Description
本発明は、一般に、癌、より詳細には、癌の診断または予後のために用いられ得るバイオマーカーに関する。
本発明は、癌患者の生存についての予後を決定するための方法を提供する。1つの方法は、以下:(a)癌患者由来の新生物細胞含有サンプルのTUCANのレベルを測定する工程、および(b)サンプル中のTUCANのレベルと、参照のTUCANのレベルとを比較する工程を包含し、ここで、サンプル中の低レベルのTUCANは、患者の増大した生存と相関する。
本発明は、タンパク質を含有するCARDドメイン(PCT公開 WO01/16170中でCARD−Xとして公知である)、TUCAN(カスパーゼ9の腫瘍アップレギュレートCARD含有アンタゴニスト(Tumor Up−regulated CARD−containing Antagonist of Caspase Nine))の発現が、独立してかまたは他のバイオマーカーと組み合わせて、癌を有する患者についての臨床的な結果を効果的に予測するために使用され得るという知見に関する。

Claims (46)
- 結腸癌患者の生存について予後を決定するための試薬であって、該試薬は、
該結腸癌患者由来の結腸癌細胞含有サンプルにおけるTUCANのレベルを測定するための、TUCANに対する選択的結合剤、TUCANと特異的に反応する抗体、またはTUCAN遺伝子と相補的な核酸プローブ;
を含み、
参照のTUCANのレベルと比較して低い該サンプル中のTUCANレベルは、該結腸癌患者の増大した生存と相関する、
試薬。 - 前記生存は、前記結腸癌患者が該癌の再発を有するかどうかにかかわらない生存である、請求項1に記載の試薬。
- 前記生存が、疾患に罹患していない生存である、請求項1に記載の試薬。
- 前記サンプルが、結腸腫瘍組織である、請求項1に記載の試薬。

- 結腸癌患者の生存についての予後を決定する試薬であって、該試薬は、
該結腸癌患者由来の結腸癌細胞含有サンプルにおける、TUCANおよび1つ以上のバイオマーカーのレベルを測定するための、
TUCANに対する選択的結合剤および/もしくは該1つ以上のバイオマーカーに対する選択的結合剤;
TUCANと特異的に反応する抗体および/もしくは該1つ以上のバイオマーカーと特異的に反応する抗体;または
TUCAN遺伝子と相補的な核酸プローブおよび/もしくは該1つ以上のバイオマーカーの遺伝子と相補的な核酸プローブ;
を含み、該1つ以上のバイオマーカーが、以下:cIAP2、Apaf1、Bcl−2、およびSmacからなる群より選択され、
TUCANおよび該1つ以上の選択されたバイオマーカーの参照のレベルと比較した該サンプルにおける、低レベルのTUCANおよび高レベルのApaf1、Bcl−2またはSmacのいずれか、あるいは低レベルのTUCANおよび低レベルのcIAP2は、該結腸癌患者の増大した生存と相関する、
試薬。 - 前記生存は、前記結腸癌患者が該癌の再発を有するかどうかにかかわらない生存である、請求項12に記載の試薬。
