Description
第7:付加体は、生理活性分子または薬物分子を介してナノ炭素材の外壁部に担持もしくは付着されていることを特徴とする上記のナノ炭素担持体の製造方法。
A)ナノ炭素材の外壁部に
B)機能性有機分子と水溶性または水分散性の化合物との付加体が
C)担持もしくは付着されている
ものであるが、ナノ炭素材については、その大きさがナノサイズ、通常は500nm以下のサイズのものであれば各種のものであってよく、たとえば代表的にはカーボンナノチューブ(NT)やカーボンナノホーン(NH)、あるいはそれらの集合体が例示される。フラーレンあるいはフラーレンチューブ等であってもよい。
<1>前記酸化処理は、酸素や、水蒸気、二酸化炭素等の1種以上による200℃〜1200℃での加熱または酸化剤による処理とする。
<2>前記酸化処理は、酸素濃度1%以上の気流中での温度200℃〜600℃の範囲の加熱処理とする。
<3>前記酸化処理は、過酸化水素、そして硝酸、塩酸等の無機酸のいずれか、もしくはその混合物を用いる液相酸化処理とする。
<4>前記還元処理は、水素または還元剤による200℃〜1200℃での加熱処理とする。
<5>前記還元処理は、水素濃度0.1%以上の気流中での温度300℃〜1000℃の範囲の加熱処理とする。
<6>前記酸化・還元処理は、酸化処理に続いての還元処理もしくは還元処理に続いての酸化処理とする。
14mgのDXR(ドキソルビシン:抗ガン活性化合物)と3.2μlのトリエチルアミンを9mlのDMF(ジメチルホルムアミド)に溶解し、この混合溶液を80mgの平均分子量5000のポリエチレングリコール誘導体(5PEG、日本油脂(株)製)を含むDMF溶液(1ml)に攪拌しながら滴下した。4℃で終夜攪拌した後、分画分子量3500の透析膜を用いて水に対して透析を行った。得られた溶液を凍結乾燥し、少量の水に溶解した後、LH−20カラム(アマシャムバイオサイエンス社製)に通塔して精製した。溶出液を凍結乾燥し、粉末状の5PEG−DXRを得た。次式は、その構造を示したものである。式中のPEGは、この実施例1では5PEGを示している。後述する実施例2では式中のPEGは20PEGを示している。

<実施例2>20PEG−DXRの合成
7mgのDXRと1.7μlのトリエチルアミンを18mlのDMFに溶解し、この混合溶液を160mgの平均分子量20000のポリエチレングリコール誘導体(20PEG、日本油脂(株)製)を含むDMF溶液(2ml)に攪拌しながら滴下した。4℃で終夜攪拌した後、分画分子量3500の透析膜を用い水に対して透析を行った。得られた溶液を凍結乾燥し、水量の水に溶解した後、LH−20カラム(アマシャムバイオサイエンス社製)に通塔した。溶出液を凍結乾燥し、粉末状の20PEG−DXRを得た。

<実施例3>PEG−DXR処理oxNHの調製
oxNH(酸化処理されたカーボンナノホーン)をDMSO(ジメチルスルホキシド)もしくはDMFに懸濁し、30秒間隔で5分間超音波処理した。このoxNH懸濁液とoxNHの4倍重量の5PEG−DXRもしくは20PEG−DXRをエッペンチューブ内で混合し、室温で1時間震盪した。得られた混合液を攪拌しながら、その9倍体積の水を滴下し、その後遮光下4℃で終夜攪拌した。得られた溶液を分画分子量100000のアミコン限外ろ過器(ミリポア社製)で濃縮した。水で希釈した後再度アミコン限外ろ過器で濃縮し、これらの操作を繰り返してPEG−DXR処理oxNH(PEG−DXR−oxNH)を得た。
<実施例4>ゲルろ過カラムを用いたPEG−DXR−oxNHの水溶液中分散性の評価
様々な条件で水に分散させたoxNHをPD−10脱塩用ゲルろ過カラム(アマシャムバイオサイエンス社製)に流し込み、ゲル内への浸透性を観察することにより水溶中分散性の評価を行った(図1)。DMSOのみで処理したoxNH、およびDXRあるいはPEG処理したoxNHでは、PD−10カラムへほとんど浸透しなかった。一方、5P−EG−DXR−oxNHもしくは20PEG−DXR−oxNHはPD−10カラム底部まで浸透し、水溶液中分散性の向上が明らかとなった。この浸透性はDXRを共存させることで阻害され、oxNHへのPEG−DXR吸着がDXR部分を介して生じていることも示唆された(図1a)。
<実施例5>動的光散乱法によるPEG−DXR処理oxNHの純水中平均粒径の測定
純水中におけるPEG−DXR−oxNH(oxNH換算で200μg/ml)の平均粒径を、濃厚系粒径アナライザーFPAR−1000(大塚電子社製)で測定した。この結果、5PEG−DXR処理oxNHの平均粒径は251nm(図2a)、20PEG−DXR処理oxNHの平均粒径は254nm(図2b)と算出された。
<実施例6>TUNEL(Terminal deoxynucleotidyl transferase biotin−dUPTP nick end labeling)染色法によるPEG−DXR処理oxNHのin vitro薬効評価
ヒト非小細胞肺癌細胞株NCI−H460の培養液に、各種条件で処理したoxNH(oxNH換算で200μg/ml)を添加し、3日後にTUNEL染色を行った。TUNEL染色に際し、In Situ Cell Death Detection Kit,Fluorescein(Roche Molecular Biochemicals社製)を用いた。10μg/ml DXR処理群(図3b)ですべての細胞がTUNEL陽性となり、DXR処理で細胞にアポトーシスが引き起こされていることを確認した。未処理群(図3a)、5PEG処理oxNH添加群(図3c)および20PEG処理oxNH添加群(図3d)においてTUNEL陽性細胞は見られなかったが、5PEG−DXR−oxNH(図3e)、および20PEG−DXR−−oxNH(図3f)添加群では明らかにTUNEL陽性細胞数が増加した。以上の結果から、PEG−DXR−oxNHは、癌細胞に対してDXR同様アポトーシス誘導活性を有していることが明らかとなった。
<実施例7>
5PEG−DXR−oxNH(5PEG−DXR−oxSWNH)、5PEG−DXR、および生理食塩水をマウスに投与したときの腫瘍サイズの減少をしらべた。投与量、検体数(n)および実験方法は以下のとおりである。

0.9%NaCl(n=4),5PEG−DXR(n=4),5PEG−DXR−oxNH(n=5)
<実験方法>
day 0,4に各種溶液(100μl)を腫瘍内に投与し、day6に腫瘍体積を測定した。(腫瘍体積=0.5×短径×短径×長径(mm3))
<結果>
day 0における腫瘍体積を1として各群の値(Relative tumor volume)を図4に示す。
5PEG−DXRと5PEG−DXR−oxNHの間には統計学的有意差があり、5PEG−DXR−oxNHを投与したときに、腫瘍サイズの減少が顕著であった。以上の結果から、PEG−DXR−oxNHは、DDS薬剤として有用であることが明らかとなった。
Claims (8)
- ナノ炭素材の外壁部に、水溶性または水分散性化合物との付加体として機能性有機分子の少くとも1種が担持または付着されているナノ炭素担持体の製造方法であって、ナノ炭素材の分散液と、生理活性分子もしくは薬物分子と水溶性もしくは水分散性化合物との付加体を混合して、ナノ炭素材の外壁部に該付加体を担持もしくは付着させることを特徴とするナノ炭素担持体の製造方法。
- ナノ炭素材は、カーボンナノチューブまたはカーボンナノホーンであることを特徴とする請求項1のナノ炭素担持体の製造方法。
- ナノ炭素材は、あらかじめ加熱処理、酸化処理および還元処理の少くともいずれかの処理がされたものであることを特徴とする請求項1または2のナノ炭素担持体の製造方法。
- 機能性有機分子は、共有結合によって水溶性もしくは水分散性化合物と付加体を形成していることを特徴とする請求項1から3のうちのいずれかのナノ炭素担持体の製造方法。
- 機能性有機分子は、生理活性分子または薬物分子であることを特徴とする請求項1から4のうちのいずれかのナノ炭素担持体の製造方法。
- 水溶性もしくは水分散性化合物は、アルキレングリコールのオリゴマーまたはポリマー、ポリビニルエーテル、ポリビニルエステル、ポリビニルピロリドン、クラウンエーテルおよびシクロデキストリンのうちの少くとも1種であることを特徴とする請求項1から5のうちのいずれかのナノ炭素担持体の製造方法。
- 付加体は、生理活性分子または薬物分子を介してナノ炭素材の外壁部に担持もしくは付着されていることを特徴とする請求項1から6のうちのいずれかのナノ炭素担持体の製造方法。
- 請求項1から7のうちのいずれかのナノ炭素担持体の製造方法で得られたナノ炭素担持体を用いてなるドラッグデリバリーシステム(DDS)薬剤。
