本発明は、ハロビニリデンハロメチルシクロヘキサン化合物、ハロビニリデンメチレンシクロヘキサン化合物、及びこれらの製造法、ならびに上記化合物を用いた活性型ビタミンD誘導体合成の重要中間体であるヒドロキシエチレンメチレンシクロヘキサン化合物の製造法に関する。

活性型ビタミンD3(1,25−ジヒドロキシコレカルシフェロール)は、小腸におけるカルシウム輸送能、骨塩動員能などの生理活性が強く、そのため人の生理機能に重要な役割を果たすことが知られている。
上記式(6)で示されるヒドロキシエチレンメチレンシクロヘキサン化合物、例えば、下記化合物1(R1=R2=R3=水素原子、W=t−ブチルジメチルシリル基、Y=t−ブチルジメチルシリルオキシ基)は、活性型ビタミンD誘導体を製造する際の最も重要な中間体の1つであるA環部分前駆体としてよく知られている。

【発明が解決しようとする課題】
しかしながら、上記スキーム1の製造法は、出発物質からの工程数が長い、収率が低い等の課題を抱えており、より実用的な製造法が望まれている。そして、現在も効率的な製造法を目指して研究が盛んに行なわれているのが現状である。本発明は、上記事情に鑑みなされたものであり、活性型ビタミンD誘導体を製造する際の重要中間体であるA環部分前駆体に効率的に変換可能な鍵中間体となるハロビニリデンハロメチルシクロヘキサン化合物、ハロビニリデンメチレンシクロヘキサン化合物及びこれらの製造法、ならびに該化合物を用いたA環部分前駆体であるヒドロキシエチレンメチレンシクロヘキサン化合物の効率的な製造法を提供することを目的とする。

【課題を解決するための手段及び発明の実施の形態】
本発明者らは、上記目的を達成するため、鋭意検討を重ねた結果、上記式(1)で示されるハロビニリデンハロメチルシクロヘキサン化合物及び上記式(2)で示されるハロビニリデンメチレンシクロヘキサン化合物が、活性型ビタミンD誘導体の製造におけるA環部分前駆体の重要な鍵中間体となり得ることを見いだすとともに、該化合物をヒドロキシメチル化することでA環部分前駆体であるヒドロキシエチレンメチレンシクロヘキサン化合物を効率的に製造できることを見いだし、本発明を完成するに至った。

本発明に係る製造法に用いられる反応溶媒としては、当該反応条件下において安定であり、かつ、不活性で目的とする反応を妨げないものであれば特に制限はない。

これらの溶媒は反応の起こり易さに従って適宜選択され、1種単独でまたは2種以上混合して用いることができる。なお、必要に応じて適当な脱水剤や乾燥剤により水分を除去し、非水溶媒として用いてもよい。
以上述べた溶媒は、本発明を実施する際の一例であって、本発明はこれらの条件に限定されるものではない。
