本発明は試料液中の特定の成分を分析するバイオセンサに関し、特に試料液を毛細管現象にて供給する検体供給路を備えたバイオセンサにおいて、試料液を供給しやすいセンサ構造に特徴があるものである。

【従来の技術】
バイオセンサは、微生物、酵素、抗体等の生物材料の分子認識能を利用し、生物材料を分子識別素子として応用したセンサである。すなわち、固定化された生物材料が、目的の基質を認識したときに起こる反応、微生物の呼吸による酵素の消費、酵素反応、発光などを利用したものである。

バイオセンサの中でも酵素センサの実用化は進んでおり、例えば、グルコース、乳酸、コレステロール、ラクトース、尿素、アミノ酸用の酵素センサは、医療計測や食品工業に利用されている。酵素センサは、検体である試料液に含まれる基質と酵素との反応により生成する電子によって電子伝達体を還元し、測定装置がその電子伝達体の還元量を電気化学的に計測することにより、検体の定量分析を行う。このようなバイオセンサの一例として、例えば、特願平11−324511号で提案されたようなセンサが知られている。

【発明が解決しようとする課題】
しかしながら、前述のような構成のバイオセンサにおいては、試料液を供給する検体供給路の入口において、検体供給路を形成する絶縁性基板とカバーとは、その平面視した端部は、同位置で同形状をしているので、試料液を供給する角度が小さかったり、また、誤って絶縁性基板の裏側(電極が形成されていない面)に試料液が付着した場合などに、再度試料液を供給しようとしても裏側に付着した試料液が邪魔をし、試料液の供給がうまくできず、測定ミスや測定誤差を誘発するという問題があった。

この発明は以上のような問題点を解消するためになされたもので、試料液の供給を正確且つ容易にできうるバイオセンサを提供することを目的とする。

【課題を解決するための手段】
本発明の請求項1にかかるバイオセンサは、第1の基板上に、切り欠け部を有するスペーサを介在させて、第2の基板を貼り合わせることにより、前記両基板間に試料液を採取しうる検体供給路を構成するとともに、この検体供給路は、前記両基板の一端部に入口を有するとともに、他端部に空気孔が設けられており、前記入口から前記空気孔に向けて毛細管現象により前記試料液が導入されるよう構成されたバイオセンサであって、
前記入口を構成する両基板の端部は、前記第1の基板を上面とする平面視において、互いに異なる形状をなしており、かつ、第1の基板上に配置される前記第2の基板は、前記入口方向に突出させたことを特徴とするものである。

【発明の実施の形態】
(実施の形態1)
以下に、本発明の実施の形態1にかかるバイオセンサについて図1を用いて説明する。なお、ここでは試料液中の特定物質と特異的に反応する分子識別素子として酵素を用いた酵素センサに関して具体的に示すものとする。

図1は本実施の形態1にかかるバイオセンサの分解斜視図及び断面図である。図1において、1は第1の絶縁性基板であり、この第1の絶縁性基板1上には、電気伝導性物質からなる測定電極2、対電極3、ならびに検知電極4が形成されている。

なお、ここでいう検知電極4は、検体量の不足を検知するための電極として機能するだけでなく、参照電極あるいは対電極の一部として用いることも可能である。

電流測定においては、本実施形態で述べた測定電極2、対電極3、検知電極4からなる三電極方式のほかにも測定電極2、対電極3のみからなる二電極方式などがあり、本発明で得られた効果を実現するためには何れの方式を用いてもよいが、3電極方式のほうがより正確な測定が可能である。
(具体例1)
ポリエチレンテレフタレートからなる第1の絶縁性基板上に、スパッタリング蒸着により、前記絶縁性基板の表面全面に約8nmの厚みのパラジウム薄膜を形成した後、YAGレーザにより、前記薄膜の一部にスリットを設けることにより、測定電極、対電極、および検知電極に電極を分割形成した。その上に酵素、電子伝達体、親水性高分子などを含んだ水溶液を前記測定電極を中心にして対電極ならびに検知電極の一部を覆うように円状に滴下し、乾燥させることで試薬層を形成した。さらにその上からポリエチレンテレフタレートからなる切り欠け部を有するスペーサと、同じくポリエチレンテレフタレートからなる空気孔を有する第2の絶縁性基板(カバー)とを貼り合わせることで、血液が導かれる毛細管となる検体供給路を形成した。

なお、本発明の効果を確認するため、ここでは前記絶縁性基板と前記スペーサおよびカバーとの平面視で見た端部のずれ(S)が各々S=0(従来センサ)、0.1、0.25、0.5、1.0、2.0mmである、計6種類の血糖値測定センサを作製した。

(表1)から明らかなように、S=0mmである従来センサでは、血液が供給される角度が小さい場合(0〜30度)には血液が吸引しなかったり、きちんと吸引させるためには数回の供給動作をする必要があることがわかる。これは、供給角度が小さい場合には、血液を検体供給路に供給する際に、先ず血液が絶縁性基板の裏側に付着してしまい、再度、血液を検体供給路に供給しようとしても、その絶縁性基板の裏に付着した血液側に引っ張られてしまうためであると推測される。

これに対し、本発明のセンサにおいては、最も距離の短いS=0.1mmの場合においても、供給角度が小さい場合には数回の供給動作をする必要はあるものの全く吸引をしないというケースは無くなり、S=0.25mm以上あれば、いかなる吸引角度においても容易に血液を吸引させれることがわかる。

以上のように本発明によれば、2枚の基板の貼り合わせにより、前記基板間に試料液を採取しうる検体供給路を構成し、その検体供給路には、前記両基板の端部に設けた開口を入口として、前記試料液が導入されるよう構成したバイオセンサであって、前記入口を構成する両基板の端部は、前記バイオセンサの平面視において、互いに異なる位置あるいは異なる形状にしたので、試料液を供給する角度が不十分でも(小さくても)、正確且つ容易に試料液を吸引させることができるという効果が得られる。さらには、第1の絶縁性基板1の裏側へ試料液が付着するのを防止する効果もあわせ持ち、仮に基板の裏側へ試料液が付着した場合でも、再度試料液を供給することで、スムーズに試料液を供給することができるという効果が得られる。
