本発明は、Gタンパク質共役型受容体(GPCR)と特定のポリペプチドとのタンパク質複合体を提供する。本明細書中で使用される場合、用語「複合体」は複数の物質があわさって一体をなしているものが意図され、「複合体化」は「一体化」と交換可能に使用される。なお、複合体を形成する複数の物質は、近接して相互作用していればよく、互いに結合していても結合していなくてもよい。好ましい実施形態において、本発明に係るタンパク質複合体は、GPCRと特定のポリペプチドとが近接して相互作用している状態であり、その結果、リガンド親和性が改変されたGPCRとして機能する。

本発明はまた、GPCRと特定のポリペプチドとのタンパク質複合体の作製方法を提供する。本発明に係るタンパク質複合体の作製方法は、GPCRおよび特定のポリペプチドを脂質膜上に共存させる工程を包含することを特徴としている。すなわち、本発明は、タンパク質複合体を含有している脂質膜およびその作製方法を提供する。
上述した脂質膜の作製方法が、タンパク質複合体の作製方法であり得、GPCRのリガンド親和性を改変する方法でもあり得ることを、当業者は容易に理解する。すなわち、本発明は、GPCRのリガンド親和性を改変する方法を提供する。
本発明はまた、GPCRとタンパク質複合体を形成するポリペプチドをノックダウンした形質転換体およびその作製方法を提供する。本発明に係る形質転換体の作製方法は、内因性のポリペプチドの発現を阻害する工程を包含することを特徴としている。
ヒトアドレナリン受容体alpha−1A遺伝子(ADRA1A)を、ヒト前立腺ライブラリからPCR法を用いてクローニングした。クローニングした遺伝子は、全長1465bpであり、ORF配列が従来報告されているヒトalpha−1A遺伝子(NCBIアクセッション番号:U03866またはNM_000680)と完全に一致した。また、その前後の配列も、Hirasawa et al. (1993)の報告のものと一致した。
通常の方法で上記のベクターを増幅/精製し、Chinese Hamster卵巣由来細胞であるCHO−K1細胞に、Lipofectamine 2000(Invitrogen社)を用いて、製造業者のプロトコールに従ってトランスフェクトした。トランスフェクトの2日後より、抗生物質ジェネティシン(G418) 1200μg/mlをDMEM培地に添加した。トランスフェクトしなかった細胞、すなわち、Lipofectamine 2000などの操作を全て同様に行い、ベクターのみを加えなかった細胞が完全に死滅するG418の濃度は、1000μg/mlであった。G418耐性細胞を1穴あたり平均0.5個の細胞が入るように希釈懸濁して、96穴プレートに分注し、単一コロニーを選択した。この操作を3回繰り返すことにより、ADRA1A遺伝子とCRELD1α遺伝子を共発現したstable clone(α1L細胞)を作製した。また、コントロールとして、ADRA1A遺伝子のみを安定発現した細胞(α1A細胞)およびCRELD1α遺伝子のみを安定発現した細胞(CRELD1α細胞)を作製した。
結合実験に用いた薬物とその正式化学名は以下の通り。silodosin, prazosin, tamsulosin, RS−17053 (N−[2−(2−cyclopropylmethoxyphenoxy)ethyl]−5−chloro−α,α−dimethyl−1H−indole−3−ethanamine hydrochloride), BMY 7378 (8−[2−[4−(2−methoxyphenyl)−1−piperazinyl]ethyl]8−azaspiro[4,5]decane7,9−dione dihydrochloride)。
α1L細胞に対して、種々の濃度(30〜500pM)の[3H]−silodosinの結合を調べた(図1)。図1は全細胞法で得られた飽和結合曲線を示す。[3H]−silodosinのα1L細胞に対する特異的結合は、最大結合量(Bmax) 134fmol/mg proteinであり、Kd値 843pM(pKD=9.1)であった。
silodosinはα1Aサブタイプとα1Lサブタイプにしか高親和性に結合しないことが知られている。ここで、高親和性とは、Kd値が1nM以下であることを意味する。α1L細胞に発現する受容体の薬理学的特性を同定するため、300pM [3H]−silodosinの結合部位に対するprazosinの競合結合実験を、全細胞結合実験法を用いて行った。対照には、α1A細胞を用いた。上記の通り、この濃度では、[3H]−silodosinはα1Aサブタイプおよびα1Lサブタイプにしか結合することができない。
さらに、α1A細胞に内因性に発現している可能性のある、CRELD1αの影響を完全に取り除くために、RNAi法を用いて、CRELD1αの発現をノックダウンした細胞(以下、KD細胞)を作製した。RNAi法を用いて遺伝子発現を抑制するために、市販のpSilencerTM4.1-CMV hygro(Ambion社)に対して、その取扱説明書に従って、chinese hamster CRELD1αのmRNAのコード領域を含んだオリゴヌクレオチド(センス配列;GATCCAGGCGACTTAGTGTTCACCTTCAAGAGAGGTGAACACTAAGTCGCCTTTA:配列番号5、アンチセンス配列;AGCTTAAAGGCGACTTAGTGTTCACCTCTCTTGAAGGTGAACACTAAGTCGCCTG:配列番号6)を挿入したベクターを作製し、上述したα1A細胞に上記と同様の手法でトランスフェクトした。このKD細胞を用いて、蛍光色素Fura−2を用いた蛍光比測光法により、ノルアドレナリンに対する細胞内Ca2+応答を調べた。KD細胞のノルアドレナリンによるCa2+応答のpEC50値は、7.9であった。一方、この細胞を、プラゾシン10−8Mにて2分間予め処置しておくと、ノルアドレナリンに対するCa2+応答の用量反応曲線は右方に移動し、プラゾシンのpKB値は9.3と計算された。このことから、KD細胞におけるノルアドレナリンに対するCa2+応答は、α1Aサブタイプの特性と一致した。
Claims (13)
- Gタンパク質共役型受容体α 1L を含有している脂質膜を作製する方法であって、
Gタンパク質共役型受容体α 1A および以下のポリペプチド:
(1)配列番号1に記載のアミノ酸配列からなるポリペプチド;
(2)配列番号1に記載のアミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;
(3)配列番号2に記載の塩基配列からなるポリヌクレオチドにコードされるポリペプチド;
(4)配列番号2に記載の塩基配列において1もしくは数個の塩基が欠失、置換もしくは付加された塩基配列からなるポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;あるいは
(5)配列番号2に記載の塩基配列の相補配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし得るポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド
を脂質膜上に共存させる工程を包含する、作製方法。 - Gタンパク質共役型受容体α 1L を発現している形質転換体を作製する方法であって、
Gタンパク質共役型受容体α 1A および以下のポリペプチド:
(1)配列番号1に記載のアミノ酸配列からなるポリペプチド;
(2)配列番号1に記載のアミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;
(3)配列番号2に記載の塩基配列からなるポリヌクレオチドにコードされるポリペプチド;
(4)配列番号2に記載の塩基配列において1もしくは数個の塩基が欠失、置換もしくは付加された塩基配列からなるポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;あるいは
(5)配列番号2に記載の塩基配列の相補配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし得るポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド
を細胞に共発現させる工程を包含する、作製方法。 - Gタンパク質共役型受容体α 1A のリガンド親和性を改変する方法であって、
Gタンパク質共役型受容体α 1A および以下のポリペプチド:
(1)配列番号1に記載のアミノ酸配列からなるポリペプチド;
(2)配列番号1に記載のアミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;
(3)配列番号2に記載の塩基配列からなるポリヌクレオチドにコードされるポリペプチド;
(4)配列番号2に記載の塩基配列において1もしくは数個の塩基が欠失、置換もしくは付加された塩基配列からなるポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;あるいは
(5)配列番号2に記載の塩基配列の相補配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし得るポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド
を脂質膜上に共存させる工程を包含する、方法。 - Gタンパク質共役型受容体α 1L の作働薬または拮抗薬をスクリーニングする方法であって、
Gタンパク質共役型受容体α 1A および以下のポリペプチド:
(1)配列番号1に記載のアミノ酸配列からなるポリペプチド;
(2)配列番号1に記載のアミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;
(3)配列番号2に記載の塩基配列からなるポリヌクレオチドにコードされるポリペプチド;
(4)配列番号2に記載の塩基配列において1もしくは数個の塩基が欠失、置換もしくは付加された塩基配列からなるポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;あるいは
(5)配列番号2に記載の塩基配列の相補配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし得るポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド
を脂質膜上に共存させてタンパク質複合体を生成する工程、ならびに
該タンパク質複合体を候補因子とともにインキュベートする工程
を包含する、方法。 - 細胞内Ca2+濃度を測定する工程、または細胞内イノシトールリン酸の代謝を測定する工程をさらに包含する、請求項4に記載の方法。
- Gタンパク質共役型受容体α 1A を発現している形質転換体を作製する方法であって、
Gタンパク質共役型受容体α 1A が発現している細胞において、以下のポリペプチド:
(1)配列番号1に記載のアミノ酸配列からなるポリペプチド;
(2)配列番号1に記載のアミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;
(3)配列番号2に記載の塩基配列からなるポリヌクレオチドにコードされるポリペプチド;
(4)配列番号2に記載の塩基配列において1もしくは数個の塩基が欠失、置換もしくは付加された塩基配列からなるポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;あるいは
(5)配列番号2に記載の塩基配列の相補配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし得るポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド
の発現を抑制する工程を包含する、方法。 - 前記ポリペプチドが内因性タンパク質である、請求項6に記載の方法。
- 前記ポリペプチドの発現を阻害する工程が、RNAi法によって行われる、請求項6に記載の方法。
- 前記RNAi法が、配列番号5に示される塩基配列からなるオリゴヌクレオチドを導入することによって行われる、請求項8に記載の方法。
- 前記細胞が、外因性のGタンパク質共役型受容体α 1A を発現させている形質転換体である、請求項6に記載の方法。
- Gタンパク質共役型受容体α 1A のリガンド親和性を改変する方法であって、
Gタンパク質共役型受容体α 1A が発現している細胞において、以下のポリペプチド:
(1)配列番号1に記載のアミノ酸配列からなるポリペプチド;
(2)配列番号1に記載のアミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;
(3)配列番号2に記載の塩基配列からなるポリヌクレオチドにコードされるポリペプチド;
(4)配列番号2に記載の塩基配列において1もしくは数個の塩基が欠失、置換もしくは付加された塩基配列からなるポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド;あるいは
(5)配列番号2に記載の塩基配列の相補配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズし得るポリヌクレオチドにコードされるポリペプチドであり、かつ複合体化したGタンパク質共役型受容体α 1A のリガンド親和性を改変する活性を有するポリペプチド
