【産業上の利用分野】
本発明は、医薬などとして有用な、β-ジガラクトシルセラミド誘導体の新規化合物、新規中間体および工業的に優れた新規製造方法に関する。

【従来技術】
天然由来のジガラクトシルセラミド(Gala1-4Galβ1-1Ceramide)は、下記化学式で表される側鎖が異なる3化合物の混合物であり、スーパー抗原(Staphylococcal enterotoxin B、以下 SEB)が、ヒト腎近位尿細管培養細胞に特異的に結合する際の結合部位であることが知られている[Glycobiology,5(3),327-333(1995).]。

ジガラクトシルセラミドはSEBと結合することから、SEBのT細胞活性化反応に何らかの影響を与えていることが推測され、スーパー抗原のT細胞阻害、サイトカインの産生抑制、あるいは好中球の増殖抑制等の薬理効果が期待されている。
さらに具体的には、例えば、以下の医薬用途等に有用である。

(1) 抗癌剤(抗癌、癌転移抑制)
最近、Phosphokinase C translocationをブロックすることによって、PhospholipaseA2を阻害し、抗癌活性を発現すると言われている物質が見つかっている(Eur.J.Pharmacol.Vol.294:555-563,1995.)。すでに、staurosporineやsphingosineなどはPhosphokinase Cを阻害することによって、carcinoma cell lineの浸潤や癌細胞のlaminineやcollagenへの接着を阻害することが知られている。これらのことはスフィンゴ糖脂質の癌転移抑制作用の可能性を示唆するものである。
糖脂質やスフィンゴミエリンの前駆物質あるいは代謝産物であるセラミドは、生体内で修飾分解され、スフィンゴシン、セラミド-1-リン酸、スフィンゴシン-1-リン酸等に変換されていく。これらの誘導体は細胞の分化、増殖に機能的に関与していることが示されつつある(薬学雑誌、Vol.114,655-668,1994.)。TNF-αによるアポトーシスの誘導のセカンドメッセンジャーとしてのセラミドの可能性が強く示唆されている(Science,Vol.259,1769,1993.)。スフィンゴ糖脂質がこれらの作用に代わる可能性により、抗癌剤としての有用性が期待される。

(2) 免疫抑制剤(臓器移植など)
Isalia sinclairii(ATCC No.24400)から発見されたスフィンゴシン類似構造を持つISP-1はサイクロスポリンAよりも強力な免疫抑制作用を示すことが示されている。その作用機序はIL-2産生経路を阻害するサイクロスポリンAとは異なり、細胞障害性T細胞のアポトーシスの誘導を起こすと考えられている。このアポトーシスの誘導はISP-1がスフィンゴ脂質生合成経路のセリンパルミトイルトランスフェラーゼを阻害することによって、スフィンゴ脂質を減少させた結果であると考えられている(生化学、Vol.68,444-452,1996.)。スフィンゴ糖脂質がこのスフィンゴ脂質生合成経路の調節に関与している可能性により、免疫抑制作用が期待される。

(3) 抗HIV剤
Galactosylceramide(GalCer)は神経ならびに結腸上皮のCD14negative細胞においてはhuman immunodeficiency virus(HIV)の侵入時のリセプターとなっている。GalCerは、HIV-1 surface envelope glycoprotein gp120のV3 regionを認識することが知られている。このV3 loopはHIV-1によるFusion processに重要な役割を担っている。
GalCerの可溶性アナローグ[CA52(N15)]が合成され、その抗HIV作用が検討された(J.Biol.Chem.Vol.272,7245-7252,1997.)。CA52(N15)はGalCerを発現するCD4 positive,negative細胞のいずれにおいてもHIVによる細胞fusionとHIVの侵入を阻害した。この結果はスフィンゴ糖脂質の抗HIV作用の可能性を示すものである。

(4) 抗毒素作用(Staphylococcus enterotoxin-B,Velo toxinなど)
Digalactosylceramide(GalGalCer)はヒト腎の近位尿細管においてStaphylococcus enterotoxin-B(SEB)のリセプターであることが示された(Glycobiology Vol,5;327-333,1995.)。このことはDigalactosylceramideならびにそのアナローグを可溶性リセプターとして使用すれば、Staphylococcus aureus感染にともなって遊離されるSEBの中和剤と成り得る可能性を示唆するものである。
また、スフィンゴ糖脂質Globotriaosylceramide(Gb3)はVerotoxinに結合することが知られており、母乳中に豊富に含まれて、乳児の感染に伴うVerotoxinによる下痢に対して防御的な作用を持つとの仮説も立てられている(J.Infect.Vol.166:832-836,1992.)。このようなことからGb3を含むスフィンゴ糖脂質はVerotoxinの有効な中和剤となり得る。

(5) ゴーシェ病
グルコシルセラミドの蓄積症であるゴーシェ病は、リソゾーム中に存在するグルコシルセラミドの分解酵素であるグルコシルセレブロシダーゼ遺伝子の変異によって発症する。この疾患では肝臓およびひ臓の肥大が主要な症状として現れるが病態によっては神経症状を呈する(ファルマシア、Vol.32:1365-1368,1996.)。従って、特異的なグルコシルセラミドの合成阻害剤はこの疾患に有効である。

【本発明が解決しようとする問題点】
しかし、従来は天然物からの抽出する以外にジガラクトシルセラミドを得る方法はなく、安価かつ大量に得ることはできなかった。

【課題を解決するための手段】
本発明者らは、上記問題点の改善を目指して鋭意研究を進めてきた。その結果、下記化学反応式で表される工程に従い、保護二糖類活性体(I)とアルコール誘導体(II)とを反応させて保護β-ジガラクトシルセラミド誘導体(III)とし、次いで脱保護することにより、目的とするβ-ジガラクトシルセラミド誘導体(IV)が、安価かつ大量に製造できることを見出し、本発明を完成するに至った。

さらに本発明により、ジガラクトシルセラミドの新規誘導体も得ることができ、より優れた薬理活性と安全性を期待することができる。従って本発明は、医薬として優れた新規誘導体を提供するものであり、さらにその製造にあたり有用な新規中間体を提供するものである。

続いて本発明について、以下に詳述する。
まず、本発明にかかる保護二糖類活性体(I)は下記一般式で表される。

式中、R1は水酸基の保護基を、Xはハロゲン原子をそれぞれ意味する。
ここで水酸基の保護基とは、通常有機合成において用いられる水酸基の保護基であれば限定されないが、具体的には、例えばアセチル基、ベンゾイル基等のアシル基、メチル基、ベンジル基等のエーテル残基等を挙げることができる。また2つのR1がジメチルケタール(イソプロピリデン)基またはフェニルアセタール(ベンジリデン)基等の環状エーテルを形成してもよい。さらにそれぞれのR1は、2種類以上の相異なる基であってもよい。
次にハロゲン原子とは具体的には、塩素原子、臭素原子、フッ素原子を意味し、フッ素原子がより好ましい。
なお、本発明にかかる保護二糖類活性体(I)には、2つのガラクトース残基間の(1→4)グリコシド結合において、α結合とβ結合の2種類の立体異性体が存在するが限定されず、いずれか一方の異性体あるいは混合物であってもよいが、α結合がより好ましい。
保護二糖類活性体(I)としてより具体的には、例えば次の化合物を挙げることができるが、これらには限定されない。
(1) O-(2',3',4',6'-テトラ-O-アセチル-α-D-ガラクトピラノシル)-(1→4)-2,3,6-トリ-O-アセチル-β/α-D-ガラクトピラノシル・フルオリド
(2) O-(2',3',4',6'-テトラ-O-アセチル-α-D-ガラクトピラノシル)-(1→4)-2,3,6-トリ-O-アセチル-β/α-D-ガラクトピラノシル・ブロミド
(3) O-(2',3',4',6'-テトラ-O-アセチル-α-D-ガラクトピラノシル)-(1→4)-2,3,6-トリ-O-アセチル-β/α-D-ガラクトピラノシル・クロリド

本発明にかかるアルコール誘導体(II)は、分子内に二重結合を有しており、(E)あるいは(Z)幾何異性体が存在するが、本発明においては限定されずいずれか一方あるいは混合物でもよいが、(E)体がより好ましい。さらにアルコール誘導体(II)は分子内に不斉炭素原子を有しており、(R)あるいは(S)光学異性体が存在するが、本発明においては限定されずいずれか一方あるいはラセミ体でもよいが、(2S,3R)異性体がより好ましい。
アルコール誘導体(II)としてより具体的には、例えば次の化合物を挙げることができるが、これらには限定されない。
(1) 1-O-ベンゾイル-2-N-パルミトイルスフィンゴシン
(2) 1-O-アセチル-2-N-パルミトイルスフィンゴシン
(3) 1-O-ベンジル-2-N-パルミトイルスフィンゴシン
(4) 3-O-ベンゾイル-2-N-パルミトイルスフィンゴシン

次に、保護β-ジガラクトシルセラミド誘導体(III)は、下記一般式で表される。本発明にかかる保護β-ジガラクトシルセラミド誘導体(III)は新規化合物であり、本発明において中間体として重要である。

本発明にかかる保護β-ジガラクトシルセラミド誘導体(III)にも、2つのガラクトース残基間の(1→4)グリコシド結合において、α結合とβ結合の2種類の立体異性体が存在するが限定されず、いずれか一方の異性体あるいは混合物であってもよいが、α結合がより好ましい。また保護β-ジガラクトシルセラミド誘導体(III)は分子内に二重結合を有しており、(E)あるいは(Z)幾何異性体が存在するが、本発明においては限定されずいずれでもよいが、(E)がより好ましい。さらに保護β-ジガラクトシルセラミド誘導体(III)は分子内に不斉炭素原子を有しており、(R)あるいは(S)光学異性体が存在するが、本発明においては限定されずいずれでもあるいはラセミ体でもよいが、(2S,3R)異性体がより好ましい。
保護β-ジガラクトシルセラミド誘導体(III)としてより具体的には、例えば次の化合物を挙げることができるが、これらには限定されない。なおここでスフィンゴシンとは、R2がペンタデカニル基、ヘプタデカニル基または8-ヘプタデセニル基のいずれかである、2-アミノ-4-オクタデセン-1,3-ジオールを意味する。

続いて本発明にかかる製造方法について詳述する。
本発明においては、保護二糖類活性体(I)と一般式ROHで表されるアルコール誘導体(II)とを反応させて保護β-ジガラクトシルセラミド誘導体(III)を得る工程(工程1)、と保護β-ジガラクトシルセラミド誘導体(III)を脱保護し目的とするβ-ジガラクトシルセラミド誘導体(IV)を得る工程(工程2)からなる(前記化学反応式[化11]参照)。

(1) 工程1
本工程はグリコシド合成の定法により行うことができるが、より具体的には、例えば Tetrahedron Lett.,27,4753-4756(1986). 等に記載された方法に従って行うことができる。

本工程は、通常有機合成において用いられる保護基の脱離法、例えば加水分解、接触還元等により行うことができる。

次に、本発明にかかるβ-ジガラクトシルセラミド誘導体(IV)あるいはβ-ジガラクトシルセラミド誘導体(V)は医薬として有用であるが、より具体的には、例えば以下の適応症を挙げることができるが、それらには限定されない。
(1) 抗癌剤(抗癌、癌転移抑制)
(2) 免疫抑制剤(臓器移植など)
(3) 抗HIV剤
(4) 抗毒素作用(Staphylococcus enterotoxin-B,Velo toxinなど)
(5) ゴーシェ病

続いて本発明を具体的に説明するため、以下に製造例および実施例を掲げるが、本発明がこれらに限定されないことは言うまでもない。

製造例1 O-(2',3',4',6'- テトラ -O- アセチル - α -D- ガラクトピラノシル )-(1 → 4)-2,3,6- トリ -O- アセチル - β / α -D- ガラクトピラノースの合成

無水酢酸(2.5ml)とピリジン(5.0ml)の混合物を氷水中で冷却し、ここに 4-O-α-D-ガラクトピラノシル-D-ガラクトピラノース 500mg(1.46mmol)と4-ジメチルアミノピリジン(DMAP, 1mg)を加え室温で17時間攪拌した。その後、反応溶媒を減圧下留去した。残査を氷水で洗浄後、塩化メチレン(100ml)に溶解し、2%クエン酸水溶液(20ml)と飽和炭酸水素ナトリウム水溶液(20ml)で洗浄後、乾燥し溶媒を減圧下留去した。得られた残査をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル系)で精製して、無色アモルファス状の標題化合物 970mg(α/β=1:1)を得た。(収率; 98%)

製造例2 O-(2',3',4',6'- テトラ -O- アセチル - α -D- ガラクトピラノシル )-(1 → 4)-2,3,6- トリ -O- アセチル - β / α -D- ガラクトピラノシル・フルオリドの合成

前製造例で得たO-(2',3',4',6'-テトラ-O-アセチル-α-D-ガラクトピラノシル)-(1→4)-2,3,6-トリ-O-アセチル-β/α-D-ガラクトピラノース 500mg(0.74mmol)のジメチルホルムアミド(5ml)溶液に、ヒドラジンアセテート 81mg(0.88mmol)を加え、50℃で2時間攪拌した。その後減圧下溶媒を留去し、得られた残査を酢酸エチル(100ml)で希釈した。有機層を水(20ml)、次いで飽和食塩水(20ml)で洗浄後、無水硫酸マグネシウムで乾燥し、減圧下溶媒を留去した。得られた残査をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル系)で精製して、無色アモルファス状の1-ヒドロキシル体 420mgを得た。(収率; 90%)
続いて、得られた1-ヒドロキシル体 420mg(0.66mmol)を1,2-ジメトキシエタン(4.2ml)に溶解し、-30℃に冷却した。ここにすばやくジエチルアミノサルファートリフルオリド[DAST, 210ml(1.6mmol)]を加え、室温で50分攪拌した。その後、再び-30℃に冷却し、メタノール(0.1ml)を加え、その温度で数分間攪拌した。その後、減圧下溶媒を留去し、残査を塩化メチレン(100ml)で希釈した。これを飽和炭酸水素ナトリウム(20ml)で洗浄し、乾燥後、溶媒を減圧下留去した。得られた残査をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル系)で精製して、無色アモルファス状の標題化合物 350mg(α/β=2:1)を得た。(収率; 83%)

280 mg (0.47 mmol) of (2S, 3R, 4E) -N-palmitoylsphingosine was dissolved in pyridine (5 ml), 260 mg (0.93 mmol) of trityl chloride was added, and the mixture was stirred at room temperature for 24 hours. Thereafter, the mixture was further stirred at 50 ° C. for 4 hours. The reaction mixture was allowed to cool to room temperature, and the solvent was distilled off under reduced pressure. The residue was diluted with chloroform (100 ml) and washed with water (20 ml). After drying over anhydrous magnesium sulfate, the solvent was distilled off under reduced pressure. 321 mg (0.41 mmol) of (2S, 3R, 4E) -1-O-trityl-2-N-palmitoylsphingosine obtained in the previous production example was dissolved in pyridine (2 ml), and stirred while cooling with ice water. To this solution, 40 mg of DMAP and 95 ml (0.82 mmol) of benzoyl chloride were added and stirred at room temperature for 24 hours. The mixture was further stirred at 55 ° C. for 1 hour. The reaction mixture was allowed to cool to room temperature and diluted with ethyl acetate (50 ml). This was washed with water (20 ml) and dried over anhydrous magnesium sulfate, and the solvent was distilled off under reduced pressure. Silver perchlorate 14 mg (0.072 mmol), tin (II) chloride 14 mg (0.069 mmol) and powdered molecular sieves 4A 300 mg were dissolved and suspended in methylene chloride (1 ml) and stirred at 0 ° C. Here, O- (2 ′, 3 ′, 4 ′, 6′-tetra-O-acetyl-α-D-galactopyranosyl)-(1 → 4) -2,3,6 obtained in Production Example 2 A solution of 20 mg (0.031 mmol) of -tri-O-acetyl-β / α-D-galactopyranosyl fluoride in methylene chloride (2 ml) was added and stirred. Ten minutes later, a solution of (2S, 3R, 4E) -1-O-trityl-2-N-palmitoylsphingosine 40 mg (0.062 mmol) obtained in Preparation Example 6 in methylene chloride (2 ml) was added and stirred for 17 hours. The reaction mixture was diluted with methylene chloride (100 ml) and filtered through celite. The filtrate was washed with a saturated aqueous sodium hydrogen carbonate solution (10 ml) and then dried over anhydrous magnesium sulfate. The solvent was distilled off under reduced pressure, and the resulting residue was purified by silica gel thin layer chromatography (hexane / ethyl acetate system) six times to obtain 5 mg of the title compound as a colorless transparent oil. (Yield; 12%)
This was used in the next reaction without purification.

O- (2 ′, 3 ′, 4 ′, 6′-Tetra-O-acetyl-α-D-galactopyranosyl)-(1 → 4) -2,3,6-triglyceline obtained in the previous example -O-acetyl-β-D-galactopyranosyl)-(1 → 3)-(2S, 3R, 4E) -1-O-benzoyl 2-N-palmitoylsphingosine 5.0 mg (4.0 mmol) in methanol (0.25 ml) and tetrahydrofuran (0.25 ml), dissolved in 28% sodium methoxide / methanol solution (7.4 ml, 38.5 mmol), and stirred at room temperature for 90 minutes. The reaction solution was diluted with methanol (8 ml), and Dowex 50W-8H (H+Type) neutralized with resin. The resin was filtered off and the filtrate was distilled off under reduced pressure. The resulting residue was purified by polymer gel Sephadex LH-20 column chromatography (methanol) to give 2.5 mg of the title compound as a white bowl. (Yield; 72%)

O- (2 ′, 3 ′, 4 ′, 6′-Tetra-O-acetyl-α-D-galactopyranosyl)-(1 → 4) -2,3,6-triglyceline obtained in the previous example -O-acetyl-β-D-galactopyranosyl)-(1 → 1)-(2S, 3R, 4E) -3-O-benzoyl-2-N-palmitoylsphingosine 5.28 mg (4.19 mmol) in methanol (0.35 ml) / tetrahydrofuran (0.35 ml) mixed solvent, 28% sodium methoxide / methanol solution 7.8 ml (40.2 mmol) was added, and the mixture was stirred at room temperature for 90 minutes.
