検出対象である物質に感応する感応性物質を含む化学センサ用膜を有する脂質膜センサ、イオンセンサ、等の化学センサに係り、特に化学センサの応答領域の広域化に関する。

【従来の技術】
ここでは脂質膜センサを例に挙げて説明する。
本願出願人は、他と共同して、先に「味覚センサ及びその製造方法」の発明につき特許出願をし(特開平3-54446 号)、その明細書及び図面によって、ある種の高分子重合体の表面マトリックス内に特定の分子配列をもって収納されたいわゆる脂質性分子群が、基本味と呼ばれる塩味、酸味、苦味、甘味に対して、感度を示すセンサとなることを示した。しかも、この種のセンサは、人間の五感の一つである味覚に代わり味を測定できるものであることを示した。

ここで、味覚センサに用いる脂質膜について説明する。
脂質膜に用いられる脂質には表1に示すようなものがある。

脂質膜1の模式図を、化学物の設計法で使われている表現方法で表したものが図8である。脂質性分子のうち円で示した球状部は親水基aすなわち親水性部位aであり、それから原子配列が長く延びる炭化水素の鎖構造b(たとえばアルキル基)がある。図ではいずれの場合も2本の鎖が延びて一つの分子を表しており、全体で分子群を構成している。この炭化水素の鎖の部分は、疎水性部位bである。このような脂質性分子群3が、膜部材4の表面のマトリクス5(表面の構造、平面的なひろがりをもったミクロな構造)の中及びマトリクス内部に溶け込ませた形(たとえば図8の2’)で収容されている。その収容のされ方は、親水性部位が表面に配列するようなものとなっている。

脂質を支持するマトリクスは、容易に入手でき、取扱いも簡単な、熱可塑性のポリ塩化ビニル( [CH2 CHCl]n ;PVC)等が用いられる。PVC以外には表2に示すような物質がある。

PVCは、テトラヒドロフラン(THF)、ニトロベンゼン、シクロヘキサノン等に溶け、可塑剤との混合比を変えることにより、軟質にも、硬質にもすることができるから、用途に応じて使いわけができる便利さがある上に、品質の安定性、成形の容易さも特徴とされる。

PVC、可塑剤、脂質を概ね2:3:1の重量比で混合する。可塑剤を添加しないと出来上がりの脂質膜が白濁していたり、不均一になったりして好ましくない。また、脂質・可塑剤の選び方、混合する比率、混合の仕方によっても、出来上がった脂質膜に白濁や不均一を生ずることがある。可塑剤としてはフタル酸ジオクチル(DOP)、ジオクチルフェニルフォスフォネート(DOPP)、あるいは、リン酸トリクレシル(TCP)が用いられる。

表1の脂質とPVCとを混合したもの約400mgを、THF10ccに溶解し、平底の容器(例えば85mmφのシャーレ)に写し、THFを揮散させて、脂質膜を形成すると、厚さがほぼ200μmの脂質膜が得られる。
脂質と可塑剤は、PVCというプラスチックの表面マトリクスの組織中に固定されていると考えられている。この脂質膜を約10mmol/lの食塩水、あるいは、塩化カリウム水溶液などの電解質溶液に1分間ほど浸すと、脂質のもつ親水基が表面に整列した分子配列が安定した状態で得られ、味覚センサとしての機能を果たすものとなる。
なお、脂質は両親媒性物質の一種であり、表3に示す脂質以外の両親媒性物質、表4に示す苦味物質を脂質の代わりに用いた感応膜も味覚センサに利用できる。

図9は前述のようにして得られた脂質膜1をセンサプローブ6に加工した概略構成を示す断面図である。
基材7として外径8mm、長さ55mm、肉厚1mmのアクリルの円筒を用いる。該円筒7の先端部7aを約60度の角度でカットする。該カットされた先端部7aに、その開口部7bを塞ぐように脂質膜1を、THFにPVCを溶解したものを接着剤として、張り付ける。直径0.5mmの銀線の先を螺旋状に巻いたものを該円筒7の上部から挿入し電極8とする。緩衝層9として電解液(3.3mmol/lの濃度の塩化カリウム溶液)を前記電極8の螺旋部分8bが浸るところまで充填する。

また、この脂質膜を用いて、マルチチャンネルのセンサとしたものが図10(a),(b)である。本図ではマルチチャンネルのアレイ電極のうち三つの感応部が示されている。
図示の例では、基材7に 0.5mmφの孔を貫通して、それに銀の丸棒を差し込み電極8とした。脂質膜1は緩衝層9を介して電極8に接触するように基材7に張りつけている。

この脂質膜を使用したセンサ(脂質膜センサ)は、味覚センサとして味の測定に使用できるだけではなく、水質の測定に使用した場合、遊離シアン、錯体シアン、金属イオンの濃度検知に有効であることも実験で確認されている。このように味の測定、水質の測定等に利用できる脂質膜中の脂質の割合は前述の例のように約1/6即ち17重量%程度であった。

また、他の化学センサについて触れれば、たとえば第4級アンモニウム塩を高分子化合物からなる支持材中に分散させた化学センサ用膜を有する陰イオン選択性電極(イオンセンサの一種)が、人間の生体液中の塩化物イオンなどの濃度測定に使用されている。この陰イオン選択性電極に使用する膜中の感応性物質の濃度は20重量%以上であるのが一般的である。

これまで使用されてきた化学センサ用膜は、膜中に感応性物質をできるだけ多く溶解させるように合成されてきた。それは膜中に感応性物質が多い方が高感度になると考えられていたためである。このため従来の化学センサ用膜には少なくとも15重量%以上の感応性物質が含まれていた。
従来の化学センサには、そのような化学センサ用膜が用いられており、或る感応性物質1種類について見れば、膜中の当該感応性物質の濃度は1種類であった。図10に示したマルチチャンネルの脂質膜センサも、用いられる複数の脂質膜は、脂質の種類が異なったものであり、同一の脂質について膜中の濃度を変えたものではない。

【発明が解決しようとする課題】
従来の化学センサで人が感じる範囲の味物質の濃度や体液中の塩化物イオンの濃度を測定する場合は、これらの濃度範囲で十分な感度が得られる。しかし特に水質の測定におけるように低濃度のイオンなどを検知しようとした場合、例えば錯体シアンを検知しようとした場合、これまでの感応性物質が15重量%以上含まれている脂質膜などの感応膜を使用したセンサでは、高濃度では感度が得られるものの1.0ppm程度の排水基準値前後の濃度では十分な感度が得られない。発明者等は膜中の感応性物質の濃度を低くすると検出対象物質が低濃度の場合の感度が向上することを発見した。しかし膜中の感応性物質の濃度を低くした膜は検出対象物質が高濃度の場合、膜中の感応性物質の濃度が高い膜に比べ感度が低い。
本発明の目的は、検出対象物質の検知可能な濃度範囲を広げ、言い換えれば広い濃度範囲に対してセンサに感度を持たせ、センサの応答領域を広域化することである。

【課題を解決するための手段】
本発明の化学センサは、前述の課題を解決するために、高分子材と、可塑剤と、検出対象である物質に感応する感応性物質としての脂質とを混合してそれぞれ個別に形成した膜を複数有する化学センサにおいて、前記複数の膜の脂質は単一且つ同一の種類であって、膜毎に前記脂質の濃度が異なるように構成されている。また、請求項2に対応する本発明の化学センサは、前記複数の膜の脂質がジオクチルフォスフェートであり、該ジオクチルフォスフェートの膜中の濃度が10重量%以下である膜と15重量%以上である膜とを含む。請求項3に対応する本発明の化学センサは、前記複数の膜の脂質がジテトラデシルメチルアンモニウムブロミドであり、該ジテトラデシルメチルアンモニウムブロミドの膜中の濃度が1.4重量%である膜と、5.3重量%である膜とを含む。

【発明の実施の形態】
化学センサには脂質膜センサ、イオンセンサなどがあるが、いずれのセンサでも感応性物質の濃度を変えて感度を向上させるという方法は適用できる。ここでは、脂質膜センサを例に挙げて説明する。

図1に本発明の第一の実施の形態である脂質膜センサの概略構成図を示す。各プローブ6,6は構造的には、従来の脂質膜センサと変わらないが、本発明の脂質膜センサは、各プローブ6,6が備えている脂質膜の膜中の脂質の濃度が互いに異なっている。

図2に本発明の第二の実施の形態である脂質膜センサの概略構成図を示す。図2(a)は正面図、図2(b)は断面図である。第一の実施の形態と同様に、構造的には、従来の脂質膜センサと変わらないが、本発明の脂質膜センサは、各チャンネルの脂質膜の膜中の脂質の濃度が互いに異なっている。

脂質の膜中の濃度は重量%で0.006〜20%の間程度で、一般的には5〜10倍の割合で脂質濃度の異なる複数の膜を作製するのが良く、これらのうち測定対象の検知に有効な範囲のものを複数組み合わせて使用する。
前記複数の膜は、図1に示すように、別々の基材(図1では円筒7)に貼り付けて、複数本のプローブを1つのセンサとしてもよく、あるいは、図2に示すように、1つの基材に貼り付けて、1本のセンサとしてもよい。
また、脂質膜の膜中の脂質の濃度が互いに異なっている複数の膜を用いるとともに、脂質の種類が異なった膜も用いることとすれば、例えば味について情報の得られる味物質の濃度範囲が広がるとともに、味についての情報も増える。

このようにして得られた脂質膜センサを用いて、錯体シアンの一例としてフェロシアン化カリウム(K4[Fe(CN)6] )の測定、およびキニーネの測定を行った。
キニーネの測定に用いた脂質膜センサの脂質膜は、脂質にジオクチルフォスフェートを用い、膜中の脂質の量(濃度)が、1.2重量%、5.3重量%、10.9重量%、20.4重量%の4種類である。また、フェロシアン化カリウムの測定に用いた脂質膜センサの脂質膜は、脂質にジテトラデシルジメチルアンモニウムブロミドを用い、膜中の脂質の量が、1.4重量%、2.7重量%、5.3重量%の3種類である。

測定系10を図3に示す。
被測定溶液11はビーカーのような容器12に入れる。被測定溶液11中に、前記センサプローブ6を入れた。使用前に、塩化カリウム1mmol/l水溶液で電極電位を安定化した。
測定の基準となる電位を発生する電極として参照電極13を用意し、それを被測定溶液11に入れる。センサプローブ6と参照電極13とは所定の距離を隔てて設置する。参照電極13の表面は、緩衝層14として、飽和塩化カリウム溶液を寒天で固化したもので覆ってある。

脂質膜1からの電気信号は、リード線15によってバッファ増幅器17に導かれる。バッファ増幅器17の出力は、A/D変換器18に加えられる。参照電極13からの電気信号もリード線16を介してA/D変換器18に加えられ、脂質膜1からの電位との差をディジタル信号に変換する。このディジタル信号はマイクロコンピュータ19で適当に処理され、またX−Yレコーダ20で表示される。

被測定溶液11を用意する。
錯体シアンについては、純水に10mmol/lのKClを添加した溶液を基準液とした。
基準液と同じ溶液にフェロシアン化カリウムをシアンの濃度で0.01ppm、0.1ppm、1.0ppm、3.0ppm、10.0ppmになるように添加してサンプル液とした。

キニーネについては、純水に30mmol/lの塩化カリウム(KCl)と3.0mmol/lの酒石酸を添加した溶液を基準液とした。
基準液と同じ溶液にキニーネを0.0010mmol/l、0.0033mmol/l、0.0100mmol/l、0.0330mmol/l、0.1000mmol/l、0.3300mmol/l、1.0000mmol/l、3.3000mmol/l、10.0000mmol/l、33.0000mmol/l添加してサンプル液とした。

第一の基準液と第二の基準液には、ともに前述の基準液を用いた。第一の基準液と第二の基準液とを用いるのはセンサの継続的なドリフトの影響と基準液を繰り返し測定することによって基準液自体が継続的に変化するという影響をできるだけ少なくするためにとられる処置である。

錯体シアン測定の手順は次の通りである。
1) 保存液(第一の基準液に同じ)に脂質膜を用いた味覚センサをほぼ10時間浸漬する。
2) 第一の基準液(洗浄用)へ味覚センサの出し入れを10回行う。第一の基準液(洗浄用)で洗浄するといってもよいし、第一の基準液に断続的に浸漬するといってもよいし、味覚センサの脂質膜の表面に刺激を与えるということもできる。
3) 第一の基準液で測定用として用意したものに浸漬し、30秒後に味覚センサの電位を測定し、測定値をV0 とする。
4) 手順2)、3)を2回以上繰り返し、測定ごとに今回の測定値V0 と前回の測定値V0 の差が所定の値以下かどうかを判断し、所定の値以下(つまりV0 が安定したら)であれば手順5)へ進む。
5) 味覚センサを第一の基準液(測定用)から出して、第二の基準液(洗浄用)で洗浄する。(前記2)と同様に10回出し入れをする。)
6) 第二の基準液(測定用)に味覚センサを浸漬し、30秒後に味覚センサの電位Vk を測定する。
7) 再度第一の基準液(洗浄用)へ味覚センサの出し入れを10回行う。第一の基準液(洗浄用)で洗浄するといってもよいし、第一の基準液に断続的に浸漬するといってもよいし、味覚センサの脂質膜の表面に刺激を与えるということもできる。
8) 第一の基準液で測定用として用意したものに浸漬し、30秒後に味覚センサの電位を測定し、測定値をV0 ´とする。
9) 手順7)、8)を2回以上繰り返し、測定ごとに今回の測定値V0 ´と前回の測定値V0 ´の差が所定の値以下かどうかを判断し、所定の値以下(つまりV0 が安定したら)であれば手順10) へ進む。
10) 味覚センサを第一の基準液(測定用)から出して、サンプル液(洗浄用)で洗浄する。(前記2)と同様に10回出し入れをする。)
11) サンプル液(測定用)に味覚センサを浸漬し、30秒後に味覚センサの電位Vs を測定する。
12) 測定の手順2)に戻り手順2)〜11) を繰り返す。所定の回数繰り返したら手順を終わる。
この手順を図4に示した。

また、キニーネ測定の手順は次の通りである。
S1 (第1の)基準液のセンサ電位V01を測定する。
バッチ式測定の場合、味覚センサを空中に一定回数出し入れした後、再度基準液のセンサ電位V01を測定する。フロー式測定の場合、味覚センサに基準液を一定時間流した後、再度基準液のセンサ電位を測定する。
S2 一回前に測定した基準液のセンサ電位V01と比較して、変化の幅が設定値以内に収まっていれば、安定したとみなし、S3へ進みサンプルのセンサ電位を測定する。変化幅が設定値以上であった場合、S1に戻る。この意味は、センサの出し入れや測定液のフローの影響をチェックし、影響がなくなるまでセンサの出し入れや測定液のフローの操作を周期的に行うものである。最終的に安定した基準液のセンサ電位をV01とする。
S3 センサが安定した所で、所定時間サンプルSi にセンサを浸漬すると共にサンプルSi のセンサ電位Vi を測る。
S4 サンプルSi の測定結果△Vi =Vi −V01を算出する。
S5 センサを洗浄する。或る程度の強さで洗浄することで、その強さに耐える吸着力をもつ物質だけがセンサに残る。
S6 (第2の)基準液のセンサ電位V02i を測定する。
S7 サンプルSi の測定結果△Vki=V02i −V01を算出する。
S8 センサを洗浄する。その後、連続してサンプルを測定する場合はS1へ進む。
この手順を図5に示した。

ジテトラデシルジメチルアンモニウムブロミドの濃度が1.4重量%の膜と5.3重量%の膜に注目して見てみると、フェロシアン化カリウムの濃度0.5ppmあたりまでは、1.4重量%の膜の方が出力、感度(曲線の傾き)ともに大きい。0.5ppmより濃度の高いところでは、5.3重量%の膜の方が感度が高くなる。したがって、このような脂質膜の組合せの場合は、フェロシアン化カリウムの濃度0.5ppmあたりまでは、1.4重量%の膜の出力を用い、0.5ppmより濃度の高いところでは、5.3重量%の膜の出力を用いてフェロシアン化カリウムの濃度を測定するようにすればよい。

このように、フェロシアン化カリウムの低濃度側は脂質濃度の低い膜、高濃度側は脂質濃度の高い膜といった様に、同一脂質の膜を使用しても脂質濃度を変えることで、感度の得られる範囲を広くすることができる。

ジオクチルフォスフェートの濃度が1.2重量%の膜と5.3重量%の膜を比べると、キニーネの濃度0.7ppmあたりまでは、1.4重量%の膜の方が出力、感度(曲線の傾き)ともに大きい。0.7ppm〜9ppmあたりでは1.4重量%の膜の方が出力は大きいが、感度(曲線の傾き)は小さくなり、9ppmより高濃度のところでは、1.4重量%の膜の方が出力、感度(曲線の傾き)ともに小さい。また、ジオクチルフォスフェートの濃度が5.3重量%の膜と20.4重量%の膜を比べると、キニーネの濃度4ppmあたりまでは、5.3重量%の膜の方が出力、感度(曲線の傾き)ともに大きい。4ppm〜40ppmあたりでは5.3重量%の膜の方が出力は大きいが、感度(曲線の傾き)は小さくなり、40ppmより高濃度のところでは、5.3重量%の膜の方が出力、感度(曲線の傾き)ともに小さい。したがって、このような脂質膜の組合せの場合は、たとえば、キニーネの濃度0.7ppmあたりまでは、1.4重量%の膜の出力を用い、0.7ppm〜4ppmでは5.3重量%の膜の出力、4ppmより濃度の高いところでは、20.4重量%の膜の出力を用いてキニーネの濃度を測定するようにすればよい。

このように、脂質膜中の脂質の濃度の異なる膜を組み合わせて使用することで、キニーネの測定でも、センサの応答領域を広くでき、また、広い濃度範囲にわたって高感度化できる。

本発明は脂質膜センサの場合は前述のような膜の合成法や測定方法となるが、このほかの化学センサについても、膜中の感応物質の濃度を変えた膜を複数使用すれば、1種類の濃度の膜で測定した場合に比べて検出対象である物質が低濃度から高濃度まで測定できるようになり、応答領域の広域化になる。また、応答領域内のある濃度について見れば、高感度化にもなる。

【発明の効果】
本発明によれば、検出対象である物質に感応する感応性物質を含んだ膜を有する化学センサで、膜中の感応性物質濃度が互いに異なる少なくとも2種類の膜を有する化学センサとすることで、検出対象物質の検知可能な濃度範囲を広げ、センサを応答領域を広域化することができる。

The applicant of the present application has previously filed a patent application regarding the invention of “taste sensor and its manufacturing method” in cooperation with others (Japanese Patent Laid-Open No. 3-54446). PVC is soluble in tetrahydrofuran (THF), nitrobenzene, cyclohexanone, etc., and can be made soft or hard by changing the mixing ratio with the plasticizer. In addition, it is characterized by quality stability and ease of molding.
