同位体の入った薬物を生体に投与した後、同位体の濃度比の変化を測定することにより、生体の代謝機能が測定できるので、同位体の分析は、医療の分野で病気の診断に利用されている。
本発明は、同位体の光吸収特性の相違に着目して、同位体ガスの濃度比を測定する同位体ガス分析測定方法に関するものである。

一般に、胃潰瘍、胃炎の原因として、ヘリコバクタピロリー(HP)と言われているバクテリアが存在することが知られている。
患者の胃の中にHPが存在すれば、抗生物質の投与による除菌治療を行う必要がある。したがって、患者にHPが存在するか否かを確認することが重要である。HPは、強いウレアーゼ活性を持っていて、尿素を二酸化炭素とアンモニアに分解する。

一方、炭素には、質量数が12のものの他、質量数が13や14の同位体が存在するが、これらの同位体の中で質量数が13の同位体13Cは、放射性がなく、安定して存在するため取り扱いが容易である。

従来、13CO2と12CO2との濃度比を求める方法として、赤外分光を用いる方法が知られている(特公昭61-42219号公報、特公昭61-42220号公報参照)。
特公昭61-42220号記載の方法は、長短2本のセルを用意し、一方のセルでの13CO2の吸収と、他方のセルでの12CO2の吸収が等しくなるようなセルの長さにし、各セルに、それぞれの分析に適した波長の光を当てて、透過光の強度を測定する。この方法によれば、自然界の濃度比での光吸収比を1にすることができ、これから濃度比がずれると、ずれた分だけ光吸収比が変化するので、濃度比の変化を知ることができる。

前記の方法では、12CO2や13CO2の吸光度を正確に測定するために、測定前に、CO2の濃度がゼロのリファレンスガス、具体的には炭酸ガス吸収剤に通した空気を、セルに満たしてリファレンス吸光測定を行っている。
