【発明の属する技術分野】
本発明は風味の改善された複合ビタミン内服用液剤に関し、さらに詳細には、有効成分としてビタミンB群またはその誘導体を含有する保存安定性に優れた、食品、医薬品または医薬部外品としての内服用液剤に関する。

【従来の技術】
ビタミンB群またはその誘導体は苦味があり、服用しにくいことはよく知られた事実である。これを改善するために、従来のビタミンB含有内服用液剤にはショ糖(白糖)、蜂蜜を配合しても服用後に苦味が残ることから、アミノ酸とアップル系香料を配合したり(特開平5−4921号)、精油または精油成分を配合したりしている(特開平5−255126号)。また、ビタミンB1誘導体にカンゾウおよび非イオン性界面活性剤を添加したり(特開平9−328429号)、チアミンまたはその塩を配合した液剤に、ジンジャー系香料を添加して(特開平11−209266号)、苦味を低減している。

【発明が解決しようとする課題】
本発明は、ビタミンB群またはその誘導体を含有する液剤の風味改善を目的とする。

【課題を解決するための手段】
本発明者らは種々検討した結果、転化型液糖がビタミンB群またはその誘導体の苦味を特異的に低減させることを見出し、本発明を完成するに至った。
すなわち、本発明は、
(1)ビタミンB群またはその誘導体を配合した液剤において、転化型液糖を配合することにより苦味が低減された内服用液剤、
(2)ビタミンB群またはその誘導体が、チアミンおよびその誘導体、リボフラビンおよびその誘導体、ピリドキシンおよびその誘導体、ビタミンB12およびその誘導体ならびにそれらの薬学的に許容される塩から選択される1種または2種以上のものである上記(1)記載の内服用液剤、
(3)ビタミンB群またはその誘導体がチアミン、その誘導体またはその薬学的に許容される塩から選択される上記(1)1記載の内服用液剤、
(4)フルスルチアミンを含有する上記(1)記載の内服用液剤、
(5)転化型液糖の配合比率が液剤全量に対して5〜30重量%である上記(1)〜(4)いずれか1項記載の内服用液剤、
(6)液剤のpHが2〜7の範囲である上記(1)〜(5)いずれか1項記載の内服用液剤、および
(7)さらに有機酸または無機酸を配合した上記(1)〜(6)のいずれか1項記載の内服用液剤を提供するものである。

【発明の実施の形態】
本発明の内服用液剤で用いるビタミンB群およびその誘導体は、この種の液剤に通常用いられるものいずれでもよく、例えば、チアミン、チアミンの薬学的に許容される塩(例、チアミン塩酸塩、チアミン硝酸塩等)、チアミン誘導体(例、プロスルチアミン、フルスルチアミン、オクトチアミン、アリチアミン、チアミンジスルフィド、O−ベンゾイルチアミンジスルフィド、チアミンモノホスフェートジスルフィド、O,S−ジベンゾイルチアミン、S−ベンゾイルチアミン、ベンホチアミン、ジセチアミン、ジクロカルボチアミン等)、チアミン誘導体の薬学的に許容される塩(例、塩酸塩、硝酸塩、リン酸、硫酸塩等の酸付加塩)、リボフラビン、リボフラビン誘導体(例、酪酸リボフラビン、リン酸リボフラビンナトリウム等)、ピリドキシン、ピリドキシンの薬学的に許容される塩(例、ピリドキシン塩酸塩等)、ピリドキシン誘導体(例、ピリドキサルリン酸、ピリドキサミンリン酸等)、ビタミンB12(例、シアノコバラミン、ヒドロキソコバラミン、酢酸ヒドロキシコバラミン、メチルコバラミン等)等から選ばれる1種または2種以上が挙げられる。

本発明で用いる転化型液糖は、サトウキビやビートの搾汁を濃縮し、ショ糖分を結晶させ、取り出した原料粗糖から製造される。この粗糖を溶解し、活性炭(骨炭)やイオン交換樹脂で精製して極めて純度の高いショ糖液とした後、さらにイオン交換樹脂で処理し、ショ糖を加水分解させて得られる。得られた転化型液糖の一般成分は、純糖率が30.0〜46.2%、還元糖分が30.0〜46.2%、転化率が40.0〜60%、糖度が74.2〜76.2%、好ましくは純糖率が35.7〜40.3%、還元糖分が35.7〜40.3%、転化率が46.0〜55.0%、糖度が74.7〜75.7%であり、一般に、液剤全量に対して5〜30重量%、好ましくは、15〜25重量%の配合比率で使用される。
本発明においては、特に限定するものではないが、転化型液糖のショ糖、ブドウ糖、果糖の混合液の比率が、ビタミンB群またはその誘導体の苦味のマスキングに関係するものと推測され、ショ糖、ブドウ糖、果糖の比率は、ショ糖30〜60%、ブドウ糖15〜30%、果糖15〜30%、好ましくはショ糖40〜60%、ブドウ糖20〜30%、果糖20〜30%であることが望ましく、さらに好ましくは、約50%転化し転化糖(ショ糖を分解して得られるブドウ糖と果糖の混合物)とショ糖の等量組成液糖が望ましい。また、ショ糖:果糖:ブドウ糖の比率は、1.3〜3.1:1:1が好ましく、さらに好ましくは、ショ糖:果糖:ブドウ糖の比率が、ほぼ2:1:1の液糖である。
このような転化型液糖は、市販のものが入手可能であり、例えば、台糖株式会社の転化型液糖DLIとして市販されている液糖が使用できる。

本発明により苦味を有するビタミンB群またはその誘導体は転化型液糖のみで苦味をマスキングすることができるが、転化型液糖以外の甘味剤を併用してもよい。このような甘味剤としては、砂糖、果糖、ブドウ糖、麦芽糖、液糖、果糖ブドウ糖液糖、ブドウ糖果糖液糖、トレハロース、パラチノース、マルチトール、ソルビトール、パラチニット、エリスリトール、キシリトール、ソーマチン、シュクラロース、ステビア抽出精製物などが挙げられる。

本発明の内服用液剤における、苦味を有するビタミンB群またはその誘導体の配合量は、好ましくは、転化型液糖1重量部に対して0.0001〜0.002重量部、より好ましくは、0.0001〜0.001重量部である。

本発明の液剤は、風味の点からpH2〜7の範囲が好ましく、pH2.5〜6.5の範囲がさらに好ましい。pHがこの範囲を外れると、ビタミンB群またはその誘導体の安定性に影響がある。

本発明の液剤はさらに有機酸または無機酸の配合により苦味を低減できる。有機酸としてはクエン酸、DL−リンゴ酸、酒石酸、乳酸、グルタミン酸、アスパラギン酸などをあげることができる。また、無機酸としては、塩酸、リン酸などをあげることができる。それらのうちで特に好ましいものとしてクエン酸、DL−リンゴ酸またはリン酸をあげることができる。

本発明の液剤には上記成分の他、通常液剤に用いることが可能な成分、例えばアミノエチルスルホン酸、アスパラギン酸、アルギニン、リジン、アスコルビン酸、ニコチン酸アミド、ビタミンAもしくはその誘導体、ビタミンEもしくはその誘導体、塩化カルニチン、コンドロイチン硫酸、カフェイン、生薬(ニンジン、トチュウ、ロクジョウ、ケイヒ、ガラナ、シャクヤク、タイソウ、ショウキョウ、カンゾウ、カイバ、ニクジュヨウ、ハゲキテン、オウギ、トウキ、ブクリョウ、ビャクジュツ、ローヤルゼリーなど)、プロピレングリコール、保存剤などを配合することができる。

本発明の液剤は、所望の成分を混合、溶解するような常法に従って製造でき、食品、医薬品および医薬部外品としての内服用液剤とすることができる。

健康な成人10人を選択し、苦味に関する官能検査を行った。官能検査は、各検体の苦味を「非常に苦い」、「苦い」、「わずかに苦い」、「苦くない」の4段階で評価した。各被験者が試験する検体の順序は、全てランダムに行った。その結果を表1に示した。

表1から明らかなように、比較例では「非常に苦い」、「苦い」、「わずかに苦い」と評価された割合が多いのに対して、本発明の実施例では「わずかに苦い」、「苦くない」の評価であった。
