基GをCに置換してL1-キャプシドを形成するように改変したもの(引用文献1)をpCMV plasmid vecter(BD Bioscience Clontech, Palo,CA)にクローニングし、PCRのテンプレートに用いた。
16L1キメラ遺伝子は組み換えバキュロウイルス系で発現させた。Bac-to-Bac baculovirus expression system(Invitrogen Corp., Carlsbad, CA)用いて組み換えウイルスを作り、sf9細胞(夜盗蛾由来細胞)で発現させた。16L1キメラ遺伝子をpFastbac1 vectorのNot IサイトにクローニングしてpFsatbac1/L1chimeraを得た。次にpFsatbac1/L1chimeraをDH10BAC大腸菌(Max Efficiency competent cell containing baculovirus DNA and helper plasmid, Invitrogen Corp., Carlsbad, CA)に導入してBacmidを作成した。 Bacmid DNAをEffectene Transfection Reagent(QIAGEN GmbH, Hilden, Germany)を用いてsf9に導入し、16L1キメラ蛋白質を発現する組み換えバキュロウイルスを得た。
各16L1キメラ蛋白質溶液中のキメラ蛋白質が集合して形成する形態は、5%から45%のショ糖密度勾配溶液(ショ糖をPBSで溶かしたもの)での沈降速度によって検討した。キメラ蛋白質溶液をショ糖勾配液の上に重層し、SW50.1 (Beckman Coulter Inc.)ローターを用いた31000rpm、4℃、2.5時間超遠心した。この条件では、キャプシドはショ糖濃度40%前後の分画に集まるが、凝集した蛋白質はショ糖層を突き抜けて沈査となる。そこで、試料を超遠心した後、底部より約200μlずつ分画し、SDS-PAGE、Western Blotting法でL1キメラ蛋白質がどの分画に含まれるかを調べた。L1キメラ蛋白質がショ糖濃度40%前後の分画に集まった場合は(図11)、透過型電子顕微鏡(Hitachi model H-7600)を使い、陰性染色法で形態を観察した(図12)。
16L1キメラ蛋白質溶液を、ジエチルエーテルで麻酔したBalb/cマウス(6週令、雌)に皮下ないし経鼻接種した。皮下接種はTiter Max Gold (TiterMax USA, Inc.)をアジュバンドとし、アジュバンドと混合することでほぼ等張とした。1回の接種量は蛋白質10μg程度とし、初回接種後、2W、4W、6Wめに追加接種し、4W、6W、8Wに下大静脈より全採血した。経鼻接種では、16L1キメラ蛋白質溶液を滅菌水で希釈して等張とし、一方の鼻腔より滴下した。1回の接種量は蛋白質12μg程度とし、初回投与から毎週4回投与し、初回投与から4週、6週、8週、10週時に下大静脈より全採血を行った。
16L1/L2-キャプシド、16L1-キャプシド、52L1/L2-キャプシド、52L1-キャプシド、58L1/L2-キャプシド、58L1-キャプシド、複数のHPV16のL2蛋白質の断片を抗原とした。キャプシドは、組み換えバキュロウイルス系(Bac-to-Bac baculovirus expression system(Invitrogen Corp., Carlsbad, CA))で発現させ、16L1キメラ蛋白質と同様に精製した。L2蛋白質の断片は、カルボキシ末端側にグルタチオン-S-トランスフェラーゼ(GST)を融合させた構造とし、このtagを使ってアフィニティカラムによる精製を行った。16L2(61-140)-GSTはHPV16L2蛋白質のアミノ酸61から142の領域を含み、16L2Δ-GSTは、16L2(61-140)-GSTのL2部分よりアミノ酸105から126部分(この領域内にL2-エピトープが存在)を欠失させた蛋白質である。16L2(18)-GST、16L2(31)-GST、16L2(6)-GST 、16L2(33)-GST、16L2(35)-GST、16L2(52)-GST、16L2(58)-GSTは、それぞれHPV18、31、6、33、35、52、58型L2蛋白質のアミノ酸105から126を16型L2蛋白質のアミノ酸105から126と置換した蛋白質である。
川名らの方法(引用文献5)に従って、マウス血清の中和活性を測定した。即ち、精製した16L1/L2-キャプシドを2-メルカプトエタノール(2-ME)で還元処理した後、pSV/β-galプラスミドDNA(Promega Corp., Madison, WI)存在下で、2-MEを透析除去し、形成された感染性偽ウイルスを塩化セシウム平衡密度遠心で精製した。感染性偽ウイルスと希釈した血清を1時間室温で反応させた後、2.5mMEDTAで培養プレートからはがしたCOS-1細胞浮遊液に加え、4℃で、60分混和してから培養プレートに播いた。48時間培養してから、β-gal発現細胞を染色した。発色した細胞数を数え、免疫前血清を用いて得られた発色細胞数と比較した。
抗体の中和活性をPastranaらの方法(Diana V. Pastrana, et al. : Reactivity of human sera in a sensitive, high throughput pseudovirus based papillomavirus neautralization assay for HPV16 and HPV18. Virology 321, 205-216, 2004.)を用いて測定した。測定方法の概略を以下に示す。HPV16型L1発現プラスミド、HPV16型L1L2発現プラスミド、分泌型アルカリフォスファターゼ発現プラスミドを混合し、293TT細胞に導入し、48時間後に細胞を回収した。細胞内で生じたHPV16型感染偽ウイルスを、細胞から界面活性剤Brij 58を含む緩衝液で抽出し、Opti Prepを担体とする密度勾配遠心法によって精製した。精製した感染性偽ウイルスを0、 0.01、 0.05、 0.1、 1μlずつ用い、それぞれに最終希釈が 1:10、 1:50、 1:100の濃度になるように抗体を加えた100μlの緩衝液を1時間、4℃に放置した。この溶液を、30000個の293TT細胞培養にかけ、72時間後に培養上清中のアルカリフォスファターゼ活性をルミノメーターで定量した。それぞれの実験は3回繰り返し行いその平均値を求めた。血清成分がアルカリフォスファターゼ活性や感染性偽ウイルスの感染性に影響を与えるために、血清からProtein Gカラムを用いてIgGのみを精製し、抗体として使用した。
1. HPV16型L2蛋白質のアミノ酸109-117領域(VAと呼ぶ)を、L1蛋白質のアミノ酸266と267の間、アミノ酸430と433の間(431と432は失われた)、及びアミノ酸56と57の間、140と141の間、266と267の間の3カ所に挿入したキメラは、キャプシド様構造を形成する(図11及び12)。これらのキメラL1蛋白質をそれぞれ、16L1266VA、16L1430VA、及び16L156/140/266VAと呼ぶ。
また、16L1430VA、または16L156/140/266VAをマウスに免疫して得た抗血清は、16L2(18)-GST、16L2(31)-GST、16L2(6)-GST、16L2(33)-GST、16L2(35)-GST、16L2(52)-GST、16L2(58)-GSTにも結合した(表3)。
5. 16L156/140/266VAをマウスに免疫して得た抗血清は、HPV16型の感染性偽ウイルスの感染を阻害した(図14)。
これまで、HPV16型L2蛋白質のアミノ酸108-120領域に結合する抗体は、HPV52型偽ウイルスとHPV11型HPVの感染阻害活性が示されていることから、L2蛋白質のこの領域に抗体が結合すれば、感染阻害が起こると考えられる。(引用文献6、7)
16L1266VA、16L1430VA、及び16L156/140/266VAは複数のHPV感染を予防するためのワクチン抗原の候補である。

- ヒトパピローマウイルス(HPV)16型L1蛋白質のループ部位である(1)アミノ酸430と433の間、但し、アミノ酸431及び432は欠失、(2)アミノ酸266と267の間、(3)アミノ酸140と141の間及びアミノ酸266と267の間、または(4)アミノ酸56と57の間、アミノ酸140と141の間及びアミノ酸266と267の間に、VEETSFIDA、 VEESGIVDV、IEETTFIES、IEESSFIDA、IEDSSVVTS、またはIEESAIINAで表されるアミノ酸配列からなる、ヒトパピローマウイルスのL2エピトープ群のうちの少なくとも1つのL2エピトープを挿入したキメラ蛋白質の集合体であるキャプシド。
- L1-キャプシドワクチンの製造に用いられる請求項1に記載のキャプシド。
