本発明者等は、前記課題を解決するために一般的に食品香料として使用されているペパーミントオイル、スペアミントオイル、各種フレーバー成分ならびに各種天然素材抽出物についてヘリコバクター・ピロリに対する抗菌効果を確認後、ペパーミントオイル、スペアミントオイル、各種フレーバー成分および各種天然素材抽出物の1種または2種以上を用いた抗菌剤を開発すべく鋭意研究したところ、カカオハスク抽出物がヘリコバクター・ピロリに対する抗菌活性を有することを見出し、本発明を完成するに至った。

【発明の実施の形態】
本発明に使用し得るペパーミントオイル、スペアミントオイルおよび各種フレーバー成分の抽出源はあらゆる天然起源のものおよび合成品を包含することを意図する。また、各種天然素材抽出物については、その産地、製法、形態、成分組成等には特に限定はない。

以下に、本発明の天然素材抽出物の抽出製造例を挙げて説明する。

各天然素材を粉砕機などの適当な粉砕手段で粉砕し、粉砕物の10倍量の抽出溶媒にて80〜100℃の温度で3〜5時間加熱還流する方法で実施する。抽出溶媒としては例えば、水、メタノール、酢酸エチル、アセトン、エーテル、エタノール、n−ヘキサン単独あるいは2種以上の混合溶媒を用いる。これらの溶媒は市販されている通常のものを使用すればよく、また混合溶媒は、これら2種以上を任意に選択し、任意の混合割合で混合して使用すればよい。

これら精油、フレーバー成分および各種天然素材抽出物単独ないし2種以上の混合物、あるいは他の医薬または任意の製剤用担体ないし希釈液と混合した任意の剤形にして医薬として利用でき、これら精油、フレーバー成分および各種天然素材抽出物の1種もしくは2種以上の混合物を有効成分として各種の食品に配合して、胃炎、胃・十二指腸潰瘍予防飲食物を製造することができる。

例えば、経口投与剤として錠剤、顆粒剤、細粒剤、硬カプセル剤、軟カプセル剤、経口用液体製剤等を例示できる。食品としてはチューインガム、キャンディ、チョコレート、ビスケット等の菓子、清涼飲料水、冷菓、乳製品、酒類および肉類等を例示することができる。

食品の利用にあたっては、各単独もしくは2種以上の組合せとなるが、香料本来の嗜好性を考え、ペパーミントオイルやスペアミントオイルをベースとし、フレーバー成分、天然素材抽出物を付加するという利用が望ましい。

抗ヘリコバクター・ピロリ剤としての本発明の精油、フレーバー成分、天然素材抽出物の投与量は、投与方法および必要な治療によって変化するので一概には規定できないが、ヒトに対して1日あたりの全投与量は50mg〜3g、好ましくは50mg〜4gである。本発明の抗ヘリコバクター・ピロリ剤を投与するにあたり投与量の最小値未満では十分な効果が望めず、また最大値を超えた投与量では効果の向上はあまり期待できないので経済的にも嗜好性の面からも好ましくなく、本発明の抗ヘリコバクター・ピロリ剤の投与量は通常食品香料として利用されているレベルである。これら素材の安全性については、ペパーミントオイル、スペアミントオイルおよびフレーバー成分は一般的に食品香料として用いられているため安全性も高く、天然素材抽出物もハーブ、生薬、食品素材として公知のものであり、安全性の高い素材である。
