発明の属する分野
本発明は、リボザイムとそれらの組み合わせに関連している。さらに詳しくは、本発明はCCR5の制御に広く関与している。

発明の背景
ウイルス感染に対する細胞内免疫を提供するための遺伝子治療の概念は数年間、考慮されてきた(Baltimore, 1998; Szydalski, 1992参照)。遺伝子治療は、近来、HIV感染の治療において、その潜在的な有用性のため、より多くの注目を受けている(Sarver and Rossi, 1993)。アンチセンスRNA、リボザイム、TARまたはRREデコイ、トランス-優勢変異HIV遺伝子、及び、条件つきで致死である毒素を含む、多数の異なる阻害剤が、そのHIV-1への耐性を与える能力に関して試験されてきた(Sarver and Rossi, 1993に概説されている)。

アンチセンスあるいはリボザイムといった、RNAを基礎とした戦略は、潜在的に免疫原となるタンパク質を生産しないだけでなく、配列特異的であり、理論上、不必要な毒性を排除するという2つの利点をもつ。一つのリボザイム分子は、結合、開裂、そして、解離という連続的サイクルにより、多数の標的RNA分子を不可逆的に不活性化できる。複数の基質のターンオーバーがなくても、リボザイムは標的RNAを、開裂を介して機能的に不活性化する(Zaug and Cech, 1986; Uhlenbeck, 1987; Castanotto et al., 1992)。

近年、CCKR-5(CCR5としても知られている)遺伝子中に32塩基のホモ接合体的欠損をもつ個体がM-向性のHIV-1 株による感染を受けにくいことが発見された。さらに、ヘテロ接合体は長期生存者であり、このことはCCR5発現の欠損がAIDSの正常な進行を妨げる可能性があることを示唆している。その32塩基が欠損した遺伝子によりコードされるタンパク質は、かなり短くなっており、細胞表面では検出できず、また、ホモ接合体の個体においては明確な表現型はない。これは細胞表面でのCCR5発現の阻害が、HIV-1の侵入に影響していることを示している。

発明の概要
本発明は、哺乳動物細胞においてCCR5を抑制制御することにより、HIV感染を治療する方法を提供する。他の観点においては、本発明はCCR5 HIV-1共受容体(co-receptor)を標的とする、新規リボザイムを提供する。

本発明はまた、抗CCR5リボザイムを発現するベクターを用いてHIV耐性細胞を作成する方法を提供する。好ましい態様において、抗CCR5リボザイムは、HIVにおいて保存的な配列を標的とした、1またはそれ以上のリボザイムの組み合わせにおいて使用される。

発明の詳細な説明
CCR5はβ-ケモカイン、MIP1-α、MIP1-β、およびRANTESに対する、7回膜貫通型の受容体である。いくつもの研究により、これらのケモカインが、CD4+-Tリンパ球へのHIV-1感染の阻害、及び、HIV感染細胞のシンシチウム形成の阻害をできることが証明されている。CCR5遺伝子に32塩基のホモ接合体の欠損を有する個体はM-向性のHIV-1 株による感染を受けにくい。さらに、ヘテロ接合体は長期生存者であり、このことはCCKR-5の発現の欠損がAIDSの正常な進行を妨げることができる可能性を高めている。その32塩基が欠損した遺伝子によりコードされるタンパク質は、かなり短くなっており、細胞表面では検出できず、また、ホモ接合体の個体においては明確な表現型はない。従って、細胞表面でのCCR5発現の阻害がHIV-1の侵入に影響していることが期待され、CCR5の発現の抑制制御が、HIV-1感染の予防と治療において、魅力的な治療上のアプローチとなった。

本発明は、他の特徴としては、リボザイムを用いてCCR5を抑制制御する新しいアプローチを提供する。本発明はその様々な観点において、CCR5受容体の発現を変化させる方法と組成物を提供する。最適には、HIVにおいて保存的な配列を標的とした、1またはそれ以上のリボザイムと組み合わせた、抗CCR5リボザイムを発現するコンビナトリアルベクターを用いて、CD34+ヒト造血前駆細胞に形質導入し、それは次にHIV耐性単核細胞を生じさせるであろう。

触媒作用を持つRNA(リボザイム)のいくつかの分類が文献に記述されてきており、そして、本発明はリボザイムのどの一つの分類にも限定されない。しかしながら、好ましい観点においては、本発明のリボザイムは“ハンマーヘッド”リボザイムである。このようなリボザイムは、mRNAといった標的核酸の少なくとも一部分に相補的な配列をもつ、一本鎖RNAにより形成された、1またはそれ以上のアームを含む、ハイブリッド形成領域(望ましい特異性を与える)を有する。ハイブリッド形成(あるいは“アンチセンス”)領域は、典型的な例では、標的の核酸との効果的なハイブリッド形成に十分な数のヌクレオチドを含む、RNA部分を含む。典型的な例では、これらの領域は少なくとも約7ヌクレオチドを含み、好ましくは、約9から約12ヌクレオチドを含む。保存的な触媒コア領域は標的とされたRNAを開裂することができる。本発明における好ましいリボザイムは、X2 がアデニン、シトシン、あるいはウラシルであり、またUがウラシルである、配列X1UX2を含む標的RNAを開裂する。好ましくはX1はグアニジンであり、そして、X1UX2は、GUC、あるいはGUAである。

リボザイムのアンチセンスアームは選択したX1UX2配列に隣接する標的CCR5 mRNA配列上のRNAと相補的であり、そしてそのためハイブリッド形成できるように合成することができる。リボザイムのアンチセンス領域が、X1UX2配列に隣接する標的RNA配列とハイブリッド形成する際には、リボザイムの触媒領域が標的RNAをX1UX2配列内で開裂する。RNA開裂は、in vitroにおいて、マグネシウム、あるいは、別の二価陽イオンの存在下、約pH7.5で促進される。

本発明の一つの態様において、図2に図示されるようなハンマーヘッドリボザイムが提供される。このリボザイムは3'-CACAGUUCAA AGCAGGUGUG CCUGAGUAGU CGUUAGAUA-5'(SEQ ID NO. 1)という配列をもつ触媒領域を含み、その配列はCCR5 AUG開始コードのすぐ下流に位置するGUC配列を認識する。特に、このリボザイムは、CCR5 mRNAのヌクレオチド67から84に由来する、二番目のGUCを標的としている。CCR5 mRNAのこの領域の配列は5'-GUGUCAAGUC CAAUCUAU-3'(SEQ ID NO. 2)である。開裂は二番目のGUCトリプレットのCの後ろで起こる(図5)。このリボザイムは、それぞれ9、及び、8ヌクレオチドの2つの短いアームを介してその標的と相互作用する。このリボザイムがケモカイン受容体ファミリーの他のメンバー、あるいは、他の内因性転写産物を標的としないことを保証するため、その塩基対がリボザイムと同じである、CCR5由来の正確な配列を、GenbankのBLASTNサーチに入力し、他のどの必須の遺伝子とも有意な相同性がないことを明らかにした。

当業者は、本発明からはずれることなく、図2のリボザイムの配列を修飾し得ることを認めるだろう。触媒領域は、CCR5 mRNA内のどのX1UX2配列をも標的とし得るが、但しX1UX2配列は、結果としてそのmRNAを開裂し、機能的CCR5分子の翻訳のための鋳型として働くことができない1またはそれ以上のRNA鎖にするように、選択されるべきである。選択されたX1UX2配列から上流、及び、下流の塩基(好ましくは7-12塩基)に、効果的に結合することができるアンチセンス領域は、そのmRNA配列の知識を基に選択されるだろう。

そのリボザイムは、さらにヌクレアーゼ耐性のRNA塩基を含むように修飾され得る。例えば、これらの修飾は、リボザイムを分解するヌクレアーゼに耐性を与えるために、ヌクレオチドのホスホロチオエート(phosphorothioate)誘導体を使用することを含む(Bratty et al., Biochem, Biophys. リボザイムはまた、ヌクレオチド塩基と共にデオキシリボヌクレオチドを含む、キメラのリボザイムの形で合成され得る。これらのキメラのリボザイムは、効果的な開裂の特性を維持すると同時に、細胞質での安定性を増加したことが示されてきた。キメラの(DNAを含む)リボザイム(“ヌクレオザイム”としても知られている)の化学は、Brattyら(上と同じ)に概説されている。もとの文献としては、Taylorら(Nucleic Acids RES., 20: 4559-4565 (1992))を参照のこと。

リボザイムは細胞内で機能するので、標的細胞によるリボザイムの取り込みは重要な問題であり、有利に最適化されている。リボザイムの外からの投与の好ましい方法は、リポソームの使用による。リポソームがリボザイムを酵素の攻撃から保護し、リポソームの液状のカプセルは細胞壁を通る輸送を促進する。リポソームは、核酸の細胞への輸送を目的として開発されてきた。例えば、Friedmann(Science, 244: 1275-1281 (1989))を参照のこと。

オリゴヌクレオチド(それらはDNA、またはRNA、またはその両方からなる)それ自体の直接の細胞への取り込みは、現在、輸送の方法としてはあまり好ましくないと考えられている。なぜなら、リボザイムやアンチセンス分子の場合、オリゴヌクレオチドの直接の投与は、それと共に、RNAseといった細胞のヌクレアーゼによる攻撃や消化という附随した問題を伴うためである。抗CCR5リボザイムの投与の一つの好ましい方法としては、目的とする標的細胞により発現されるような、目的とするリボザイムの配列をコードする遺伝子の輸送を促進する、既知のベクターを利用する。このようなベクターはプラスミド、及び、ウイルス(アデノウイルス、レトロウイルス、及び、アデノ随伴ウイルス)[及び、リポソーム]及び、それらを修飾したもの(例えばポリリジン修飾アデノウイルス[Gao et al., Human Gene Therapy, 4:17-24 (1993) ]、陽イオンリポソーム[Zhu et al., Science, 261:209-211(1993)]及び、リポソームで覆われた修飾アデノ随伴ウイルスプラスミド[Phillip et al., Mol. Cell. Biol., 14:2411-2418 (1994)])を含む。リボザイムRNAの発現は、RNAポリメラーゼII及びIIIといった遺伝子要素により行われる。

本発明のリボザイムは、当技術分野においてRNA分子の合成についての既知の方法により、調製できる。特に、本発明のリボザイムは、機能可能であるようにプロモーターに連結した、相当するDNA配列(転写によりリボザイムを生じるDNAであり、当技術分野においてそれ自体でDNAの合成についての既知の方法に従って合成できる)から調製できる。本発明のリボザイムに相当するDNAは、プラスミド、バクテリオファージDNA、あるいは、ウイルスDNAといった、DNA輸送ベクター内にライゲーションすることができる。その後、本発明に従って、リボザイムが宿主細胞内で転写されるように、機能可能であるようにプロモーターに連結したリボザイムに相当する遺伝子の材料を含む適当な輸送ベクターを用いて、原核細胞、または、真核細胞(哺乳動物の移植された細胞も含む)にトランスフェクトできる。リボザイムは輸送ベクターから直接に転写することができるか、または、より大きなRNA分子の一部として転写されて、その後、開裂されて目的とするリボザイム分子を産生できる。目的とするリボザイムを産生するために細胞を形質転換する様々な方法が、本明細書中に記載されているが、哺乳動物細胞における、非天然の(non-native)(組換え型の)遺伝子発現に関する一般分野における当業者は、CCR5産生細胞においてリボザイムの発現を提供、または、最適化するための、さらなる手段や方法を提供するために、既知の技術を応用することができる。

図5の配列をコードするリボザイムを化学的に合成し、四つの異なる発現ベクターにクローン化した。初めの二つのベクターは、Bertrandら(1997)、及び、Goodら(1997)に記載されているヒトU6遺伝子から由来している(図1)。これは、転写される配列の5'側にプロモーターがあるPol IIIカセットである。この二つの構築物は、RNA転写産物に含まれるU6配列の量に違いがある。このRNAの初めの19塩基は安定化ステムループを形成する(Bertrand et al.,1997)が、キャップ形成のための情報は欠いている(図8)。U6+27に含まれる追加の8塩基は、結果としてγ-メチルリン酸をもつRNAのキャップ形成を生じる(Singh, Gupta and Reddy, 1990; Goode et al., 1997)(図9)。U6+27配列はもっぱら核にあるのに対して、U6+19は主として核にあるが、様々な程度で細胞質にも見い出すことができる(Bertrand et al., 1997)。これらの両方のプロモーターカセットで転写される安定化3'ステムループ構造は、リボザイムの配列に都合良く付加できる。5'配列及び3'配列が付加されたリボザイムの開裂の比活性を評価するため、U6+19-CCR5及び、U6+27 CCR5リボザイムカセットの両方からのRNAをPCRを使用して調製した。これらのリボザイムは、バクテリアファージT7プロモーターを使用する転写の鋳型から作成されたPCRから調製された。その転写産物は、U6プロモーターから転写されるだろうものの、(U6+27にあるキャップを除いては)正確な模倣体を産生した。これらの異なる二つのRNAに仲介される、in vitroでの開裂反応を図3に示す。U6+19、及び、U6+27を付加したリボザイムは、CCR5標的を同程度の見かけ効率で開裂する。

抗CCR5リボザイムの機能的な発現について試験した、他の二つのプロモーターは、(LNレトロウイルスベクター中の)MoMLV LTRプロモーターと、アデノウイルスVA1プロモーターである(図6、及び、7)。MoMLVプロモーター構築物はリボザイム転写産物上にキャップとポリA配列を提供し、また、細胞培養系における研究、及び、前臨床試験の両方において、抗tat及びtat/revリボザイムを首尾よく転写するのに使用されてきた(Zhou, et al., 1994; Bertrand et al, 1997; Bauer et al., 1997)。Pol IIIプロモーターである、アデノウイルスVA1プロモーターは細胞質に局在するRNAを産生する。ほとんどのPol IIIプロモーターと同様に、そして、U6プロモーターとは異なり、その調節領域はコード配列の内部にある。このシステムの利点は、VA配列が、細胞質に非常に長期間存在し得る、高度に安定した構造を与えることである。図2に、コンピューターにより予測された、熱力学的に最も安定なVA1-CCR5リボザイムの構造を示す。ステムループ構造の上端にリボザイムを挿入することにより、リボザイムの構造が維持される。図2に示したVA1-CCR5リボザイム構築物はin vitro においてリボザイムの開裂活性について試験した。直鎖状にしたDNA鋳型から、バクテリオファージT3ポリメラーゼを介して転写した転写産物を使用して、VA1-CCR5リボザイムRNA全体をin vitroにて転写した。VA1 RNA中にとどめられているにも関わらず、このリボザイムがCCR5 基質を開裂できることが、図4のデータから読み取れる。

実施例 I
U6+19 CCR5rz と U6+27 CCR5rz の in vitro における開裂反応
放射標識した103ヌクレオチドであるCCR5標的(S)を、以下に示す条件のもと、リボザイムの存在下、37℃でインキュベートした(図3)。開裂反応産物を6%ポリアクリルアミド、7 M尿素変性ゲルにより、分析した。パネルAは、37℃、20 mMマグネシウム存在下(+)(レーン2及び3)あるいは非存在下(-)(レーン1及び4)、そして、5分間(レーン1及び2)あるいは1時間(レーン3及び4)における、放射標識した103ヌクレオチドCCR5基質(S)のU6+19 CCR5rzによる、in vitroでの開裂反応を示している。

実施例 II
VA CCR5rz 及び VACCR5rzm の in vitro における開裂反応
放射標識した103ヌクレオチドCCR5標的(S)を、VArz1及び2(同じリボザイム構築物の異なる調製物)(図4、レーン1〜4)、あるいはその機能を失わせた(crippled)ものである、VArzm1及び2(図4、レーン5〜8)の存在下、20 mM MgCl2存在下(+)あるいは非存在下(-)、2時間、37℃でインキュベートした。その後、6%ポリアクリルアミド、7 M尿素変性ゲルにより、開裂反応を分析し、結果を図4に示している。

一過性にトランスフェクションした、293細胞由来のRNAに対するプライマー伸長により、RNA分析を行った(図6)。293細胞にそれぞれVA1プラスミド、あるいは、VA1抗CCR5プラスミドのいずれかをトランスフェクションした。トランスフェクションした2日後、RNAを調製し、VA1 RNAの3'端に特異的なプローブを用いたプライマー伸長に使用した。

一過性にトランスフェクションした 293 細胞からのノザンブロット
pVA1(レーン1)、pVA1抗CCR5リボザイム(レーン2、及び、3)、及び、pVA1抗CCR5変異体(レーン4、及び、5)をトランスフェクションした293細胞由来のRNA(図7)。使用したプローブは、VA1 RNAの3'端に特異的であった。

実施例 V
細胞培養系における CCR5 受容体の抑制制御
HOS-CD4-CCR5細胞は、国立衛生研究所(Bethesda, Maryland, U.S.A.)より入手した。実施例I、及びIIに記載した、様々な構築物をこれらの細胞に一過性にトランスフェクションした(リポフェクション)。トランスフェクションした48時間後、ヨウ素処理したリガンドであるMIP-1βを用いて結合アッセイを行った。

細胞は、4℃で、1 nMの125I-MIP-1β中で、100 nMの非標識I-MIP-1β存在下、あるいは、非存在下にて、2時間インキュベートした。その後、リン酸緩衝溶液で細胞を3回洗浄し、細胞ペレットのカウントを評価した。100倍過剰の非放射標識リガンド存在下、バックグラウンドのカウントを測定した。結果は、図10に図表で示している。

- 請求項1記載のリボザイム構築物をコードする核酸を含有する発現ベクターであって、哺乳動物細胞で前記核酸が発現するように指令させる調節シグナルに機能可能に連結する、前記発現ベクター。
- 請求項1記載のリボザイム構築物をコードする核酸が、発現カセットU6+19に挿入されて存在する、請求項2記載の発現ベクター。
- 請求項1記載のリボザイム構築物をコードする核酸が、発現カセットU6+27に挿入されて存在する、請求項2記載の発現ベクター。
- ベクターがMoMLV LTRプロモーターを含有するLNレトロウイルスベクターである、請求項2記載の発現ベクター。
