Cell Biol 119:55−68)。hop−1における変異は目に見える表現型を全く示さないが、その変異とsel−12の変異とを組み合わせると、合成的な致死性の表現型がもたらされる(Li,X.et al.1997,Proc Natl Acad Sci USA 94:12204−12209)。sel−12は、ヒトPS1およびPS2に最もよく似た遺伝子であり、C.elegansにおいて最もよく研究されたプレセニリン遺伝子でもある。特定のsel−12変異体は産卵異常を示し、さらに産卵口発達における形態学的変化も示すということがわかっている(Levitan,D.et al.1995,Nature 377:351−4)。sel−12はヒトプレセニリンと50%同一であり、sel−12変異体における産卵異常は、sel−12プロモーターからPS1またはPS2が遺伝子導入により発現させることにより相殺される(Baumeister et al.,1997,Genes and Function 1:149−159)。これは、PS1およびPS2が、sel−12と機能的に相同であることを示している。当初は、sel−12変異は、lin−12およびglp−1ノッチ受容体遺伝子における機能獲得型突然変異体の複数の産卵口の表現型のサプレッサーとして同定された。ノッチ受容体は、多くの多細胞生物における細胞運命の決定を制御する。加えて、sel−12変異体の表現型が、lin−12の弱い機能喪失型変異体の表現型と類似していることが示されている。要約すれば、哺乳動物など多数の生物におけるノッチシグナル伝達においてプレセニリンは重要な役割を果たすと言える。

として用いることができる物質を同定および/または特徴付ける方法における、この同じトランスジェニックC.elegans生物の使用に関する。ヒト細胞との類似性を考慮し、さらに、C.elegansおよびヒトにおいて実証されたノッチシグナル経路およびプレセニリン機能の機能的類似性という背景において、FADプレセニリン変異体の機能不全は、ヒトにおけるses−1と相同または機能的に同等な遺伝子に変異を入れることにより同じような方法で相殺され得ると考えられる。それゆえに、本発明の内容において、ヒトses−1相同体または機能的に同等な遺伝子における欠陥により、FAD変異により影響を受けないプレセニリン遺伝子の発現が増加すると予想される。

配列番号7:pBy1015の配列である;このセグメントは、ses−1ヌル変異体の欠陥を補足する。
公開された方法(elegans.swmed.edu)に従ってEMSおよびUV/TMP変異誘発を行った。sel−12サプレッサーを同定するために、L4段階のsel−12変異体を突然変異誘発要因(例えばエチルメタンスルホネート)に晒し、再回復期の後、4または5のグループで、9cmのペトリ皿にプレーティングした。全ての試験
において、変異誘発後のF2段階で動物の産卵を分析した。Sel−12ヌル変異体は、通常は全く産卵しなかった。単離された動物のF3世代を再び試験し、両方の世代において成体動物あたり少なくとも5個の卵を産んだ動物を単離した。この方法において、ホモ接合型サプレッサー系を得た。バックグラウンド変異を除去するために、すべての変異を野生型動物と数回戻し交雑した。強力なサプレッサー変異により、sel−12ヌル変異体がホモ接合型であるにもかかわらず野生型動物の産卵行動に匹敵する動物の産卵行動を示した。このようなサプレッサー変異の一つをses−1変異と名付けた。一般的に、ses−1/ses−12二重変異体は、動物あたり約250−350個の卵を産む。
ses−1における変異は、sel−12変異体の産卵異常を完全に抑制する。立体顕微鏡検査によれば、sel−12産卵異常の3つの観点を区別することができる。全てのsel−12(ar171)動物の約75%が顕著な産卵口の隆起を示し、これは、隆起した産卵口の表現型(Pvl)として知られている。この欠陥の正確な理由は報告されていない。実質的に全てのsel−12(ar171)動物は、同等の野生型動物が有するより多くの卵を子宮に有しており、すなわち同等の野生型動物の一定の卵の数は約15であるのに対し、sel−12の場合の卵の数は〜60個である。これにより動物が膨張するが、すなわちこれはEgl表現型と説明される。卵は子宮に蓄積され、胚は子宮内で孵化し、母体で発生を続けるが、全てのケースの実質的に100%において、これらはこれに関連して成熟前に死亡する。表現型は、「bag−of−worms」表現型と名付けられた。非常に珍しいホモ接合型sel−12培養において野生型動物はほとんどめったに観察されない。ses−1変異は、このsel−12(ar171)表現型の全ての3つの観点を実質的に完全に抑制する。sel−12(ar171);ses−1動物は、野生型動物よりわずかに小さく、正常な動物よりも子宮中の卵は少ない(弱いEgl−構成的表現型)。これは、二重変異体における産卵が、わずかに過剰刺激されていることを示す。ses−1対立遺伝子動物はまた、説明された全てのその他のsel−12変異の産卵異常を抑制する。これは、ses−1変異が対立遺伝子特異的な方法で作用しないことを証明する。
ses−1を染色体X左腕においてマッピングした。これに関して、sel−12(ar171)ses−1系を、遺伝子マーカー変異を用いて位置付け、産卵欠陥(Egl)組換え動物を単離し、試験していくつのこれら動物がマーカー変異を受け入れたかを決定した。ses−1をより詳細にマッピングするために、系におけるses−1変異の存在/非存在をsel−12(ar171)産卵異常の抑制により調べた。これに関して、sel−12(ar171)ses−1の雌を、AB二重変異体系(AおよびBは、遺伝子地図上でsel−12の右に位置する2つの変異であり、Bは、2つのマーカーの右側にある)と交雑した。次に、Bのみの組換え体でありA変異を有さない組換え体を探した。この組換え法の結果として、全ての組換え体は、二重組換えを有する組換え体を除いて、sel−12(ar171)を含むはずである。一連の交雑実験により、ses−1は、dpy−23(e840)とlin−2(e678)との間、dpy−23(e840)からlon−2までの距離の約1/5にマッピングされた。すべての実験により、ses−1変異が機能の損失を引き起こすことが示された。これを実証するために、2つの自由な重複(free duplication)によりses−1変異を位置付けた。ses−1対立遺伝子by108およびby109変異は互いに補足しあわないが、両方とも染色体X左腕に2つの異なる遺伝子における変異を示す可能性があった。この理由として、当初、by108およびby109は、それぞれ独立してマッピングされた。マッピングを確かめるために、2つの重複、すなわちmnDp31およびmnDp32を、系sel−12(ar171)ses−1(by108)lon−2(e678)にトランスファーさせ、産卵について試験した。
導入遺伝子を、系sel−12(ar171)ses−1(by108)に注入し、トランスフェクトされたクローンの抗サプレッサー活性を試験した。形質転換マーカーとして、1μlあたり100ngのpRF4、1μlあたり20ngのpBY218と共に、すべてのクローンを濃度20ng/μlで注入した。コスミドF46H6の注入によりses−1表現型は、完全に相殺された(レスキューされた)。その後、ses−1遺伝子の同一性を明らかにするために、このコスミドからのF46H6サブクローン、精製された制限断片およびPCR産物を試験した。
様々な制限酵素でF46H6を切断した。サブフラグメントをQiaquickゲルエクストラクションキットを用いて精製した。21.5kbのNcoI断片および0.2kbのBamHI断片(プラスミドpBY1015中)を直接注入し、(両方の)表現型をレスキューした。最小のレスキューを示す断片は、4kbの配列に減少された。このセグメントは、多数のジンクフィンガーを有するタンパク質をコードする1つの遺伝子のみを含む。
ses−1の2つのcDNAを同定したところ、1.9kbおよび2.1kbの配列であった。2つのcDNAは、同一のDNA配列およびエキソン−イントロン境界を含み、ただし以下の点において異なる:大きいほうのクローンは追加のイントロンを有するが、小さいほうのクローンは6塩基だけ長い5’末端を有する(これについては図3を参照)。
ses−1融合遺伝子をバキュロウイルスSF9細胞系で発現させ、免疫染色で位置を同定した。ses−1は、細胞核に位置しており、これは、その予想された転写因子としての役割に一致する。
ノーザンブロット分析によれば、ses−1転写物が、全ての発生段階で発現されることが示された(図2)。
by131は、ses−1機能を全く示すことができない変異体である(遺伝子全体の欠失)。しかしながら、この欠失はまた、隣の遺伝子も欠失している。by135は、説明されたようにリーディングフレームシフトを起こし、それゆえに、最も強力なses−1変異体であるだろう。ses−1(by135)およびby131動物を調査した。両方の系は、形態学的に、かつそれらの産卵行動に基づき、両方とも野生型の表現型である。2種の動物の子孫の数は、野生型よりいくらか少ない。両方の対立遺伝子は、全ての対立遺伝子のsel−12表現型を完全に抑制し、さらに野生型の子孫数を有するsel−12対立遺伝子をもたらす。それゆえに、ses−1は、sel−12の特異的なサプレッサーであると結論付けることができる。sel−12変異体またはsel−12、hop−1二重変異体の全ての欠陥は、lin−12およびglp−1ノッチシグナル伝達経路におけるこれらプレセニリンの効果による可能性がある。
Claims (14)
- hop−1の発現を抑制するか、および/またはその除去がhop−1の発現の増加をもたらすタンパク質をコードする単離された核酸分子であって、該核酸分子は配列番号3、配列番号4、配列番号5または配列番号6で示されるヌクレオチド配列を有する、核酸分子。
- DNA分子またはRNA分子である、請求項1に記載の単離された核酸分子。
- DNA分子がcDNA分子またはゲノム分子である、請求項2に記載の単離された核酸分子。
- タンパク質が、配列番号1または配列番号2で示されるアミノ酸配列と同一であるアミノ酸配列を有する、請求項1〜3のいずれか一項に記載の単離された核酸分子。
- 請求項1〜4のいずれか一項に記載の単離された核酸分子の変異体であって、以下の変異:
a) 配列番号1の少なくとも494位から前方のアミノ酸を欠失している、C末端部が短縮されるようにコードされたタンパク質を生じさせる変異;
b) 配列番号3の核酸の+1784位におけるGからAへの変換;および
c) 配列番号1のコードされたタンパク質の595位でCの代わりにYが存在するようにする変異;
の1またはそれ以上を含み、該変異はsel−12変異体の産卵異常を修復する、核酸分子。 - 変異が:
a)配列番号1のコードされたタンパク質の209、388、465または494位において翻訳停止が導かれる変異;
b)配列番号3の核酸の+1393位におけるCからTへの変換;
c)配列番号3の+1481位におけるヌクレオチドCAAからAACへの転換を生じさせる変異;
d)配列番号3の核酸の+1784位におけるGからAへの変換;
e)配列番号3の+549位におけるAの除去;または
f)配列番号3の核酸の+1163位におけるTからAへの変換;
である、請求項5に記載の単離された核酸分子。 - 請求項1〜6のいずれか一項に記載の単離された核酸分子を含むベクター。
- 線虫C.elegansの細胞における発現のためのプロモーターを含む、請求項7に記載のベクター。
- プラスミド、コスミド、バクミド、ウイルスまたはバクテリオファージである、請求項7または8に記載のベクター。
- hop−1の発現を抑制するか、および/またはその除去がhop−1の発現の増加をもたらす単離されたタンパク質であって、配列番号1または配列番号2で示されるアミノ酸配列を有するか、または配列番号3、配列番号4、配列番号5、配列番号6または配列番号7の配列の1つを有する核酸によりコードされる、単離されたタンパク質。
- 請求項1〜6のいずれか一項に記載の核酸分子を含むトランスジェニック非ヒト生物。
- C.elegansである、請求項11に記載のトランスジェニック非ヒト生物。
- 神経変性疾患を治療するための医薬を製造するための、請求項1〜6のいずれか一項に記載の単離された核酸分子の使用。
- 神経変性疾患の調査および解明のための非ヒトモデル生物をトランスジェニック方法により作製するための、請求項1〜6のいずれか一項に記載の単離された核酸分子の使用。
