Description
【0001】
[Amaduci, L.; Fratiglioni, L."Epidemiology of AD:Impact on the treatment", in Alzheimer Disease : Therapeutic Strategies, E. Giacobini and R. Becker, Eds., Birhauser, EEUU, 1994, pp. 8]。
【0004】
["Alzheimer Disease: From molecular biology to therapy", E. Giacobini and R. Becker, Eds., Birhauser, EEUU, 1996].
【0005】
これらは主に、異常に過剰リン酸化された状態の微小管結合タンパク質(MAP)タウを含む[Grundke-Iqbal, I.; Iqbal, K.; Tung, Y. C.; Quinlan, M.; Wisniewski, H. M.; Binder, L. I.,"Abnormal phosphorylation of the microtubule-associated protein tau in Alzheimer cytoskeletal pathology", Proc. Natl. Acad. Sci. USA, 1986,83, 4913-4917; Grundke-Iqbal, I. ; Iqbal, K.; Quinlan, M.; Tung, Y. C.; Zaidi, M. S.;Wisniewski, H. M.,"Microtubule-associated protein tau. A component of the Alzheimer paired helical filaments", J Biol. Chem., 1986,261,6084-6089; Greenberg, S. G.; Davies, P.; Schein, J. D.; Binder, L. I.,"Hydrofluoric acid-treated tau PHF proteins display the same biochemical properties as normal tau.", J. Biol. Chem., 1992,267,564-569]。タウのこうした異常なリン酸化は、様々なタンパク質キナーゼ及びホスファターゼの効果によって決定されるものであるが、微小管に結合してこれを安定化するというその能力と折衷しているようであり、これはADの病理に貢献しうる[Moreno, F. J.; Medina, M.; Perez, M.; Montejo de Garcini, E.; Avila, J.,"Glycogen sintase kinase 3 phosphorylation of different residues in the presence of different factors: Analysis on tau protein", FEBS Lett., 1995,372,6568]。
グリコーゲンシンターゼキナーゼ3β(GSK-3β)が、脳内において生体内タウキナーゼであることは疑いがない[Lovestone, S.; Hartley, C. L.; Pearce, J.; Anderton, B. H.,"Phosphorylation of tau by glycogen synthase-3 in intact mammalian cells: the effects on the organization and stability of microtubules", Neuroscience, 1996,73,1145-1157; Wagner, U.; Utton, M.; Gallo, J. M.; Miller, C. C.,"Cellular phosphorylation of tau by GSK-3β influences tau binding to microtubules and microtubule organisation", J. Cell. Sci., 1996,109,1537-1543; Ledesma, M.; Moreno, F. J.; Perez, M. M.; Avila, J., "Binding of apolipoprotein E3 to tau protein: effects on tau glycation, tau phosphorylation and tau-microtubule binding, in vitro", Alzheimer Res., 1996,2,85-88]。
【0011】

BACKGROUND OF THE INVENTION
The present invention relates to enzyme inhibitors, and in particular to glycogen synthase kinase 3β, GSK-3.
[0002]
[Prior art]
Alzheimer's disease (AD) is a neurofibrillary tangle that is a progressive deterioration of cholinergic function, neuropathological lesions as senile plaques formed by fibrotic β-amyloid, and bundles of spiral fibrils Is a neurodegenerative pathological condition characterized by cognitive impairment.
[0003]
Generally speaking, AD is limited to groups over 60 years of age and is the most common cause of dementia in the elderly population. Today, AD affects 230,000 people around the world. As lifespan increases, the number of AD cases is estimated to exceed three times by 2050 [Amaduci, L .; Fratiglioni, L. "Epidemiology of AD: Impact on the treatment", in Alzheimer Disease: Therapeutic Strategies, E. Giacobini and R. Becker, Eds., Birhauser, EEUU, 1994, pp. 8].
[0004]
In AD brain with neuronal loss, two major histological lesions, neurofibrillary tangles and senile plaques, are observed at intracellular and extracellular levels, respectively. ["Alzheimer Disease: From molecular biology to therapy", E. Giacobini and R. Becker, Eds., Birhauser, EEUU, 1996].
[0005]
Nerve fibrils are structures formed by paired spiral fibrils (PHFs). These mainly contain microtubule-associated protein (MAP) tau that is abnormally hyperphosphorylated [Grundke-Iqbal, I .; Iqbal, K .; Tung, YC; Quinlan, M .; Wisniewski, HM; Binder, LI, "Abnormal phosphorylation of the microtubule-associated protein tau in Alzheimer cytoskeletal pathology", Proc. Natl. Acad. Sci. USA, 1986, 83, 4913-4917; Grundke-Iqbal, I .; Iqbal, K .; Quinlan, M .; Tung, YC; Zaidi, MS; Wisniewski, HM, "Microtubule-associated protein tau. A component of the Alzheimer paired helical filaments", J Biol. Chem., 1986, 261, 6084-6089; Greenberg, SG; Davies, P .; Schein, JD; Binder, LI, "Hydrofluoric acid-treated tau PHF proteins display the same biochemical properties as normal tau.", J. Biol. Chem., 1992, 267, 564-569]. This abnormal phosphorylation of tau, which is determined by the effects of various protein kinases and phosphatases, seems to be a compromise with its ability to bind to and stabilize microtubules. Can contribute to the pathology of AD [Moreno, FJ; Medina, M .; Perez, M .; Montejo de Garcini, E .; Avila, J., "Glycogen sintase kinase 3 phosphorylation of different residues in the presence of different factors : Analysis on tau protein ", FEBS Lett., 1995,372,6568]. And blocking this hyperphosphorylation process can be the primary goal for disrupting the pathogenic cascade. Selective inhibitors of tau kinase may be new effective drugs for the treatment of AD.
[0006]
[Problems to be solved by the invention]
The search for tau kinase inhibitors is an area of great interest. Tau can be phosphorylated by several proline specific protein kinases (PDKs) and non-PDKs.
[0007]
However, in AD, the exact role of any of these kinases in aberrant hyperphosphorylation of tau has not yet been found to date, and the activity of these kinases has not been found to be up-regulated. . There is no doubt that glycogen synthase kinase 3β (GSK-3β) is an in vivo tau kinase in the brain [Lovestone, S .; Hartley, CL; Pearce, J .; Anderton, BH, “Phosphorylation of tau by glycogen synthase -3 in intact mammalian cells: the effects on the organization and stability of microtubules ", Neuroscience, 1996,73,1145-1157; Wagner, U .; Utton, M .; Gallo, JM; Miller, CC," Cellular phosphorylation of tau by GSK-3β influences tau binding to microtubules and microtubule organization ", J. Cell. Sci., 1996,109,1537-1543; Ledesma, M .; Moreno, FJ; Perez, MM; Avila, J.," Binding of apolipoprotein E3 to tau protein: effects on tau glycation, tau phosphorylation and tau-microtubule binding, in vitro ", Alzheimer Res., 1996, 2,85-88]. These discoveries pave the way for the use of GSK-3β inhibitors as therapeutic agents in the treatment of AD. At present, few compounds are known to have this enzyme inhibition property.
[0008]
Lithium behaves as a specific inhibitor of the GSK-3 family of protein kinases in vivo and in whole cells [Munoz-Montano, JR; Moreno, FJ; Avila, J .; Diaz-Nido, J., "Lithium inhibits Alzheimer's disease-like tau protein phosphorylation in neurons ", FEBS Lett., 1997,411,183-188].
[0009]
Finally, it has been observed that insulin inactivates GSK-3 and it has been shown that non-insulin dependent diabetes occurs with activation of this enzyme. Thus, GSK-3 inhibitors may be a new treatment for non-insulin dependent diabetes.
[0010]
Our team recently discovered a new group of small synthetic heterocyclic molecules with GSK-3β inhibitory properties at the micromolar level.

[0035]
Exemplary compounds of the present invention selectively inhibit GSK-3β without inhibiting other protein kinases such as PKA, PKC, CK-2, and CdK2, which can eliminate the widespread effects. GSK-3β is involved in the pathogenesis of AD and is responsible for abnormal hyperphosphorylation of tau protein. The selective inhibitors disclosed herein can be useful therapeutic agents for the treatment of neurodegenerative diseases associated with tau protein pathology, particularly for AD which forms part of the present invention. The inhibitory action of these compounds on GSK-3β enables the design of drugs that can prevent the formation of neurofibrillary tangles, one of the evidence that exists in this neurodegenerative process.
[0036]
These compounds may also be useful for the treatment of other pathologies involving GSK-3β, such as non-insulin dependent diabetes.
[0037]
Furthermore, these compounds are hyperproliferative, such as dysplasia or metaplasia of various tissues, psoriasis, arteriosclerosis, restenosis, and cancer, by inhibiting these cell cycles that form part of the present invention. Can be useful for the treatment of disease.
[0038]
Accordingly, the present invention further provides pharmaceutical compositions comprising a compound of the present invention together with a pharmaceutically acceptable carrier or diluent. Appropriate dosage forms and administration rates can be devised in view of conventional examples.

Example 1 Enzyme Inhibition of a Compound of the Invention
GSK-3β inhibition:
By incubating a mixture of GSK-3 enzyme (Sigma), phosphate source, and GSK-3 substrate in the presence and absence of the corresponding test compound and measuring the GSK-3 activity of this mixture, GSK-3 activity was determined.
[0040]
Specifically, GSK-3 activity was determined by supplementing the enzyme with 15 μM (final concentrated) synthetic peptide GS 1 [Woodgett, JR “Use of peptides for affinity purification of protein-serine kinases”, Anal. Biochem ., 1989,180,237-241], final volume of 12 μl buffer as substrate (50 mM tris, pH = 7.5, 1 mM EDTA, 1 mM EGTA, 1 mM DTT, 10 mM Cl 2 Mg), 15 μM ATP, 0.2 Determined by incubation for 20 minutes at 37 ° C. in μCi [γ- 32 P] ATP and various concentrations of test compound. The reaction is quenched using phosphocellulose p81 paper and adding an aliquot of the reaction mixture. These papers are washed three times with 1% phosphoric acid and the radioactivity contained in the GS1 peptide is measured with a liquid scintillation counter.

The compounds shown in Table 3 are representative examples of the GSK-3 inhibitory active substance of the present invention. IC 50 (indicating the concentration at which enzyme inhibition is 50%) values are summarized in Table 5 below.
[Table 5]
[0042]
GSK-3 inhibition:
Inhibition experiments were performed at various concentrations of ATP (up to 50 μM) and in all cases the same IC 50 value was obtained. Thus, it can be said that thiadiazolinedione does not compete with ATP for binding to GSK-3.
The first four compounds were assayed for inhibition of another enzyme.
[0043]
Protein kinase A (PKA) inhibition:
Possible inhibition of this enzyme is assessed by measuring esthatmine phosphorylation by protein kinase A (PKA). Ezatomin was purified according to the procedure disclosed by Belmont and Mitchinson (Belmont, LD; Mitchinson, TJ "Identification of a protein that interacts with tubulin dimers and increases the catastrophe rate of microtubule", Cell, 1996, 84, 623-631).
[0044]
Specifically, purified PKA (Sigma, catalyzed subunit from bovine heart (p 2645)) and 10-15 μg substrate (ezatomin), 25 μl total volume containing 20 μM (γ- 32 P) ATP. In buffer solution. cAMP kinase protein (100 ng / 1 single reaction), of 50 [mu] l, were 25mM Hepes, MgCl 2, 2 mM EGTA, the PH7.4,20MM, 2 mM dithiothreitol, to act in 0.5mM of Na 3 V0 4 . After the reaction took place, quenching buffer was added, the reaction mixture was boiled at 100 ° C. for 5 minutes, and the phosphorylated protein was characterized by gel electrophoresis and quantified by autoradiography.
Under these conditions, none of the assayed compounds showed any PKA inhibition.
[0045]
Protein kinase C (PKC) inhibition:
Possible inhibition of this enzyme is assessed by measuring phosphorylation of the peptide PANKTPPKSPGEPAK by protein kinase C (PKC) using phosphatidylserine as a stimulant (Woodgett, JR "Use of peptides for affinity purification of protein-serine kinases ", Anal. Biochem., 1989, 180, 237-241). The subsequent method is the same as described above for GSK-3.
[0046]
Specifically, PKC purified from rat brain according to the method disclosed by Walsh (Walsh, MP; Valentine, KA; Nagi, PK; Corruthers, CA; Hollenberg, MD Biochem. J., 1984, 224, 117-127). And 1-10 mM substrate was used in 25 μl total volume of optimized buffer solution containing 10 μM (γ- 32 P) ATP.
Under these conditions, none of the assayed compounds showed any PKC inhibition.
[0047]
Casein kinase 2 (CK-2) inhibition:
The phosphorylation activity of this enzyme against ezatomin was determined by the method disclosed by Alcazar (Alcazar, A .; Marin, E .; Lopez-Fando, J .; Salina, M. "An improved purification procedure and properties of casein kinase II from brain ", Neurochem. Res., 1988,13,829-836), CK-2 purified from bovine brain with 3.6 μM substrate and 20 μM (γ- 32 P) ATP containing a total volume of 25 μl Measured using in buffer solution. CK-2 assay using ezatomin as substrate (see PKA measurement), 50 μl, 25 mM Hepes, pH 7.4, 20 mM MgCl 2 , 2 mM EGTA, 2 mM dithiothreitol, 0.5 mM Na 3 VO 4 And 100 ng of purified CK-2. After the reaction took place, the same method as described for PKA was performed.
Under these conditions, none of the compounds assayed showed any CK-2 inhibition.
[0048]
Cyclin-dependent protein kinase 2 (Cdc2) inhibition:
The phosphorylation activity of this enzyme against histone H1 was determined by the method disclosed by Kobayashi (Kobayashi, H .; Stewart, E .; Poon, RY; Hunt, T. "Cyclin A and cyclin B dissociate from p34cdc2 with half-times of 4 and 15 h, respectively, regardless of the phase of the cell cycle ", J BioL Chem., 1994, 269, 29153-29160), and Cdc2 (Calbiochem) with 20 μM (γ- Used in a suitable buffer solution of 25 μl total volume containing 32 P) ATP. Cdc2 assay using histone H1 as substrate (see PKA measurement), 50 gl buffer, pH 7.5, 50 mM Tris-HCI, 10 mM Cl 2 Mg, 1 mM DTT, 1 mM EGTA, lOOμM ATP was performed in 0.01% BRIJ-35. After the reaction took place, the same method as described for PKA was performed.
Under these conditions, none of the assayed compounds showed any Cdc2 inhibition.
[0049]
Example 2-Analysis of neurites growth after drug treatment
Cells were maintained in Dulbecco medium (DEMEM) with 10% fetal bovine serum, glutamine (2 mM), and antibiotics. For analysis of possible in vivo GSK-3 inhibition, the mouse neuroblastomas N 2 A culture (Garcia-Perez, J .; Avila, J .; Diaz-Nido, J. “Lithium induces morphological differentiation of mouse neuroblastoma ", J. Neurol. Res., 1999, 57, 261-270) was used. Test compounds were added to these cell cultures. This cell line is characterized by a certain neuronal phenotype (neuritic elongation) after the addition of the known GSK-3 inhibitor lithium chloride (10 mM). After 2-3 days of culture, the effects of the test compounds were confirmed and summarized in Table 3. It was observed that neuritic extension occurred at the same extension as when lithium chloride was added. This fact supports GSK-3 inhibition of the compounds of the present invention in vivo.
[0050]
Example 3-Cell cycle block
In parallel, the cell cycle interference that these compounds can exert was studied in N 2 A cells. Cell cultures were maintained in Dulbecco medium (DEMEM) with 10% fetal bovine serum, glutamine (2 mM), and antibiotics.
[0051]
The first four compounds of general formula (I) summarized in Table 5 were assayed under the described conditions and showed the ability to inhibit the cell cycle at inhibitor concentrations from 100 nM to 1 μM. Cell block was first observed at a concentration of 100-200 nM and was fully effective at 1 μM.
The test compound was nontoxic in static fibroblast culture MRC-5 after 10 days of continuous exposure to the inhibitor.
[0052]
Example 4-GSK-3 inhibition of further compounds
[Table 6]
[0053]
GSK-3 inhibitors:
For compounds belonging to group D, further experiments with GSK-3 inhibitors were carried out at various concentrations of ATP (up to 50 μM), and in all cases the same IC 50 value was obtained. Therefore, it can be said that these compounds did not compete with ATP in binding to GSK-3.
[0054]
Example 5-Cell cycle block
IC 50 values are summarized in Table 7 below for some of the compounds tested in N 2 A cell culture.
