【発明の属する技術分野】
本発明は、α−リポイルアミノ酸またはその薬理学的に許容できる塩を含有する水性液剤の安定化法並びに安定化されたα−リポイルアミノ酸またはその薬理学的に許容できる塩を含有する水性液剤に関する。

【従来の技術】
α−リポ酸(別名:チオクト酸または6,8−ジチオオクタン酸)はミトコンドリアに存在する補酵素で、抗酸化能を有し、酸化ストレスによる種々の病態の治療、たとえば動脈硬化症および白内障の治療薬として注目されている。また、その還元型のジメルカプトオクタン酸は酸化型のグルタチオンやビタミンCなどを還元再生させる作用がある。

一方、α−リポ酸誘導体として、α−リポ酸にグリシン、メチオニン、グルタミン酸、バリンなどがそれぞれ結合したα−リポイルアミノ酸が知られている(特公昭42-1286号)。また、特開2000-169371号公報にはα−リポイルアミノエチルスルホン酸のイミダゾール塩などのジチオラン誘導体が記載されており、これら化合物は酸化的ストレスによって起こる疾患、たとえば糖尿病や白内障などに有効であることが開示されている。

しかし、これらα−リポイルアミノ酸の水溶液は経時的に硫化水素またはメルカプタンの不快臭を発生し、製剤化が困難であるという欠点がある。

【発明が解決しようとする課題】
本発明はα−リポイルアミノ酸またはその薬理学的に許容できる塩を含有する水性液剤の安定化法並びに安定化されたα−リポイルアミノ酸またはその薬理学的に許容できる塩を含有する水性液剤を提供する。

【課題を解決するための手段】
上記のような状況下、本発明者らは種々検討を重ねた結果、α−リポイルアミノ酸およびそれらの薬理学的に許容される塩が亜硫酸塩またはメタ重亜硫酸塩を添加することにより安定化できることを見出し、さらに研究を進めて本発明を完成させた。

すなわち、本発明は、
(1)α−リポイルアミノ酸またはその薬理学的に許容できる塩と亜硫酸塩またはメタ重亜硫酸塩を含有する水性液剤、
(2)α−リポイルアミノ酸がN−α−リポイル−α−アミノ酸、N−α−リポイル−β−アミノ酸、N−α−リポイル−γ−アミノ酸、N−α−リポイル−δ−アミノ酸およびN−α−リポイル−ε−アミノ酸からなる群から選ばれるものである、上記(1)記載の水性液剤、
(3)α−リポイルアミノ酸がN−α−リポイルアミノエタンスルホン酸、N−α−リポイルアスパラギン酸、N−α−リポイルグリシン、N−α−リポイル−γ−アミノ−n−酪酸、N−α−リポイル−6−アミノヘキサン酸、N−α−リポイルフェニルアラニン、N−α−リポイル−5−ハイドロキシトリプトファン、N−α−リポイルアントラニル酸、N−α−リポイルメチオニンおよびN−α−リポイルシステインからなる群から選ばれるものである、上記(1)記載の水性液剤、
(4)亜硫酸塩が亜硫酸ナトリウム、亜硫酸カリウム、亜硫酸水素ナトリウムまたは亜硫酸水素カリウムであり、メタ重亜硫酸塩がメタ重亜硫酸ナトリウムまたはメタ重亜硫酸カリウムである上記(1)記載の水性液剤、
(5)注射剤である上記(1)〜(4)のいずれかに記載の水性液剤、

本発明に用いられるα−リポイルアミノ酸はα−リポ酸とアミノ酸がアミド結合した構造を有している。本発明において、アミノ酸とは同一分子内にカルボキシル基とアミノ基を有する、いわゆるα−アミノ酸、β−アミノ酸、γ−アミノ酸、δ−アミノ酸、ε−アミノ酸、並びにアミノメチルシクロヘキサンカルボン酸およびアントラニル酸、並びに同一分子内にスルホン酸基とアミノ基を有するアミノエタンスルホン酸(タウリン)をいう。α−アミノ酸としては、たとえばグリシン、アラニン、バリン、ロイシン、イソロイシン、セリン、トレオニン、チロシン、システイン、メチオニン、アスパラギン酸、アスパラギン、グルタミン酸、グルタミン、アルギニン、リジン、ヒスチジン、フェニルアラニン、チロシン、トリプトファン、ヒドロキシトリプトファンなどが例示される。β−アミノ酸としてはβ−アラニンが挙げられ、γ−アミノ酸としてはγ−アミノ−n−酪酸(GABA)やカルニチンが挙げられ、δ−アミノ酸としては5−アミノレプリン酸や5−アミノ吉草酸、ε−アミノ酸としては6−アミノヘキサン酸が挙げられる。

本発明に用いられるα−リポイルアミノ酸の薬理学的に許容できる塩としては、ナトリウム塩やカリウム塩などのアルカリ金属塩およびカルシウム塩やマグネシウム塩などのアルカリ土類金属塩が挙げられるが、これら以外の塩であっても本発明の安定化法により適宜安定化することができる。

本発明に用いられるα−リポイルアミノ酸は、例えば次の合成法により、またはこれに準じて適宜合成することができる。

α−リポイルアミノ酸の合成法としては、アミノ酸の酸性基のカルボン酸を保護基としてエステル化し、α−リポ酸と脱水縮合剤で酸アミドとし、最後にケン化するのが一般的である。しかし、アミノエチルスルホン酸などの場合、この方法では合成するのが困難であるが、混合酸無水物法(MA法)により合成すると目的化合物のα−リポイルアミノ酸を高収率で得ることができる。すなわち、α−リポ酸を有機溶媒(たとえばクロロホルム、テトラヒドロフラン、アセトニトリルなど)に溶かし、これに3級アミン(トリエチルアミン、トリブチルアミンやN−メチルモルホリン(NMM)など)の存在下−15℃〜−5℃でハロゲン化炭酸エステル(クロル炭酸エチル、クロル炭酸ブチルなど)、イソブチルオキシカルボニルクロリド、塩化ジエチルアセチルまたは塩化トリメチルアセチルなどの混合酸無水物化試薬を反応させてα−リポ酸の混合酸無水物とする。反応時間は1〜2分から数10分程度である。さらにアミノ酸をアルカリ(水酸化ナトリウム、水酸化カリウムなど)存在下でアルコール、水またはそれらの混液などの溶媒に溶かしたものを加えて反応させた後、適当な溶媒、たとえば水またはアルコールから再結晶させると、高収率でα−リポイルアミノ酸を得ることができる。

本発明の水性液剤に使用される溶媒としては、精製水、特に注射用蒸留水が挙げられる。

本発明の水性液剤には、通常用いられる緩衝剤、等張化剤、溶解補助剤、防腐剤、粘稠剤、キレート剤、芳香剤等の各種添加剤を適宜添加してもよい。

本発明に用いられるα−リポイルアミノ酸を上記のような疾患の予防・治療剤として使用する際の投与量は、使用するα−リポイルアミノ酸の種類、患者の体重や年齢、対象とする疾患の種類やその状態および投与方法などによっても異なるが、たとえば注射剤の場合成人1日1回約1mg〜約30mg程度投与するのがよい。また、点眼剤の場合は、成人1日数回、1回数滴、濃度が約0.01〜5(w/v)%の点眼剤を投与するのがよい。

本発明のα−リポイルアミノ酸を含有する水性液剤には、本発明の目的に反しない限り、その他の同種または別種の薬効成分を適宜含有させてもよい。

また、本発明の水性液剤は紫外線吸収(紫外線照射による紅斑の抑制、日焼け防止など)、美肌、美白(しみ、そばかすなどの原因であるメラニン色素の沈着防止など)および抗シワを目的とし、またはその他の化粧品成分の安定化を目的として、ローション剤や化粧水などの化粧品としても使用される。化粧品として本発明の水性液剤を用いる場合、通常化粧品に用いられる成分を適宜添加させることができる。

参考例1 N−α−リポイルアミノエタンスルホン酸ナトリウム
DL−α−リポ酸6.2gをクロロホルム60mLに溶かし、トリエチルアミン3.2gを加えて−5℃に冷却して置き、これにクロル炭酸エチル3.3gを徐々に滴下し、滴下終了15分後にこれにアミノエタンスルホン酸4.5gおよび水酸化ナトリウム1.5gをメタノール60mLに溶かしたものを一挙に加えて、その温度で15分間、さらに室温に戻して1時間攪拌させた。次に、これに水酸化ナトリウム1.5gをメタノール50mLに溶かした溶液を加えて、減圧下で溶媒を1/3になるまで濃縮させ、これにエタノール60mLを加えて析出した結晶を濾取した。これを水−メタノールから再結晶させて、目的化合物のナトリウム塩の白色結晶5.8gを得た。融点235〜237℃。TLC、Rf=0.53(n−ブタノール:酢酸:水=4:1:2)

DL−α−リポ酸4.2gおよびトリエチルアミン2.2gをテトラハイドロフラン60mLに溶解して−5℃に冷却して置き、攪拌下、これにクロル炭酸エチル2.3gを徐々に滴下し、滴下終了後、さらに6分後にL−システイン2.6gおよびトリエチルアミン2.5gを水20mLに溶かした溶液を加えて、参考例1と同様に反応させた後、塩酸酸性として酢酸エチルで抽出し水洗後、酢酸エチルを留去させ、残渣をエタノールに溶かし、これに水酸化ナトリウム/メタノール溶液を徐々に加えてpH7として析出する白色結晶を濾取し、水/エタノールから再結晶させて、目的化合物の白色結晶4.3gを得た。融点150℃付近から徐々に分解。TLC、Rf=0.72(n−ブタノール:酢酸:水=4:1:2)
